Development of monoclonal antibody and lateral test strip for sensitive detection of clenbuterol and related β2-agonists in urine samples

Gabriel Wafula Khaemba,Bitange Nipa Tochi,Daniel Mukunzi,Isanga Joel,Lingling Guo,Steven Suryobrobowo,Shanshan Song,Hua Kuang,Chuanlai Xu

Being the most used and preferred β2-agonist, clenbuterol (CLB) has been reported to be illegally abused in animal production and breeding as it improves growth rate while inhibiting fat synthesis and deposition leading to buildup of lean muscle. Due to its potential hazard to human health, causing food poisoning that can be fatal, it has been forbidden as a growth promoter for food-producing animals in most countries including China. Therefore, frequent monitoring of CLB abuse is necessary for consumer safety against its residues. We therefore report development of a monoclonal antibody and lateral flow test strip for sensitive detection of CLB in urine samples. In this investigation, male BALB/C mice were first immunized for antibody production. Thereafter, gold nanoparticles were utilized for antigen and antibody immobilization during construction of the strip. Under optimized conditions, the cut-off limit of the test strip was found to be 2.5 and 5 ng/g in phosphate-buffered saline (PBS) and urine using the wet method. When the dry method was used for CLB detection, the cut-off limit of the test strip in PBS and urine was found to be 3 ng/g. Each test was evaluated within 6 min. The data obtained show that the strip developed is sensitive and has an advantage of fast test results and simplicity and hence can be adopted for screening of CLB residues in urine samples for mitigation of CLB residue effects to human health.

Novel Stir Bar Array Sorptive Extraction Coupled With Gas Chromatography–Mass Spectrometry for Simultaneous Determination of Three β2-Agonist Residues in Pork

Liu Haibo,Ousmane Diebate,Gan Ning,Wu Dazhen,Li Tianhua

Abstract This study aimed to simultaneously determine three β2-agonists (clenbuterol, salbutamol, and ractopamine) in pork samples using novel stir bar array sorptive extraction coupled with gas chromatography–mass spectrometry. Pencil refill was employed as the substrate of stir, which was covered with carboxyl graphene sheets and polyluminol coating. The coating was simply prepared by electrochemical deposition of luminol at a voltage of 0–0.6 V. Then, an automatic device with the stir bar array for extraction was used. The conditions for the extraction were optimized. Under the optimum extraction conditions, with toluene as the extraction solvent, the extraction and desorption times were 20 min. A good linearity of three β2-agonists was obtained in the range of 0.5–100 ng g −1 by gas chromatography–mass spectrometry detection. The recovery of clenbuterol, salbutamol, and ractopamine in spiked pork samples ranged from 88.7 to 93.2%, 85.5–94.2%, and 85.7–93.9%, respectively, with the relative standard deviations less than 10%. The detection limits were in the range of 0.022–0.091 ng g −1 for pork samples. The extraction coating with high extraction capacity was simply fabricated by the electrochemical method, which could simplify the pretreatment procedures. Moreover, the array could extract several samples simultaneously and improve the pretreatment efficiency. Graphical Abstract

Fabrication of a molecularly imprinted polymer immobilized membrane with nanopores and its application in determination of β2-agonists in pork samples

Xiuzhen Qiu,Xian-Yan Xu,Yong Liang,Yongbiao Hua,Huishi Guo

In this paper, a method for the synthesis of ractopamine molecularly imprinted polymers (MIPs) nanotube membranes on anodic alumina oxide (AAO) nanopore surface by atom transfer radical polymerization (ATRP) was presented, in which methacrylic acid (MAA) was selected as functional monomer with a polymerization rate of 1:6 between ractopamine and MAA by the computational investigations. The morphology of MIPs nanotube membranes characterized by scanning electron microscope (SEM) suggested a well growth in the AAO nanopore surface. A series of adsorption experiments revealed that the MIPs nanotube membranes showed better extraction capacity and good selectivity for ractopamine and its analogues than that of non-imprinted polymers (NIPs) nanotube membranes. In order to evaluate the usability of the MIPs nanotube membranes, a methodology by combining MIPs nanotube membranes extraction couple with high performance liquid chromatography (HPLC) detection for the determination of β 2 -agonists in complex samples was developed. The linear ranges were 10–1000   μg/L for ractopamine, 100–1000   μg/L for clenbuterol, epinephrine and dopamine, and 200–1000   μg/L for terbutaline. The detection limits were within the range of 0.074–0.25   μg/L and the RSDs ( n   =   3) were from 2.8% to 4.3%. The method was successfully applied to the analysis of β 2 -agonists in spiked real samples, The recoveries of all the β 2 -agonists at the two concentration levels were found to be within the range of 86.3–97.0% and 82.8–95.7%, respectively. The RSDs were within 2.7–5.7%. The results demonstrated that the proposed method is very suitable for the determination of β 2 -agonists in pork samples.

Novel method for the rapid and specific extraction of multiple β2-agonist residues in food by tailor-made Monolith-MIPs extraction disks and detection by gas chromatography with mass spectrometry

Haibo Liu,Ning Gan,Yinji Chen,Qingqing Ding,Jie Huang,Saichai Lin,Yuting Cao,Tianhua Li

A quick and specific pretreatment method based on a series of extraction clean-up disks, consisting of molecularly imprinted polymer monoliths and C 18 adsorbent, was developed for the specific enrichment of salbutamol and clenbuterol residues in food. The molecularly imprinted monolithic polymer disk was synthesized using salbutamol as a template through a one-step synthesis process. It can simultaneously and specifically recognize salbutamol and clenbuterol. The monolithic polymer disk and series of C 18 disks were assembled with a syringe to form a set of tailor-made devices for the extraction of target molecules. In a single run, salbutamol and clenbuterol can be specifically extracted, cleaned, and eluted by methanol/acetic acid/H 2 O. The target molecules, after a silylation derivatization reaction were detected by gas chromatography-mass spectrometry. The parameters including solvent desorption, sample pH, and the cycles of reloading were investigated and discussed. Under the optimized extraction and clean-up conditions, the limits of detection and quantitation were determined as 0.018–0.022 and 0.042–0.049 ng/g for salbutamol and clenbuterol, respectively. The assay described was convenient, rapid, and specific; thereby potentially efficient in the high-throughput analysis of β 2 -agonists residues in real food samples.
