Metagenomic insights into the enhanced methane production by hydrochar at varied propionate concentrations

Taoyang Jiang,Huu Hao Ngo,Meichen Sun,Chao Zhang,Shicheng Zhang,Zhijian Shi,Gang Luo

Propionate is a crucial intermediate product in anaerobic digestion (AD), but its degradation is thermodynamically unfavorable. Its accumulation in the AD system can significantly reduce methane production efficiency and may lead to severe acidification. The present study investigated the methane production at various propionate concentrations (15, 30 and 60 mM) with and without hydrochar and also explored the microbial shifts by metagenomic analysis. The findings revealed that higher propionate concentrations reduced methane production and propionate degradation efficiency, leading to acetate accumulation. The addition of hydrochar mitigated the inhibitory effects of increased propionate concentrations, notably reduced the lag phase by 20.13 % and enhanced the methane production rate by 138.43 % under 30 mM propionate. This also resulted in faster degradation of propionate and acetate. Hydrochar impacted the microbial communities’ functional potentials, enhancing methane metabolic potential in all conditions. Genomic-centric analysis revealed that hydrochar selectively enriched syntrophic propionate-oxidizing bacteria (SPOB) candidate MAG79 Syntrophopropionicum sp., which could participate in DIET, across different concentrations of propionate, with the most pronounced effect at 30 mM propionate. This was distinct from the dominant species in the control groups, MAG63 Syntrophosphaera sp. and MAG69 Syntrophobacteraceae sp.. Hydrochar further supported the growth of another DIET candidate MAG4 Methanosarcina mazei and increased the abundance of proposed syntrophic acetate-oxidizing bacterium (SAOBs), across all propionate concentrations. These effects collectively led to more efficient propionate degradation and methane production.

Propionate and Butyrate Inhibit Biofilm Formation of Salmonella Typhimurium Grown in Laboratory Media and Food Models

Jiaxiu Liu,Wenxiu Zhu,Ningbo Qin,Xiaomeng Ren,Xiaodong Xia

Salmonellais among the most frequently isolated foodborne pathogens, and biofilm formed bySalmonellaposes a potential threat to food safety. Short-chain fatty acids (SCFAs), especially propionate and butyrate, have been demonstrated to exhibit a beneficial effect on promoting intestinal health and regulating the host immune system, but their anti-biofilm property has not been well studied. This study aims to investigate the effects of propionate or butyrate on the biofilm formation and certain virulence traits ofSalmonella. We investigated the effect of propionate or butyrate on the biofilm formation ofSalmonella entericaserovar Typhimurium (S.Typhimurium) SL1344 grown in LB broth or food models (milk or chicken juice) by crystal violet staining methods. Biofilm formation was significantly reduced in LB broth and food models and the reduction was visualized using a scanning electron microscope (SEM). Biofilm metabolic activity was attenuated in the presence of propionate or butyrate. Meanwhile, both SCFAs decreased AI-2 quorum sensing based on reporter strain assay. Butyrate, not propionate, could effectively reduce bacterial motility. Bacterial adhesion to and invasion of Caco-2 cells were also significantly inhibited in the presence of both SCFAs. Finally, two SCFAs downregulated virulence genes related to biofilm formation and invasion through real-time polymerase chain reaction (RT-PCR). These findings demonstrate the potential application of SCFAs in the mitigation ofSalmonellabiofilm in food systems, but future research mimicking food environments encountered during the food chain is necessitated. Keywords:propionate;butyrate;biofilm;antimicrobial activity;SalmonellaTyphimurium

Fe3O4 nanoparticles promote methanogenesis in propionate acclimated system

Fangzhou Wang,Ge Song,Mou Zhang,Shunan Zhao,Tuo Wang,Kai Zhao,Xin Wang,Ruiping Liu

