Qualitative and quantitative analysis of β-sitosterol marker in virgin camellia oil and virgin olive oil

Li Yang,Wu Minyu,Zhai Liang,Zhang Hui,Shen Lirong

Camellia oil and olive oil with superior economic value are easily adulterated with other, cheaper oils. It is difficult to identify both oils by traditional methods because of their similar fatty acid profiles. In the present study, a novel method for qualitative and quantitative analysis of β-sitosterol using GC/MS and GC/FID was developed to identify camellia oil and olive oil. The method validation of β-sitosterol analysis showed good linearity and satisfactory values for recovery, accuracy, precision, and repeatability. The linear regression coefficient (R2) of the calibration curve was 0.9985. An acceptable limit of detection (0.36 mg/100 g) and limit of quantification (1.20 mg/100 g) were achieved. The spiked recoveries were 95.0% to 100.3%. The relative standard deviation (RSD) of within-day precision was less than 3.26%, and the RSD of retention times and peak areas for repeatability were within 0.03% and 1.08%, respectively. The contents of β-sitosterol in virgin camellia oil and virgin olive oil were in the range of 14.1–30.2 mg/100 g and 94.3–173.2 mg/100 g, respectively, indicating that the β-sitosterol content in the former is seven times that in the latter, and β-sitosterol could be a potential marker for the authentication and adulteration detection of both oils.

Structure and thermal properties of β-sitosterol-beeswax-sunflower oleogels

Min Pang,Xiuxiu Wang,Lili Cao,Zhaojuan Shi,Zheng Lei,Shaotong Jiang

Summary Oleogels of β-sitosterol (Sit) and beeswax (BW) were combined at varying ratios (w/w) and added to sunflower oil (SFO) at concentrations of 10 g per 100 g and 20 g per 100 g oil to prepare oleogels (Sit/BW/SFO). Structural and thermal properties were characterised and results showed that the hardness and enthalpy of oleogels were affected by the amount of β-sitosterol and beeswax in the oleogelator combination (Sit/BW, w/w). Oleogels with beeswax as the only oleogelator (Sit0/BW10) had the highest hardness and maximum enthalpy change. Gel network form was influenced by the crystalline behaviour of the oleogelator, and Sit0/BW10-oleogel was densely packed, spherical and white while Sit10/BW0-oleogel displayed a needle shape. X-ray diffraction patterns showed that the oleogel width of the crystals and D- spacing increased with increasing amounts of β-sitosterol and the FTIR spectra revealed that oleogels formed via non-covalent bonding and may be stabilised with physical entanglements.

Beta-sitosterol regulates PTGS1 to inhibit gastric cancer cell proliferation and angiogenesis

Jindao Wang,Minghui Zhou,Qiuli Zhou,Guangyang Sun,Yu Zhang,Feng Tao,Minfeng Ye

Background Gastric cancer (GC) is the third leading culprit of cancer-related deaths around the world. Beta-sitosterol (BS) is an important phytosterol that has been proven to have anti-proliferative effects on GC and other tumors. However, mechanisms and targets of BS in cancer are rarely explored. Methods In this investigation, the targets of BS in the treatment of GC were analyzed by network pharmacology. Molecular docking and cellular thermal shift assay were introduced to validate the binding relationship between BS and PTGS1. The impacts of BS on GC cell viability, half maximal inhibitory concentration, proliferation ability, apoptosis level, and angiogenesis ability were detected by using Cell Counting Kit-8, clone formation assay, flow cytometry, and angiogenesis experiment, respectively. In addition, the expression levels of angiogenic factors (VEGF, FGF, PAI-1) were detected by using western blot. Results In this project, through cell experiments, PTGS1 was identified as a protein that directly binds to BS. In vitro cell experiments revealed that BS promoted apoptosis and inhibited GC cell proliferation and angiogenesis. Importantly, treatment with BS attenuated the promoting influence of PTGS1 overexpression on GC cell proliferation and angiogenesis. Conclusion This investigation highlighted PTGS1 as the target of BS in GC cells. BS can regulate PTGS1 to inhibit GC cell proliferation and angiogenesis, providing new evidence for the potential use of BS as a therapeutic agent for GC.

