生化机理 Lapatinib is an antineoplastic agent used in breast cancer research research. It appears to be a tyrosine kinase inhibitor which interrupts the Neu/ ErbB2 and EGFR (epidermal growth factor receptor) pathways, thus inhibiting ErbB-overexpressing tumors. Lapatinib binds to the ATP-binding pocket of the protein kinase domains on EGFR/ErbB, preventing self-phosphorylation in solid tumor cells. Lapatinib is an inhibitor of EGFR and Neu/ ErbB2.Cell-permeable, potent dual tyrosine kinase inhibitor of EGFR and HER2 (ERBB2)\xa0(IC 50 values are\xa03 nM\xa0and 13 nM respectively).\xa0Also inhibits HER4 (IC 50 = 347 nM).\xa0Reduces tyrosine phosphorylation of EGFR and HER2
