Osteosarcoma (OS) is a highly fatal bone tumor characterized by high degree of malignancy and early lung metastasis. Traditional chemotherapy fails in improving the efficacy and survival rate of patients with OS. Butyrate (NaBu) has been reported as a new antitumor drug for inhibiting proliferation and inducing apoptosis in various cancer cells. However, the effect of NaBu on the ferroptosis of osteosarcoma is still unknown. This study aimed to investigate whether NaBu promotes erastin-induced ferroptosis in OS cells and to uncover the underlying mechanism. Here, we found that NaBu significantly enhanced erastin-induced ferroptosis in vitro . Compared with the group that erastin used alonely, pre-treating with NaBu exacerbated erastin-meditated GSH depletion, lipid peroxidation, and mitochondrial morphologic changes in OS cells. In a subcutaneous OS model, NaBu combined with erastin significantly reduced tumor growth and increased the levels of 4-HNE. Mechanistically, NaBu downregulated SLC7A11 transcription via regulating ATF3 expression. Overexpression of ATF3 facilitated erastin to induce ferroptosis, while ATF3 knockdown attenuated NaBu-induced ferroptosis sensitivity. In conclusion, our findings revealed a previously unidentified role of NaBu in erastin-induced ferroptosis by regulating SLC7A11, suggesting that NaBu may be a potential therapeutic agent for OS treatment.

Background Chemotherapy-induced oral mucositis (OM) is a common complication of cancer treatment that significantly impacts patients' quality of life. Butyrate, a short-chain fatty acid, has been shown to inhibit inflammation and mitigate intestinal mucosal damage. How, its effect in treating OM remains unclear. Material and Methods OM model in mice pretreated with 5-FU solution was established by injecting 20% acetic acid into oral mucosa. Sodium butyrate was given for treatment. H&E staining was used to observe histopathological changes, and qRT-PCR to assess inflammatory factor level changes in ulcer tissues after treatment. Also, qRT-PCR and immunofluorescence staining were used to evaluate the expression and distribution of tight junction protein in ulcer tissues. Results Sodium butyrate treatment improved the weight loss in mice caused by OM and promoted the repair of oral mucosa in a time-dependent manner. In addition, sodium butyrate significantly inhibited the mRNA levels of inflammatory factors such as tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), interleukin-6 (IL-6), and interleukin-18 (IL-18) in the ulcer tissue. Moreover, sodium butyrate promoted the mRNA and protein expressions of tight junction protein-1 (ZO-1) and Claudin-1 in the epithelial cells of the ulcer tissue. Conclusions Butyrate promotes OM healing by reducing inflammation and increasing the expression of tight junction proteins in ulcer tissues. Key words: Butyrate, chemotherapy, oral mucositis, inflammation, tight junction proteins.

Simple SummaryThis study examines the impact of sodium acetate and sodium butyrate supplementation on the compositional profile, blood biochemical parameters, and volatile flavor compounds of lactating mares’ milk. By employing comprehensive analytical techniques, including Gas Chromatography–Ion Mobility Spectrometry (GC-IMS), this study elucidates the effects of these additives on both the chemical composition and sensory attributes of the milk. The findings offer critical insights into optimizing its flavor profile, enhancing overall quality, market appeal, and nutritional value. This study aims to explore the impact of adding sodium acetate and sodium butyrate on the composition, blood biochemical parameters, and volatile flavor compounds of lactating mares’ milk. By assessing the influence of these additives on milk flavor enhancement, the findings provide scientific evidence for optimizing flavor characteristics and offer new strategies for improving the sensory attributes of mare milk products. Eighteen lactating Yili mares were randomly assigned to three groups: a control group, a sodium acetate group (85 mg/kg·BM−1), and a sodium butyrate group (85 mg/kg·BM−1). The experiment lasted 90 days, with milk yield recorded on days 0, 30, 60, and 90. Milk samples were collected on day 60 (peak lactation) for compositional analysis, and GC-IMS was employed to identify and quantify volatile compounds. Additionally, blood samples were drawn from the jugular vein before morning feeding on day 60 using heparinized tubes to assess key biochemical markers, including glucose, triglycerides, total cholesterol, and urea. The results revealed the following findings: (1) Milk yield and composition: The addition of sodium acetate and sodium butyrate had no significant effect on milk yield. However, both treatment groups exhibited significantly or extremely significantly higher milk fat content compared to the control group, whereas milk protein and lactose levels remained largely unchanged. (2) Blood biochemical indicators: The sodium butyrate group showed an extremely significant increase in urea levels compared to the sodium acetate and control groups. Glucose levels in the sodium acetate group were also significantly higher than in the control group. Moreover, triglyceride levels were markedly elevated in the sodium butyrate group compared to the sodium acetate group, while total bilirubin concentrations were significantly higher in the sodium acetate group than in the control group. (3) Volatile compounds: The addition of these additives led to a significant increase in the diversity and concentration of volatile compounds in mare milk. Notably, esters, aldehydes, and ketones showed substantial enrichment in both treatment groups. The relative abundance of esters such as butyl acetate, L-lactic acid ethyl ester, 1-pentene-3-ol, pentanol, and 3-pentanone increased, alongside a significant rise in aldehydes and ketones, including 2-heptenal and 3-pentanone. In conclusion, sodium acetate and sodium butyrate enhance milk flavor by modulating milk composition and metabolic parameters, providing a scientific foundation for improving the quality of mare milk products.

