A laminin mimetic peptide SIKVAV-conjugated chitosan hydrogel promoting wound healing by enhancing angiogenesis, re-epithelialization and collagen deposition

Shixuan Chen,Min Zhang,Xuebing Shao,Xueer Wang,Lei Zhang,Pengcheng Xu,Wen Zhong,Lu Zhang,Malcolm Xing,Lin Zhang

Angiogenesis and re-epithelialization are critical factors in skin wound healing. Growth factors and stem cells have demonstrated their active roles in promoting these two processes. Peptides show similar effects with growth factors with lower cost and controllable properties. Here we report a biomimetic fragment of the laminin –Ser-Ile-Lys-Val-Ala-Val (SIKVAV)-conjugated chitosan hydrogel that can promote skin regeneration. In vitro we found that this peptide-conjugated hydrogel significantly promoted BMSC adhesion and proliferation. In vivo, this hydrogel accelerated wound contraction. The subcutaneous implantation test and H&E staining results revealed that the peptide-modified chitosan hydrogel dramatically led to the formation of new blood vessels. Moreover, Masson staining showed that many newborn collagen fibers appeared in the peptide hydrogel group, while only a few newborn collagen fibers were found in control and chitosan hydrogel groups. The peptide chitosan hydrogel also re-epithelialized quickly, while the control and chitosan hydrogel took more time to complete. These results suggest that the SIKVAV peptide is an effective motif to significantly improve the function of chitosan in angiogenesis and re-epithelialization of skin.

A biomimetic hyaluronic acid-silk fibroin nanofiber scaffold promoting regeneration of transected urothelium

Yuqing Niu,Massimiliano Galluzzi,Fuming Deng,Zhang Zhao,Ming Fu,Liang Su,Weitang Sun,Wei Jia,Huimin Xia

This study was designed to investigate the regulatory effect of hyaluronic acid (HA)—coating silk fibroin (SF) nanofibers during epithelialization of urinary tract for urethral regeneration. The obtained electrospun biomimetic tubular HA-SF nanofiber scaffold is composed of a dense inner layer and a porous outer layer in order to mimic adhesion and cavernous layers of the native tissue, respectively. A thin layer of HA-gel coating was fixed in the inner wall to provide SF nanofibers with a dense and smooth surface nano-topography and higher hydrophilicity. Compared with pure SF nanofibers, HA-SF nanofibers significantly promoted the adhesion, growth, and proliferation of primary urothelial cells, and up-regulate the expression of uroplakin-3 (terminal differentiation keratin protein in urothelium). Using the New Zealand male rabbit urethral injury model, the scaffold composed of tubular HA-SF nanofibers could recruit lumen and myoepithelial cells from the adjacent area of the host, rapidly reconstructing the urothelial barrier in the wound area in order to keep the urinary tract unobstructed, thereby promoting luminal epithelialization, smooth muscle bundle structural remodeling, and capillary formation. Overall, the synergistic effects of nano-topography and biophysical cues in a biomimetic scaffold design for effective endogenous regeneration.

3D printed mold-based capsaicin candy for the treatment of oral ulcer

Heliu Jiang,Xiang Yu,Rongzhen Fang,Zhimei Xiao,Yiguang Jin

Oral ulcer is one common mucosal disease with high prevalence. Here, capsaicin candies were prepared based on the stereolithographically (SLA) 3D printed molds. The molds can be freely designed depending on the needs of patients, involving symmetric shapes (e.g., round, four-lead clover and cube), asymmetric shapes (e.g., car) and various color (e.g., blue, red and yellow). A two-part-combined mold was filled with the xylitol-based material and separated to obtain hard candies. Capsaicin was amorphous in the candies according to the differential scanning calorimetry and X-ray diffraction. Poloxamer 188 improved the release of capsaicin from the candies. Rat oral ulcer models were established on the tongue with phenol liquids. The blank candy, 0.05% capsaicin candy and dexamethasone were respectively administered on the ulcer once daily. On Day 7, a healing rate of 97.8% was achieved by the capsaicin candy, much higher than those in the other groups. Moreover, the blank candy also showed the remarkable ulcer healing effect due to the presence of xylitol and poloxamer. Capsaicin remarkably enhanced the reepithelialization of ulcer tissues and showed strong anti-inflammatory effect by reducing the expressions of THF-α and IL-6. 3D printing-based capsaicin candies provide an interesting therapeutic choice for the people with oral ulcer.

