Design and construction of self-hidden and pH-reversed targeting drug delivery nanovehicles via noncovalent interactions to overcome drug resistance

Dan Zhao,Xiaoqing Yi,Jiaqi Xu,Gongdao Yuan,Renxi Zhuo,Feng Li

Using the host–guest interaction between β-cyclodextrin (β-CD) and adamantane (Ad), and borate formation between phenylboronic acid (PBA) and cis-diols, a smart pH-responsible targeting drug delivery nanovehicle, PBA–PEG–CD/Ad-lys(Diol)–PCL, was prepared via one-step self-assembly. Under physiological conditions, the targeted PBA function could be restrained by binding PBA with diol components located at the interface of self-assemblies, and hydrophilic PEG segments could be shielded simultaneously. When the environmental pH decreased, the PBA groups could be unbound and exposed on the surface of self-assemblies, leading to recovery of its targeted function, as shown using fluorescence spectroscopy, in vitro cell toxicity, and uptake. Under acidic conditions, PBA–PEG–CD/Ad-lys(Diol)–PCL/Dox showed significantly increased uptake and toxicity toward HepG2 cells in comparison with the control group. The smart vehicles were further utilized to test their efficiency in overcoming drug resistance in chemotherapy. Compared with free Dox, PBA–PEG–CD/Ad-lys(Diol)–PCL delivered six times more Dox into MCF-7/ADR cells and showed greater toxicity toward the ADR cells. As a result, this may be a facile strategy toward constructing efficient targeting vehicles through the rational utilization of noncovalent interactions.

Super-Assembled Periodic Mesoporous Organosilica Frameworks for Real-Time Hypoxia-Triggered Drug Release and Monitoring

Yingnan Liu,Lei Xie,Meng Gao,Runhao Zhang,Jingchen Gao,Jiangdong Sun,Qingdong Chai,Tong Wu,Kang Liang,Pu Chen,Qi Wan,Biao Kong

Hypoxia, induced by inadequate oxygen supply, is a key indication of various major illnesses, which necessitates the need to develop new nanoprobes capable of sensing hypoxia environments for the targeted system monitoring and drug delivery. Herein, we report a hypoxia-responsive, periodic mesoporous organosilica (PMO) nanocarrier for repairing hypoxia damage. β-cyclodextrin (β-CD) capped azobenzene functionalization on the PMO surface could be effectively cleaved by azoreductase under a hypoxia environment. Moreover, the nanosystem is equipped with fluorescence resonance energy transfer (FRET) pair (tetrastyrene derivative (TPE) covalently attached to the PMO framework as the donor and Rhodamine B (RhB) in the mesopores as the receptor) for intracellular visualization and tracking of drug release in real-time. The design of intelligent nanocarriers capable of simultaneous reporting and treating of hypoxia conditions highlights a great potential in the biomedical domain.

Supramolecular chemistry in cyclodextrin inclusion complexes: The formation rules of terpenes/β-cyclodextrin inclusion complexes

Xingran Kou,Xiwei Xu,Nan Gao,Yunchong Zhang,Xin Huang,Feng Chen,Qinfei Ke,Qingran Meng

Supramolecular chemistry of cyclodextrins (CDs) inclusion complexes is a hot research topic at present, but current studies often focus on the host-guest interaction between CDs and hydrophobic substances, while ignoring the self-assembly behavior of the host and guest during the inclusion process. Based on the spatial structures, six representative terpenes were selected in this study, and the stoichiometric ratio between terpenes and β-cyclodextrin (β-CD) was investigated by isothermal titration calorimetry (ITC) method. Meanwhile, the constructed inclusion complexes (ICs) were characterized by nuclear magnetic resonance , X-ray diffraction, scanning electron microscope , etc. On this basis, the assembly process of β-CD and terpenes was investigated firstly by molecular simulation method, and the inclusion process between β-CD and terpenes was further studied in terms of the formation of individual ICs and ICs aggregates. Results showed that hydrophobic interaction and van der Waals force were the main driving forces of the reaction between terpenes and β-CD, and the formation of ICs, β-CD molecule aggregation, terpene aggregation, and ICs aggregation occurred simultaneously during the inclusion process of terpenes and β-CD. To sum up, this study may provide certain thinking and reference for the subsequent supramolecular chemical research on CDs.

