Flexible tactile sensors (FTS) can be integrated into various parts of the human body to achieve physiological signal detection and sensitive tactile perception. The microstructure design effectively improves the change rate of the contact area at the contact interface and achieves high sensitivity and a wide detection range of FTS. However, the attenuation of sensitivity with increasing pressure is yet to be fully resolved. Herein, inspired by the oriented arrangement of lamprey teeth, an FTS (noted as FTSC) with high-pressure sensitivity (60.38 kPa–1), linear response (R2 = 0.998) over a broad pressure range (10 Pa–200 kPa), and high stability over 5000 cycles is designed. The strategy of cooperative stress transfer of concentric rows of an inwardly curved barb array (CBA) mitigates the saturation rate of the contact area in the high-pressure region, resulting in high sensitivity over a wide linear range. Furthermore, the great application potential of the FTSC in healthcare monitoring and information interaction is demonstrated. This work not only advances the sensing performance of tactile sensors but also provides a promising strategy for developing next-generation soft electronics.

ABSTRACT Root caries is the main cause of oral pain and tooth loss in the elderly. Protecting root lesions from environmental disturbances, resisting pathogens, and facilitating remineralization over time are essential for addressing root caries, but are challenging due to the irregular root surface and the complex oral environment. Hagfish secretes slime when facing danger, which converts into gels upon contact with seawater, suffocating the predators. Inspired by hagfish's defense mechanism, a fluid-hydrogel conversion strategy is proposed to establish a mechanical self-regulating multifunctional platform for root caries treatment. The fluid system (silk fibroin-tannic acid-black phosphorene-urea, ST-BP-U), in which urea disrupts the hydrogen bonds between silk fibroin and tannic acid, can easily spread on the irregular root surface and permeate into dentinal tubules. Upon contact with the surrounding water, urea diffuses, prompting the hydrogel re-formation and creating intimate attachments with micromechanical inlay locks. Meanwhile, BP increases the crosslinking of the re-formed hydrogel network, resulting in reinforced cohesion for robust wet adhesion to the tooth root. This process establishes a structured platform for effective antimicrobial phototherapy and dentin remineralization promotion. This water-responsive fluid-hydrogel conversion system adapts to the irregular root surface in the dynamic wet environment, holding promise for addressing root caries. Statement of Significance Root caries bring a heavy burden to the aging society, but the irregular root surface and dynamic moist oral environment always hinder non-surgical therapeutic effects. Here, we propose a water-responsive fluid-hydrogel conversion strategy aimed at mechanical self-regulation on the irregular and wet root interface to construct a functional structural platform. The liquid system (ST-BP-U) that prebreak intermolecular hydrogen bonds can easily spread on irregular surfaces and dentin tubules. When encountering water, hydrogen bonds re-form, and BP increases the crosslinking of the hydrogel formed in situ. Based on this firm wet-adhesion platform, it provides powerful phototherapy effects and promotes dentin remineralization. This fluid-hydrogel conversion system turns the disadvantages of wet environment into advantages, offering a promising strategy for root caries.

As one of the representatives of jawless vertebrates, lamprey is an important animal model for research on the evolution of adaptive immune system. Although it is widely accepted that the key characteristic of the immune response in jawed vertebrates is the functional cooperation between different immune effector cell lineages, whether immune cells of lamprey can communicate with each other is still unclear. Recently, mounting evidence has revealed the emerging role of exosomes in mediating intercellular communication. In this study, by means of ultrafiltration followed by size exclusion chromatography, exosomes are purified from conditioned growth medium of the primary supraneural myeloid body cells (SMB cells) in LPS-immunized and control Lampetra japonica (Japanese lamprey). The proteomic profiling and qualitative comparison are performed between protein components enriched in exosomes released by SMB cells under quiescent and activated conditions. Our results demonstrate that SMB cells can release exosomes with well-defined characteristics comparable to their mammalian counterparts on their size, morphology and protein markers, which supports exosomes are evolutionarily conserved between lamprey and other higher vertebrate species. In terms of comparison with exosomes released by quiescent SMB cells, activated SMB cell-derived exosomes contain more and significantly different protein components. The unique exosomal protein ‘fingerprint’ might reflect that exosomes from the SMB cells with different states of activation have distinguishing functional roles as well as targeting properties. Further bioinformatic analysis suggests that compared to quiescent exosomes, activated SMB cell-derived exosomes mainly participate in distinctive biological processes including activation of cellular component biogenesis and regulation of cell proliferation. Moreover, activated exosomes may function through the specific signaling pathways associated with the regulation of cell mitosis and immune response. Our results give valuable clues about the immunomodulatory functions of SMB cell-derived exosomes and provide the foundation for further investigation in the regulation mechanism of the adaptive immune response in lamprey.

Ferritin, one of the key regulators of iron homeostasis, is widely present throughout almost all species. The vertebrate ferritin family, which originates from a single gene in the ancestral invertebrates, contains the widest variety of ferritin subtypes among all animal species. However, the evolutionary history of the vertebrate ferritin family remains to be further clarified. In this study, genome-wide identification of the ferritin homologs is performed in lampreys, which are the extant representatives of jawless vertebrates that diverged from the future jawed vertebrates more than 500 million years ago. Molecular evolutionary analyses show that four members of the lamprey ferritin family, L-FT1–4, are derived from a common ancestor with jawed vertebrate ferritins prior to the divergence of the jawed vertebrate ferritin subtypes. The lamprey ferritin family shares evolutionarily conserved characteristics of the ferritin H subunit with higher vertebrates, but certain members such as L-FT1 additionally accumulate some features of the M or L subunits. Expression profiling reveals that lamprey ferritins are highly expressed in the liver. The transcription of L-FT1 is significantly induced in the liver and heart during lipopolysaccharide stimulation, indicating that L-FTs may play a role in the innate immune response to bacterial infection in lampreys. Furthermore, the transcriptional expression of L-FT1 in quiescent and LPS-activated leukocytes is up- and down-regulated by the lamprey TGF-β2, an essential regulator of the inflammatory response, respectively. Our results provide new insights into the origin and evolution of the vertebrate ferritin family and reveal that lamprey ferritins may be involved in immune regulation as target genes of the TGF-β signaling pathway.
