Bone defect repair is a hot topic in tissue engineering, and the development of economical and effective bone scaffold materials to achieve the goal of repairing bone defects is an important task for researchers. In this experiment, fresh bovine cancellous bone was used as a xenogeneic
bone material. After two high-temperature calcinations, β-tricalcium phosphate (β-TCP) material was obtained. An in-situ co-precipitation method was employed to prepare hydroxyapatite+graphene oxide (HA+GO) composite material. Then, the β-TCP material
was immersed in the HA+GO composite material solution to prepare β-TCP+HA+GO composite material. The surface modification of β-TCP material with HA and GO improved the elastic modulus of the raw material. In vitro cell showed that the composite material was non-toxic,
biocompatible, and promoted the proliferation of MC3T3-E1 cells on the β-TCP+HA+GO material. Finally, it was found that β-TCP+HA+GO composite material effectively stimulated the actin and mineralization behavior of MC3T3-E1 cells, and promoted the expression of bone-related
factors such as Alkaline phosphatase (ALP), Collagen I (COLI), Osteocalcin (OCN), Bone Morphogenetic Protein-2 (BMP-2), and Bone Morphogenetic Protein-7 (BMP-7) in MC3T3-E1 cells In conclusion, β-TCP+HA+GO composite material can be applied in the biomedical field as a bone substitute
or repair material.

Cartilage repairing grafts have been widely studied, and osteochondral replacement hydrogels have proven to be an excellent method in research and clinical fields. However, it has been difficult to simultaneously solve three main issues in osteochondral replacement preparation: surface lubrication, overall mechanical support and good simulations of cell regeneration. A novel integrated bilayered hydrogel osteochondral replacement was constructed by blending polyvinyl alcohol (PVA) and β-tricalcium phosphate (β-TCP) in this study. Separated nano-ball milling with ultrasound dispersion prepared β-TCP demonstrated suitable properties of tiny particle size, high purity and ideal distribution, improving the mechanical properties of the novel integrated hydrogel, and providing a cartilage-like lubrication effect and high biocompatibility, including cytocompatibility and osteogenesis. The reinforcement of β-TCP and integrated molding technology enabled the hydrogel to demonstrate excellent component compatibility and good bonding between the two layers, which promoted the strengthening of the compression modulus and tensile modulus up to three times by mechanical testing. The surface lubrication properties of the novel osteochondral hydrogel were similar to the natural cartilage by friction coefficient characterization. The two layers of the novel integrated graft provided a considerable bio-function by co-culturing with chondrocytes and synovium mesenchymal stem cells: chondrocytes promoted adherence achieved by the upper density layer and better osteogenesis performance of the porous lower layer. The design of the bilayered β-TCP/PVA osteochondral hydrogel is promising for use in articular cartilage repair.

Piezoelectricity is reported to be able to promote bone scaffolds with excellent osteogenic performance. Herein, barium titanate/β-tricalcium phosphate (BTO/β-TCP) piezoelectric composite scaffolds were 3D printed, and their osteogenic performances were investigated in detail. The fabrication of BTO/β-TCP piezoelectric composite scaffolds employed cutting-edge DLP 3D printing technology. The scaffolds, featuring a triply periodic minimal surface (TPMS) design with a porosity of 60%, offered a unique structural framework. A comprehensive assessment of the composition, piezoelectric properties, and mechanical characteristics of the BTO/β-TCP scaffolds was conducted. Notably, an increase in the BTO volume fraction from 50 to 80 vol % within the scaffolds led to a reduction in compressive strength, decreasing from 2.47 to 1.74 MPa. However, this variation was accompanied by a substantial enhancement in the piezoelectric constant d33, soaring from 1.4 pC/N to 21.6 pC/N. Utilizing mouse osteoblasts (MC3T3-E1) in a live/dead cell staining assay, under the influence of external ultrasound, demonstrated the commendable biocompatibility of these piezoelectric composite ceramic bone scaffolds. Furthermore, thorough analyses of alkaline phosphatase (ALP) activity and polymerase chain reaction (PCR) findings provided compelling evidence of the scaffolds’ superior osteogenic properties, underpinning their effectiveness at the cellular protein and gene levels. In conclusion, this study offers a groundbreaking strategy for the employment of BTO/β-TCP piezoelectric composite scaffolds in bone implant applications, harnessing their unique blend of biocompatibility, piezoelectricity, and osteogenic potential.