Cell-Membrane-Anchored Upconversion Nanoprobe for Near-Infrared Light Triggered Cell–Cell Interactions

Yujing Kong,Fang Yuan,Fei Yang,Cuiling Zhang,Yuezhong Xian

Manipulating cell–cell interactions is of great significance in cell communication and cell-based therapies. Although efforts have been made to construct cell–cell assembly by stimuli-responsive host–guest interactions, controllable cell–cell interactions by near-infrared (NIR) light triggered reversible assembly remain a challenge. Herein, we develop a NIR-controlled system based on β-cyclodextrin (β-CD) modified upconversion nanoparticles (UCNPs) for reversible and noninvasive manipulation of cell assembly and disassembly, which is realized by host–guest interactions between E/Z-photoisomerization of arylazopyrazole (AAP) and β-CD under the NIR irradiation. UCNPs can convert NIR to ultraviolet light, which leads to the transformation of AAP from the E-isomer to the Z-isomer. And it can be reverted back to the E-isomer under visible light irradiation. This reversible photoisomerization can modulate the host–guest interaction between β-CD and AAP, thus leading to reversible cell assembly and disassembly. Furthermore, by precise regulating cell–cell interactions by NIR light, cell–cell communication and molecular transportation can be realized. Given the diversity of host and guest molecules and the advantages of NIR light in biological applications, reversible cell–cell assembly has great potential for the regulation of cell behaviors and cell-based therapies.

Multicolor Iridium(III) Complexes with Host–Guest Recognition Motifs for Enhanced Electrochemiluminescence and Modular Labeling

Xinrui Yang,Yingying Xu,Xiaojin Huang,Junmeng Hang,Weiliang Guo,Zhihui Dai

Cyclometalated Ir(III) complexes with high electrochemiluminescence (ECL) efficiency and appropriate bioconjugation sites are urgently needed in ECL immunoassays (ECLIA). Herein, we report the synthesis, photophysics, electrochemistry, and ECL of six new Ir(III) complexes bearing naphthyl (nap) or adamantane phenyl (adap) substitutions, four of which emit cyan, green, or red light and display 1.7- to 7.5-fold increases in ECL intensity. In combination with DFT/TDDFT calculations, this enhancement is rationalized to the augmented radiative rate that arises from both the strengthened spin-orbit coupling (SOC) and the increased transition dipole moment. In addition, the adap-based Ir(III) complex shows high binding affinity with β-cyclodextrin (β-CD) due to the strong hydrophobic interaction, which enables us to develop a modular strategy for the labeling of Ir(III) complexes with biomolecules and to use hydrophobic luminophores in the aqueous-phase detection. As demonstrated, a novel ECLIA is built up and exhibits a wide linear range from 1 ng/mL to 10 μg/mL and a detection limit of 72 pg/mL for the determination of C-reactive protein (CRP). These findings provide new insights into the design, synthesis, and bio-labeling of highly emissive Ir(III) complexes and pave the way for the development of novel ECLIA based on host–guest recognition motifs.

Reactive oxygen species (ROS)-responsive size-reducible nanoassemblies for deeper atherosclerotic plaque penetration and enhanced macrophage-targeted drug delivery

Jianhua He,Wenli Zhang,Xiaoju Zhou,Fengfei Xu,Jiahui Zou,Qiqi Zhang,Yi Zhao,Hongliang He,Hu Yang,Jianping Liu