Propionate accumulation is common in anaerobic digestion systems, while Fe 3 O 4 nanoparticles (Fe 3 O 4 NPs) show potential in steering direct interspecies electron transfer (DIET) to facilitate methanogenesis from propionate. However, the effect of Fe 3 O 4 NPs in stable microbial communities remains unclear. This study demonstrated that 1.0 g/L Fe 3 O 4 NPs enhanced propionate degradation and methane production by 220 % and 55 % of the propionate-acclimated microbial consortia post-shock loading, as evidenced by batch-test. High propionate concentrations suppressed Geobacter , yet Fe 3 O 4 NPs enabled electron syntrophy between alternative DIET-participants ( Arcobacter , Syntrophobacter ) and methanogens ( Methanothrix , Methanobacterium ) serving as electron conduits. Network analysis further revealed that Fe 3 O 4 NPs activated interactions among potential electroactive microbes, highlighting the potentially ubiquitous presence of electron syntrophy. Even in propionate-stressed microbial communities, such mutualism may be rapidly activated by Fe 3 O 4 NPs, offering a practical solution for mitigating propionate accumulation and enhance digester performance.

Effects of surfactants on the wettability of sodium propionate aqueous deacidification agent

Ge Ge,Jianan Liu,Yunfei Liao,Deli Zeng,Hongyan Mou,Huiming Fan

To address the challenges of difficulty and low penetration speed of aqueous deacidification agents in paper, we aimed to enhance the wetting performance of these agents by incorporating a surfactant. We selected four surfactants for our study: anionic sodium dodecyl benzene sulfonate (SDBS), cationic cetyltrimethylammonium bromide (CTAB), nonionic polysorbate-80 (TW80), and polyether-modified trisiloxane (TRSE). The wetting properties of these surfactants on sodium propionate aqueous deacidification agent, the alkali stability of the solution, and the effects of moist heat aging on paper durability were explored. The results demonstrated that TRSE exhibited superior performance compared to the other three surfactants, effectively reducing the surface tension of sodium propionate solution to 21.02 mN/m. In alkaline conditions with a pH range of 8.00–10.00, the surface tension of SDBS, CTAB, and TW80 solutions remained stable for 30 days. Similarly, under weakly alkaline conditions with a pH value below 9, the surface tension of TRSE solutions exhibited stability. It should be noted that SDBS accelerated the decline of paper tensile index and whiteness during the heat and humidity aging process; at the same time, CTAB, TW80, and TRSE had no noticeable adverse effects on paper aging resistance.

Continuous electromethanogenesis of propionate wastewater: Effect of external voltage and hydraulic retention time

Dong Feng,Ao Xia,Yun Huang,Xianqing Zhu,Xun Zhu,Pau-Loke Show,Qiang Liao

Conventional anaerobic digestion (AD) may be inhibited by the accumulation of intermediate metabolites, thereby disrupting the stability of the AD process. The emerging electromethanogenesis system opens up a new opportunity to enhance the efficiency of biogas technology. In this study, the potential mechanism of the combined impact of applied external voltage and hydraulic retention time on continuous propionate digestion was investigated for the first time. The methane yield was increased up to 98.6 %, while the methane content was enhanced up to 48.0 % with an external voltage of 1.0 V, compared with the control. The cyclic voltammetry results suggested that the external voltage facilitated faster growth, stronger resistance to hydraulic shock, and higher electrochemical activity of electroactive biofilms with a thickness of approximately 10 μm. Further analysis via a three-dimensional excitation-emission matrix confirmed the external voltage-boosted EPS secretion comprised of protein-like substances, acting as the electron mediator between Smithella and Methanosaeta .

