Distinct response patterns of bacterial communities in Ag- and ZnO-rGO nanocomposite-amended silt loam soils

Tao Li,Yuhua Li,Xike Zhang,Jiawei Yuan,Junkang Guo,Pan Wang,Gehong Wei,Chun Chen

The widespread application of metal-based nanoparticle (MNPs)/reduced graphene oxide (rGO) composites inevitably leads to their release into soils. However, we lack a detailed understanding of the bacterial community response to MNPs-rGO exposure in farmland soils. Here, we conducted a soil microcosm experiment to analyze the potential impact of MNPs-rGO on bacterial communities in two field soils via high-throughput sequencing. The change in alpha diversity of bacterial communities was more susceptible to Ag-rGO and ZnO-rGO treatments than CuO-rGO. In both soils, MNPs-rGO significantly changed the bacterial community structure even at a low dose (1 mg kg −1 ). The bacterial community structure was most strongly affected by Ag-rGO at 30 days, but the greatest changes occurred in ZnO-rGO at 60 days. The differences in soil properties could shape bacterial communities to MNPs-rGO exposure. Distance-based redundancy analysis and functional annotation of prokaryotic taxa showed that some bacterial species associated with nitrogen cycling were greatly influenced by Ag-rGO and ZnO-rGO exposure. In sum, Ag-rGO and ZnO-rGO may potentially affect bacterial communities and nitrogen turnover under long-term realistic field exposure. These findings present a perspective on the response of bacterial communities to MNPs-rGO and provide a fundamental basis for estimating the ecological behavior of MNPs-rGO.

Waterborne Cr3+ and Cr 6+ exposure disturbed the intestinal microbiota homeostasis in juvenile leopard coral grouper Plectropomus leopardus

Hengzhen Ye,Yanjie Zhang,Lu Wei,Hao Feng,Qiongyao Fu,Zhiqiang Guo

Background Chromium (Cr) mainly has two stable forms: Cr 3+ and Cr 6+ . Cr and its compound are widely used in the printing, dyeing, leather making, and metallurgy industry. They are evitably released into the environment and pose a significant threat to creatures, for instance, the excessive chromium (Cr) burden in the marine ecosystem is often harmful to fish. Intestinal microbiota greatly affects fish performance, but how waterborne Cr affects fish intestinal microbiota is unclear. To test the hypothesis that the waterborne Cr exposure could significantly affect fish’ intestinal microbiota homeostasis, and the effect was highly dependent on Cr concentration and speciation, the juvenile leopard coral grouper Plectropomus leopardus were exposed to waterborne Cr 3+ and Cr 6+ (0.1, 0.5 ppm) for 7 days, and the intestinal microbiota was determined by Amplicon sequencing of the 16S rRNA. Results In all Cr treatment groups, the alpha diversity of intestinal microbiota communities of P. leopardus was decreased. The phyla Proteobacteria, Firmicutes, Actinobacteria, Chloroflexi and Bacteroidetes were the dominant intestinal microbiota. The Chao index diversity significantly declined in Cr treatment group, indicating the intestinal microbiota community structure was changed. Among the dominant intestinal microbiota, Proteobacteria was most sensitive to Cr exposure, and it increased after xposure. The PICRUSt predicted that 0.5 ppm Cr 3+ expousure caused metabolism disordered in the intestinal of P. leopardus . Conclusions Waterborne Cr 3+ and Cr 6+ significantly disturbed intestinal microbiota homeostasis in P. leopardus , including their diversity, composition, and community structure. The metabolism level of intestinal microbiota in P. leopardus was decreased by Cr 3+ exposure. High concentrations of Cr 3+ may pose potential risks to the intestinal homeostasis of P. leopardus .