Nanoparticle-based therapeutics represent potential strategies for treating atherosclerosis; however, the complex plaque microenvironment poses a barrier for nanoparticles to target the dysfunctional cells. Here, we report reactive oxygen species (ROS)-responsive and size-reducible nanoassemblies, formed by multivalent host-guest interactions between β-cyclodextrins (β-CD)-anchored discoidal recombinant high-density lipoprotein (NP 3 ST ) and hyaluronic acid-ferrocene (HA-Fc) conjugates. The HA-Fc/NP 3 ST nanoassemblies have extended blood circulation time, specifically accumulate in atherosclerotic plaque mediated by the HA receptors CD44 highly expressed in injured endothelium, rapidly disassemble in response to excess ROS in the intimal and release smaller NP 3 ST , allowing for further plaque penetration, macrophage-targeted cholesterol efflux and drug delivery. In vivo pharmacodynamicses in atherosclerotic mice shows that HA-Fc/NP 3 ST reduces plaque size by 53%, plaque lipid deposition by 63%, plaque macrophage content by 62% and local inflammatory factor level by 64% compared to the saline group. Meanwhile, HA-Fc/NP 3 ST alleviates systemic inflammation characterized by reduced serum inflammatory factor levels. Collectively, HA-Fc/NP 3 ST nanoassemblies with ROS-responsive and size-reducible properties exhibit a deeper penetration in atherosclerotic plaque and enhanced macrophage targeting ability, thus exerting effective cholesterol efflux and drug delivery for atherosclerosis therapy.

Friction-induced supramolecular nanovalves for the potential treatment of osteoarthritis

Q. Wang,X. Yu,Y. Zhao,Y. Wang,K. Chen,Y. Liu,B. Wang,H. Zhang,Y. Sun

Owing to the overuse of rough articular cartilage, it increases the friction of joint and further leads to inflammation in joint cavities. However, the pathological changes also bring a new opportunity to achieve the treatment of osteoarthritis (OA). The increasing friction from damaged articular cartilage can convert more mechanical energy to heat. The higher localized temperature can reduce supramolecular interactions in general. Therefore, friction becomes another new activation to operate nanovalve systems for the targeting treatment of OA. Here, we designed and prepared a friction-induced nanovalve system constructed with β-cyclodextrins (β-CD)-modified mesoporous silica nanoparticles (MSN). Besides, a copolymer containing two components – guest molecules (adamantane, AD) and poly(2-methacryloyloxyethyl phosphorylcholine) (PMPC) – has been self-assembled on the surface of β-CD-modified MSN (MSN-CD) by supramolecular host–guest interactions. In this design, the charged PMPC on the MSN surface can form a dense hydration layer by ion–dipole interactions, switch the friction from surfaces to water molecules to achieve hydration lubrication , and sharply reduce the coefficient of friction (COF). Besides, the higher localized temperature from the mechanical conversion can weaken the supramolecular host–guest interactions and turn on the nanovalves to accomplish the targeted release near rough articular cartilage of OA. Even when PMPC is peeled off, the hydrophilic surface of β-CD on MSN surface can still perform the acceptable lubricating properties. The following testings of cytotoxicity by CCK-8 and live/dead staining indicated the excellent biocompatibility. Further analysis of protein expression levels has also shown the potential application in OA.

Multistage Anticoagulant Surfaces: A Synergistic Combination of Protein Resistance, Fibrinolysis, and Endothelialization

Jian Feng,Jinghong Wang,Huanhuan Wang,Xinyin Cao,Xiaoliang Ma,Yu Rao,Huimin Pang,Sulei Zhang,Yuheng Zhang,Lei Wang,Xiaoli Liu,Hong Chen