Probiotic Limosilactobacillus reuteri DSM 17938 Alleviates Acute Liver Injury by Activating the AMPK Signaling via Gut Microbiota–Derived Propionate

Zhao Yuting,Xie Weiqi,Duan Jingyi,Li Fei

Limosilactobacillus reuteri DSM 17938 ( L. reuteri DSM 17938) was one of the most widely used probiotics in humans for gastrointestinal disorders, but few studies have investigated its role in drug-induced liver injury (DILI). Here, we evaluated the efficacy of L. reuteri DSM 17938 using a mouse model of DILI induced by triptolide. Pregavage of L. reuteri DSM 17938 for 1 week remarkably lowered hepatic inflammatory cytokines level and oxidative stress, with diminished serum alanine transaminase and aspartate aminotransferase levels. Metabolomics and RT-qPCR analysis confirmed its ability in ameliorating TP-disrupted hepatic fatty acid β oxidation. Genome annotation of L. reuteri showed its ability to modulate energy metabolism. Targeted metabolomics demonstrated that L. reuteri DSM 17938 modified the short fatty acid profiles in cecum, especially enhancing propionate levels. Further experiments found that L. reuteri DSM 17938 can activate AMPK signaling by upregulating gut microbiota–derived propionate level, thus restoring impaired mitochondrial biogenesis and energy supply processes to recover energy homeostasis, which leads to diminished ROS production and oxidative stress injury in hepatocytes. Besides, AMPK inhibitor dorsomorphin abolished all the effects on propionate protecting mitochondria and energy metabolism. This study established probiotic therapy of L. reuteri DSM 17938 as a preventive intervention for DILI in clinical. We also revealed that L. reuteri DSM 17938 can activate AMPK signaling by propionate, facilitating a deeper understanding of the action mechanism of L. reuteri DSM 17938 against acute liver injury and contributing to the development of its postbiotics and wider applications. Graphical Mechanism illustration of Limosilactobacillus reuteri DSM 17938 alleviating TP-induced liver injury by enhancing AMPK signaling pathway via propionate (Created with Biorender. com).

Promoting biomethane production from propionate with Fe2O3@carbon nanotubes composites

Zhi-Man Yang,Rong-Bo Guo,Xiao-Huan Dong

Using a batch anaerobic system constructed with 60 mL serum bottles, potential of a composite material with Fe 2 O 3 nanoparticles decorated on carbon nanotubes (CNTs) to enhance biomethane production was investigated. The composites (Fe 2 O 3 @CNTs) with well dispersed Fe 2 O 3 nanoparticles (4.5 nm) were fabricated by a facile thermal decomposition method in a muffle furnace under nitrogen atmosphere. Compared with Fe 2 O 3 , Fe 2 O 3 @CNTs showed a large specific surface area and good electrical conductivity. Supplementation of Fe 2 O 3 @CNTs to the propionate-degrading enrichments enhanced the methane production rate, which was 10.4-fold higher than that in the control experiment without material addition. The addition of Fe 2 O 3 @CNTs also not only showed a clearly electrochemical response to flavin and cytochrome C, but also reduced the electron transfer resistance when compared to the control. Comparative analysis showed that Fe 2 O 3 in Fe 2 O 3 @CNTs played a key role in initiating electrochemical response and triggering rapid methane production, while CNTs functioned as rapid electron conduits to facilitate electron transfer from iron-reducing bacteria (e.g., Acinetobacter , Syntrophomonas , and Geobacter ) to methanogens (e.g. Methanosarcina ).

Metabolic rescue of α-synuclein-induced neurodegeneration through propionate supplementation and intestine-neuron signaling in C. elegans

