Investigating the mechanisms of Triclocarban-induced liver injury: A comprehensive analysisbased on network toxicology and in vivo experiments

Xinyu Fang,Jinfeng Zhao,Simin Wu,Peng Liao,Guiping Guan

Triclocarban (TCC) is a widely used antibacterial agent with chemical stability and lipophilicity, posing potential health risks due to environmental accumulation and bioaccumulation. This research explored the toxicological features and molecular pathways underlying TCC-induced liver damage through a combination of network toxicology, molecular docking, and in vivo studies. Network toxicology identified 269 potential targets and nine core genes, associated with TCC-induced liver damage. Molecular docking studies indicated significant binding affinities between TCC and the core proteins, implying potential functional interference. In vivo experiments with weaned piglets showed significant liver toxicity at high TCC doses, characterized by decreased liver index, elevated serum ALT and AST, and histopathological damage. High-dose TCC also inhibited antioxidant enzyme activities (GSH-PX and CAT), reduced T-AOC and T-SOD, and increased MDA levels. RT-qPCR analysis further confirmed that the mRNA levels of key target genes—including AKT1, HIF1A, EGFR, ALB, MMP9, BCL2L1, ANXA5, and PTGS2—were significantly up-regulated in the liver, whereas no significant change was observed for the core gene ESR1.This indicates the activation of oxidative stress and inflammatory pathways. These findings provide new insights into TCC-induced hepatotoxicity and highlight the need for further research on intervention targets and comprehensive health risk assessments across different exposures and species.

Responses of microbial communities in coastal sediments exposed to triclocarban and triclosan

Jingwei Wang,Pan Zhao,Jiaxin Wang,Shuzhen Li,Qiao Ma

Triclocarban (TCC) and triclosan (TCS) are applied in a wide range of pharmaceutical and personal care products to prevent or reduce bacterial growth. Due to their extensive application, they are frequently detected in marine environments. In this study, marine sediment systems exposed to different concentrations of TCC and TCS were established to evaluate their effects on microbial communities. It was found that TCC and TCS increased catalase and protease activities on Day 1, but inhibited after 15 days. Microbial activity, as indicated by increased dehydrogenase activity and polysaccharide production, should be enhanced after a 15-day adaptation period. High-throughput sequencing revealed resilient α-diversity but significant shifts in community structures were observed, particularly on Day 15. Function prediction analysis confirmed that most functional profiles remained stable, and network analysis indicated that TCC and TCS enhanced the complexity of the microbial community. This study provides new insights into the impacts and risks of TCC and TCS on the marine environment.

Metabolic network and recovery mechanism of Escherichia coli associated with triclocarban stress

Qiao Zhong,Ying Deng,Huaming Qin,Huase Ou,Yanfen Qu,Jinshao Ye

Although the toxicity of triclocarban at molecular level has been investigated, the metabolic networks involved in regulating the stress processes are not clear. Whether the cells would maintain specific phenotypic characteristics after triclocarban stress is also needed to be clarified. In this study, Escherichia coli was selected as a model to elucidate the cellular metabolism response associated with triclocarban stress and the recovery metabolic network of the triclocarban-treated cells using the proteomics and metabolomics approaches. Results showed that triclocarban caused systematic metabolic remodeling. The adaptive pathways, glyoxylate shunt and acetate-switch were activated. These arrangements allowed cells to use more acetyl-CoA and to reduce carbon atom loss. The upregulation of NH 3 -dependent NAD + synthetase complemented the NAD + consumption by catabolism, maintaining the redox balance. The synthesis of 1-deoxy-D-xylulose-5-phosphate was suppressed, which would affect the accumulation of end products of its downstream pathway of isoprenoid synthesis. After recovery culture for 12 h, the state of cells returned to stability and the main impacts on metabolic network triggered by triclocarban have disappeared. However, drug resistance caused by long-term exposure to environmentally relevant concentration of triclocarban is still worthy of attention. The present study revealed the molecular events under triclocarban stress and clarified how triclocarban influence the metabolic networks.