Anticoagulant surface modification of blood-contacting materials has been shown to be effective in preventing thrombosis and reducing the dose of anticoagulant drugs that patients take. However, commercially available anticoagulant coatings, that is, both bioinert and bioactive coatings, are typically based on a single anticoagulation strategy. This puts the anticoagulation function of the coating at risk of failure during long-term use. Considering the several pathways of the human coagulation system, the synergy of multiple anticoagulation theories may provide separate, targeted effects at different stages of thrombosis. Based on this presumption, in this work, negatively charged poly(sodium p-styrenesulfonate-co-oligo(ethylene glycol) methyl ether methacrylate) and positively charged poly(lysine-co-1-adamantan-1-ylmethyl methacrylate) were synthesized to construct matrix layers on the substrate by electrostatic layer-by-layer self-assembly (LBL). Amino-functionalized β-cyclodextrin (β-CD-PEI) was subsequently immobilized on the surface by host–guest interactions, and heparin was grafted. By adjusting the content of poly(oligo(ethylene glycol) methyl ether methacrylate) (POEGMA), the interactions between modified surfaces and plasma proteins/cells were regulated. This multistage anticoagulant surface exhibits inertness at the initial stage of implantation, resisting nonspecific protein adsorption (POEGMA). When coagulation reactions occur, heparin exerts its active anticoagulant function in a timely manner, blocking the pathway of thrombosis. If thrombus formation is inevitable, lysine can play a fibrinolytic role in dissolving fibrin clots. Finally, during implantation, endothelial cells continue to adhere and proliferate on the surface, forming an endothelial layer, which meets the blood compatibility requirements. This method provides a new approach to construct a multistage anticoagulant surface for blood-contacting materials.

On-demand bactericidal and self-adaptive antifouling hydrogels for self-healing and lubricant coatings of catheters

Pan Ran,Bo Qiu,Huan Zheng,Shuang Xie,Guiyuan Zhang,Wenxiong Cao,Xiaohong Li

Catheter-related infections are one of the most common nosocomial infections with increasing morbidity and mortality, and robust antibacterial or antifouling catheter coatings remain great challenges for long-term implantation. Herein, multifunctional hydrogel coatings were developed to provide persistent and self-adaptive antifouling and antibacterial effects with self-healing and lubricant capabilities. Polyvinyl alcohol (PVA) with β-cyclodextrin (β-CD) grafts (PVA-Cd) and 4-arm polyethylene glycol (PEG) with adamantane and quaternary ammonium compound (QAC) terminals (QA-PEG-Ad) were crosslinked through host-guest recognitions between adamantane and β-CD moieties to acquire PVEQ coatings. In response to bacterial infections, QACs exhibit reversible transformation between zwitterions (pH 7.4) and cationic lactones (pH 5.5) to generate on-demand bactericidal effect. Highly hydrophilic PEG/PVA backbones and zwitterionic QACs build a lubricate surface and decrease the friction coefficient 10 times compared with that of bare catheters. The antifouling hydrated layer significantly inhibits blood protein adsorption and platelet activation and reveals negligible hemolysis and cytotoxicity. The dynamic host-guest crosslinking achieves full self-healing of cracks in PVEQ hydrogels, and the mechanical profiles were recovered to over 90% after rejuvenating the broken hydrogels, exhibiting a long-term stability after mechanical stretching, twisting, knotting and compression. After subcutaneous implantation and local bacterial infection, the retrieved PVEQ-coated catheters display no tissue adhesion and 3 log folds lower bacterial number than that of bare catheters. PVEQ coatings effectively prevent the repeated bacterial infections and there are few inflammatory reactions in the surrounding tissue, while substantial lymphoid infiltration and inflammatory cell aggregation occur in muscle tissues around the bare catheter. Thus, this study demonstrates a catheter coating strategy by on-demand bactericidal, self-adaptive antifouling, self-healing and lubricant hydrogels to address medical devices-related infections. Statement of Significance It is estimated over two billion peripheral intravenous catheters are annually used in hospitals around the world, and catheter-associated infection has become a great clinical challenge with rapidly rising morbidity and mortality. Surface coating is considered a promising approach, but substantial challenges remain in the development of coatings that simultaneously satisfy both anti-fouling and antibacterial attributes. Even more, few attempts have been made to design mechanically robust coatings and reversible antibacterial or antifouling capabilities, which are critical for long-term medical implants. To address these challenges, we propose a concise strategy to develop hydrogel coatings from commercially available poly(ethylene glycol) and polyvinyl alcohol. In addition to self-healing and lubricant capabilities, the reversible conversion between zwitterionic and cationic lactones of quaternary ammonium compounds enables on-demand bactericidal and self-adaptive antifouling effects.