Salmonellais among the most frequently isolated foodborne pathogens, and biofilm formed bySalmonellaposes a potential threat to food safety. Short-chain fatty acids (SCFAs), especially propionate and butyrate, have been demonstrated to exhibit a beneficial effect on promoting intestinal health and regulating the host immune system, but their anti-biofilm property has not been well studied. This study aims to investigate the effects of propionate or butyrate on the biofilm formation and certain virulence traits ofSalmonella. We investigated the effect of propionate or butyrate on the biofilm formation ofSalmonella entericaserovar Typhimurium (S.Typhimurium) SL1344 grown in LB broth or food models (milk or chicken juice) by crystal violet staining methods. Biofilm formation was significantly reduced in LB broth and food models and the reduction was visualized using a scanning electron microscope (SEM). Biofilm metabolic activity was attenuated in the presence of propionate or butyrate. Meanwhile, both SCFAs decreased AI-2 quorum sensing based on reporter strain assay. Butyrate, not propionate, could effectively reduce bacterial motility. Bacterial adhesion to and invasion of Caco-2 cells were also significantly inhibited in the presence of both SCFAs. Finally, two SCFAs downregulated virulence genes related to biofilm formation and invasion through real-time polymerase chain reaction (RT-PCR). These findings demonstrate the potential application of SCFAs in the mitigation ofSalmonellabiofilm in food systems, but future research mimicking food environments encountered during the food chain is necessitated. Keywords:propionate;butyrate;biofilm;antimicrobial activity;SalmonellaTyphimurium

The present study evaluated the effect of dietary sodium butyrate (NaBT) supplementation on the growth performance, intestinal microbiota composition, and liver health of largemouth bass Micropterus salmoides fed high-fat diets. Four isonitrogenous and isolipid diets were formulated: a high-fat diet and the high-fat diet supplemented by 0.05% NaBT, 0.1% NaBT, or 0.2% NaBT. Each diet was randomly distributed to triplicate tanks (200 L ) containing 30 fish (initial weight 4.0 ± 0.1 g) per tank. The experimental fish were fed to apparent satiation twice daily for 60 days. The orthogonal polynomial contrasts showed that the weight gain rate (WGR) and specific growth rate (SGR) increased linearly ( P < 0.05) with increasing dietary NaBT levels (P < 0.05). No significant differences were found for feed intake (FI), feed conversion ratio (FCR), condition factor (CF), viscerosomatic index (VSI), and hepatosomatic index (HSI). There were no differences in the alpha diversity indices (including Operational Taxonomic Units, Chao index, and Simpson index) of intestinal microbiota between the various dietary groups ( P > 0.05). For the intestinal microbiota composition, the proportion of Proteobacteria were linearly and cubically increased, whereas the proportion of Tenericutes and Mesomycoplasma were linearly and cubically decreased with the increase of dietary NaBT levels ( P < 0.05). The increasing dietary NaBT levels led to significantly quadratic decrease of alanine aminotransferase (ALT) ( P < 0.05), linear and quadratic decrease of aspartate aminotransferase (AST) ( P < 0.05), and linear, quadratic, and cubic decrease of diamine oxidase (DAO) ( P < 0.05). Hepatic triglyceride (TG) and total cholesterol (TCHO) contents decreased linearly and quadratically with the increase of dietary NaBT levels ( P < 0.05). Increasing dietary NaBT levels linearly, quadratically, and cubically decreased the malondialdehyde (MDA) contents ( P < 0.05). Total superoxide dismutase (T-SOD) and glutathione peroxidase (GPx) activity levels increased linearly with the increasing dietary NaBT levels ( P < 0.05). As the dietary NaBT levels gradually increased, the relative expression of carnitine palmitoyl transferase (CPT1) and peroxisome proliferator-activated receptor α (PPARα) increased linearly, quadratically, and cubically ( P < 0.05), while the relative expression of sterol regulatory element binding protein 1 (SREBP 1) decreased linearly, quadratically, and cubically ( P < 0.05). The relative expression of tumor necrosis factor α (TNF-α) and cysteinyl aspartate specific protease 3 (Caspase 3) decreased linearly and quadratically with the increasing NaBT levels ( P < 0.05). Pearson correlation analysis showed that fish growth was closely correlated with microbiota composition, hepatic lipid accumulation, hepatic antioxidant-related parameters, and hepatic gene expression ( P < 0.05). These results indicated that NaBT could be used to alleviate the negative influence of high-fat diets on fish growth, intestinal microbiota homeostasis , and liver health.

