Molecular imprinted polymers (MIPs) were prepared using rutin as the template, different reagents as the functional monomer and different reagents as the cross-linker by solution polymerization. Several parameters that would influence the performance of MIPs were investigated including the type of functional monomer (single or double) and cross-linker (single or double), and the molar ratio of the template, the functional monomer and the cross-linker. The optimum synthesis conditions of MIPs were found to be bi-monomers (acrylamide-co-2-vinyl pyridine, 3:1) and bi-crosslinker (ethylene glycol dimethacrylate-co-divinylbenzene, 3:1). The ratio of the template, the functional monomer and the cross-linker was found to be 1:6:20. MIPs synthesized under these conditions were filled into the cartridges as the adsorbents of solid-phase extraction (SPE). A competition test was conducted to authenticate the selectivity and the specificity of molecularly imprinted solid-phase extraction (MISPE) for rutin using the mixture solution of standard rutin and its structural analogs including quercetin, naringenin and kaempferol. Compared with purchased SPE including C 18 , silica and PCX, MISPE showed better selectivity and enrichment property for rutin in the extracted solutions of Chinese medicinal plants than any others. The mean recoveries were 85.93% (RSD: 3.04%, n = 3) for Saururus chinensis (Lour.) Bail and 88.61% (RSD: 3.36%, n = 3) for Flos Sophorae, respectively, which indicated that the optimized rutin -MIPs possess the value of practical application.

An ionic liquid-based microwave-assisted extraction (ILMAE) method has been developed for the effective extraction of rutin from Chinese medicinal plants including Saururus chinensis (Lour.) Bail. (S. chinensis) and Flos Sophorae. A series of 1-butyl-3-methylimidazolium ionic liquids with different anions were investigated. The results indicated that the characteristics of anions have remarkable effects on the extraction efficiency of rutin and among the investigated ionic liquids, 1-butyl-3-methylimidazolium bromide ([bmim]Br) aqueous solution was the best. In addition, the ILMAE procedures for the two kinds of medicinal herbs were also optimized by means of a series of single factor experiments and an L 9 (3 4 ) orthogonal design. Compared with the optimal ionic liquid-based heating extraction (ILHE), marinated extraction (ILME), ultrasonic-assisted extraction (ILUAE), the optimized approach of ILMAE gained higher extraction efficiency which is 4.879 mg/g in S. chinensis with RSD 1.33% and 171.82 mg/g in Flos Sophorae with RSD 1.47% within the shortest extraction time. Reversed phase high performance liquid chromatography (RP-HPLC) with ultraviolet detection was employed for the analysis of rutin in Chinese medicinal plants. Under the optimum conditions, the average recoveries of rutin from S. chinensis and Flos Sophorae were 101.23% and 99.62% with RSD lower than 3%, respectively. The developed approach is linear at concentrations from 42 to 252 mg L −1 of rutin solution, with the regression coefficient ( r ) at 0.99917. Moreover, the extraction mechanism of ILMAE and the microstructures and chemical structures of the two researched samples before and after extraction were also investigated. With the help of LC–MS, it was future demonstrated that the two researched herbs do contain active ingredient of rutin and ionic liquids would not influence the structure of rutin.

