We investigated the effects of dietary supplementation with sodium propionate (SP) and tributyrin (TB) on hepatic lipid deposition and antioxidant capacity of spotted seabass (Lateolabrax maculatus) via an 8-week feeding experiment and a hypoxia stress experiment. The fish were fed five experimental diets: a control diet (CON), a diet supplemented with 2 g/kg SP (SP-0.2%), 4 g/kg SP (SP-0.4%), 2 g/kg TB (TB-0.2%), or 4 g/kg TB (TB-0.4%). No significant difference in growth performance was presented among the groups (P > 0.05). The SP-0.4% and TB-0.2% groups presented significantly lower hepatosomatic and viscerasomatic indexes compared with the CON group. Then, the SP-0.4% and TB-0.2% groups presented stronger resistance to hypoxic stress than the other groups and were analyzed further. The hepatic histology and triglyceride levels revealed that SP-0.4% and TB-0.2% reduced hepatic lipid deposition. Similarly, the downregulation of malondialdehyde and the upregulation of total antioxidant capacity, superoxide dismutase, and catalase activities and the related gene expression levels revealed that SP-0.4% and TB-0.2% improved the antioxidant capacity. Additionally, the RNA sequencing demonstrated that SP-0.4% and TB-0.2% regulated gene expression to a similar extent. Among the 117 differentially expressed genes, 67 genes were enriched in the same pattern, and involved the FoxO signaling, PI3K-Akt signaling, and insulin-related pathways. In conclusion, supplementing SP-0.4% and TB-0.2% as feed additives effectively improved hepatic lipid metabolism, antioxidant capacity, and hypoxic stress resistance of spotted seabass.

Short-chain fatty acids (SCFAs) produced by the colonic bacterial fermentation of dietary fiber contribute a significant proportion of daily energy requirement. Furthermore, these compounds are modulators of macrophage function and potential targets for the development of new drugs. The aims of this study were to evaluate the effects of three types of SCFAs (sodium acetate (NaAc), sodium propionate (NaP), and sodium butyrate (NaB)) on the production of NO and inducible nitric oxide synthase (iNOS) and proinflammatory and antiinflammatory cytokines (tumor necrosis factor-α (TNF-α) and interleukin (IL-1, IL-6, and IL-10)) and to observe the effect of NaAc on inhibiting lipopolysaccharide (LPS)-induced NF-κB activation in LPS-stimulated RAW264.7 cells. The results show that three types of SCFAs (acetate, propionate, and butyrate) reduced the production of proinflammatory factors, including TNF-α, IL-1β, IL-6, and NO, and inhibited the vitality of iNOS. Meanwhile, SCFAs enhanced the production of antiinflammatory cytokine IL-10 in lower concentrations (1–1,200 μmol/L). Like NaB, NaAC inhibited LPS-induced NF-κB activation. These results may hold promise on the role that SCFAs have on the prevention and treatment of various inflammatory conditions.

AimsThe aims were to examine whether oral sodium propionate supplementation regulate lipid metabolism through modulating gut microbiota. Methods and ResultsICR male mice (26·98 ± 0·30 g) were randomly assigned to three groups (n = 10) and fed control diet (Con), high‐fat diet (HFD) and HFD plus propionate (Pro) respectively. In this study, we found that HFD increased the weight of final body, inguinal white adipose tissues (iWAT), epididymal white adipose tissue (eWAT) and perirenal white adipose tissue (pWAT), as well as the adipocyte mean area of iWAT and eWAT in mice (P < 0·05), whereas sodium propionate treatment reduced the weight of iWAT and pWAT as well as adipocyte mean area of iWAT in mice fed a HFD (P < 0·05). Moreover, in the iWAT, the mRNA expression of lipogenesis genes, including peroxisome proliferator activated receptor γ, acetyl‐CoA carboxylase and carnitine palmitoyl transferase‐1β, was upregulated by HFD challenge (P < 0·05), and the elevation of these genes was nearly reversed to the level of control diet‐fed mice by sodium propionate treatment. Meanwhile, sodium propionate treatment increased the hormone‐sensitive lipase mRNA expression in the iWAT of HFD‐fed mice (P < 0·05). High‐throughput pyrosequencing of the 16S rRNA demonstrated that sodium propionate treatment significantly recovered the gut microbiota dysbiosis in HFD‐fed mice, including the richness and diversity of microbiota and the ratio of Firmicutes to Bacteroidetes. Furthermore, the HFD‐induced reductions in colonic levels of butyrate and valerate were reversed by sodium propionate treatment, which also normalized the serum LPS level seen in HFD‐fed mice to the levels of the control diet‐fed mice.ConclusionsCollectively, these results indicated that sodium propionate treatment could improve lipid metabolism in HFD‐fed mice, and the potential mechanisms might be via regulating gut microbiota.Significance and Impact of the StudyWe demonstrated for the first time that oral sodium propionate significantly improved HFD‐induced dysbiosis of gut microbiota, indicating that the mitigative effect of propionate for HFD‐induced lipid dysmetabolism might be mediated by gut microbiota in mice.