Response of bacterial communities to benzalkonium chloride exposure in marine sediment systems

Wang Jingwei,Jiang Yingze,Jia Quan,Zhao Pan,Kang Haowu,Huang Huatao,Deng Yubin,Yu Hang,Xu Dan,Ma Qiao

Benzalkonium chloride (BAC) is widely employed as a biocide due to its low toxicity, high efficacy, and broad-spectrum antibacterial properties, making it frequently detected across diverse environmental settings. However, the impacts of BAC on the marine microbial community remain poorly documented. In this study, the coastal sediment systems exposed to varying concentrations of BAC (0, 0.001, 0.1, and 10 mg/L) were constructed, and microbial responses were investigated. Results demonstrated that the activities of enzymes, including soil catalase, dehydrogenase, fluorescein diacetate hydrolyase, and neutral protease, increased on Day 1 following exposure to BAC. BAC increased the α-diversity of the microbial community on both Day 1 and Day 14 and played a deterministic role in community assembly. The microbial community structures were significantly shifted. Celeribacter , Fusibacter , and Flavobacterium increased especially on Day 1, while Gillisia , Sulfitobacter , and Marinobacter significantly decreased especially on Day 14. Functional prediction revealed that oxidative phosphorylation was promoted, suggesting that BAC might enhance the metabolic activity of the microbial communities. Network analysis showed that BAC reduced the complexity and potential correlations of the community, and genera from Pseudomonadota were highly connected within the community. Our findings revealed that BAC could substantially affect the microbial metabolism, composition, and interaction of marine sediment communities, thereby enhancing our understanding of the environmental impacts of BAC on marine ecosystems.

Antibiotics can cause weight loss by impairing gut microbiota in mice and the potent benefits of lactobacilli

Miao Zhonghua,Cheng Ruyue,Zhang Yujie,Liang Huijing,Jiang Fengling,Shen Xi,Chen Gong,Zhang Qisheng,He Fang,Li Ming

This study assessed whether antibiotics could alter gut microbiota to affect host growth and the possibility of alleviation by lactobacilli. We divided four-week-old BABL/c mice into control (Ctrl), antibiotic exposure (Abx), Lactobacillus plantarum PC-170 (PC), and Lactobacillus rhamnosus GG (LGG) group and the Abx, LGG, and PC group received an one-week antibiotic/antibiotic + probiotic treatment. The fecal microbiota and the expression of splenic cytokines were determined. Following the ceftriaxone treatment, the body weight gain of Abx was delayed compared with others. The ceftriaxone treatment significantly decreased the alpha-diversity of the fecal microbiota and altered the fecal microbiota but LGG and PC can partly alleviate the effect. At the end of the study, the microbial community of LGG and PC group were more similar to Ctrl compared with Abx group. The results indicated that ceftriaxone could significantly alter intestinal microbiota. Lactobacilli might alleviate the side effects of antibiotics by stabilizing the intestinal microbiota.

TiO2 nanoparticles exert an adverse effect on aquatic microbial communities

Bingfeng Chen,Yizhou Pan,Yiling Chen,Zhenyan Zhang,Zhihan Yang,Meng Zheng,Tao Lu,Liying Jiang,Haifeng Qian

Titanium dioxide nanoparticle (n-TiO 2 ) is a widely used nanomaterial, which is inevitably released as a residue into aquatic ecosystems during material production and usage. However, the effects of n-TiO 2 on aquatic microbial communities have not been completely elucidated. This study examined the toxic effects of n-TiO 2 on eukaryotic and prokaryotic microbial communities in freshwater environments. We determined that n-TiO 2 had a greater inhibitory effect on the growth of eukaryotic algae than cyanobacteria in monocultures. A similar phenomenon was observed in a microcosm experiment, revealing that n-TiO 2 slightly reduced the content of chlorophyll-a but evidently increased the phycocyanin content. Moreover, the alpha diversity of the eukaryotic community was not affected, whereas its beta diversity increased with exposure to n-TiO 2 . Although n-TiO 2 altered the composition of freshwater microbial communities, it did not change the functions of the prokaryotic community, which might be attributed to the functional redundancy of microbiota. Co-occurrence network analysis indicated that n-TiO 2 destabilized the freshwater community, especially the eukaryotic community, and potentially disturbed the aquatic ecosystem. Our study revealed that the ecological risk of n-TiO 2 on aquatic microbial communities is complex; hence, rational utilization of n-TiO 2 should be emphasized.

