The use of degradable chelating agents to facilitate phytoextraction is a promising low-cost method for the remediation of heavy metal-contaminated soils. However, there are few studies on how plants and soils respond to the chelating agents. In this study, the responses of French marigold ( Tagetes patula L.) and soil cadmium (Cd) to the chelator tetrasodium glutamate (GLDA) was investigated in a 180 d field trial. Five GLDA treatments (0, 292.5, 585, 1170, and 2340 kg hm −2 ) were carried out in a Cd-contaminated soil (0.47 mg kg −1 ) under French marigold plantation. The results showed that the application of GLDA promoted the transformation of other forms of Cd in soil to exchangeable state, and the exchangeable Cd and Fe–Mn oxide bound state increased by 42.13% and 32.97% ( p < 0.05), respectively. The cell wall Cd accumulations significantly increased 9.39% ( p < 0.05) and the percentages of soluble fractions increased by 460.33% ( p < 0.05). Furthermore, increases occurred in soil pH, as well as DOC and DTPA-Cd contents with increasing the total amount of GLDA. The composite application of GLDA (2340 kg hm −2 ) with French marigold reduced the total soil Cd content by 7.59% compared with the soil background. Altogether, results of this study suggested that the application of GLDA can effectively activate soil Cd and enhance the capability of French marigold for the remediation of Cd-contaminated soils.

Enzyme-assisted aqueous two-phase extraction (EA-ATPE) using ethanol/ammonium sulfate system was investigated for total polyphenol (TP) and lutein from marigold flowers. The important factors were investigated by single factor experiment and response surface methodology combined with Box–Behnken design to optimize the operating parameters of EA-ATPE. The maximum yields of TP and lutein were 83.56 ± 0.69 mg g−1 and 5.59 ± 0.13 mg g−1, respectively. Compared with aqueous two-phase extraction and Soxhlet extraction (SE), the extraction yield of TP by EA-ATPE is 64.91% higher and the extract of EA-ATPE has better antioxidant activity. The pretreatment effect was also researched by scanning electron microscopy. Thus, EA-ATPE is an efficient method for extracting bioactive components from plants.

In recent years, synthetic antioxidants that are widely used in foods have been shown to cause detrimental health effects, and there has been growing interest in antioxidants realised from natural plant extracts. In this study, we investigate the potential effects of natural antioxidant components extracted from the forage plant marigold on the oxidative stability of soybean oil. First, HPLC-Q-TOF-MS/MS was used with 1,1-diphenyl-2-picrylhydrazyl (DPPH) to screen and identify potential antioxidant components in marigold. Four main antioxidant components were identified, including quercetagetin-7-O-glucoside (1), quercetagetin (2), quercetin (3) and patuletin (4). Among them, quercetagetin (QG) exhibited the highest content and the strongest DPPH radical scavenging activity and effectively inhibited the production of oxidation products in soybean oil during accelerated oxidation, as indicated by reductions in the peroxide value (PV) and acid value (AV). Then, the fatty acids and volatile compounds of soybean oil were determined with gas chromatography–mass spectrometry (GC-MS) and headspace solid-phase microextraction–gas chromatography–mass spectrometry (HS-SPME-GC-MS). A total of 108 volatile components, including 16 alcohols, 23 aldehydes, 25 ketones, 4 acids, 15 esters, 18 hydrocarbons, and 7 other compounds, were identified. QG significantly reduced the content and number of aldehydes and ketones, whereas the formation of acids and hydrocarbons was completely prevented. In addition, the fatty acid analysis demonstrated that QG significantly inhibited oxidation of unsaturated fatty acids. Consequently, QG was identified as a potential, new natural antioxidant that is believed to be safe, effective and economical, and it may have potential for use in plant extracts feed additives. Keywords:Tagetes erectaL.;quercetagetin;DPPH-HPLC-Q-TOF/MS;HS-SPME-GC-MS;oxidative stability