Effect of β-sitosterol and palmitic acid mass ratio on structural, physicochemical, and rheological properties of rice bran oil-based oleogel

Shu Yang,Xueping Zhang,Ahmed S.M. Saleh,Lishuang Wang,Yumin Duan,Zhigang Xiao

Edible oil-based oleogels provide a viable and healthier alternative to traditional saturated fats. This study aimed to develop oleogels using rice bran oil combined with β-Sitosterol (Sit) and Palmitic Acid (PA). Structural, physicochemical, and rheological properties of the prepared oleogels were investigated. The results showed that Sit alone was insufficient to create a stable oleogel. However, when Sit was combined with PA, a firm and stable oleogel was obtained. The gel network was formed and stabilized through physical non-covalent interactions such as hydrogen bonding. Additionally, x-ray diffraction analysis indicated that the oleogel molecules rearranged and underwent a homogeneous polycrystalline transition due to the different gelator combinations, revealing the coexistence of β and β′ crystal types in the oleogels. Moreover, the firmness and oil binding capacity of the oleogels increased with higher PA content in the gelator mixture. Furthermore, the storage modulus (G′) and loss modulus (G″) were frequency-independent, with G′ significantly higher than G". The results suggest that fat-like oleogel can be produced using a combination of Sit and PA at mass ratios of Sit2:PA3 or Sit1:PA4 (w/w).

Functional Characteristics of Oleogel Prepared from Sunflower Oil with β-Sitosterol and Stearic Acid

Yang Shu,Li Guode,Saleh Ahmed S. M.,Yang Hongli,Wang Na,Wang Peng,Yue Xiqing,Xiao Zhigang

β-Sitosterol (Sit) and stearic acid (SA) were combined at varying ratios (w/w) and added to sunflower oil (SFO) at the concentration of 20 g/100 g oil for preparing edible fat-like oleogel. The oleogel was characterized using an optical microscope, Fourier transform infrared spectroscopy (FTIR), X-ray diffractometer (XRD), differential scanning calorimeter, and texture analyzer. The oil-binding capacity, melting temperature, and firmness of the oleogel increased with the increase in the amount of SA in the gelator combination (Sit:SA, w/w). The microscopic analysis showed that the gel network formed based on the crystallization and self-organization of gelator molecules, and both gelators showed an independent crystalline behavior in the oleogel. In addition, the FTIR spectra showed that the gel network formed via physical entanglements and was stabilized by non-covalent interactions such as hydrogen bonding. Furthermore, XRD diffraction patterns indicated high lateral packing of molecular layers in oleogel prepared with the Sit and SA combination compared with oleogel prepared with a single gelator. On the other hand, for studying the effect of varying concentrations of gelator combinations, the Sit3:SA2 (w/w) combination was added to SFO at concentrations of 10, 15, 20, 25, and 30 g/100 g oil. Specific characteristics such as the oil-binding capacity and firmness of the oleogel improved as the concentration of the gelator combination (Sit3:SA2) increased from 10 up to 30 g/100 g oil. Therefore, it can be concluded that the saturated fat alternative oleogel can be prepared from SFO with a specific Sit and SA combination ratio and concentration.

Monolithic Material Prepared with Nanodiamond as Monomer for the Enrichment of β-Sitosterol in Edible Oil

Cui Beijiao,Liu Haiyan,Yu Huan,Pang Xiaoya,Yan Hongyuan,Bai Ligai

In this paper, a hybrid monolithic column with modified nanodiamond and 1-dodecene as co-monomers was prepared and applied to the online enrichment and purification of β-sitosterol in edible oil. The obtained monolithic column was characterized by scanning electron microscopy (SEM) and nitrogen adsorption–desorption isotherm measurement, which indicated that the monolith possessed characteristics of porous structure and high permeability. Under the optimum conditions for extraction and determination, the linear regression coefficient for β-sitosterol was 0.998; the limit of detection (LOD) and the limit of quantitation (LOQ) were 3.0 μg mL −1 and 10.0 μg mL −1 , respectively; the relative standard deviation (RSD) of intra-day and inter-day assays for four edible oils were 2.33–6.58% and 3.24–6.94%, respectively. The recovery was in the range of 89.6–105.5%. The results showed that the hybrid monolithic column with high selectivity and good permeability were successfully used as online solid-phase extraction (SPE) column for determination of β-sitosterol in edible oil.