Biodegradable polymer/bioceramic composite scaffolds can overcome the limitations of conventional ceramic bone substitutes such as brittleness and difficulty in shaping. A novel degradable scaffold material for bone tissue repairing was developed by combining 1,4-butanediamin-modified poly(lactide-co-glycolide) (BMPLGA) with nano-biaoactive glass and nano-β-tricalcium phosphate (NBAG-β-TCP) . The effects of the inorganic filler content on the morphology, water adsorption, and compressive strength of the nanocomposite scaffolds were investigated. The nanocomposite scaffolds showed an interconnected pore structure with the pore size ranging from 50 to 200 μm. The compressive strength and the water adsorption ability were enhanced with increasing content of the inorganic fillers. The cell proliferation of fibroblast was significantly improved after three and seven days of culture on BMPLGA/NBAG-β-TCP films in comparison with BMPLGA films. The BMPLGA/NBAG-β-TCP composites have a wide application as a potential biomaterial in medical field.

Biphasic calcium phosphate (BCP) compositions consisting of β-tricalcium phosphate (β-TCP) and calcium pyrophosphate (CPP) are potential biodegradable ceramics for bone regeneration. The present work demonstrates the formation of such dense ceramics by first preparing the precursors of nano-sized, amorphous , and equiaxed calcium pyrophosphate particles, and then sintering the precursors at 900 °C to transform them into desired BCP. However, if the complex of calcium tripolyphosphate was used, only CPP could be generated. It was also observed that the incorporation of Mg 2+ had several effects on the resulting products including: (1) promoting the generation of meso-porous precipitates; (2) favoring the formation of β-TCP instead of CPP; (3) reducing the grain size and increasing the density of the sintered ceramics, and (4) enhancing the negative electric charge of the BCP surface. Thus, the as-prepared BCP ceramics can serve as potential bone substitute materials in orthopedic applications.

Objectives Vascularization is an essential step in successful bone tissue engineering. The induction of angiogenesis in bone tissue engineering can be enhanced through the delivery of therapeutic agents that stimulate vessel and bone formation. In this study, we show that cucurbitacin B (CuB), a tetracyclic terpene derived from Cucurbitaceae family plants, facilitates the induction of angiogenesis in vitro. Methods We incorporated CuB into a biodegradable poly (lactide-co-glycolide) (PLGA) and β-tricalcium phosphate (β-TCP) biomaterial scaffold (PT/CuB) Using 3D low-temperature rapid prototyping (LT-RP) technology. A rat skull defect model was used to verify whether the drug-incorporated scaffold has the effects of angiogenesis and osteogenesis in vivo for the regeneration of bone defect. Cytotoxicity assay was performed to determine the safe dose range of the CuB. Tube formation assay and western blot assay were used to analyze the angiogenesis effect of CuB. Results PT/CuB scaffold possessed well-designed bio-mimic structure and improved mechanical properties. CuB was linear release from the composite scaffold without affecting pH value. The results demonstrated that the PT/CuB scaffold significantly enhanced neovascularization and bone regeneration in a rat critical size calvarial defect model compared to the scaffold implants without CuB. Furthermore, CuB stimulated angiogenic signaling via up-regulating VEGFR2 and VEGFR-related signaling pathways. Conclusion CuB can serve as promising candidate compound for promoting neovascularization and osteogenesis, especially in tissue engineering for repair of bone defects. The translational potential of this article This study highlights the potential use of CuB as a therapeutic agent and strongly support its adoption as a component of composite scaffolds for tissue-engineering of bone repair.