MXenes-integrated microneedle combined with asiaticoside to penetrate the cuticle for treatment of diabetic foot ulcer

Wang Pei,Wang Yun,Yi Yang,Gong Yan,Ji Haoran,Gan Yuci,Xie Fei,Fan Jinchen,Wang Xiansong

Patients with diabetic foot ulcers usually suffer from inefficient epithelisation and angiogenesis accompanied by chronic wound healing. Diabetic foot ulcers remain a major challenge in clinical medicine; however, traditional treatments are incapable of transdermal drug delivery, resulting in a low drug delivery rate. We report the development of Ti 2 C 3 MXenes-integrated poly-γ-glutamic acid (γ-PGA) hydrogel microneedles to release asiaticoside (MN-MXenes-AS). Asiaticoside was loaded into PGA-MXenes hydrogel to facilitate cell proliferation while regulating angiogenesis. The characterisation and mechanical strength of the microneedles were investigated in vitro, and the wound-healing efficacy of the microneedles was confirmed in diabetic mice. MXenes significantly improved the mechanical strength of microneedles, while γ-PGA hydrogels provided a moist microenvironment for wound healing. Mice treated with MN-MXenes-AS demonstrated obvious improvements in wound healing process. We successfully fabricated an MXenes-integrated microneedle that possesses sufficient rigidity to penetrate the cuticle for subcutaneous drug delivery, thereby accelerating diabetic wound healing. We demonstrated that MN-MXenes-AS is effective in promoting growth both in vivo and in vitro. Collectively, our data show that MN-MXenes-AS accelerated the healing of diabetic foot ulcers, supporting the use of these microneedles in the treatment of chronic wounds. Graphical Abstract

A Bilayer Polyurethane Patch with Sustained Growth Factor Release and Antibacteria for Re-epithelization of Large-Scale Oral Mucosal Defects

Qiao Zhang,Jinlin Chen,Tianyu Zhang,Dan Liu,Xirui Long,Jiehua Li,Lu Jiang,Yanchao Wang,Hong Tan

In the field of oral and maxillofacial surgery, extensive oral soft-tissue injuries occur repeatedly in clinical practice; however, effective restorative materials are lacking. In this study, a biodegradable waterborne polyurethane patch featuring a mucosa bionic bilayer structure is presented. This patch consists of a porous scaffold layer that faces the lesion, incorporating a polydopamine coating to achieve sustained release of epidermal growth factors (EGFs) for mucosal defect reconstruction. Additionally, there is a dense barrier layer toward the oral cavity loaded with silver nanoparticles, which prevents bacteria from entering the wound and simultaneously acts as a physical barrier. This patch can sustainably release EGF in vitro for 2 weeks, thereby facilitating the proliferation and migration of HaCaT and L929 cells, while effectively killing common oral cavity bacteria. In a rabbit buccal mucosal full-thickness defect model, the patch demonstrates better efficacy than the clinical benchmark, decellularized extracellular matrix (dECM). It effectively reduces wound inflammation and significantly upregulates gene expression associated with epithelialization by activating the EGF/epidermal growth factor receptor (EGFR) pathway. These mechanisms promote the proliferation, differentiation, and migration of epithelial/keratinocyte cells, ultimately expediting mucosal defect healing and wound closure.

The diameter factor of aligned membranes facilitates wound healing by promoting epithelialization in an immune way

Chenbing Wang,Chenyu Chu,Xiwen Zhao,Yang Yang,Chen Hu,Li Liu,Jidong Li,Yili Qu,Yi Man

Topographical properties, such as pattern and diameter, of biomaterials play important roles in influencing cell activities and manipulating the related immune response during wound healing. We prepared aligned electrospinning membranes with different fiber diameters, including 319 ± 100 nm (A300), 588 ± 132 nm (A600), and 1048 ± 130 nm (A1000), by adjusting the distance from the tip to the collector, the injection rate, and the concentration of the solution. The A300 membranes significantly improved cell proliferation and spreading and facilitated wound healing (epithelization and vascularization) with the regeneration of immature hair follicles compared to the other membranes. Transcriptomics revealed the underlying molecular mechanism that A300 could promote immune-related processes towards a pro-healing direction, significantly promoting keratinocyte migration and skin wound healing. All the results indicated that wound healing requires the active participation of the immune process, and that A300 was a potential candidate for guided skin regeneration applications.