An ionic liquid-based microwave-assisted extraction (IL-MAE) technique has been successfully developed for the simultaneous effective extraction of quercetin and kaempferol from Toona sinensis and Rosa chinensis which are Chinese medicinal plants. Twenty different types of ionic liquids (ILs) with different cations and anions have been synthesized and investigated. Several significant operating parameters of IL-MAE were optimized by a series of single factor experiments and orthogonal design L9 (34) experiments, respectively. Compared with ionic liquid-based maceration extraction (IL-ME), ionic liquid-based heat extraction (IL-HE) and ionic liquid-based ultrasound-assisted extraction (IL-UAE), the optimized approach was demonstrated as the optimal technique with the highest extraction efficiency and the shortest extraction time. Reversed phase high performance liquid chromatography (RP-HPLC) with ultraviolet-detection (UV-DAD) was employed for the simultaneous analysis of kaempferol and quercetin. The results indicated that the extraction amounts of quercetin from Toona sinensis extracted using [Bmim]Br and from Rosa chinensis extracted using [Omim]Br were 176.5 mg g−1 with RSD 1.2% and 134.3 mg g−1 with RSD 0.63%. And the extraction amounts of kaempferol from Toona sinensis extracted using [Bmim]Br and from Rosa chinensis extracted using [Omim]Br were 16.12 mg g−1 with RSD 0.24% and 9.070 mg g−1 with RSD 0.15%. The developed approach for analyzing quercetin was linear between the concentrations of 32.10 mg L−1 to 233.3 mg L−1 in Toona sinensis and 25.70 mg L−1 to 160.5 mg L−1 in Rosa chinensis with the regression coefficients (R) of 0.9998 and 0.9999, respectively. And for kaempferol, the developed approach was linear between the concentrations of 0.73 mg L−1 to 23.20 mg L−1 in Toona sinensis and 0.19 mg L−1 to 15.00 mg L−1 in Rosa chinensis with the regression coefficients (R) of 0.9993 and 0.9996, respectively. The recoveries of quercetin extracted by the developed methods were 96.76–104.1% in Toona sinensis and 98.40–103.3% in Rosa chinensis, and the recoveries of kaempferol were 95.75–106.3% in Toona sinensis and 98.55–102.9% in Rosa chinensis with the RSDs of lower than 3.0%. The LODs were 1.87–2.26 μg mL−1. The microstructures and chemical structures of these two researched samples before and after different extraction procedures were also investigated.

The preparation of magnetic molecularly imprinted polymers (MMIPs) which can be used for the separation and purification of rutin from Chinese medicinal plants has been proposed. By applying the improved co-precipitation method, magnetic Fe3O4 particles were easily prepared, followed by the modification of TEOS and functionalization with –CHCH2. Using functionalized Fe3O4 particles as the magnetic cores, rutin as the template, and acrylamide as the functional monomer, MMIPs were synthesized by surface-imprinted polymerization under the protection of nitrogen gas and successive mechanical stirring at 60 °C for 24 h. Magnetic non-molecularly imprinted polymers (MNIPs) were also prepared with the same synthesis procedure as with MMIPs only without the presence of rutin. Magnetic particles were characterized by FT-IR, XRD, and TG analysis. And the selectivity of MMIPs was also investigated in detail. In addition, the performance of the MMIPs for the adsorption of rutin in the analysis of Chinese medicinal plants was assessed. The mean recoveries were 84.33% (RSD: 3.22%, n = 3) for Saururus chinensis (Lour.) Bail and 85.20% (RSD: 3.58%, n = 3) for Flos Sophorae, respectively, which showed that the prepared MMIPs with many advantages possess the value of practical application.

A new method has been developed for separation of chelerythrine and sanguinarine in medicinal plants used in traditional Chinese medicine (TCM). The separation is achieved by microchip electrophoresis (CE) using laser-induced fluorescence detection. The CE separation is achieved by using a hydro-organic medium as the electrolyte buffer. The experimental results are consistent with the prediction by theory in terms of resolution and migration speed because of the low Joule heat generated in microchip CE . In addition, formamide was found to have a potential for separation of molecules with similar chemical structures. Based on these findings, a run buffer containing 50% formamide was used to separate chelerythrine (CHE) and sanguinarine (SAN). The influencing factors, such as solvent of run buffer, pH of buffer, separation distance, and separation voltage, were optimized. Baseline separation of chelerythrine and sanguinarine was achieved within 120 s under an electrical voltage of 1.8 kV. Good linearity was observed in the concentration range of 0.15–550 μg mL −1 ( r =0.9993) for CHE and in the range of 0.3–600 μg mL −1 ( r =0.9998) for SAN. A low limit of detection (LOD) was achieved because of the high sensitivity achieved by laser-induced fluorescence detection (i.e. 5.0 ng mL −1 and 2.0 ng mL −1 for CHE and SAN, respectively). The contents of CHE are found to be 641.8±7.5 and 134.0±2.3 mg/kg in extracts of Macleaya cordata and Chelidonium majus , respectively, with good recovery of above 99%. The corresponding values for SAN found in these Chinese herbal extracts are 681.8±7.9 mg/kg and 890.5±8.9 mg/kg, respectively.