High-fat diet (HFD) promotes adipose tissue senescence, which in turn disrupts insulin-mediated glycemic homeostasis. The underlying mechanisms remain unclear. Through clinical survey data, animal models, and primary adipose-derived mesenchymal stem cells (ADSC), we investigated how dietary patterns influence adipocyte senescence. We found that elevated fatty acid levels enhance the interaction between the E3 ubiquitin ligase TRIP12 and Cyclin-dependent kinase 4 (CDK4) in ADSCs, triggering CDK4 ubiquitination and degradation. As a process associated with this disruption in cell cycle progression, cellular senescence may represent a key outcome. Consequently, senescent ADSC-derived mature adipocytes (ADSC-MA) exhibit impaired insulin-stimulated GLUT4 membrane translocation and reduced glucose uptake. In contrast, within an HFD setting, dietary fiber supplementation is associated with the reversal of cellular senescence. The gut microbiota–short-chain fatty acids (SCFAs) axis may be involved in the restoration of cell cycle progression and the amelioration of ADSC senescence, correlating with a partial recovery of glucose uptake capacity in ADSC-MAs. Our study highlights potential strategies to reverse cellular senescence and identifies promising therapeutic targets for impaired glucose tolerance.

The utilization of waste-derived volatile fatty acids (VFAs) as carbon sources for polyhydroxyalkanoate (PHAs) biosynthesis offers a promising route toward sustainable bioplastics. However, reliance on single VFAs often limits product diversity and structural tunability. In this study, we developed and optimized a co-feeding strategy using a ternary mixture of formate, acetate, and propionate in Cupriavidus necator to enable tailored PHAs production. Using response surface methodology for multi-objective optimization, a VFA ratio (formate:acetate:propionate = 0:0.38:0.62) was identified that balances PHA titer (1.33 ± 0.13 g/L) with a 3-hydroxyvalerate (3-HV) content of 62.02% (in mol). Formate exerted no beneficial effect on the biosynthesis of PHA or 3-HV under mixed substrate conditions. Fermentation kinetic analysis further revealed that acetate primarily promoted cell growth and PHAs backbone synthesis, whereas propionate enhanced 3-HV formation while competitively inhibiting acetate uptake. The resulting copolymer exhibited improved thermal processability, as evidenced by a lower glass transition temperature (46.81 °C ± 0.01) and a reduced primary melting temperature (81.05 °C ± 0.14). Collectively, this work provides a viable strategy for producing tunable bioplastics from renewable mixed VFAs, advancing circular bio-manufacturing.

