Ethanol precipitation serves an important separation technology used for the manufacturing of more than 200 traditional Chinese medicines (TCM). In the present study, a membrane dispersion micromixer was employed for the ethanol precipitation of Codonopsis Radix concentrates to reduce the coating loss of active components. As membrane pore size decreased, mixing chamber width decreased, or mixing chamber depth decreased, lobetyolin recovery increased and dry matter removal decreased. Higher dry matter contents of concentrates (DM C ) or lower flow rates of two phases could lead to reduced lobetyolin recovery and increased dry matter removal. The mass ratio of ethanol solution to concentrate (ECR, g/g) showed little effect on lobetyolin recovery. Higher dry matter removal was obtained when ECR was higher. Higher continuous phase Reynolds number ( Re c ) or dispersed phase Reynolds number ( Re d ) resulted in higher lobetyolin recovery and lower dry matter removal. Lobetyolin recovery could be higher than 95% when using micromixing technology, which was about 10% higher than that of stirring. Membrane dispersion technology is promising to reduce the coating loss of active compounds in TCM ethanol precipitation.

Patulin (PAT) is a kind of mycotoxin which must be monitored for the sake of quality and safety in traditional Chinese medicine (TCM) owing to its harm to human health. On this account, a rationally designed ratiometric fluorescent aptasensor was developed based on the study of the interaction mechanism between PAT and its aptamer (PAT-APT). Firstly, CD spectroscopy, molecular docking, and molecular dynamic simulation were applied to investigate the details on how PAT-APT bind with its target molecule. The results indicated that the structure of PAT-APT changed to a certain extent and was stabilized after binding with PAT. C-11, C-37 and C-38 were the key sites for the recognition and interaction between PAT-APT and its target. Secondly, based on these results, a ratiometric aptasensor made use of fluorescence resonance energy transfer (FRET) and synchronous fluorescence spectroscopy was designed. The complementary sequence (cDNA) to the aptamer with appropriate length and hybridization position was obtained through rationally design and optimization. Both PAT-APT and cDNA were labeled by a pair of fluorophores, which could generate FRET when the two single-stranded oligonucleotides hybridized. The accurate detection of PAT could be realized according to the change ratio of fluorescence intensity at the corresponding wavelength of the two fluorophores before and after assay. The aptasensor achieved an ultralow limit of detection 0.16 nM, perfect selectivity, and satisfactory practibility in complex TCM sample. To our knowledge, this is the first aptasensor for PAT designed through the interaction mechanism between aptamer and target molecule. Moreover, the assay for PAT is cost-effective, does not need complicated pretreatment and only takes less than an hour. Summarily, this study makes contribution to safety control of TCM, and provides a thinking mode from mechanism to rational design to conquer the problem of sensitive aptasensing of one component in a complex system.

Lung cancer is known as the leading cause which presents the highest fatality rate worldwide; non-small-cell lung cancer (NSCLC) is the most prevalent type of lung carcinoma with high severity and affects 80% of patients with lung malignancies. Up to now, the general treatment for NSCLC includes surgery, chemotherapy, and radiotherapy; however, some therapeutic drugs and approaches could cause side effects and weaken the immune system. The combination of conventional therapies and traditional Chinese medicine (TCM) significantly improves treatment efficacy in lung cancer. Therefore, it is necessary to investigate the chemical composition and underlying antitumor mechanisms of TCM, so as to get a better understanding of the potential natural ingredient for lung cancer treatment. In this study, we selected 78 TCM to treat NSCLC cell line (A549) and obtained 92 transcriptome data; differential expression and WGCNA were applied to screen the potential natural ingredient and target genes. The sample which was treated with A. pierreana generated the most significant DEG set, including 6130 DEGs, 2479 upregulated, and 3651 downregulated. KEGG pathway analyses found that four pathways (MAPK, NF-kappa B, p53, and TGF-beta signaling pathway) were significantly enriched; 16 genes were significantly regulated in these four pathways. Interestingly, some of them such as EGFR, DUSP4, IL1R1, IL1B, MDM2, CDKNIA, and IDs have been used as the target biomarkers for cancer diagnosis and therapy. In addition, classified samples into 14 groups based on their pharmaceutical effects, WGCNA was used to identify 27 modules. Among them, green and darkgrey were the most relevant modules. Eight genes in the green module and four in darkgrey were identified as hub genes. In conclusion, we screened out three new TCM (B. fruticose, A. pierreana, and S. scandens) that have the potential to develop natural anticancer drugs and obtained the therapeutic targets for NSCLC therapy. Our study provides unique insights to screen the natural components for NSCLC therapy using high-throughput transcriptome analysis.