In this study, a novel method of simultaneous extraction and enrichment of total polyphenol (TP) and lutein from marigold flower was developed by microwave and enzyme co-assisted aqueous two-phase extraction (ATPE) (MEAATPE). In this study, ethanol/ammonium sulfate was chosen to construct aqueous two-phase system (ATPS) due to its fine partition and recycling behaviors. The optimal MEAATPE condition was obtained, that is, 28% (w/w), ethanol/20% (w/w) ammonium sulfate, 0.45 U/g enzyme concentration, enzymolysis time of 150 min, enzymolysis temperature of 45 °C, microwave time of 120 s, and microwave power of 270 W, by investigating the effects of ATPS composition and other factors on the yield of target products. Under this optimal condition, the yields of TP and lutein reached 84.61 and 7.32 mg/g with the corresponding recovery rates of 95.35% and 99.85%, respectively. Compared with enzyme-assisted ATPE, microwave-assisted ATPE, ATPE, and Soxhlet extraction (SE), the extraction yield of TP by MEAATPE was the highest, which is 71.14% higher than that of SE. Scanning electron microscopy analysis further showed an optative pretreatment effect of marigold flower powders. Thus, MEAATPE was an efficient, rapid, and environmental-friendly method used for the extraction of natural products.

In present study, a novel, efficient and environmental friendly microemulsion extraction method was established to extract lutein from marigold petals powder (MPP). The optimum microemulsion formulation was obtained by combining the screening of oils, co-surfactants and ternary phase diagram. Based on the saturation solubility of lutein, isoamyl acetate and n-propanol were selected as oil phase and co-surfactant, respectively. A 5 mL microemulsion consisting of Tween80/n-propanol/isoamyl acetate/water (36:36:18:10 wt%) was used for the extraction of lutein from 20 mg MPP, with the highest extraction efficiency of 95.17%. For lutein microemulsion, the average droplet size was 26.44 ± 0.17 nm, the polydispersity index was 0.37 ± 0.00 and Zeta potential was − 11.20 ± 0.26 mV. Compared with ethanol extract (EE), MPP microemulsion extract (LME) had higher stability under different temperature, pH, and storage time conditions. This study would provide a new alternative method for lutein extraction, and also could be expected to be applied for other poorly water-soluble ingredients.

Quercetagetin is one of the main active flavonoids in a well-known herbal medicine of Tagetes erecta Linn ( T. erecta ) flower. Selective enrichment method of quercetagetin from T. erecta flower and blood analysis method of it is lacking and need to be developed. In present study, quercetagetin based molecularly imprinted polymers (MIPs) and restricted access molecularly imprinted polymers (RAMIPs) were synthesized to separately enrich quercetagetin from deep eutectic solvent (DES) extract of T. erecta flower and plasma for the first time. Firstly, MIPs were synthesized by using quercetagetin, 2-vinylpyridine (2-VP), acetonitrile, ethylene glycol dimethylacrylate (EGDMA) and azobisisobutyronitrile (AIBN) as template, functional monomer, porogen, cross-linker and initiator, respectively. Further, RAMIPs were fabricated by introducing BSA on the surface of MIPs. The MIPs and RAMIPs were characterised using scanning electron microscopy, Fourier-transform infrared spectroscopy, and thermogravimetric analyses, and the results demonstrated the successful preparation of them. The selective adsorption properties of two kinds of polymers were further analyzed using static, dynamic and selectivity adsorption experiments. The results indicated that MIPs and RAMIPs presented good adsorption capacities (27.2 mg·g −1 and 23.1 mg·g −1 ) and selectivity (Imprinting factor, IF = 2.57 and 2.09 for MIPs and RAMIPs, respectively) for quercetagetin. Moreover, macromolecular exclusion experiment results revealed that almost all selected protein can be completely exclused by RAMIPs, revealing the outstanding macromolecular exclusion properties of RAMIPs. In addition, in order to environmental protection and more effective extraction of quercetagetin from T. erecta flower, organic solvent was substituted by DES. To obtain the highest extraction efficieny, the extraction parameters, including the types of DESs, solid-to-liquid ratio, extraction temperature and time were systematically optimized. By using DES-9 (L-menthol- pyruvic acid with the molar rito of 1:1) as extraction solvent, quercetagetin could be effectively extracted under the conditions of solid to liquid ratio of 5:1, extraction temperature of 30 °C and extraction time of 50 min. Following, coupled with HPLC-DAD analysis, MIPs were successfully applied as selective adsorbents for quercetagetin extraction from DES extract of T. erecta flower. Besides, in comparison with the traditional precipitation method, the absorption of quercetagetin can be more effectively elucidated by using RAMIPs.