Core–Shell Structured Magnetic Covalent Organic Framework Nanocomposites for Triclosan and Triclocarban Adsorption

Yanxia Li,Hongna Zhang,Yiting Chen,Lu Huang,Zian Lin,Zongwei Cai

Triclosan (TCS) and triclocarban (TCC) are widely used as bactericides in personal-care products. They are frequently found in environmental water and have the potential to cause a number of environmental and human health problems. In this study, we investigated adsorption and magnetic extraction for efficient removal of TCS and TCC from water and serum samples by core–shell structured magnetic covalent organic framework nanocomposites (Fe3O4@COFs). The as-prepared Fe3O4@COFs was fabricated on the Fe3O4 nanoparticles in situ growth strategy at room temperature via condensation reaction of 1,3,5-tris(4-aminophenyl) benzene (TAPB) and terephthaldicarbox-aldehyde (TPA) in the presence of dimethyl sulfoxide (DMSO). The whole process of adsorption was monitored by ultrahigh performance liquid chromatography–tandem mass spectrometry (UHPLC-MS/MS) analysis with high sensitivity. The adsorption behaviors showed high adsorption capacity and fast adsorption. Furthermore, the adsorption performance through Langmuir and Freundlich isotherms showed multilayer adsorption through the interactions of space embedding effect, van der Waals forces, and benzene ring π–π stacking at a low concentration range and monolayer adsorption through strong π–π stacking at a high concentration range between the interface of TCS or TCC and Fe3O4@COFs at a high concentration range. Results indicated that the adsorption of TCS and TCC onto Fe3O4@COFs can be better represented by the pseudo-second-order model. Good removal efficiencies (82.3∼95.4%) and recoveries (92.9∼109.5%) of TCS and TCC in fetal bovine serum (FBS) and reusability at least 10 times were achieved. The Fe3O4@COFs exhibited high stability and excellent performance for the removal of TCS and TCC from water and biological samples. The results presented here thus reveal the exceptional potential of COFs for high-efficient environmental remediation.

Molecular mechanism of adsorbing triclocarban by the activated sludge in wastewater treatment systems

Hao Wang,Hanlin Cui,Xiaodan Ma,Carolyn R. Cornell,Liying Zhang,Yijun Ren,Minghan Li,Ying Liu,Shu-Hong Gao,Zhiling Li,Ai-Jie Wang,Jizhong Zhou,Bin Liang

The ubiquitous use of antimicrobial triclocarban (TCC) has raised widespread concern about the potential impacts on wastewater treatment systems, the environment, and the human population. As important constituents of activated sludge secretions, extracellular polymeric substances (EPS) play essential roles in removing micropollutants during wastewater treatment. Here, the adsorption of TCC by sludge and EPS in a multistage biological process was investigated. The EPS was enriched upon TCC stress, and the adsorption by sludge and EPS showed multilayer heterogeneity. The adsorption constant K f (6.3794–7.6061) of EPS was over 10 times higher than that of sludge (0.5661–0.6200) in all stages, potentially owing to the loose multilayer structure and substantial functional groups (tryptophan-protein as the main binding site) in EPS. TCC significantly inhibited the biological removal of ammonia nitrogen. Notably, TCC once reaching the cell surface, formed a stable complex with ammonia transporter (Amt, binding energy: −73.1 ± 16.6 kJ·mol −1 ) and bound to the selectivity filter vestibule of Amt (tryptophan 144 and serine 227), inhibiting the transport of ammonia. Our work drew molecular-level insights into the interaction between TCC and activated sludge, and evaluated the adverse impacts on treatment performance through an in silico approach, providing new information on the fate and risk of TCC in wastewater treatment systems.

Photodegradation of the antimicrobial triclocarban in aqueous systems under ultraviolet radiation

Ding Shi-Ling,Wang Xi-Kui,Jiang Wen-Qiang,Meng Xia,Zhao Ru-Song,Wang Chen,Wang Xia

This work aimed to investigate the effectiveness of ultraviolet (UV) radiation on the degradation of the antimicrobial triclocarban (TCC). We investigated the effects of several operational parameters, including solution pH, initial TCC concentration, photocatalyst TiO 2 loading, presence of natural organic matter, and most common anions in surface waters (e.g., bicarbonate, nitrate, and sulfate). The results showed that UV radiation was very effective for TCC photodegradation and that the photolysis followed pseudo-first-order kinetics. The TCC photolysis rate was pH dependent and favored at high pH. A higher TCC photolysis rate was observed by direct photolysis than TiO 2 photocatalysis. The presence of the inorganic ions bicarbonate, nitrate, and sulfate hindered TCC photolysis. Negative effects on TCC photolysis were also observed by the addition of humic acid due to competitive UV absorbance. The main degradation products of TCC were tentatively identified by gas chromatograph with mass spectrometer, and a possible degradation pathway of TCC was also proposed.