Chenyin Wang,Meigui Yang,Dongyao Liu,Chaogu Zheng

Summary Microbial metabolites that can modulate neurodegeneration are promising therapeutic targets. Here, we found that the short-chain fatty acid propionate protects against α-synuclein-induced neuronal death and locomotion defects in a Caenorhabditis elegans model of Parkinson’s disease (PD) through bidirectional regulation between the intestine and neurons. Both depletion of dietary vitamin B 12 , which induces propionate breakdown, and propionate supplementation suppress neurodegeneration and reverse PD-associated transcriptomic aberrations. Neuronal α-synuclein aggregation induces intestinal mitochondrial unfolded protein response (mitoUPR), which leads to reduced propionate levels that trigger transcriptional reprogramming in the intestine and cause defects in energy production. Weakened intestinal metabolism exacerbates neurodegeneration through interorgan signaling. Genetically enhancing propionate production or overexpressing metabolic regulators downstream of propionate in the intestine rescues neurodegeneration, which then relieves mitoUPR. Importantly, propionate supplementation suppresses neurodegeneration without reducing α-synuclein aggregation, demonstrating metabolic rescue of neuronal proteotoxicity downstream of protein aggregates. Our study highlights the involvement of small metabolites in the gut-brain interaction in neurodegenerative diseases.

Relieving ammonia inhibition by zero-valent iron (ZVI) dosing to enhance methanogenesis in the high solid anaerobic digestion of swine manure

Xiaoshan Meng,Qianwen Sui,Jibao Liu,Dawei Yu,Yawei Wang,Yuansong Wei

Relieving from ammonia inhibition and enhancing the utilization of thermodynamically unfavorable propionate are crucial for methane harvest in the high solid anaerobic digestion (HSAD) of ammonia-rich swine manure. In this study, the potential of dosing zero-valent iron (ZVI, 150 um) for enhancing the methanogenesis to resist total ammonia (TAN) over 5.0 g-N·L −1 was investigated via batch experiments under mesophilic condition. The cumulative methane production was enhanced by 22.2% at ≥160 mM ZVI dosage and the HSAD duration was further shortened by 50.6% at ≥320 mM ZVI dosage. The enhanced methanogenesis was mainly resulted from the full utilization of propionate and the accelerated collapse of posterior-biodegradable organics which might be driven by ZVI. Results of microbial community and qPCR ( mcrA ) showed that ZVI might trigger the blooming of Methanosarcina (from 27.9% to 78.3%) and Syntrophomonas (0.5% to 3.7%) and attribute to their possible direct interspecies electron transfer (DIET) to enhance propionate utilization. Besides, the main methanogenesis might remain in the effective aceticlastic pathway even under free ammonia (FAN) almost 1.0 g-N·L −1 because syntrophic acetate oxidizing bacteria ( SAOB ) decreased to almost none at 320 mM ZVI dosage. Dosing ZVI could relieve HSAD from TAN inhibition and more dosage was required to resist FAN inhibition.

Bifidobacterium longum relieves constipation by regulating the intestinal barrier of mice

Linlin Wang,Mao Chai,Jialiang Wang,Qiangqing YU,Gang Wang,Hao Zhang,Jianxin Zhao,Wei Chen

Constipation is a major health concern worldwide, requiring effective and safe treatment options. This study mainly focused on three species and nine strains of bifidobacteria from different sources to study their abilities to relieve constipation induced by loperamide in BALB/C mice. By monitoring constipation-related indicators, it was found that only Bifidobacterium longum (B. longum) relieved constipation, which indicated that bifidobacteria had inter-species differences in relieving constipation. Furthermore, through the detection of biological, chemical, mechanical, and immune barriers in mice, it was discovered that B. longum upregulates the relative abundance of 22 genera that were positively related to faecal water content, small intestinal propulsion rate, acetate, propionate, and intestinal mechanical barrier and negatively correlated with inflammatory factors, AQP8 and the time of first black stool and downregulates the relative abundance of Akkermansia. Furthermore, it increased the level of acetate in faeces and reduced the expression of AQP8 in the colon. This enhances intestinal motility and improves water and electrolyte metabolism. Meanwhile, it inhibited inflammation and prevented loperamide-induced intestinal barrier damage in constipated mice by upregulating occludin and downregulating IL-1β and TNF-α. In summary, B. longum relieved constipation by regulating the intestinal barrier in constipated mice.