Objective: This study aimed to investigate the therapeutic effects of sodium propionate in combination with Sishen Pill in the treatment of diarrhea with kidney-yang deficiency syndrome in mice, with a focus on its influence on intestinal microbiota, enzyme activity, and associated therapeutic outcomes. Methods: A model of diarrhea with kidney-yang deficiency syndrome was established via adenine combined with Folium sennae administration. The model group was randomly assigned to the following treatment groups: natural recovery group, 100% Sishen Pill group, 75% Sishen Pill + 60 mg/kg sodium propionate group, 50% Sishen Pill + 120 mg/kg sodium propionate group, 25% Sishen Pill + 240 mg/kg sodium propionate group, and 480 mg/kg sodium propionate group. A variety of parameters, including general symptoms, body weight, rectal temperature, intestinal microbiota composition, microbial activity, and enzyme activity, were assessed. Results: Compared with natural recovery group, the 480 mg/kg sodium propionate group presented significant improvements in mental state, anal temperature, fecal water content, the thymus index, and Bifidobacterium counts (p < 0.01). Compared with those in normal group, the fecal water content and Escherichia coli counts in the 100% Sishen Pill group were significantly different (p < 0.01), but no significant differences were observed compared with those in natural recovery group (p > 0.05). The 75% Sishen Pill + 60 mg/kg sodium propionate group showed improvements in mental state, food and water intake, body weight, rectal temperature, fecal water content, spleen, and thymus index, Bifidobacterium counts, total bacterial count, E. coli count, microbial activity, and lactase activity, which were close to normal levels, and significant differences were observed when compared to the natural recovery group (p < 0.01). The 50% Sishen Pill + 120 mg/kg sodium propionate group and 25% Sishen Pill + 240 mg/kg sodium propionate group also exhibited significant differences in mental status, microbial activity, and Lactobacillus count relative to those of normal group (p < 0.01). Furthermore, the 50% Sishen Pill + 120 mg/kg sodium propionate group presented significant changes in fecal water content (p < 0.01), whereas 25% Sishen Pill + 240 mg/kg sodium propionate group presented significant differences in the spleen index, total bacterial count, E. coli count, protease activity, lactase activity, and xylanase activity compared with those of normal group (p < 0.01).Conclusion: The 75% Sishen Pill + 60 mg/kg sodium propionate can improve the symptoms of kidney-yang deficiency syndrome, promote the growth and development of mice, inhibit excessive bacterial proliferation, support the growth of beneficial bacteria, and enhance intestinal enzyme activity. Its effects are superior to the use of sodium propionate or Sishen Pill alone. These results suggest that this therapeutic ratio may optimize the efficacy of Sishen Pill in the treatment of diarrhea with kidney-yang deficiency syndrome. However, further research is necessary to confirm whether this combination represents the most effective treatment regimen for this condition in mice.

Propionate accumulation is common in anaerobic digestion systems, while Fe 3 O 4 nanoparticles (Fe 3 O 4 NPs) show potential in steering direct interspecies electron transfer (DIET) to facilitate methanogenesis from propionate. However, the effect of Fe 3 O 4 NPs in stable microbial communities remains unclear. This study demonstrated that 1.0 g/L Fe 3 O 4 NPs enhanced propionate degradation and methane production by 220 % and 55 % of the propionate-acclimated microbial consortia post-shock loading, as evidenced by batch-test. High propionate concentrations suppressed Geobacter , yet Fe 3 O 4 NPs enabled electron syntrophy between alternative DIET-participants ( Arcobacter , Syntrophobacter ) and methanogens ( Methanothrix , Methanobacterium ) serving as electron conduits. Network analysis further revealed that Fe 3 O 4 NPs activated interactions among potential electroactive microbes, highlighting the potentially ubiquitous presence of electron syntrophy. Even in propionate-stressed microbial communities, such mutualism may be rapidly activated by Fe 3 O 4 NPs, offering a practical solution for mitigating propionate accumulation and enhance digester performance.