Influence of pH, inorganic anions, and dissolved organic matter on the photolysis of antimicrobial triclocarban in aqueous systems under simulated sunlight irradiation

Ding Shi-Ling,Wang Xi-Kui,Jiang Wen-Qiang,Zhao Ru-Song,Shen Ting-Ting,Wang Chen,Wang Xia

The photolysis of the antimicrobial triclocarban (TCC) in aqueous systems under simulated sunlight irradiation was studied. The effects of several abiotic parameters, including solution pH, initial TCC concentration, presence of natural organic matter, and most common inorganic anions in surface waters, were investigated. The results show that the photolysis of TCC followed pseudo-first-order kinetics. The TCC photolysis rate constant increased with increasing solution pH and decreasing the initial TCC concentration. Compared with the TCC photolysis in pure water, the presence of aqueous bicarbonate, nitrate, humic acids, and its sodium salt decreased the TCC photolysis rate, but fulvic acid increased the TCC photolysis rate. The electron spin resonance and reactive oxygen species scavenging experiments indicated that TCC may undergo two different types of phototransformation reactions: direct photolysis and energy transfer to generate 1 O 2 . The main degradation products were tentatively identified by gas chromatography interfaced with mass spectrometry (GC-MS), and a possible degradation pathway was also proposed.

Binding studies of triclocarban with bovine serum albumin: Insights from multi-spectroscopy and molecular modeling methods

Jiao Guan,Xin Yan,Yajing Zhao,Yinhe Sun,Xin Peng

The antimicrobial triclocarban (TCC) is frequently found in various personal care products (PCPs), and recent studies have demonstrated that it shows a high unintended biological activity on humans and wildlife. To evaluate the toxicity of TCC at the protein level, the effect of TCC on bovine serum albumin (BSA) has been investigated using various spectroscopic methods in combination with molecular modeling. Analysis of fluorescence quenching data of BSA revealed the formation of a ground state BSA-TCC complex with a binding constant of 2.58 × 10 4  M −1 at 298 K. The values of the thermodynamic parameters suggested that the binding of TCC to BSA was driven mainly by hydrophobic interaction and hydrogen bond . Site marker competitive experiments coupled with molecular docking studies confirmed that site I was the main binding site for TCC on BSA. Furthermore, TCC binding to BSA led to conformational and structural alterations of BSA as revealed by multi-spectroscopic studies. In addition, the stability of BSA and BSA-TCC complex were well analyzed by the molecular dynamics studies . In short, this work indicated that TCC could interact with BSA and impact the conformation of BSA, which could provide valuable information to understand the toxicity mechanism of TCC.

Co-occurrence of antiseptic triclocarban and chiral anti-inflammatory ibuprofen in environment: Association between biological effect in sediment and risk to human health

Han Qu,Holly Barrett,Bin Wang,Jiajun Han,Fang Wang,Wenwen Gong,Junxue Wu,Wei Wang,Gang Yu

Residues of antiseptics and drugs have been ubiquitously detected in aquatic water-sediment systems, and are thus considered emerging contaminants that threaten our global environment. To investigate the potential risk of ibuprofen and triclocarban in sediment, effects of enzyme activity on the enantioselective degradation in sediment were investigated. Enantioselective fate of rac-ibuprofen was observed in sediment with R-enantiomer exhibiting preferential degradation. Enzyme evidence showed that high levels of triclocarban could significantly inhibit activities of catalase and urease activities in sediment, as well as increase the half-life of ibuprofen (from 5.8 d to 10.1 d). Cytotoxicity data suggested that cell growth processes were significantly affected by ibuprofen and triclocarban co-exposure, which was consistent with apoptosis results. Additionally, the expression of several proteins (Cyto-c, Nrf2, p62, Keap1, NQO1 , and Pink1) were markedly induced upon exposure to ibuprofen in the presence of triclocarban. In conclusion, these findings illustrated that co-occurrence of ibuprofen and triclocarban residues have synergistic adverse effects to the environment and synergistically threaten human health.

Functional Characterization of a Novel Amidase Involved in Biotransformation of Triclocarban and its Dehalogenated Congeners in Ochrobactrum sp. TCC-2

