Rhamnogalacturonan I (RG-I) pectin are regarded as strong galectin-3 (Gal-3) antagonist because of galactan sidechains. The present study focused on discussing the effects of more structural regions in pectin on the anti-Gal-3 activity. The water-soluble pectin (WSP) recovered from citrus canning processing water was categorized as RG-I pectin. The controlled enzymatic hydrolysis was employed to sequentially remove the α-1,5-arabinan, homogalaturonan and β-1,4-galactan in WSP. The Gal-3-binding affinity KD (kd/ka) of WSP and debranched pectins were calculated to be 0.32 μM, 0.48 μM, 0.56 μM and 1.93 μM. Moreover, based on the more sensitive cell line (MCF-7) model, the IC 30 value of WSP was lower than these of modified pectins, indicating decreased anti-Gal-3 activity. Our results suggested that the total amount of neutral sugar sidechains, the length of arabinan and cooperation between HG and RG-I played important roles in the anti-Gal-3 activity of WSP, not the Gal/Ara ratio or RG-I/HG ratio. These results provided a new insight into structure-activity relationship of citrus segment membrane RG-I as a galectin-3 antagonist and a new functional food.

BACKGROUND Tea dregs, typically generated during the production of instant tea or tea beverages, have conventionally been regarded as waste material and routinely discarded. Nevertheless, contemporary research endeavors are concentrating on discovering efficient methods for utilizing the potential of this discarded resource. RESULTS In this study, we employed a sequential extraction method using chemical chelating agents to extract and isolate four distinct cell wall polysaccharides, designated as CWTPS-1 through CWTPS-4, from the tea dregs of Liubao brick tea. A comprehensive investigation into their physicochemical, structural, and hypoglycemic properties was conducted. The analysis of chemical composition and physicochemical characteristics revealed that all four CWTPSs were characterized as acidic polysaccharides, albeit with varying chemical compositions and physicochemical attributes. Specifically, the xyloglucan fractions, CWTPS-3 and CWTPS-4, were found to be rich in glucose and xylose, displaying a more uniform molecular weight distribution, greater structural stability, and a more irregular surface compared to the others. Moreover, they exhibited a higher diversity of monosaccharide residues. Importantly, our research unveiled that all four CWTPSs exhibited the capacity to modulate key glucose-regulated and antioxidant enzyme activities within HepG2 cells via the IRS-1-PI3K/Akt signaling pathway, thereby ameliorating cellular insulin resistance. Furthermore, our correlation analysis highlighted significant associations between monosaccharide composition and neutral sugar content with the observed hypoglycemic activity of CWTPSs. CONCLUSIONS This study highlights the potential of utilizing tea dregs as a valuable resource, making a significant contribution to the advancement of the tea industry. Furthermore, CWTPS-4 exhibits promising prospects for further development as a functional food ingredient or additive. This article is protected by copyright. All rights reserved.

An Antarctic yeast was cultivated to produce and enzymatic extract with polygalacturonase activity, whose biochemical properties were studied. It assisted pectin extraction from lime pomace at 20 °C. The extract produced by Tausonia pullulans 8E had an optimum temperature of 40 °C and optimum pH of 5.0. At 20 °C, it displayed 54% of relative activity. A good pH-stability and thermostability were observed. Hg +2 and Co +2 inhibited its activity in 40 and 11%, respectively. Pectin was obtained from lime pomace at 20 °C, with 15% of yield after 120 min extraction. Uronic acid (46%) and neutral sugars (53%) were determined. Pectin's molecular weight was estimated in the order of 100,000 g mol −1 . By anion-exchange chromatography, pectin-linked and free neutral sugars were observed. The high-methoxyl pectin was used to prepare low-calorie gels. It was demonstrated that this cold-active enzyme allows enzymatic-assisted pectin extraction at 20 °C.

In this work, a novel core–shell structured gold@graphitized mesoporous silica nanocomposite (Au@GMSN) was synthesized by in situ graphitization of template within the mesochannels of mesoporous silica shell on gold core and demonstrated to be promising nanomaterials for surface-assisted laser desorption/ionization time-of-flight mass spectroscopy (SALDI-TOF MS). The integration of the graphitized mesoporous silica with the gold nanoparticles endowed Au@GMSN with large surface areas of graphitic structure, good dispersibility, and strong ultraviolet (UV) absorption. Au@GMSN exerted the synergistic effect on the efficient detection of small-molecular-weight analytes including amino acids, neutral saccharides, peptides, and traditional Chinese medicine. The Au@GMSN-assisted laser desorption/ionization exhibited the following superiorities: high ionization efficiency, low fragmentation interference, favorable salt tolerance, and good reproducibility. Moreover, because of the large hydrophobic inner surface area of the graphitized mesoporous silica shell, the Au@GMSN demonstrated its promising capacity in the pre-enrichment of aromatic analytes prior to SALDI-TOF MS, which favored rapid and sensitive detection.