Being one of the three elements of tissue engineering, three-dimensional porous structure scaffold plays an important role in tissue engineering. As it not only prvovide cells for the life, but also serves as a template to guide tissue regeneration and control of organizational structure and other functions. In this study, hyaluronic acid and gelatin are successfully cross-linked by 1-ethyl- (3-dimethylaminopropyl) -carbodiimide hydrochloride (EDC) , and compound β-TCP microspheres to prepare porous hydrogel scaffolds. The microspheres were analyzed by X-ray diffraction (XRD). The scaffolds were characterized by scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR). At the same time, the compressive strength, swelling ratio, degradation of the scaffold were tested. To assess the in vitro cell compatibility of the scaffolds, mouse L929 fibroblasts were seeded onto scaffolds for cell morphology and cell viability studies. The results showed that the pore size of the porous scaffold can be adjusted by changing the ratio of gelatin to hyaluronic acid (HA), increasing the proportion of hyaluronic acid in a certain range, pore size will be significantly increased. With the increase of the proportion of hyaluronic acid in the scaffold, the swelling ratio and the degradation rate also increased. The compressive strength of the scaffold increased with the increase of the proportion of gelatin. The appropriate ratio of β-TCP can promote cell growth and proliferation.

Segmental bone defects caused by trauma, tumor resection or congenital malformations are often reconstructed with autologous, allogeneic bone grafts or artificial bone materials, of which, about 5% ~ 10% will have delayed healing or even nonunion of fractures. The loss of periosteum and excessive accumulation of ROS in fracture site leads to the aging of osteoblasts and the decline of their proliferation and differentiation, thus affecting the fracture healing process. In this study, we prepared a functional modified artificial periosteum β-TCP/MnO 2 /PCL(β-TMP) by electrospinning with a function of catalyzing decomposition of H 2 O 2 . We examined the surface morphology of β-TMP, the concentration of Ca, P and Mn of β-TMP, as well as the diameter distribution range of nanofibers on β-TMP. Through X-ray diffraction patterns and Fourier transform infrared spectra, β-TMP was characterized and its hydrophilicity was tested. The release of Mn 2+ and Ca 2+ of 0.1 and 0.05% β-TMP in different pH values (7.4 and 5.5) determined by ICP. We also identified that β-TMP could reduce the level of ROS in cells by lowering the level of H 2 O 2 . 0%, 0.05% and 0.1% β-TMP displayed good cell compatibility, cell adhesion and cellular morphology in the condition with or without H 2 O 2 . 0.5% β-TMP showed compromised cell compatibility in normal condition, however, the compromised phenotypes could be partially rescued in the present of H 2 O 2 . Compared with 0%, 0.05% and 0.1% β-TMP displayed higher osteoblastic differentiation with or without H 2 O 2 in BMSCs as well as in MG-63. In sum, β-TMP helped osteogenesis and promoted repair of bone defects.

Although β-TCP bioceramics possess favorable biocompatibility and bioactivity, their inherent brittleness restricts further applications. In this study, we aimed to enhance the mechanical performance and biocompatibility of β-TCP by incorporating 15% Akermanite (AK) into the structure. The fabrication process involved the use of digital light processing (DLP) technology to fabricate ceramic green bodies, followed by high-temperature debinding and sintering to obtain the ceramic scaffold. Subsequently, we employed an in-situ growth technique to create micro/nano surface topography on the scaffolds, facilitating synergistic regulation of cell behavior in conjunction with the controlled release of bioactive agents. Scanning electron microscopy (SEM) revealed that the addition of AK enlarged the grain size and reduced the number of micropores in the composite scaffold. Furthermore, after the in-situ growth treatment, a significant increase in micro/nano surface features was observed. Mechanical testing demonstrated that the incorporation of AK effectively improved the mechanical properties of the composite scaffold, resulting in an increase in strength of approximately 20%. Cell experiments indicated that AK supplementation further enhanced the material's cell compatibility, and the presence of micro/nano structures effectively promoted cell adhesion, proliferation, and expression of osteogenic genes. Consequently, we propose a strategy for fabricating porous bioceramics that stimulate bone growth, exhibit high strength, and ameliorate the degradation microenvironment. This approach opens up promising avenues for future applications in bone tissue engineering.

