Main conclusion A novel cytochrome P450 from Tripterygium wilfordii , CYP81AM1, specifically catalyses the C-15 hydroxylation of dehydroabietic acid. This is the first CYP450 to be found in plants with this function. Abstract Cytochrome P450 oxygenases (CYPs) play an important role in the post-modification in biosynthesis of plant bioactive terpenoids. Here, we found that CYP81AM1 can catalyze the formation of 15-hydroxydehydroabietic acid by in vitro enzymatic reactions and in vivo yeast feeding assays. This is the first study to show that CYP81 family enzymes are involved in the hydroxylation of abietane diterpenoids. At the same time, we found that CYP81AM1 could not catalyse abietatriene and dehydroabietinol, suggesting that the occurrence of the reaction may be related to the carboxyl group. Through molecular docking and site mutations, it was found that some amino acid sites (F104, K107) near the carboxyl group had an important effect on enzyme activity, also suggesting that the carboxyl group played an important role in the occurrence of the reaction.

Background
Ischemia–reperfusion (I/R) injury to the kidney occurs commonly in organ transplantation from donation after cardiac death, involving many pathologic processes. In this study, we used rat model to assess whether tripterysium glycosides (TG) preconditioning could exert protective effects in renal I/R injury. Materials and methods All male SD rats were randomly divided into four groups (6 each): sham group, TG group, I/R group and TG + I/R group. Groups TG and TG + I/R were pretreated with TG at 0.1 mg/kg for 14 days; groups sham and I/R were administered with the same dosage of normal saline. Groups TG + I/R and I/R underwent 45 min of renal ischemia of left kidney after right nephrectomy, and then, they were subjected to 72-h reperfusion. Groups sham and TG were only received right nephrectomy. The indicators of apoptosis, fibrosis and inflammation were analyzed to evaluate the effect of tripterysium glycosides preconditioning on renal I/R injury. Results Pretreatment with TG significantly inhibited the levels of serum creatine and blood urea nitrogen and improved histologic lesions induced by I/R injury. Moreover, for the apoptosis signal pathway, pretreatment with TG markedly decreased the expression of caspase-3 and Bax and increased the level of Bcl-2. HMGB1, which was regarded as one of inflammation marker molecule, it was inhibited in the TG + I/R group. For the fibrosis signal pathway, the pretreatment with TG before I/R could down-regulate the expression level of typical molecules of fibrosis (TGF-β1, Smad3, p-Smad3). Conclusions Pretreatment with tripterysium glycosides exhibited protective effect on kidney ischemia/reperfusion injury, which might be related to the alleviation of inflammation, fibrosis and the reduction in apoptosis.

Acute lung injury (ALI) is a critical respiratory syndrome significantly impacting patient health. Tetrastigma hemsleyanum Diels et Gilg (Sanyeqing, SYQ) is a traditional Chinese medicine and its polysaccharides (SYQP) have demonstrated efficacy in counteracting lipopolysaccharide-induced ALI. This study characterized the structure of SYQP and synthesized the SYQP@ZIF-8 composite using biomimetic mineralization, evaluating encapsulation and release efficiency. The biocompatibility of SYQP@ZIF-8 in vitro was assessed by the CCK-8 colorimetric assay and hemolytic activity. Inflammatory cytokine was measured to evaluate the therapeutic effect. The efficacy of SYQP@ZIF-8 in lung injury was assessed using a mice ALI model. Characterization showed SYQP as a homogeneous α-type polysaccharide, comprising galactose, mannose, glucuronide, glucose, galacturonide, and arabinose, with a molecular weight of 516.94 kDa. SYQP@ZIF-8 exhibited high encapsulation rate (> 90 %), rapid pH-responsive release (within 60 min up to ~100 %), low toxicity and favorable hemolytic characteristics. Furthermore, it demonstrated reduced inflammatory cytokine secretion compared to SYQP, along with a superior inhibitory effect. The outcomes of in vivo experiments, including a decrease in the W/D ratio and LDH activity, further confirmed the efficacy of SYQP@ZIF-8 in treating LPS-induced ALI. In conclusion, SYQP@ZIF-8 released SYQP in acidic inflammatory conditions, outperforming SYQP alone in treating ALI.
