Adopting the concept and operation of hybridization-based associative toehold activation (HATA) concatemer induced catalyzed hairpin assembly (CHA), a novel colorimetric biosensor was developed for detecting target DNA and human alpha-thrombin. The assemble of separate DNA hairpin components can be triggered by specific target binding that are otherwise unable to spontaneously happen. In the presence of four ingeniously designed hairpin structures, the target molecule can trigger two cascade recycling reactions: HATA concatemer reaction and CHA reaction. In the HATA concatemer reaction, the toehold and branch migration domain overhang at the 5′ end and 3′ end of hairpin probe H1, respectively. Upon the addition of target molecule, plenty of toeholds and branch migration domains can be brought into close proximity via DNA self-assemble. Then, the toeholds and branch migration domains of HATA products serve as the trigger of CHA. In this concatemer recycling, numerous guanine rich DNA duplexes can be produced through the highly effective use of target molecule. The proposed sensing system exhibits a remarkable analytical performance and an ultrahigh discrimination capability for proximal mismatch. Under optimal conditions, the calibration curves have a good linearity from 1 pM to 75 nM and 1 pM to 2.5 nM for target DNA and human alpha-thrombin, respectively. The corresponding detection limits are 0.14 pMand 0.68 pM Therefore, this new biosensor has a great potential for further applications in biomedical research and demonstrates excellent versatility for different analytes.

In this study, we report a metallobioassay for ultrasensitive electrochemical detection of prostate-specific antigen (PSA) based on DNA hybridization chain reaction (HCR) for amplifying the signal, which is derived from silver nanoparticles (Ag NPs) on DNA concatemers. The assay mainly consists of primary antibody (Ab1), secondary antibody (Ab2) with primer, and a signal probe. In the presence of PSA, a sandwich structure with DNA concatemers was formed, and numerous Ag NPs were loaded on the DNA concatemers, resulting in a strong signal, which appeared within the applied potential (−0.2 V to 0.3 V) in the phosphate-buffered saline (PBS). Differential pulse voltammetry (DPV) was employed to evaluate the analytical performance. Under optimal conditions, the DPV peak current of Ag NPs at about +0.09 V (vs. SCE) increased linearly as the logarithm of PSA concentration increased from 0.1 pg mL−1 to 75 ng mL−1, and the detection limit of PSA was estimated to be 0.033 pg mL−1 at the signal to noise ratio of 3. In addition, the assay was evaluated with human serum samples, and satisfying results were obtained, indicating that the assay can achieve PSA detection in the serum sample.

Ethylene production via chemical looping oxidative dehydrogenation (CL-ODH) of ethane faces a critical challenge in balancing conversion efficiency and selectivity due to competing oxygen species requirements. Here, we present a bifunctional composite catalyst (CrH/BiCZ) by physically integrating a Cr-HZSM5 (Cr-H) dehydrogenation (DH) catalyst with a Bi 2 O 3 -Ce 0.4 Zr 0.6 O 2 (Bi-CZ) selective hydrogen combustion (SHC) catalyst. This tandem system decouples ethane activation and hydrogen combustion, promoting the DH process of ethane through the in situ hydrogen combustion of the SHC catalyst. The Ce-Zr solid solution in Bi-CZ acts as an oxygen reservoir, enabling complete H 2 combustion within 3 min and suppressing carbon deposition. This rapid hydrogen combustion shifts the reaction equilibrium towards ethylene production. At 650 °C, the optimized CrH/BiCZ catalyst achieves 84.1 % ethylene selectivity at 43.0 % ethane conversion, and maintains exceptional stability over 36 redox cycles. This work establishes a scalable chemical looping strategy for efficient ethylene synthesis.

A novel surface-enhanced Raman scattering/surface plasmon resonance (SERS/SPR) dual-mode biosensor prepared on a silver nanorod-covered silver nanohole (Ag NR-NH) array by surface modification of tetrahedral DNA probes is proposed for highly sensitive detecting nucleic acids by a special signal amplification strategy of DNA supersandwich. The Ag NR-NH with a large area and uniformly arrayed nanostructure possesses excellent anisotropic extraordinary optical transmission and strong localized surface plasmon resonance, which lead to sensitive SPR response to the change of a local refractive index and strong localized electric fields for excellent SERS activity. To obtain high sensitivity and specificity, smart tetrahedral DNA probes are immobilized onto the Ag NR-NH array and the DNA supersandwich sensing strategy, including the signal amplification of DNA concatamers, is used. About 10 times signal enhancement for SPR and 4 times for SERS are achieved by this sensing strategy. In the detection of the target DNA in the human serum, the two sensing modes have complementary performances, i.e., the limit of detection for the SPR array is high (0.51 pM), while for SERS, it is low (0.77 fM), but the specificity for SPR is much higher than that of SERS. This improves the robustness of the DNA sensors, and subsequent recovery tests also confirm good reliability of the biosensor. The proposed SERS/SPR dual-mode biosensor has a great potential for high performance and reliable detection of trace disease-related nucleic acid biomarkers in the serum and is a powerful sensing platform for early-stage disease diagnosis.

A novel capacitance immunosensor based on DNAzyme concatemer-amplified signal-generation tags was developed for the sensitive detection of interleukin-6 (IL-6) on an interdigitated micro-comb electrode. To construct such an immunosensor, monoclonal mouse anti-human IL-6 antibodies were covalently conjugated onto a bovine serum albumin (BSA)-coated electrode using the glutaraldehyde cross-linker. Gold nanoparticles heavily functionalized with primer DNA (for the subsequent rolling cycle amplification; RCA) and polyclonal rabbit anti-human IL-6 antibodies were used for the development of a sandwich-type immunoassay on the modified interdigitated electrode in the presence of the target IL-6. Thereafter, the attached primer DNA with the immunocomplex triggered the RCA reaction for the formation of numerous DNAzyme concatemers with the assistance of hemins and G-rich DNA probes. DNAzyme concatemers catalyzed 4-chloro-1-naphthol into insoluble benzo-4-chlorohexadienone and were deposited onto the electrode, thus resulting in the local capacitance variation of the interdigitated micro-comb electrode. The performance and external factors affecting the analytical properties were studied in detail. Under optimum conditions, the capacitance variations of the immunosensor were proportional to the IL-6 concentration in the dynamic range of 1.0–1000 pg mL−1 at a detection limit of 0.68 pg mL−1. Good reproducibility, high specificity and acceptable storage stability were acquired for the analysis of the target IL-6. Moreover, the capacitance immunosensor was utilized to determine IL-6 in human serum specimens and gave well-matched analytical results in reference to the human IL-6 ELISA kit.