Collagen membrane loaded with doxycycline through hydroxypropyl chitosan microspheres for the early reconstruction of alkali-burned cornea

Jia Liu,Yingni Xu,Yongrui Huang,Xiaomin Sun,Yuehai Peng,Wenjing Song,Jin Yuan,Li Ren

Corneal alkali burn is one of the most devastating ophthalmic emergencies correlated with remarkable morbidity resulting in severe visual impairment. Appropriate intervention in the acute phase determines the eventual outcome for later corneal restoration treatment. Since the epithelium plays an essential role in inhibiting inflammation and promoting tissue repair, sustained anti-matrix metalloproteinases (MMPs) and pro-epithelialization are the prior remedies during the first week. In this study, a drug-loaded collagen membrane (Dox-HCM/Col) that could be sutured to overlay the burned cornea was developed to accelerate the early reconstruction. Doxycycline (Dox), a specific inhibitor of MMPs, was encapsulated in collagen membrane (Col) through hydroxypropyl chitosan microspheres (HCM) to develop Dox-HCM/Col, affording a preferable pro-epithelialization microenvironment and an in-situ controlled release. Results showed that loading HCM into Col prolonged the release time to 7 days, and Dox-HCM/Col could significantly suppress the expression of MMP-9 and -13 in vitro and in vivo . Furthermore, the membrane accelerated the corneal complete re-epithelialization and promoted early reconstruction within the first week. Overall, Dox-HCM/Col was a promising biomaterial membrane for treating alkali-burned cornea in the early stage, and our attempt may provide a clinically feasible method for the ocular surface reconstruction.

ECM-inspired stem cell secretome sustained releasing composite nanofibrous membranes for accelerated wound healing

Jiutao Cao,Shengchang Luo,Wanling Huang,Xiaochang Lu,Ranjith Kumar Kankala,Shibin Wang,Peiyao Xu,Aizheng Chen

Designing dressings that mimic the composition, structure, and function of native skin is of great significance. Despite commercial decellularized extracellular matrix (dECM)-based wound dressings promoted skin regeneration process, reconstructing the fibrous microstructure of native dermis remains a significant challenge. In this work, a dECM-based composite nanofibrous membranes loaded with stem cell secretomes (SCS/dECMM) have been developed for accelerating wound repair, which comprehensively mimicked the dermal tissue in terms of composition, structure, and function. In detail, dermal dECM was prepared by eco-friendly supercritical carbon dioxide (SC-CO 2 ) technology and then mixed with SCS to obtain SCS/dECMM by using electrospinning technology. The resulting nanofibrous membrane with 555.19 nm diameter that maintained SCS bioactivity while enabling sustained SCS release. SCS/dECMM significantly enhanced cell adhesion, proliferation, migration, and angiogenesis, thereby continuously promoting wound repair. In vivo wound healing results revealed that SCS/dECMM accelerated wound healing by promoting re-epithelialization, collagen deposition, and vascularization. SCS/dECMM offered a novel strategy for accelerating wound healing by replicating the composition, structure, and function of native skin.

Chenodeoxycholic acid triggers gastric mucosal injury by inducing apoptosis and FXR activation

Shuaijun Peng,Ruiqin Sun,Hui Liu,Baoying Wang,Yucheng Li

Gastric mucosal injury can lead to significant gastrointestinal disorders, including inflammation, ulcers, and intestinal metaplasia in severe cases. Although bile acids are known contributors to mucosal damage due to their acidity, the role of chenodeoxycholic acid (CDCA), a primary bile acid, in gastric mucosal injury remains unclear. This study investigates the role of CDCA in inducing gastric mucosal injury and its underlying mechanisms, with a focus on apoptosis and FXR activation, to identify potential therapeutic targets. Mice were administered varying doses of CDCA to evaluate changes in body weight, food intake, and gastric mucosal pathology using hematoxylin-eosin (HE) and alcian blue-periodic acid-Schiff (AB-PAS) staining. Expression levels of apoptosis-related genes (BAX, BCL-2) and the intestinal metaplasia marker gene (CDX2) were analyzed via RT-PCR and Western blotting (WB). GES-1 cells were treated with different CDCA concentrations to assess cell viability via the CCK-8 assay. Apoptosis rates were measured using flow cytometry, and FXR inhibitors were applied to examine their impact on CDCA-induced effects. CDCA administration in mice resulted in weight loss, reduced food and water intake, gastric epithelial shedding, and intestinal metaplasia. CDCA exposure upregulated Bax and CDX2 expression, while reducing BCL-2 levels. In vitro, CDCA inhibited GES-1 cell viability and increased apoptosis rates, effects that were reversed by FXR inhibition. CDCA induces gastric mucosal injury through apoptosis and FXR activation. These findings provide insights into the mechanisms of bile acid-mediated mucosal damage and highlight FXR as a potential therapeutic target for gastric mucosal disorders.

