Remodeling of Gut Microbiota by Probiotics Alleviated Heat Stroke-Induced Necroptosis in Male Germ Cells

Hui Cai,Dezhe Qin,Yundie Liu,Xinrui Guo,Yang Liu,Baohua Ma,Jinlian Hua,Sha Peng

Scope Systemic heat stress (or heatstroke; HS) induces germ cell death and spermatogenesis disorders in men and male mammals. Also, it affects the immune environment of the circulatory system promoting gut inflammation and intestinal permeability, leading to pathogenic infection. In this study, the crosstalk between the gut and testis (gut-testis axis) under HS is explored, by examining the effects of intestinal immune status on the health of the male reproductive system in mice. Methods and results A mouse model of systemic heat stress is established to investigate the effect of probiotics on testis health. The results reveal that pro-inflammatory factor receptor activation pathway and pathogen infection response pathway are significantly upregulated in HS testes, leading to necroptosis, while pro-inflammatory factor and endotoxin are detected locally in the intestine and then entered the blood. The study then uses probiotics to intervene in gut microbiota, which results in milder gut microbial changes, lower inflammatory responses in the HS gut, and less necroptosis in the HS testes. Conclusion Probiotics-based remodeling of gut microbiota (GM) reduces the proliferation of abnormal bacteria and decreases the spread of gut-derived inflammatory mediators into the blood circulation under long-term systemic heat stress, which relieves inflammation on germ cells.

A Sensitive Fluorescent Probe with Large Stokes Shift for Real-Time Tracking Lysosomal pH Changes in Live Cells

Siyuan Li,Xianrong Yu,Linlin Zeng,Yuhan Xu,Xiaolan Zhao,Wei Tang,Xinrui Duan

Graphical A lysosomal-targeted ratiometric ICT fluorescent probe TCF-LysopH with ideal p K a value (6.12), large Stokes shift (110 nm), and good photostability was developed for real-time tracking lysosomal pH changes during heatstroke. The lysosome is the typical acidic organelle involved in many biological processes, the fluctuation of its pH influences its normal digestive function and even the fate of the cells. To conveniently detect and visualize lysosomal pH changes during heatstroke, we designed and synthesized a lysosomal-targeted ratiometric pH-sensitive fluorescent probe TCF-LysopH. TCF-LysopH showed an ideal p K a value (p K a =6.12) in the lysosomal pH range during heatstroke (5.00–7.00), rapid response for the changes of pH in real-time, reversible pH switching ability, good selectivity, low cytotoxicity, and excellent lysosome-targeting ability. The large Stokes shift of 110 nm allows us to apply the probe for fluorescence imaging in MCF-7 cells. The probe has much better photostability (only less than 20 % decrease in intensity for 15 min irradiation) than LysoTracker Green. The fluorescence intensity became higher with the increase of pH, which is consistent with the tendency of the lysosomal pH change during heatstroke. Importantly, TCF-LysopH was applied to measure the change of lysosomal pH during heatstroke in live cells, the results prove once again that the pH of the lysosome is increased during heatstroke. Thus, TCF-LysopH was successfully developed as a practical and useful tool to provide a better understanding of the complicated mechanisms between lysosome pH change and heatstroke.

Quantitative Proteomics Provided Insights into the Protective Effects of Heat Acclimation on the Rat Hypothalamus after Exertional Heatstroke

Fan Xv,Li-zhen Ma,Xin Li,Jin-bao Zhao,Shu-yuan Liu,Han-ding Mao,Jun Ma,Ling Xing,Li-feng Wang,Wei-jia Zhi,Qing Song

Background: The effects of heat acclimation (HA) on the hypothalamus after exertional heatstroke (EHS) and the specific mechanism have not been fully elucidated, and this study aimed to address these questions. Methods: In the present study, rats were randomly assigned to the control, EHS, HA, or HA + EHS groups (n = 9). Hematoxylin and eosin (H&E) staining was used to examine pathology. Tandem mass tag (TMT)-based proteomic analysis was utilized to explore the impact of HA on the protein expression profile of the hypothalamus after EHS. Bioinformatics analysis was used to predict the functions of the differentially expressed proteins. The differential proteins were validated by western blotting. An enzyme-linked immunosorbent assay was used to measure the expression levels of inflammatory cytokines in the serum. Results: The H&E staining (n = 5) results revealed that there were less structural changes in hypothalamus in the HA + EHS group compared with the EHS group. Proteomic analysis (n = 4) revealed that proinflammatory proteins such as argininosuccinate synthetase (ASS1), high mobility group protein B2 (HMGB2) and vimentin were evidently downregulated in the HA + EHS group. The levels of interleukin (IL)-1β, IL-1, and IL-8 were decreased in the serum samples (n = 3) from HA + EHS rats. Conclusions: HA may alleviate hypothalamic damage caused by heat attack by inhibiting inflammatory activities, and ASS1, HMGB2 and vimentin could be candidate factors involved in the exact mechanism.

A dual-targeted organelles SO2 specific probe for bioimaging in related diseases and food analysis

Yongfei Huang,Yongbin Zhang,Fangjun Huo,Jianbin Chao,Caixia Yin

Sulfur dioxide (SO 2 ), as the main member of reactive sulfur species (RSS), it is mainly produced in mitochondria and involved in a variety of activities at the cellular level. Excessive inhalation of SO 2 could cause respiratory tract damage, cardiovascular disease and even cancer. The mitochondrial and lysosomal as two important organelles, their interactions (fusion and contact) are important for maintaining eukaryotic homeostasis. Some study indicated that SO 2 is active in the above interaction process. In this work, benzopyranium salt unit was constructed as fluorophore I, where O + was the mitochondrial target site, and naphthalimide was linked to morpholine to construct fluorophore II, where morpholine was the lysosomal target group. Studies have shown that the probe has a good performance in targeting both mitochondria and lysosomes. And after the reaction with SO 2 , FRET was broken, resulting in red quenching, and green emitting, which made it can detect SO 2 with a ratiometric response. Cells cultured with nystatin showed decreased red and green fluorescence increased, indicating that the autophagy of mitochondria produced SO 2 . In addition, synthesized control reagents (targeting lysosomes alone and responding to SO 2 ) from no signal to gradually enhanced blue fluorescence, indicating that SO 2 produced by mitochondria is transferred to lysosomes. What's more, it could monitor the changes of SO 2 concentration under heat stroke. More importantly, it could quantitatively detect sulfite concentration in Yuba and crystal sugar.