Effect of β-sitosterol on the curcumin-loaded liposomes: Vesicle characteristics, physicochemical stability, in vitro release and bioavailability

Kedong Tai,Michael Rappolt,Xiaoye He,Yang Wei,Shaoxin Zhu,Jingbo Zhang,Like Mao,Yanxiang Gao,Fang Yuan

In this work, the effect of β-sitosterol (Sito) on vesicle characteristics, physicochemical stability as well as the in vitro release and bioavailability of curcumin-loaded liposomes (Cur-LP) was studied. When 20–33 mol% of Sito was incorporated, encapsulation efficiency of curcumin was improved due to the high amount of liquid-ordered domains in membranes. At 50 mol% Sito a lower encapsulation efficiency was observed possibly due to membrane defects. The physical, thermal and photo stability of curcumin in liposomes were markedly improved with increasing the amount of Sito. First-order kinetics fitted best the curcumin release dynamics of Sito containing liposomes, clearly showing that sustained release improved with increasing amounts of Sito in liposomes. Simulated digestion studies suggested that Sito concentration of about 20–33 mol% improved the bioavailability of curcumin in liposomes. These study shows that Sito is an applicable and potential route in forming healthier cholesterol-free curcumin-loaded liposomes for functional supplements.

β-sitosterol protects against ANIT-induced hepatotoxicity and cholestasis via FXR activation

Yuhui Yan,Wenyu Wang,Aiwen Yan,Haonan Zhu,Qiang Meng

Cholestasis, a condition marked by bile acid accumulation in the liver and body systems, leads to liver dysfunction and cirrhosis. Currently, ursodeoxycholic acid (UDCA) and obeticholic acid (OCA) are the only two FDA-approved drugs for Cholestasis. Thus, new therapeutic approaches need to be developed. In this study, we validated the liver-protective properties of β-sitosterol (SIT), a key bioactive element abundant in plants, against hepatic toxicity and cholestasis induced by alpha-naphthylisothiocyanate(ANIT), while elucidating its mechanisms of action both in vivo and in vitro. SIT's FXR activation was confirmed via molecular docking and dual-luciferase assays. In the mechanisms of SIT hepatoprotection, the expression levels of bile salt export pump (Bsep) and multidrug resistance protein2 (Mrp2) which are bile acid efflux transporter, and sulfate transferase 2a1 (Sult2a1) which is a bile acid metabolizing enzyme were all increased by SIT, whereas the expression of uptake transporter sodium taurocholate transporting polypeptide (Ntcp), bile acid synthesis enzyme cholesterol 7α-hydroxylase (Cyp7a1) and oxysterol 12α-hydroxylase (Cyp8b1) was decreased by SIT. In addition, SIT alleviated liver inflammation by suppressing inflammatory factor expression. However, FXR antagonist guggulsterone and FXR siRNA abolished SIT's improvements in liver histology, bile acid transporters, and enzymes. Conclusively, through activating FXR, SIT provides a protective effect against hepatotoxicity and cholestasis. SIT might serve as a new potential therapeutic strategy for the treatment of cholestatic liver diseases.

Self-assembled fibrillar networks induced by two methods: a new unmodified natural product gel

Ning He,Kangkang Zhi,Xin Yang,Haitian Zhao,Hua Zhang,Jing Wang,Zhenyu Wang

To date, low molecular weight gels based on natural products have attracted much attention due to their excellent biological activities. Recently, some natural products with new features (e.g., natural product gelators, NPGs) have been reported to directly self-assemble in different solvents to form soft material-gels, i.e., natural product gels. Unlike synthetic low molecular weight gelators, NPGs come from organisms and therefore, their structures and properties are unknown. Herein, a general and efficient method for obtaining NPGs from organisms, which provides specific and important guidance for researchers to easily discover new NPGs from organisms, has been reported. We report a new NPG, β-sitosterol (Sito), isolated from Arisaema heterophyllum Blume. SEM, UV-vis, FT-IR, CAT (contact angle text), XRD, and theoretical calculations were used to explain the properties, morphology, driving force, and molecular packing models of the gels. Sito self-assembled into different fibrillar networks via heat–cool and ultrasound methods. Then, we demonstrated that hydrogen bonding plays a major role in the formation of these nanoaggregates. In addition, in vitro experiments showed that the nanoaggregates have similar or better ability to inhibit the growth of cancer cells as Sito. Therefore, the Sito nanoaggregates are expected to be potential drug delivery materials.