Fabrication of bio-engineered chitosan nanoformulations to inhibition of bacterial infection and to improve therapeutic potential of intestinal microflora, intestinal morphology, and immune response in infection induced rat model

Xiao Wan,Liu Liu,Lu Ding,Zhiqiang Zhu

Overdosage of antibiotics used to prevent bacterial infections in the human and animal gastrointestinal tract would result in disturbing of intestinal barrier, significant misbalancing effects of intestinal microflora and persuading bacterial resistance. The main objective of the present investigation is to design and develop novel combinations of organic curcumin (Cur) and antimicrobial peptide (Amp) loaded chitosan nanoformulations (Cur/Amp@CS NPs) to improve significant effects on antibacterial action, immune response, intestine morphology, and intentional microflora. The antibacterial efficiency of the prepared nanoformulations was evaluated using Escherichia coli (E. coli) induced bacterial infections in GUT of Rat models. Further, we studied the cytocompatibility, inflammatory responses, α-diversity, intestinal morphology, and immune responses of treated nanoformulations in rat GUT models. The results indicated that Cur/Amp@CS NPs are greatly beneficial for intestinal microflora and could be a prodigious alternative of antibiotics.

Comparison and Correlation Analysis of Immune Function and Gut Microbiota of Broiler Chickens Raised in Double-Layer Cages and Litter Floor Pens

Bochen Song,Shaojia Yan,Peng Li,Guang Li,Mingkun Gao,Lei Yan,Zengpeng Lv,Yuming Guo

This study aimed to compare the immune function and gut microbiota between double-layer caged and litter floor pen-raised broiler chickens. Eighty meaty male chicks were selected and divided into cage group and litter floor group, with 20 replicates in each group. The broilers were raised in the same chicken house. The rearing density of the two rearing systems was same. The broilers were sampled on days 13 and 34. The results showed that compared with the cage group, the litter floor broilers had worse growth performance (23.24% increase in feed conversion ratio) in the early stage; better slaughter performance at day 42; stronger peripheral immune function (including higher lysozyme activity, T-cell ratio, Th-cell ratio, Tc-cell ratio, CD4/CD8, IL-10, B-cell ratio, IgG and IgA levels; and spleen immune-related gene expression); and stronger intestinal immune function (including higher ileum CD80, AvBD2, Mucin2, NF-κB, IL-8, IFN-γ/IL-4, and IgA mRNA expression levels and ileal mucosa sIgA levels). Compared with the cage group, the alpha diversity of ileum microbiota of the litter floor broilers was higher, and the relative abundance levels of litter breeding bacteria (Facklamia, Globicatella, and Jeotgalicoccus) and potential pathogenic bacteria (Streptococcus and Staphylococcus) were higher (P < 0.05). Through Spearman correlation analysis, it was found that enriched microbes in the ileum of litter floor broilers were positively correlated with immune function. In summary, compared with cage broilers, litter floor broilers had more potential pathogenic bacteria and litter breeding bacteria in the ileum, which may be one of the important reasons for the stronger immune function status.IMPORTANCE In China, the three-dimensional rearing system (cage) for broilers has gradually become a trend. In production, it was found that the incidence of disease in broiler chickens raised in cage systems was significantly higher than that of ground litter. Given that broilers raised on ground litter systems may be exposed to more environmental microbes, it is important to understand whether the rearing environment affects the function and status of the host immune system by altering the gut microbiota. In this study, rearing environment-derived gut microbes associated with stronger immune function in ground litter broilers were provided, which will provide new insights into strategies to target gut microbes to promote immune function and status in broilers raised in cages.