At the present stage, the incorporation of modified biochar materials into anaerobic digestion (AD) systems has emerged as a pivotal strategy for the treatment of high-concentration organic wastewater and the promotion of methane production. This study proposes a novel methodology for a simple one-pot method, utilizing deep eutectic solvents (DES, urea/FeCl₃) to modify the preparation of nitrogen/iron co-doped straw biochar (DES BC), and it was applied for the first time with the objective of enhancing methane production in brewery wastewater AD. The incorporation of 1 g/L DES 1:1, 1:2, 1:3 BC into mesophilic AD systems resulted in increase the maximum methane production by a factor of 1.31, 2.84, and 1.50, respectively, in comparison with the control group that without DES BC. The enhancement mechanisms are attributed to three key aspects: (1) DES BC accelerated substrate conversion, as evidenced by rapid volatile fatty acid (VFA) consumption and microbial activity promotion, facilitated by its high specific surface area and pore volume providing attachment sites for functional microorganisms (e.g., Georgenia and Anaerolinea ). (2) the redox-active functional groups (─ OH, Fe O, N O) on DES BC enhanced electron exchange capacity (EEC), electron donating capacity (EDC), and electron accepting capacity (EAC), directly improving electron transfer efficiency, and (3) DES BC potentially promoted direct interspecies electron transfer (DIET) between syntrophic bacteria (e.g., Geobacteraceae , Desulfomicrobium ) and methanogenic archaea (e.g., Methanobacterium , Methanothrix ), optimizing methane yield without solely relying on microbial abundance. Metagenomic analysis further confirmed that DES BC increased the relative abundances of key enzymes in hydrogenotrophic and acetate decarboxylation methanogenesis pathways. This work provides innovative strategies for modifying biochar to advance brewery wastewater treatment and energy recovery.

Liver fibrosis remains a critical health concern with limited therapeutic options. Kaempferol (Kae) is a natural flavonoid widely present in natural plants, yet its role in modulating gut–liver axis interactions during fibrosis is unexplored. This study investigates the hepatoprotective effects of Kae on alleviating carbon tetrachloride (CCl4)-induced liver fibrosis, and its underlying mechanisms, focusing on oxidative stress, gut microbiota, and short-chain fatty acids (SCFAs), are revealed. A mouse model of hepatic fibrosis was built by the subcutaneous injection of CCl4. Meanwhile, Kae was administered by gavage at doses of 25, 50, and 100 mg/kg body weight. Serum biomarkers, liver histopathology, oxidative damage markers, and nuclear factor erythroid 2-related factor 2 (Nrf2)/kelch-like ECH-associated protein 1 (Keap1)/heme oxygenase 1 (HO-1) signaling were analyzed. AML12 hepatocytes were pretreated with Kae or SCFAs (acetate, propionate, butyrate) before H2O2-induced oxidative injury. The changes in gut microbiota and the levels of SCFAs were assessed via 16S rRNA sequencing and GC-MS, respectively. Kae effectively alleviated the destruction of the liver morphology and tissue structure, reduced the infiltration of inflammatory cells, collagen deposition in the liver, and the expression of fibrotic factors, and downregulated the oxidative stress level in the liver of mice with liver fibrosis by activating the Nrf2/Keap1/HO-1 pathway (p< 0.05 or 0.01). In vitro, Kae significantly mitigated H2O2-induced cytotoxicity and oxidative damage (p< 0.05 or 0.01). Furthermore, Kae restored gut microbiota diversity, increased beneficial genera (e.g.,Lactobacillus), and elevated both intestinal and hepatic SCFA levels (p< 0.01). The discrepant SCFA pretreatment similarly protected AML12 cells by activating Nrf2 signaling (p< 0.05 or 0.01). Our research suggests that Kae could inhibit CCl4-induced liver fibrosis by restoring the levels of intestinal metabolite SCFAs to reduce oxidative damage.

Gestational diabetes during pregnancy is associated with an increased risk of developmental abnormalities in offspring, but the underlying mechanisms remain unclear. It is not known how maternal metabolism and gut microbes influence kidney development in the fetus. Here we show that gestational diabetes alters maternal gut microbiota and reduces the production of key fatty acids that normally support kidney development in offspring. We find that these changes impair a molecular pathway involving the receptor GPR43 , which promotes the growth and migration of kidney cells. In a mouse model, restoring short-chain fatty acids or transferring gut bacteria from healthy donors improves kidney development in offspring, while blocking GPR43 reverses this effect. This study reveals a previously unknown link between maternal gut metabolism and fetal kidney formation and may guide future strategies to prevent congenital kidney disorders in children born to mothers with gestational diabetes.

