μPADs on Centrifugal Microfluidic Discs for Rapid Sample-to-Answer Salivary Diagnostics

Shixian Liu,Yuting Hou,Zirui Li,Chenyu Yang,Guozhen Liu

A fully integrated device for salivary detection with a sample-in-answer-out fashion is critical for noninvasive point-of-care testing (POCT), especially for the screening of contagious disease infection. Microfluidic paper-based analytical devices (μPADs) have demonstrated their huge potential in POCT due to their low cost and easy adaptation with other components. This study developed a generic POCT platform by integrating a centrifugal microfluidic disc with μPADs to realize sample-to-answer salivary diagnostics. Specifically, a custom centrifugal microfluidic disc integrated with μPADs is fabricated, which demonstrated a high efficiency in saliva treatment. To demonstrate the capability of the integrated device for salivary analysis, the SARS-CoV-2 Nucleocapsid (N) protein, a reliable biomarker for SARS-CoV-2 acute infection, is used as the model analyte. By the chemical treatment of the μPAD surface, and by optimizing the protein immobilization conditions, the on-disc μPADs were able to detect the SARS-CoV-2 N protein down to 10 pg mL–1 with a dynamic range of 10–1000 pg mL–1 and an assay time of 8 min. The integrated device was successfully used for the quantification of the N protein of pseudovirus in saliva with high specificity and demonstrated a comparable performance to the commercial paper lateral flow assay test strips.

Semi-quantitative analysis of nickel: counting-based μPADs built via hand drawing and yellow oily double-sided adhesive tape

Jian Wang,Tong Yang,Zhengjia Li,Kecen Zhou,Bo Xiao,Peng Yu

Excess exposure to a high environmental level of nickel can cause serious harm to human health. Therefore, it is particularly necessary to develop a low-cost, fast and sensitive method for nickel assay. In this paper, novel counting-based microfluidic paper-based devices (μPADs) were prepared by hand drawing and yellow oily double-sided adhesive tape. The dissolved adhesive tape was used for the first time to make the hydrophobic “ink”. The marker filled with the “ink” drew the desirable layout on paper followed by a drying process. The μPADs were constituted of one circular sample introduction zone (diameter 4.5 mm) and four circular detection zones (diameter 3 mm). The adjoining detection zones were connected by a strip channel (1.2 mm × 2 mm). The fabrication conditions were optimized and the barriers created with the marker revealed good reproducibility. The analytical performance of the developed devices was investigated for nickel assay. The Ni2+ standard was added to the sample introduction zone, and subsequently moved into the detection zones containing dimethylglyoxime (DMG), where it reacted with Ni2+ and formed a reddish pink Ni–DMG complex. Through counting the number of colored dots, the developed μPADs realized the semi-quantitative analysis of nickel. More importantly, the fabrication of the developed counting-based μPADs only used a marker and adhesive tape, possessing the advantages of instrument-free simplicity and low cost. Our method has great potential for enabling μPADs to be easily implemented in laboratories or research centers with limited resources.

Portable detection of trace metals in airborne particulates and sediments via μPADs and smartphone

Jia Yuan,Dong Hui,Zheng Jianping,Sun Hao

Particulate matter (PM), a key indicator of air pollution by natural and anthropogenic activities, contributes to a wide spectrum of diseases that lead to a shortening of life expectancy. It has been recognized that trace metals in airborne PM are highly toxic and can be correlated with lesion in respiratory, gastrointestinal, immunological, and hematological systems. Traditional methods for trace metal assay require sophisticated instrumentations and highly trained operators in centralized laboratories. In this work, by integrating the technologies of microfluidic paper-based analytical devices, additive manufacturing, smartphone, and colorimetric sensing, we developed the first smartphone based paper microfluidic platform for portable, disposable, and quantitative measurements of cobalt (Co), copper (Cu), and iron (Fe) in ambient air and street sediments. On a single A4-sized paper, 48 devices were fabricated in under 30 s with a total cost of ∼$1.9. On each device, 12 reaction units were patterned and used for colorimetric tests. Particulate samples from urban ambient air and street sediments were collected, processed, and analyzed. Signals of the on-chip complexation product were recorded using a smartphone camera and processed by a self-developed app on an iOS system. For precisely controlling the object distance, chip position, and luminance, a hand-held 3D cellphone housing was designed and printed. The detection limits of Co, Cu, and Fe were determined to be 8.2, 45.8, and 186.0 ng, while the linear dynamic ranges were calculated to be 8.2–81.6, 45.8–4.58 × 10 2 , and 1.86 × 10 2 –1.86 × 10 3  ng, representing a practically relevant device performance with a significant reduction in the detection cost and time consumption. Trace metals in ambient air and sediments of two cities in China have been quantified portably, thus demonstrating the utility of our system in improving strategies for air pollution control in low-resource settings.