Effects of long-term exposure to the herbicide nicosulfuron on the bacterial community structure in a factory field

Qingyun Ma,Hao Tan,Jinlong Song,Miaomiao Li,Zhiye Wang,Rebecca E. Parales,Lin Li,Zhiyong Ruan

This study aims to investigate the effects of long-term nicosulfuron residue on an herbicide factory ecosystem. High-throughput sequencing was used to investigate the environmental microbial community structure and interactions. The results showed that the main contributor to the differences in the microbial community structure was the sample type, followed by oxygen content, pH and nicosulfuron residue concentration. Regardless of the presence or absence of nicosulfuron, soil, sludge , and sewage were dominated by groups of Bacteroidetes, Actinobacteria, and Proteobacteria. Long-term exposure to nicosulfuron increased alpha diversity of bacteria and archaea but significantly decreased the abundance of Bacteroidetes and Acidobateria compared to soils without nicosulfuron residue. A total of 81 possible nicosulfuron-degrading bacterial genera, e.g., Rhodococcus , Chryseobacterium , Thermomonas , Stenotrophomonas , and Bacillus , were isolated from the nicosulfuron factory environmental samples through culturomics. The co-occurrence network analysis indicated that the keystone taxa were Rhodococcus , Stenotrophomonas , Nitrospira , Terrimonas , and Nitrosomonadaceae_MND1. The strong ecological relationship between microorganisms with the same network module was related to anaerobic respiration, the carbon and nitrogen cycle , and the degradation of environmental contaminants. Synthetic community (SynCom), which provides an effective top-down approach for the critical degradation strains obtained, enhanced the degradation efficiency of nicosulfuron. The results indicated that Rhodococcus sp. was the key genus in the environment of long-term nicosulfuron exposure.

Oral supplementation with Bifidobacterium infantis and 2′-fucosyllactose revives gut microbiota perturbation and intestinal and immune developmental delay following early-life antibiotic challenge in BALB/c mice

Yating Luo,Jinxing Li,Simou Wu,Wen Jia,Zhimo Zhou,Meixun Liu,Fengling Jiang,Ting Huang,Xi Shen,Yun Li,Fang He,Ruyue Cheng

ABSTRACT Probiotics and synbiotics can mitigate the negative health consequences of early-life antibiotic exposure. This study aimed to determine whether supplementation with Bifidobacterium longum ssp. infantis 79 (B79) or synbiotics composed of B79 and 2′-fucosyllactose (2′-FL) could mitigate the negative impact of ceftriaxone exposure in early-life. We found that antibiotic-treated mice exhibited lower body weight, crypt depth, short-chain fatty acid content, and α-diversity indices at weaning, while they increased the relative abundance of opportunistic pathogens (such as Enterococcus and Staphylococcus ) and decreased the relative abundance of intestinal commensal bacteria. Supplementation with B79 and 2′-FL revived these antibiotic-induced negative effects and reduced the mRNA expression of IL-6, IL-12p40, and TNF-α in the spleen at weaning. Moreover, B79 and 2′-FL supplementation persistently improved crypt depth, propionic acid synthesis, and IgG and sIgA production and revived the gut microbiota structure and composition in adulthood. In conclusion, our study suggests that early-life supplementation with B79 alone or in combination with 2′-FL can mitigate ceftriaxone-induced negative effects on the gut microbiota and intestinal and immune development of mice, and these improvements can partially last into adulthood.

