Occurrence and modeling of disinfection byproducts in distributed water of a megacity in China: Implications for human health

Zhen Pang,Peifeng Zhang,Xinyi Chen,Feilong Dong,Jing Deng,Cong Li,Junping Liu,Xiaoyan Ma,Andrea M. Dietrich

Disinfection byproducts (DBPs) are initially formed in the process of chlorination in the drinking water treatment plants (DWTPs), then further formed in the distribution system due to the presence of residual chlorine and reactive organic matters. However, in China, DBPs are monitored in the effluent from the DWTPs, but less is known about concentrations of DBPs in tap water since they are usually monitored once per half a year. The smart water service system is establishing real-time monitoring of water indices, although DBPs are an urgent need, they are difficult to monitor in real-time due to their diversity and complicated detection methods. If the correlation between DBP concentration and routinely real-time monitored water quality parameters (e.g., pH value, residual chlorine, ammonia) can be evaluated, the concentration of DBPs can be predicted, which will strengthen the control of tap water safety. This article comprehensively assessed the physicochemical parameters and the occurrence of DBP formation in the tap water with an 18-month investigation in Z city (China). DBP formation in tap water of different seasons and different water sources were compared. Based on the relationship between DBPs and physicochemical parameters, linear prediction and nonlinear prediction models of trihalomethanes (THMs), haloacetonitriles (HANs) and haloacetic acids (HAAs) were established, and the accuracy of these models was verified by measured data. Finally, the toxicity and carcinogenic and non-carcinogenic health risk assessment of DBPs in tap water were analyzed.

Rapid and Sensitive Detection of Water Toxicity Based on Photosynthetic Inhibition Effect

Min Chen,Gaofang Yin,Nanjing Zhao,Tingting Gan,Chun Feng,Mengyuan Gu,Peilong Qi,Zhichao Ding

To achieve rapid and sensitive detection of the toxicity of pollutants in the aquatic environment, a photosynthetic inhibition method with microalgae as the test organism and photosynthetic fluorescence parameters as the test endpoint was proposed. In this study, eight environmental pollutants were selected to act on the tested organism,Chlorella pyrenoidosa, including herbicides (diuron, atrazine), fungicides (fuberidazole), organic chemical raw materials (phenanthrene, phenol, p-benzoquinone), disinfectants (trichloroacetonitrile uric acid), and disinfection by-products (trichloroacetonitrile). The results showed that, in addition to specific PSII inhibitors (diuretic and atrazine), other types of pollutants could also quickly affect the photosynthetic system. The photosynthetic fluorescence parameters (Fv/Fm, Yield, α, and rP) could be used to detect the effects of pollutants on the photosynthetic system. Although the decay rate of the photosynthetic fluorescence parameters corresponding to the different pollutants was different, 1 h could be used as an appropriate toxicity exposure time. Moreover, the lowest respondent concentrations of photosynthetic fluorescence parameters to diuron, atrazine, fuberidazole, phenanthrene, P-benzoquinone, phenol, trichloroacetonitrile uric acid, and trichloroacetonitrile were 2 μg·L−1, 5 μg·L−1, 0.05 mg·L−1, 2 μg·L−1, 1.0 mg·L−1, 0.4 g·L−1, 0.1 mg·L−1, and 2.0 mg·L−1, respectively. Finally, diuron, atrazine, fuberidazole, and phenanthrene were selected for a comparison of their photosynthetic inhibition and growth inhibition. The results suggested that photosynthetic inhibition could overcome the time dependence of growth inhibition and shorten the toxic exposure time from more than 24 h to less than 1 h, or even a few minutes, while, the sensitivity of the toxicity test was not weakened. This study indicates that the photosynthetic inhibition method could be used for rapid detection of the toxicity of water pollutants and that algae fluorescence provides convenient access to toxicity data. Keywords:photosynthetic inhibition;photosynthetic fluorescence parameter;Chlorella pyrenoidosa;toxic water pollutants;rapid detection;biotesting

Triboelectric-photocatalytic coupling enhanced photogenerated electrons and holes utilization for efficient algal inactivation

Feilong Dong,Peifeng Zhang,Jiayi Cheng,Jianing Chen,Tao Liu,Xiaoyan Ma,Shuang Song,Shuangxi Nie