Scaffolds prepared by 3D printing are increasingly used in the field of bone tissue repair. However, on traditional 3D printed bone tissue engineering scaffolds, cells can only grow on the fiber surface and form bone. We designed a scaffold with a cross-scale structure of PCL/β-TCP, which contains thick fibers with a diameter of 500 μm printed by FDM. And in the pores of the coarse fiber, the ultra-high precision fine fiber grid with a diameter of about 10 μm is filled by MEW mode. In cell experiments, cells can not only grow on the thick fiber surface of the cross-scale scaffold. At the same time, the mesh structure of fine fibers provides a bridge for cell growth, allowing cells to pass through the pores of thick fibers and grow in the pores and gradually cover the pores of the scaffold. In the osteoinduction experiment, β-TCP in the PCL/β-TCP composite provides Ca 2+ and PO 4 3− to the scaffold, which effectively promotes the osteogenic differentiation of cells on the scaffold. Compared with traditional scaffolds, the osteogenic performance of cross-scale scaffolds is greatly improved. Not only did bone form on the surface of the scaffold, but also obvious ALP expression and effective calcium precipitation appeared in the pores of the scaffold. This can effectively speed up the repair of bone defects. We believe that the 3D printed PCL/β-TCP cross-scale scaffold with high-precision fibers has great application prospects in the field of bone tissue engineering.

Background: Three-dimensional (3D) printed bone tissue engineering scaffolds have been widely used in research and clinical applications. β-TCP is a biomaterial commonly used in bone tissue engineering to treat bone defects, and its multifunctionality can be achieved by co-doping different metal ions. Magnesium doping in biomaterials has been shown to alter physicochemical properties of cells and enhance osteogenesis. Methods: A series of Mg-doped TCP scaffolds were manufactured by using cryogenic 3D printing technology and sintering. The characteristics of the porous scaffolds, such as microstructure, chemical composition, mechanical properties, apparent porosity, etc., were examined. To further study the role of magnesium ions in simultaneously inducing osteogenesis and angiogenesis, human bone marrow mesenchymal stem cells (hBMSCs) and human umblical vein endothelial cells (HUVECs) were cultured in scaffold extracts to investigate cell proliferation, viability, and expression of osteogenic and angiogenic genes. Results: The results showed that Mg-doped TCP scaffolds have the advantages of precise design, interconnected porous structure, and similar compressive strength to natural cancellous bone. hBMSCs and HUVECs exhibit high proliferation rate, cell morphology and viability in a certain amount of Mg 2+ . In addition, this concentration of magnesium can also increase the expression levels of osteogenic and angiogenic biomarkers. Conclusion: A certain concentration of magnesium ions plays an important role in new bone regeneration and reconstruction. It can be used as a simple and effective method to enhance the osteogenesis and angiogenesis of bioceramic scaffolds, and support the development of biomaterials and bone tissue engineering scaffolds.

Porous Hyaluronan/β-tricalcium phosphate composite scaffolds were synthesized through lyophilizing and subsequent heterogeneous crosslinking method. The morphology of the composite scaffolds were investigated by scanning electron microscopy (SEM). The swelling behavior, mechanical property, degradation behavior and cell adhesion ability of samples were also studied. The results revealed that hyaluronan mainly contributed to the polymer matrix and water adsorption, whereas β-TCP acted as a reinforcement to strengthen the porous structure, while too much β-TCP would make the structure collapse. The pose size of obtained scaffolds ranges from100μm to 200μm and the porosity decreased with the increase of β-TCP content. The degradation behavior and cell adhesion test indicated that increasing hyaluronan concentration can effectively improve the degradability of scaffolds and the incorporation of β-TCP improved the cell adhesion performance. Thus a simple way to prepare hyaluronan-based composite scaffolds was provided, which could be potentially used as an tissue engineering material.