Digital Quantification Method for Sensitive Point-of-Care Detection of Salivary Uric Acid Using Smartphone-Assisted μPADs

Kexin Fan,Jiayang Zeng,Chenyu Yang,Gonglei Wang,Kai Lian,Xiuhong Zhou,Yaping Deng,Guozhen Liu

Uric acid (UA) is an important biomarker for many diseases. A sensitive point-of-care (POC) testing platform is designed for the digital quantification of salivary UA based on a colorimetric reaction on an easy-to-build smartphone-assisted microfluidic paper-based analytical device (SμPAD). UA levels are quantified according to the color intensity of Prussian blue on the SμPAD with the aid of a MATLAB code or a smartphone APP. A color correction method is specifically applied to exclude the light effect. Together with the engineering design of SμPADs, the background calibration function with the APP increases the UA sensitivity by 100-fold to reach 0.1 ppm with a linear range of 0.1–200 ppm. The assay time is less than 10 min. SμPADs demonstrate a correlation of 0.97 with a commercial UA kit for the detection of salivary UA in clinical samples. SμPADs provide a sensitive, fast, affordable, and reliable tool for the noninvasive POC quantification of salivary UA for early diagnosis of abnormal UA level-associated health conditions.

A novel combination of quick response code and microfluidic paper-based analytical devices for rapid and quantitative detection

Wang Tengfei,Xu Guangning,Wu Wenyuan,Wang Xiaoying,Chen Xueliang,Zhou Shumin,You Fan

Nowadays, there is great interest in the use of microfluidic paper-based analytical devices (μPADs) for low-cost diagnostics. In most cases, the test equipment is needed. Here we report a new type of device-independent detection method based on timer-paper-based analytical devices, which can be tested by smartphone, and its application for cholesterol detection. The Quick Response code was designed as the timer component of this device. Wax printing method was employed to print the pattern on filter paper. The color and enzyme reagents have been immobilized in the hydrophilic channel to complete the colorimetric detection for cholesterol. Under laminar flow conditions of the cellulose network, the liquid volume of detection zone has been quantified by monitoring the fluid residence time on different area of the timer-paper-based devices. The precise monitoring of detection time can promote the accuracy of colorimetric detection. This is very important for the quantitative detection of paper-based analytical devices. One significant outcome of this report is that simple and accurate timer can be used for detection process self-clocking. The factors of total detection time have been investigated. The linear range of the calibration curve was 3.0 ~ 6.0 mmol L −1 , with correlation coefficient of 0.9956. With the characteristic of easy to use, low cost and accurate monitoring of detection time, this kind of timer-paper-based devices can be applied to cholesterol or other substances rapid detection.

Isoelectric focusing on microfluidic paper-based chips

Yu Siyang,Yan Chunfang,Hu Xianqiao,He Baoqi,Jiang Yan,He Qiaohong

Isoelectric focusing (IEF), a powerful technique for protein separation and enrichment, was successfully integrated into microfluidic paper-based analytical devices (μPADs) in this work. The μPADs for isoelectric focusing were fabricated by octadecyltrichlorosilane (OTS) silanization and subsequent region-selective plasma treatment. The system of IEF on μPADs could be easily assembled. And a series of conditions of the system were investigated, including the suitable concentration of ampholyte to create good pH gradient, the effect of polyvinylpyrrolidone (PVP) on electroosmotic flow (EOF) suppression, and focusing voltage applied on the paper channel. After optimization, simultaneous separation and enrichment of protein sample containing myoglobin and cytochrome C was successfully demonstrated. Besides, parallel IEF on multichannels were also achieved for the separation of multiple protein samples on one single chip, and their performance was compared with that of the conventional gel-IEF system. The developed IEF on μPADs exhibits appealing features such as low cost, simplicity, and disposability and are believed to have great application potentials.