Algal blooms have become a challenging problem in conventional water treatment processes due to numerous negative effects on human health and ecosystems. Photocatalysis has turned out to be a promising solution with the environmental and economic benefits beheld, but the rapid recombination of photogenerated electron and hole inhibited the application in practice. This study proposed a rotating triboelectric nanogenerator (R-TENG) as a self-powered bias for photocatalysis to enhance the algal removal efficiencies. R-TENG harvest waste water wave energy from coagulation tank to convert it into stable DC voltage (31 V) and DC current (27 μA). TiO 2 nanotubes (TNT) serving as the anode in the photocatalysis with TENG system achieved complete inactivation of algae (>97 %) after 7 h of reaction, which was increased by 13.9 % compared with photocatalysis. This enhanced performance was attributed to the improvement of active species generation (e.g., •O 2 - , •OH and h + ). During algal inactivation, the active species destroyed photosynthetic activity (decline of Chlorophyll a), antioxidant system (decrease of superoxide dismutase, catalase and malondialdehyde) and membrane in algal cells. Given the ubiquitous existence of algal cells in surface water, this work offers a novel method to develop high-performance electro-assisted photocatalysis for algal removal.

Rapid and visual detection of dichloroacetonitrile in water

Jiaben Zhong,Hangyan Zhang,Yina Cai,Xiuping Chen,Zhiyuan Fang,Dun Deng

Dichloroacetonitrile (DCAN) is a common biotoxic disinfection by-product (DBP) of chlorine. The current methods used for detecting DCAN are tedious and heavily instrument-dependent, and are not suitable for on-site detection. In the present study, we developed a colorimetric assay for rapid detection of DCAN. DCAN in water acted as a complexing agent that formed a complex with cuprous species. The cuprous species was then extracted by chloroform and visualized using dithizone. The visual detection limit for DCAN was 20 ng mL−1, while fluorescence quantification could detect DCAN at a concentration as low as 8.75 ng mL−1. Moreover, haloacetonitriles (HANs) derived from chlorine disinfection and structurally similar to DCAN, including TCAN, BCAN, and DBAN, could also be detected using this method. Other DBPs at concentrations as high as 200 ng mL−1 did not affect the detection process. The low cost and instrument-independence characteristic of the present method enables its routine determination of the concentration of DCAN in water.

A new sensitive response index for detecting water toxicity based on microalgal fluorescence kinetics

Gan Tingting,Yin Gaofang,Zhao Nanjing,Tan Xiaoxuan,Wang Ying,Sheng Ruoyu,Ye Ziqi

Microalgal fluorescence kinetics method, as a promising technique for rapidly detecting water toxicity, still lacks a consistent response index with sensitive response characteristics to different types of toxic pollutants. In order to solve this problem, in this study, the influences of six typical toxic pollutants belonging to different types, including the heavy metal cadmium, the herbicide atrazine, organic chemical raw materials 1,4-benzoquinone and 1,4-dichlorobenzene, and the disinfection by-products chloroacetic acid and trichloroacetonitrile on the fluorescence rise kinetics (OJIP) curve of microalga Chlorella pyrenoidosa were investigated. According to the same characteristics of changes in the OJIP curves of different toxic pollutant treatments, a new response index for detecting the comprehensive toxicity of water (RI ct ) was constructed. By comparing with the maximum photochemical quantum yield of photosystem II (PSII) (F v /F m ), the potential quantum yield of PSII (F v /F o ), the performance index of PSII (PI ABS ) and the quantum yield of electron transport of PSII (φ Eo ), the feasibility and sensitivity of quantitative response of RI ct to the toxicity of each pollutant and polluted water containing multiple pollutants were verified. The results indicated that all the six toxic pollutants had the same influence on parameters F v(m-j) /F o and F v(m–o) /F o contained in the OJIP curve, and RI ct constructed based on F v(m-j) /F o and F v(m–o) /F o had a good logistic curve concentration–response relationship with each pollutant. By comparison, the response sensitivity of RI ct to all pollutants was significantly superior to F v /F m , F v /F o and φ Eo , and also had better stability in the toxicity response to each pollutant compared to PI ABS , indicating that RI ct was a suitable response index for rapidly and sensitively detecting water comprehensive toxicity based on microalgal fluorescence kinetics. This study provides an important basis for the development of rapid and sensitive detection methods for water toxicity.