Plectasin, a novel antimicrobial peptide, has the potential to disrupt bacterial cell walls and alter the rumen fermentation mode, making it a superior alternative to antibiotics. However, there is limited research on the effects of plectasin on rumen microbiota. This study aimed to evaluate the effects of plectasin (0.057 μmol/L) on in vitro rumen fermentation characteristics and select groups of rumen bacterial communities in comparison with monensin (5 μmol/L), one of the most commonly used ionophores in ruminants, and as a control treatment with the basal substrate. Unlike monensin, plectasin was found to increase the molar proportions of butyrate and acetate/propionate ratio (p< 0.001) while decreasing pH and the molar proportions of propionate (p< 0.05). Principal component analysis of bacterial 16S rRNA gene amplicons clearly showed a separation between the bacteria shaped by plectasin and monensin. Comparative analysis also revealed differences in the relative abundance of certain bacteria in different taxa between plectasin and monensin. The divergent effects of plectasin and monensin on bacterial communities are likely responsible for the differences in their ability to alter rumen fermentation. Plectasin may have advantages over monensin in modulating ruminal bacterial communities and increasing the butyrate and the acetate/propionate ratio. Therefore, it may be considered as a potential additive for ruminant feed.

An 8-week experiment was conducted to investigate the effects of sodium butyrate (NaBT) inclusion in high-fat (HF) diets on growth performance, liver health, and disease resistance in Grass Carp Ctenopharyngodon idella . Three diets (Control diet containing crude lipid at 58 g/kg, HF diet with 108-g/kg crude lipid, and NaBT diet with 108-g/kg crude lipid and 1-g/kg NaBT) were randomly assigned to nine tanks with 30 fish (9.50 ± 0.06 g) in each tank. After the feeding trial, disease resistance was assessed by injecting the fish with Aeromonas hydrophila . Compared to the Control diet group, the HF diet group showed lower specific growth rate, feed efficiency, and survival rate (15.7%) after the A. hydrophila challenge; significantly higher activity levels of alanine aminotransferase and aspartate aminotransferase in plasma; higher malondialdehyde content; higher messenger RNA (mRNA) expression of interleukin-8, cysteinyl aspartate specific protease (caspase) 9, and caspase 3; lower activity level of glutathione peroxidase; and lower mRNA expression of nuclear factor erythroid 2-related factor 2 in liver. However, the NaBT diet significantly increased fish growth performance and survival rate (39.7%) after the A. hydrophila challenge and reduced hepatic oxidative stress, inflammation, and apoptosis compared to the HF diet. In conclusion, NaBT can ameliorate the detrimental effects of HF diets on fish growth performance and fish health.

Drug combination is considered to be an effective approach to improve the efficacy of cancer therapy and chemoprevention. Selenite, a representative of inorganic form of selenium, and butyrate, a major short-chain fatty acid, are two well-documented colon cancer dietary chemopreventive agents with distinct molecular mechanisms. We hypothesized that combination of selenite and butyrate might produce improved outcome against colon cancer. This hypothesis was tested using both HCT116 human colon cancer cells and its xenograft mouse model in the present study. The in vitro study showed a synergistically inhibitory effect on HCT116 colon cancer cells but not on NCM460 normal human colon mucosal epithelial cells. Consistent with the in vitro study, results of the xenograft mouse model further demonstrated that combination of selenite and butyrate led to improved efficacy in comparison with each agent alone. Mechanistically, the induction of alanine-serine-cysteine transporter 2 (ASCT2) by selenite repressed its inhibitory effect on colon cancer cells, which was reversed by its co-treatment with butyrate. The findings of the present study denote the likely potential for developing selenite/butyrate combination remedy to combat against colon cancer.