Rapid hemostatic and mild polyurethane-urea foam wound dressing for promoting wound healing

Xiangyu Liu,Yuqing Niu,Kevin C. Chen,Shiguo Chen

A novel rapid hemostatic and mild polyurethane-urea foam (PUUF) wound dressing was prepared by the particle leaching method and vacuum freeze-drying method using 4, 4-Methylenebis(cyclohexyl isocyanate), 4,4-diaminodicyclohexylmethane and poly (ethylene glycol) as raw materials. And X-ray diffraction (XRD), tensile test, differential scanning calorimetry (DSC) and thermogravimetry (TG) were used to its crystallinity, stress and strain behavior, and thermal properties, respectively. Platelet adhesion, fibrinogen adhesion and blood clotting were performed to evaluate its hemostatic effect. And H&E staining and Masson Trichrome staining were used to its wound healing efficacy. The results revealed the pore size of PUUF is 50–130   μm, and its porosity is 71.01%. Porous PUUF exhibited good water uptake that was benefit to adsorb abundant wound exudates to build a regional moist environment beneficial for wound healing. The PUUF wound dressing exhibit better blood coagulation effect than commercial polyurethane dressing (CaduMedi). Though both PUUF and CaduMedi facilitated wound healing generating full re-epithelialization within 13   days, PUUF was milder and lead to more slight inflammatory response than CaduMedi. In addition, PUUF wound dressing exhibited lower cytotoxicity than CaduMedi against NIH3T3 cells. Overall, porous PUUF represents a novel mild wound dressing with excellent water uptake, hemostatic effect and low toxicity, and it can promote wound healing and enhance re-epithelialization.

Co-assembling of natural drug-food homologous molecule into composite hydrogel for accelerating diabetic wound healing

Yiyang Hou,Haibo Huang,Wei Gong,Ran Wang,Wanying He,Xinchuang Wang,Jiangning Hu

Diabetic wound healing is a major clinical challenge due to its vulnerability to bacterial infection and the prolonged inflammation in the wound. Traditional dressings for the healing of diabetic wounds are often suffered from unsatisfactory efficacy and frequent dressing changes which may cause secondary damage. Therefore, it is necessary to find a wound dressing that balances material functionality, degradation, safety, and tissue regeneration. Our recent studies demonstrated that gallic acid (GA) could spontaneously form supramolecular hydrogels at a relatively high concentration. However, a single network of GA hydrogel is prone to degradation, poor adhesion, and poor swelling, and may not be suitable for wound healing dressings. In this study, a composite hydrogel (GAK) was constructed by introducing konjac glucomannan (KGM) into the gel system of gallic acid (GA) and applied to promote diabetic wound healing. The composite hydrogel (GAK) with superior surface adhesion, stability, and swelling properties than the single-network of GA hydrogel. Moreover, in vitro experiments showed that GAK hydrogel had excellent biocompatibility and exhibited antibacterial activity against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ). Additionally, the GAK hydrogel could significantly accelerate angiogenesis, collagen deposition, and re-epithelialization during wound healing in diabetic mice, reducing the expression of related inflammatory proteins interleukin-1β (IL-1β), tumor necrosis factor-alpha (TNF-α), and cyclooxygenase-2 (COX-2), and improving the wound closure rate. The findings of this study suggest that this composite hydrogel (GAK) can be an ideal dressing material for accelerating diabetic wound healing.

The effect of Bletilla striata polysaccharide on the physical and healing properties of curdlan-based hydrogel for wound healing

Jin Shang,Liangliang Duan,Weimin Zhang,Qibin Zhuang,Xiaomei Ren,Dale Gu

Bletilla striata polysaccharide (BSP) was added to curdlan to form a blend hydrogel through a simple heating-cooling procedure to improve the hydrophilicity and healing efficacy of curdlan-based hydrogel used in wound healing. We explored the interplay between BSP and curdlan, studied how BSP concentration affects the physical properties and microstructures of hydrogels, and examined the biocompatibility and healing properties of the blend hydrogel. It was proved that the hydrogel framework was primarily formed by ordered arranged curdlan molecules, with BSP uniformly dispersed and intertwined with curdlan through hydrogen bonding. This effectively improved its hydrophilicity and strengthened the microstructure. Curdlan was found to be compatible with BSP. The blend hydrogel B3Cd3 (containing 1.5% BSP and 1.5% curdlan, w/v) was identified as the optimal formulation based on its higher water adsorption, water retention, thermal stability and interconnected microstructure, and was thus selected for further research. In vitro experiments revealed the highest cell viability of L929 in B3Cd3 extracts compared to those extracts of single-component curdlan hydrogel (Cd). In vivo, animal studies indicated that the B3Cd3 accelerated wound healing compared to the control group by improving re-epithelialization and blood vessel regeneration. On Days 3 and 11, the therapeutic benefits of B3Cd3 exceeded those of the Cd group, and no significant differences were observed in wound healing rates between the B and B3Cd3 groups from Day 7. The study proves that BSP enhances the physical and healing properties, as well as cell proliferation, of the curdlan-based hydrogel. The blend hydrogel B3Cd3, with its exceptional properties, holds potential for future application as a material for non-infected wound healing.