Synthesis and Cytotoxic Property of Annonaceous Acetogenin Glycoconjugates

Jing-Fang Shi,Ping Wu,Xiao-Li Cheng,Xiao-Yi Wei,Zi-Hua Jiang

Background Annonaceous acetogenins (ACGs) are secondary metabolites produced by the Annonaceae family and display potent anticancer activity against various cancer cell lines. Squamocin and bullatacin are two examples of ACGs that show promising antitumor activity; however, preclinical data are not sufficient partly due to their being highly lipophilic and poorly soluble in water. These compounds also display high toxicity to normal cells. Due to these disadvantageous properties, the therapeutic potential of squamocin and bullatacin as antitumor agents has not been fully evaluated. Methods In order to enhance their water solubility and potentially improve their cancer targeting, squamocin and bullatacin were conjugated to a glucose or galactose to yield glycosylated derivatives by direct glycosylation or the Cu(I)-catalyzed azide-alkyne 1,3-dipolar cycloaddition (CuAAC) reaction (the click reaction). The synthesized compounds were evaluated for their anticancer property against HeLa, A549 and HepG2 cancer cell lines using MTT assay. Results Nine glycosyl derivatives were synthesized and structurally characterized. Most of them show comparable in vitro cytotoxicity against HeLa, A549 and HepG2 cancer cell lines as their parent compounds squamocin and bullatacin. It appears that the type of sugar residue (glucose or galactose), the position at which the sugar residue is attached, and whether or not a linking spacer is present do not affect the potency of these derivatives much. The solubility of galactosylated squamocin 13 in phosphate buffer saline (PBS, pH = 7) is greatly improved (1.37 mg/mL) in comparison to squamocin (not detected in PBS).Conclusion The conjugation of a glucose or galactose to squamocin and bullatacin yields glycosyl derivatives with similar level of anticancer activity in tested cell lines. Further studies are needed to demonstrate whether or not these compounds show reduced toxicity to normal cells and their therapeutic potential as antitumor agents.

Cholesterol moieties as building blocks for assembling nanoparticles to achieve effective oral delivery of insulin

Yu Ding,Qiaochu Wang,Guangqu Liu,Yaqian Feng,Wei Zhou

Although nanoparticles (NPs) have emerged as a promising strategy to improve oral absorption of insulin, most of them still have minimal oral bioavailability due to the harsh gastrointestinal (GI) environment and epithelial barriers. A novel platform of oral insulin delivery is herein proposed based on amphiphilic cholesterol-phosphate conjugate (CHP). The CHP was synthesized with a cholesterol moiety as a hydrophobic block and a phosphate group as a hydrophilic block. The structural characteristics of CHP made cholesteryl moieties to point outward to form a shell coated on the surface of NPs, and phosphate groups toward inside as the core via the water-in-oil (W/O) emulsion method. The additional Ca2+ could chelate to phosphate groups in the core to bind adjacent phosphate groups and further increased the stability of NPs (CP-Ca NPs). Phospholipid and glycocholate were modified on the surface of CP-Ca NPs to form hydrophilic corona coated CP-Ca NPs (Hc@CP-Ca NPs) and didn't change the structure of CP-Ca NPs. The NPs exhibited high encapsulation efficiency (>95%) and had high stability in the GI tract by reducing insulin release and protecting insulin from enzymatic degradation. Compared to free insulin, Hc@CP-Ca NPs demonstrated 31-fold higher cellular uptake in Caco-2 cells for 1 h incubation. Furthermore, the transepithelial transport efficiency of Hc@CP-Ca NPs was 12-fold higher than that of free insulin. Moreover, CP-Ca NPs and Hc@CP-Ca NPs generated a rapid-onset and long-lasting hypoglycemic effect following gavage in type 1 diabetic mellitus (T1DM) rats. The pharmacological activity and the relative bioavailability were increased to 13.5% and 12.2%, respectively. Thus, these results indicated that CHP assembled NPs may provide a promising potential for oral insulin delivery by protection and absorption promotion.

Spatio-temporal variability of halogenated disinfection by-products in a large-scale two-source water distribution system with enhanced chlorination

Feilong Dong,Zhen Pang,Jianquan Yu,Jing Deng,Xueyan Li,Xiaoyan Ma,Andrea M. Dietrich,Yang Deng