A three-dimensional-printed SPION/PLGA scaffold for enhanced palate-bone regeneration and concurrent alteration of the oral microbiota in rats

Lu Jia,Zukun Yang,Liuxu Sun,Qian Zhang,Yu Guo,Yulian Chen,Yang Dai,Yang Xia

Scaffold implantation for the repair of oral bone defects involves an interplay between the scaffold biomaterial and the microenvironment. However, previous studies on this subject have only considered the effects of the immune system and largely ignored those of the oral microbiota. Accordingly, in the present study, we prepared composite scaffolds comprising a three-dimensional poly( l -lactide- co -glycolide) matrix with a superparamagnetic iron oxide nanoparticle (SPION) coating and used a rat model to evaluate their palate-bone-regenerating effects and their interaction with the oral microbiota. It was found that the SPION coated scaffold induced better bone regeneration than that achieved by the controls. Furthermore, it significantly decreased the operational taxonomic units (OTU) numbers as determined by 16 s rRNA gene sequencing, and also resulted in decreased Chao and ACE alpha diversity indexes compared with those of the controls. However, it had no effect on beta diversity. SPION coated scaffolds caused a shift in oral bacterial composition characterized by a decrease in the Clostridium spp. population, and the dominant flora being Proteobacteria . Furthermore, SPION coated scaffolds upregulated the concentration of serum iron, hepcidin, and P1NP. Thus, SPION coated scaffolds enhanced bone regeneration, and this effect was partly related to alteration of the oral microbiota by the antibacterial effects of SPION. Our findings provide a better understanding of the role of oral microbiota in oral bone regeneration and how SPION coated scaffolds can be used to enhance it.

Effects of Dietary Astragalus Polysaccharide Supplementation on the Th17/Treg Balance and the Gut Microbiota of Broiler Chickens Challenged With Necrotic Enteritis

Bochen Song,Peng Li,Shaojia Yan,Yan Liu,Mingkun Gao,Huiyuan Lv,Zengpeng Lv,Yuming Guo

This study aimed to investigate the effects of dietary astragalus polysaccharide (APS) supplementation on the immune function, gut microbiota and metabolism of broiler chickens challenged with necrotic enteritis (NE). Two hundred forty Arbor Acres broiler chicks (one day old) were randomly assigned using a 2 × 2 factorial arrangement into two groups fed different levels of dietary APS (0 or 200 ppm of diet) and two disease challenge groups (control or NE challenged). The results showed that NE infection significantly increased FCR, mortality rate, Th17/Treg (Th17 cells% in blood and ileum, Th17/Treg, IL-17 and IL-17/IL-10 in blood), NO, lysozyme activity and IL-1β in blood, intestinal immune cell proportion and activity (Tc%, Treg% and monocyte phagocytic activity in ileum), intestinal inflammatory cytokines (TLR2, NF-κB, TNF-α and IL- 6) gene expression levels, and the number of Clostridium perfringens in cecum. NE infection significantly reduced body weight gain, thymus index, lymphocyte proliferation activity in blood and ileum, villus height and V/C in jejunum, Th cells% and Mucin2 gene expression in ileum. Dietary APS supplementation significantly increased body weight, feed intake, proportion of immune cells (T cells in blood and Tc, Treg in ileum), lymphocyte proliferation activity, V/C in jejunum, and ZO-1 gene expression in ileum. Dietary APS supplementation significantly reduced FCR and mortality rate, Th17/Treg, Th17%, intestinal pathology scores, intestinal inflammatory cytokine gene expression levels, and the number of Clostridium perfringens in cecum. In addition, broilers challenged with NE significantly increased Staphylococcus and Turicibacte r and reduced α diversity of microbiota in ileum. Dietary APS supplementation significantly increased α diversity, Romboutsia , Halomonas , propionic acid, butyric acid, formononetin, taurine, cholic acid and equol and downregulated uric acid, L-arginine and serotonin in ileum. Spearman’s correlation analysis revealed that Romboutsia, Turicibacter, Staphylocpccus, Halomonas, Streptococcus, Escherichia-Shigella, Prevotella , uric acid, L-arginine, jerivne, sodium cholate and cholic acid were related to inflammation and Th17/Treg balance. In conclusion, APS alleviated intestinal inflammation in broilers challenged with NE probably by regulating intestinal immune, Th17/Treg balance, as well as intestinal microbiota and metabolites.