Controlling release of astaxanthin in β-sitosterol oleogel-based emulsions via different self-assembled mechanisms and composition of the oleogelators

Shujie Wang,Yuyue Qin,Yaping Liu,Guoqin Liu,Guiguang Cheng,Thanapop Soteyome

In this study, three types of β -sitosterol-based oleogels ( β -sitosterol +  γ -oryzanol oleogels, β -sitosterol +  lecithin , oleogels and β -sitosterol + monostearate oleogels), loaded with astaxanthin, were employed as the oil phase to create oleogel-based emulsions (SO, SL, and SM) using high-pressure homogenization. The microstructure revealed that fine-scale crystals were dispersed within the oil phase of the droplets in the β -sitosterol oleogel-based emulsion. The bioaccessibility of astaxanthin was found to be 58.13 %, 51.24 %, 36.57 %, and 45.72 % for SM, SL, SO, and the control group, respectively. Interestingly, the release of fatty acids was positively correlated with the availability of astaxanthin (P = 0.981). Further analysis of FFAs release and kinetics indicated that the structural strength of the oil-phase in the emulsions influenced the degree and rate of lipolysis. Additionally, the micellar fraction analysis suggested that the nature and composition of the oleogelators in SM and SL also impacted lipolysis and the bioaccessibility of astaxanthin. Furthermore, interfacial binding of lipase and isothermal titration calorimetry (ITC) measurements revealed that the oleogel network within the oil phase of the emulsion acted as a physical barrier, hindering the interaction between lipase and lipid. Overall, β-sitosterol oleogel-based emulsions offer a versatile platform for delivering hydrophobic molecules, enhancing the bioavailability of active compounds, and achieving sustained release.

Controlled volatile release from β-sitosterol-based oleogels based on different self-assembly mechanisms

Shujie Wang,Guoqin Liu

This study examined how the self-assembly mechanisms of β -sitosterol-based oleogels influenced the release of volatile compounds. Microscopy, X-ray diffraction (XRD) and small-angle X-ray scattering (SAXS) measurements showed that the three β -sitosterol-based oleogels ( β -sitosterol +  γ -oryzanol oleogels (SO), β -sitosterol + lecithin oleogels (SL) and β -sitosterol + monostearate oleogels (SM)) had significant differences in their microstructures, which were formed via different self-assembly mechanisms. SO exhibited the highest oil binding capacity (OBC), complex modulus (G*) and apparent viscosity. Dynamic and static headspace analyses suggested that network structure of β -sitosterol-based oleogels affected the release of volatile components. SO showed the strongest retention effect, followed by SL and SM. The release of volatile compounds mainly related to structural strength and compositions of oleogels. These results indicated that β -sitosterol-based oleogels formed with different self-assembly mechanisms have the potential to serve as effective delivery systems for controlling the release of volatile compounds.

Effect of the water and oleogelator content on characteristics and stability of BC-loaded oleogel-based emulsion

Shu Yang,Ahmed S.M. Saleh,Qiang Yang,Xiaotong Cui,Yumin Duan,Zhigang Xiao

Oleogel samples were prepared from sunflower oil with β-Sitosterol (Sit) and Stearic acid (SA) mixture. Water was added to prepare an oleogel-based emulsion. The oleogel-based emulsion showed a white and smooth appearance without separation after storage at 5 ο C for 24 h. The firmness of emulsion reduced as the water content increased, while it was increased as the oleogelator concentration increased. Additionally, diffraction patterns and thermal properties of emulsion varied depending on water content. Furthermore, all the oleogel-based emulsion samples showed a positive slope of storage modulus (G′) and loss modulus (G″) with the increase in frequency. On the other hand, the stability of β-carotene (BC) -loaded emulsion to different conditions was evaluated. A greater BC content was found for samples stored in the dark and under natural light than that of samples stored under UV light. Oleogel and oleogel-based emulsion showed a good BC release rate in simulated gastrointestinal fluids. The obtained results indicate that functional BC-fortified emulsion can be successfully prepared using β-Sitosterol and Stearic acid mixture with sunflower oil and water.