Development of a paper-based microfluidic analytical device by a more facile hydrophobic substrate generation strategy

Yuan-Yuan Xue,Wen-Tao Zhang,Meng-Yue Zhang,Li-Zhi Liu,Wen-Xin Zhu,Ling-Zhi Yan,Jing Wang,Yan-Ru Wang,Jian-Long Wang,Dao-Hong Zhang

Microfluidic paper-based analytical devices (μPADs) have a significant potential in developing portable and disposable point-of-care testing (POCT). Herein, a facile, rapid, cost-effective and environment friendly strategy for μPADs fabrication is proposed. Specifically, the substrate paper was hydrophobized by coating with trimethoxysilane (TOS), and then the selected area was hydrophilized by treating with surfactant. The whole fabrication process was implemented within 7 min, with no need for complex pre-treatment, high-temperature and special equipment. As a proof-of-concept application, the as-prepared μPAD was applied to determination of the glucose content in human serum samples. The results agreed well with those obtained by a glucometer. We believe that the μPADs fabrication method proposed here could provide a facile, rapid and low-cost reference for other related studies.

Abnormal Liquid Chasing Effect in Paper Capillary Enables Versatile Gradient Generation on Microfluidic Paper Analytical Devices

Yu Liu,Jin-Wen Shangguan,Bi-Yi Xu,Xiao-Dong Yu,Jing-Juan Xu,Hong-Yuan Chen

Microfluidic paper analytical devices (μPADs) are smart and accessible substituents to traditional counterparts in point-of-care tests (POCT), which exploited delicate control over passive fluid in microscale for rich functions. In this work, we are extending such control by introducing novel ways to generate 1D and 2D gradients on μPADs. It is achieved by using paper-capillary-based serial sampling. The paper capillary is composed of a concaved paper channel sealed with tape, with test pads properly distributed aside. In such a structure, the liquid can not only quickly and automatically flow along the capillary but also be continuously consumed by the peripheral test pads. Thus, when we do serial sampling, an abnormal liquid chasing effect can be observed, where the later liquid sample chases and surpasses the earlier parts in the paper capillary, leading to reversely ordered sample distribution compared with that in a typical glass capillary. This specialty allows for fast, ordered, and tunable sequential sampling and enables efficient generation of 1D and 2D concentration gradient with one, two, and even three components on μPADs. Besides, we verified the applicability of this technique for arrayed assays, including 1D serial dilution-based metal ion colorimetry and a 2D bacterial antibiotic susceptibility test for synergic effect evaluations, which paves the way for high-throughput sample analysis and information-rich condition screening on μPADs.

A Three-Dimensional Paper-Based Isoelectric Focusing Device for Direct Analysis of Proteins in Physiological Samples

Jicheng Niu,Zhihui Bao,Zining Wei,Jasmine Xinze Li,Bin Gao,Xiaofan Jiang,Fei Li

On-site protein analysis is crucial for disease diagnosis in community and family medicine in which microfluidic paper-based analytical devices (μPADs) have attracted growing attention. However, the practical applications of μPADs in protein analysis for physiological samples with high complexity is still limited. Herein, we developed a three-dimensional (3D) paper-based isoelectric focusing (IEF) platform, which is composed of power supply, reservoirs, and separation channel and made by the origami and stacking method, to simultaneously separate and enrich proteins in both low-salt and high-salt samples. Under the optimized experimental conditions, standard proteins (bovine hemoglobin (BHb) and phycocyanin (Phy)) were separated within 18 min under a 36 V power supply and obtained a 10-fold enrichment using the 3D paper-based IEF platform. Then, the capability of the 3D paper-based IEF platform for direct pretreatment of high-salt samples using a 12 V battery as power supply was measured through separating three standard proteins in saline (0.9% NaCl) with separation resolution (SR) > 1.29. Through further coupling with colorimetric and lateral flow strip measurements, the 3D paper-based IEF platform was applied to directly pretreat and quantitatively analyze microalbuminuria and C-reactive proteins in clinical urine and serum samples with analytical results with relative deviations of <8.4% and < 13.1%, respectively, to the clinical test results. This work proposes a new strategy to minimize the difficulty of directly processing high-salt samples with the traditional IEF system and provides a versatile, miniaturized, and low voltage demand analytical platform for on-site analysis of proteins in physiological samples.