Distributions of halogenated disinfection byproducts (DBPs) in a two-source water distribution system (WDS) with enhanced chlorination were investigated. The WDS was divided into different sub-service areas based on different electrical conductivity of two water sources. Results clearly show that the principal halogenated DBPs were trihalomethanes (THMs) (5.06–82.69 μg/L), varying within the concentration range as 2–5 times as the levels of haloacetic acids (HAAs) (1.41–61.48 μg/L) and haloacetonitriles (HANs) (0.21–15.13 μg/L). Different water sources, treatment trains, and enhanced chlorination within the WDS had significant effects on seasonal and spatial variations of the DBP distributions over water conveyance. THM and HAA formation followed the sequence of summer > autumn > winter > spring. On the other hand, the DBP spatial distributions were visualized using the ArcGIS enabled Inverse distance weighting technique. The superposition of different DBP spatial distributions allowed for the identification of the high-risk THMs and HAAs areas based on the average values of THMs (27.49 μg/L) and HAAs (14.06 μg/L). Beyond the comprehensive analyses of DBP distribution in a municipal WDS , the project proposed and validated an innovative methodology to locate the DBP high-risk areas and to reveal the effects of different factors on DBPs distribution in a two-source WDS.

Ferrate(VI) pre-treatment and subsequent chlorination of blue-green algae: Quantification of disinfection byproducts

Feilong Dong,Jiaqi Liu,Cong Li,Qiufeng Lin,Tuqiao Zhang,Kejia Zhang,Virender K. Sharma

Algal organic matter (AOM) from seasonal algal blooms may be an important precursor of disinfection byproducts (DBPs) in drinking water. This paper presents the effect of ferrate(VI) treatment on two blue-green algae, Chlorella sp. and Pseudanabaena limnetica , in eutrophic water. The results demonstrated that Fe(VI) removed the algal cells by causing cell death, apoptosis, and lost integrity, and decreased AOM (in terms of total organic carbon) in water via oxidation and coagulation. Chlorination of the Fe(VI) pre-oxidized algal water samples generated halogenated DBPs (including trihalomethanes, haloacetic acids, haloketones, chloral hydrate, haloacetonitriles, and trichloronitromethane), but the concentrations of DBPs were lower than those formed in the chlorinated samples without pre-treatment by Fe(VI). Higher Fe(VI) dose, longer oxidation time, and alkaline pH were beneficial in controlling DBPs. In bromide-containing algal solutions, negligible amount of bromo-DBPs were generated in the Fe(VI) pre-oxidation, and halogenated DBPs were mainly formed in the subsequent chlorination.

Novel MIL-53(Fe)@C Magnetic Composite Electrode for Efficient Dechlorination of Disinfection By-Product Trichloroacetic Acid in Water Treatment

Xiaoyan Ma,Rongbin Quan,Wenqing Cao,Weijie Zhang,Su Jiang,Jiao Feng,Jiulong Wang,Stefanos Giannakis

Electrochemical reduction is a promising strategy for the dechlorination of halogenated organic compounds, offering advantages such as enhanced electron transfer efficiency and increased hydrogen atom concentration. It has garnered significant attention for application in mitigating halogenated disinfection by-products (DBPs) in drinking water, owing to its high efficiency and simple operation. In this study, trichloroacetic acid (TCAA), a representative DBP, was selected as the target contaminant. A novel composite cathode comprising a metal–organic framework MIL-53(Fe)@C supported on an Nd magnet (MIL-53(Fe)@C-MAG) and its dechlorination performance for TCAA were systematically investigated. The innovative aspect of this study is the magnetic attachment of the MOF catalyst to the carbonized cathode surface treated through carbonization, which fundamentally differs from conventional solvent-based adhesion methods. Compared to the bare electrode, the MIL-53(Fe)@C-MAG achieved a TCAA removal efficiency exceeding 96.03% within 8 h of contact time. The structural characterization revealed that the α-Fe0crystalline phase serves as the primary active center within the MIL-53(Fe)@C catalyst, facilitating efficient electron transfer and TCAA degradation. The scavenger experiments revealed that TCAA reduction involves a dual pathway: direct electron transfer and atomic hydrogen generation. The modified MIL-53(Fe)@C-MAG electrode exhibited robust electrolytic performance over a broad pH range of 3–7, with TCAA removal efficiency showing a positive correlation with current density within the range of 10–50 mA/cm2. Furthermore, the electrode maintained exceptional stability, retaining more than 90% removal efficiency after five consecutive operational cycles. The versatility of the system was further validated by the rapid and efficient dechlorination of various chlorinated DBPs, demonstrating the broad applicability of the electrode. The innovative magnetic composite electrode demonstrates a significant advancement in electrochemical dechlorination technology, offering a reliable and efficient solution for the purification of drinking water contaminated with diverse halogenated DBPs. These results provide valuable insights into the development of electrolysis for dechlorination in water treatment applications.