Background: Ferroptosis has been proven to contribute to the progression of myocardial ischemia/reperfusion (I/R) injury and can be inhibited or promoted by ATF3. Short-chain fatty acids (SCFAs) have shown benefits in various cardiovascular diseases with anti-inflammatory and antioxidant effects. However, the impact of SCFAs on ferroptosis in ischemic-stimulated cardiomyocytes remains unknown. This study aimed to investigate the effect of SCFAs on cardiomyocyte ferroptosis, the expression of ATF3, and its potential upstream regulators. Methods and results: The expression of ATF3, ferroptosis pathway geneset (FPG), and geneset of potential regulators for ATF3 (GPRA, predicted by the PROMO database) was explored in the public human myocardial infarction single-cell RNA-seq (sma) dataset. Cardiomyocyte data was extracted from the dataset and re-clustered to explore the FPG, ATF3, and GPRA expression patterns in cardiomyocyte subclusters. A dose-dependent toxic experiment was run to detect the suitable dose for SCFA treatment. The erastin-induced ferroptosis model and hypoxia-reoxygenation (H/R) model (10 h of hypoxia followed by 6 h of reoxygenation) were adopted to assess the effect of SCFAs via the CCK8 assay. Gene expression was examined via RT-PCR and western blot. Ferroptosis markers, including lipid peroxides and Fe2+, were detected using the liperfluo and ferroOrange probes, respectively. In the sma dataset, upregulated ferroptosis pathway genes were mainly found in the infarction-stimulated cardiac cells (border zone and fibrotic zone), particularly the cardiomyocytes and adipocytes. The ATF3 and some of its potential transcription factors (VDR, EGR3, PAX5, and SP1) can be regulated by SCFA. SCFA can attenuate erastin-induced lipid peroxidation in cardiomyocytes. SCFA treatment can also reverse erastin-induced Fe2+ increase but may strengthen the Fe2+ in the H/R model. We also precisely defined a ferroptosis subcluster of cardiomyocytes (CM09) that highly expressed FPG, ATF3, and GPRA.Conclusion: The ATF3 and the ferroptosis pathway are elevated in cardiomyocytes of injury-related cardiac regions (border zone, ischemic zone, and fibrotic zone). SCFA can attenuate cardiomyocyte ferroptosis and regulate the expression of ATF3. Our study offers novel insights into the potential targets of SCFAs in the cardiovascular system.

Objective To investigate the potential mechanisms underlying the combined therapeutic effects of sodium propionate and Sishen Pill and to provide experimental evidence supporting their mode of action. Methods The method utilized involved the induction of a mouse model of Diarrhea with Kidney-Yang Deficiency Syndrome by administering adenine combined with Folium sennae. After the successful establishment of the model, the mice in the model group were randomly assigned to one of the following treatment groups: natural recovery group, sodium propionate group, 75% Sishen Pill combined with 60 mg/kg sodium propionate group, or Sishen Pill group. Results The 75% Sishen Pill combined with 60 mg/kg sodium propionate demonstrated significantly better therapeutic effects compared to the Sishen Pill alone and sodium propionate alone groups. The combined treatment significantly improved the behavioral indices of mice (p < 0.05); increased the levels of MUC2 and sIgA (p < 0.01); reduced IL-6 levels (p < 0.05); and improved structural damage in the kidneys and small intestines. Intestinal microbiota analysis showed that 75% Sishen Pill combined with 60 mg/kg sodium propionate significantly increased beneficial bacteria such as Lactobacillus (p < 0.05), while Prevotellamassilia and Maribacter were significantly enriched in this group. Correlation analysis revealed that Lactobacillus and Pediococcus were positively correlated with MUC2 and sIgA, while negatively correlated with IL-6.Conclusion The 75% Sishen Pill combined with 60 mg/kg sodium propionate significantly alleviates symptoms related to Diarrhea with Kidney-Yang Deficiency Syndrome, enhances the efficacy of Sishen Pill by regulating the intestinal microbiota, boosts intestinal immune function, and reduces intestinal inflammation, providing a new approach for treating Diarrhea with Kidney-Yang Deficiency Syndrome.