Preconception maternal gut dysbiosis affects enteric nervous system development and disease susceptibility in offspring via the GPR41–GDNF/RET/SOX10 signaling pathway

Cunzheng Zhang,Yuzhu Chen,Ruqiao Duan,Yiming Zhang,Haonan Zheng,Jindong Zhang,Tao Zhang,Jingxian Xu,Kailong Li,Fei Pei,Liping Duan

Maternal health, specifically changes in the gut microbiota, can profoundly impact offspring health; however, our understanding of how gut microbiota alterations during the preconception period influence the offspring remains limited. In this study, we investigated the impact and mechanisms of preconception maternal gut dysbiosis on the development of the enteric nervous system (ENS) in mice. We found that preconception maternal exposure to antibiotics led to the abnormal development of the ENS in offspring, increasing their susceptibility to water avoidance stress at the adult stage. Metagenomic, targeted metabolomic, and transcriptomic analyses revealed that preconception antibiotic exposure disrupted the expression of genes crucial for embryonic ENS development by altering maternal gut microbiota composition. Multi-omics analysis combined with Limosilactobacillus reuteri and propionate gestational supplementation demonstrated that the maternal gut microbiota and metabolites may influence embryonic ENS development via the GPR41–GDNF/RET/SOX10 signaling pathway. Our findings highlight the critical importance of maintaining a healthy maternal gut microbiota before conception to support normal ENS development in offspring. Graphical Preconception administration of antibiotics to female mice reduces the abundance of Limosilactobacillus reuteri in the maternal gut microbiota during pregnancy, subsequently affecting propionate levels. Decreased propionate levels downregulated the expression of Gdnf / Ret / Sox10 mediated by GPR41, leading to abnormal development of the enteric nervous system during the embryonic period. This dysplasia results in colonic dysmotility, impaired colonic epithelium, and increased susceptibility to water-avoidance stress in offspring. Highlights Preconception maternal antibiotic exposure results in the abnormal development of the enteric nervous system (ENS) and intestinal mucosa in mouse offspring, thereby increasing their susceptibility to water avoidance stress in adulthood. Preconception maternal antibiotic exposure alters the composition of the maternal gut microbiota, leading to abnormal embryonic ENS development via the modulation of gene expression. Multi-omics analysis and supplementation with Limosilactobacillus reuteri and propionate during pregnancy indicate that the maternal gut microbiota and metabolites affect embryonic ENS development through the GPR41–GDNF/RET/SOX10 signaling pathway.

High-Yield Production of Propionate from 1,2-Propanediol by Engineered Pseudomonas putida KT2440, a Robust Strain with Highly Oxidative Capacity

Ya’nan Shi,Rongshan Li,Jie Zheng,Yubin Xue,Yong Tao,Bo Yu

Bio-based propionate attracts increasing attention owing to its green nature and specific food additive market. To date, the time-consuming and costly fermentation process by strict anaerobes makes propionate production not ideal. In this study, we designed a new route for propionate production, in which 1,2-propanediol was first dehydrated to propionaldehyde and then to propionate by taking advantage of the robust oxidization capacity of the Pseudomonas putida KT2440 strain. The high atom economy (0.97 g/g) in this proposed pathway is more advantageous than the previous l-threonine-derived route (0.62 g/g). The molecular mechanism of the extraordinary oxidation capacity of P. putida KT2440 was first deciphered. The propionate production was realized in P. putida KT2440 by screening suitable glycerol dehydratases and optimizing the expression to eliminate the formation of 1-propanol and the accumulation of the intermediate propionaldehyde. The engineered strain produced propionate with a molar conversion rate of >99% from 1,2-propanediol. A high titer of 46.5 g/L pure propionic acid with a productivity of 1.55 g/L/h and a mass yield of 0.96 g/g was achieved in fed-batch biotransformation. Thus, this study provides another idea for the production of high-purity bio-based propionate from renewable materials with high atom economy.