Hui Yun,Bin Liang,Jiguo Qiu,Long Zhang,Youkang Zhao,Jiandong Jiang,Aijie Wang

Haloaromatic antimicrobial triclocarban (3,4,4′-trichlorocarbanilide, TCC) is a refractory contaminant which is frequently detected in various aquatic and sediment environments globally. However, few TCC-degrading communities or pure cultures have been documented, and functional enzymes involved in TCC biodegradation hitherto have not yet been characterized. In this study, a bacterial strain, Ochrobactrum sp. TCC-2, capable of degrading TCC under both aerobic and anaerobic conditions was isolated from a sediment sample. A novel amidase gene (tccA), responsible for the hydrolysis of the two amide bonds of TCC and its dehalogenated congeners 4,4′-dichlorocarbanilide (DCC) and carbanilide (NCC) to more biodegradable chloroaniline or aniline products, was cloned and characterized. TccA shares low amino acid sequence identity (27 to 38%) with other biochemically characterized amidases and contains the conserved catalytic triad (Ser-Ser-Lys) of the amidase signature enzyme family. TccA was stable over a pH range of 5.0 to 10.0 and at temperatures lower than 60 °C, and it was constitutively expressed in strain TCC-2. In contrast to the halogenated TCC and DCC, the nonchlorinated NCC was the preferred substrate for TccA. TccA also had hydrolysis activity to a broad spectrum of amide bonds in herbicides, insecticides, and chemical intermediates. The constitutive expression and broad substrate spectrum of TccA suggested strain TCC-2 may be potentially useful for bioremediation applications.

Short-term exposure to triclocarban alters microbial community composition and metabolite profiles in freshwater biofilms

Yitong Yan,Jin Qian,Yin Liu,Jing Hu,Bianhe Lu,Shasha Zhao,Shuai Jin,Yuxuan He,Kailin Xu

Triclocarban (TCC), an emerging contaminant in water environments, its effects on freshwater biofilms remain insufficiently understood. This study investigates the effects of TCC exposure (at concentrations of 10 μg L −1 and 10 mg L −1 ) on mature freshwater biofilms. TCC was found to inhibit biofilm activity as evidenced by changes in surface morphology and the ratio of live/dead cells. Moreover, both concentrations of TCC were observed to modify the structure of the biofilm community. Metabolomics analysis revealed an overlap in the toxicity mechanisms and detoxification strategies triggered by various concentrations of TCC in biofilms. However, the higher toxicity induced by 10 mg L −1 TCC resulted from the downregulation of proline betaine , disrupting the homeostasis of cellular osmotic pressure regulation in biofilms. Notably, lipid and lipid-like molecules showed high sensitivity to different concentrations of TCC, indicating their potential as biomarkers for TCC exposure. Annotation of the differential metabolites by KEGG revealed that alterations in amino acid and carbon metabolism constituted the primary response mechanisms of biofilms to TCC. Moreover, the biofilm demonstrated enhanced nucleic acid metabolism , which bolstered resistance against TCC stress and heightened tolerance. Furthermore, elevated TCC concentrations prompted more robust detoxification processes for self-defense. Overall, short-term exposure to TCC induced acute toxicity in biofilms, yet they managed to regulate their community structure and metabolic levels to uphold oxidative homeostasis and activity. This research contributes to a deeper comprehension of TCC risk assessment and policy control in aquatic environments .

Triclocarban exposure at environmentally relevant concentrations perturbs the gut microbiota and metabolic profile in Rana taihangensis (Anura, Ranidae) tadpoles

Ruinan Zhao,Pengyuan Liu,Yaoyang Wu,Hongyan Bi,Jiaoying He,Jie Zhang,Yanfu Qu,Xiaohong Chen,Zhuo Chen

Introduction The antibacterial agent triclocarban (TCC) poses a significant threat to aquatic ecosystems and its impact on amphibians remain poorly understood. Methods Here, we investigated its acute and chronic effects on Rana taihangensis tadpoles at environmentally relevant concentrations (i.e., 5, 15 and 45 μg/L) using an integrated approach combining morphology, gut microbiome, and non-targeted metabolomic analyses. Results and Discussion The acute toxicity tests for 96 h revealed that TCC had a lethal concentration (LC50) value of 169.863 μg/L for R. taihangensis tadpoles. Chronic exposure resulted in reduced body condition score across all the three TCC-treated groups compared to the control. Gut microbiome analysis revealed that TCC exposure significantly altered the community composition at both phylum (e.g., Pseudomonadota and Fusobacteriota) and genus (e.g., Cetobacterium and Citrobacter ) levels. In addition, several metabolites (e.g., 20-carboxy-leukotriene B4, 11b-PGF2a, and leukotriene E4) associated with immune response and neural signaling were significantly perturbed in TCC-exposed tadpoles. Interestingly, correlation analysis indicated a significant relationship between specific metabolite changes and shifts in gut microbiota. Overall, our findings demonstrated that TCC exposure adversely affects the growth indexes, gut microbial composition and metabolites in R. taihangensis tadpoles, and the present study will provide new insights into the ecotoxicological risks of TCC and enhance the understanding of its mechanisms of toxicity in aquatic organisms.