Ligand-functionalization can increase the affinity of nanoparticles (NPs) with targeted cells. However, one major defect of ligands still exists in oral administration: limited receptor recognition. The hindrance of mucus network and deactivation of enzymes severely challenge the targeting efficiency of macromolecular ligands. Inspired by “molecular exchange” between intestinal microbiota and host cells, we anchored microbiota metabolite butyrate on classical “mucus-inert” polyethylene glycol (PEG) NPs. Butyrate has unique advantages of low molecule weight, high hydrophilicity and chemical stability. Interestingly, in vitro mucus-permeability and in vivo mucus distribution of PEG NPs were not impaired by butyrate-functionalization. Enhanced cellular uptake was achieved via specific interaction between butyrate and the monocarboxylate transporter (MCT) on cell membranes , which subsequently ameliorated transepithelial transport and intestinal absorption in the ileum. In vitro safety assessment validated the non-toxicity of butyrate-modification. Finally, insulin-loaded Bu-PEG NPs generated a stronger hypoglycemic response on diabetic rats and 2.87-fold higher oral bioavailability compared with bare PEG NPs. This study demonstrated that butyrate-functionalization could improve the intestinal absorption of macromolecules by overcoming multiple obstacles in the gastrointestinal tract, providing a promising active targeting strategy for oral administration.

Anaerostipes hadrus (A. hadrus) is a dominant species in the human gut microbiota and considered a beneficial bacterium for producing probiotic butyrate. However, recent studies have suggested that A. hadrus may negatively affect the host through synthesizing fatty acid and metabolizing the anticancer drug 5-fluorouracil, indicating that the impact of A. hadrus is complex and unclear. Therefore, comprehensive genomic studies on A. hadrus need to be performed. We integrated 527 high-quality public A. hadrus genomes and five distinct metagenomic cohorts. We analyzed these data using the approaches of comparative genomics, metagenomics, and protein structure prediction. We also performed validations with culture-based in vitro assays. We constructed the first large-scale pan-genome of A. hadrus (n = 527) and identified 5-fluorouracil metabolism genes as ubiquitous in A. hadrus genomes as butyrate-producing genes. Metagenomic analysis revealed the wide and stable distribution of A. hadrus in healthy individuals, patients with inflammatory bowel disease, and patients with colorectal cancer, with healthy individuals carrying more A. hadrus. The predicted high-quality protein structure indicated that A. hadrus might metabolize 5-fluorouracil by producing bacterial dihydropyrimidine dehydrogenase (encoded by the preTA operon). Through in vitro assays, we validated the short-chain fatty acid production and 5-fluorouracil metabolism abilities of A. hadrus. We observed for the first time that A. hadrus can convert 5-fluorouracil to α-fluoro-β-ureidopropionic acid, which may result from the combined action of the preTA operon and adjacent hydA (encoding bacterial dihydropyrimidinase). Our results offer novel understandings of A. hadrus, exceptionally functional features, and potential applications.

As the adsorption receptor of bacteriophage tail protein, bacterial lipopolysaccharide (LPS) is a main culprit responsible for nonalcoholic fatty liver disease (NAFLD) caused by high-fat diets. However, few studies have focused on how the interaction between intestinal bacteriophages and bacterial LPS affects the development and progression of NAFLD. Herein, we determined that excessive fat intake significantly increases the levels of endogenous LPS, while the administration of beneficial metabolites of the intestinal microbiota (specifically butyrate) alleviated hepatic injury in rats. The beneficial mechanism of butyrate was attributed to the reprogramming of the structure of the intestinal DNA virome (primarily, phageome). Butyrate possesses the potential to augment bacteriophagic microbial diversity, thereby potentially facilitating interactions between intestinal bacteriophages and bacterial LPS (in the case of homologous phage), further improving mitochondrial dysfunction and reactive oxygen species production, which, in turn, lowered HepG2 cell damage. Likewise, fecal phage transplantation further confirmed that intestinal phages from rats that received butyrate could effectively interact with bacterial LPS to reduce liver damage in rats. Taken together, modifying the intestinal phageome is a promising treatment option for high-fat diet-related NAFLD.
