An ammonia-based etchant for attaining copper nanoclusters with green fluorescence emission

Hao-Hua Deng,Ke-Lin Li,Quan-Quan Zhuang,Hua-Ping Peng,Qiong-Qiong Zhuang,Ai-Lin Liu,Xing-Hua Xia,Wei Chen

Luminescent copper nanoclusters (CuNCs) constitute a very active research topic due to their unique properties and lower cost than gold and silver NCs. In this study, we report a new, facile, and rapid top-down etching method for synthesizing luminescent CuNCs, using Cu nanoparticles (CuNPs) as the precursor and ammonia (NH3) as the etchant. The etching mechanism is systematically investigated and the optical and structural properties of the obtained CuNCs are carefully studied. The NH3-triggered etching process is very fast and the newly generated CuNCs can emit strong green fluorescence with a high quantum yield. Moreover, by coupling the urease-catalyzed hydrolysis of urea with the NH3-induced etching of CuNPs, we developed a novel fluorescence turn-on assay for urea. The linear range for urea detection is from 0.25 to 5 mM, and the limit of detection is 0.01 mM. This novel sensing approach, with good sensitivity and excellent selectivity, is then successfully utilized to detect urea in human serum samples, demonstrating its great potential in clinical diagnosis. In addition, the proposed coupling method can be extended to monitor other analytes that influence the size-focusing etching process, allowing metal NCs to be used to construct diverse chemosensors and biosensors.

Profiling of Homocysteine Metabolic Pathway Related Metabolites in Plasma of Diabetic Mellitus Based on LC-QTOF-MS

Chanyi Li,Jiaying Qin,Wuping Liu,Bo Lv,Ning Yi,Jinfeng Xue,Zhigang Xue

Background: Homocysteine (Hcy) has been found to be closely related to the occurrence of diabetes mellitus (DM) and is considered as one of the risk factors of DM. However, Hcy alone is not enough as a factor to predict DM, and our study analyzed and determined the relationship between the main metabolites involved in the Hcy metabolic pathway and DM. Methods: A total of 48 clinical samples were collected, including 18 health control samples and 30 DM samples. All standards and samples were detected by LC-QTOF-MS. Multivariate statistical analysis and k-means cluster analysis were performed to screen and confirm the metabolites significantly correlated with DM. Results: A total of 13 metabolites of the Hcy metabolic pathway were detected in the samples. The content of Hcy, cysteine, taurine, pyridoxamine, methionine, and choline were significantly increased in the DM group (p< 0.05). Hcy, choline, cystathionine, methionine, and taurine contributed significantly to the probabilistic principal component analysis (PPCA) model. The odds ratios (OR) of Hcy, cysteine, taurine, methionine, and choline were all greater than one. K-means cluster analysis showed that the Hcy, taurine, methionine, and choline were significantly correlated with the distribution of glucose values (divided into four levels: 10.5–11.7 mmol/L, 7.7–9.7 mmol/L, 6.0–6.9 mmol/L, and 5.0–5.9 mmol/L, respectively). Conclusion: Hcy, taurine, methionine, and choline can be used as risk factors for diabetes diagnosis and are expected to be used for the assessment of diabetes severity. Keywords:homocysteine metabolism;diabetes;LC-MS;clinical diagnosis

A compact liquid chromatography-mass spectrometry instrument for the quantitation of immunosuppressants

Hao Gu,Guoxin Dai,Siyu Liu,Yanbing Zhai,Qian Xu,Hongqiang Wu,Wei Xu

Liquid chromatography tandem mass spectrometry (LC-MS/MS) plays an important role in clinical diagnostics. Although LC-MS/MS is superior in terms of accurately quantifying molecules in complex matrices, instrument footprint, operation and maintenance complexity also hinder its expansion as the analytical technique of choice. In this study, a compact LC-MS instrument was developed, in which an assembled liquid chromatograph was coupled with a miniature ion trap mass spectrometer. The overall instrument has a footprint of 69 cm × 31 cm × 31 cm, and it requires no gas supply as well as minimum maintenance. Furthermore, the use of LC-MS is in accord with conventional clinical diagnostic protocols, and the choice of ion trap offers tandem MS performance. The results showed that the use of LC could improve both mixture analysis capability and detection sensitivity of the miniature mass spectrometer. After optimization, feasibility of this instrument in clinical practice was demonstrated by the quantitation of four widely used immunosuppressants in blood samples. Relatively good linearities were obtained, which spanned the reference ranges of effective therapeutic concentrations of each immunosuppressant. Intra-day and inter-day accuracy and precision of analytical method were also assessed. This work showed that a compact LC-MS instrument could be used in clinical diagnosis, either to replace conventional lab-scale instruments or to be used in POCT applications.

Nonequilibrium hybridization-driven CRISPR/Cas adapter with extended energetic penalty for discrimination of single-nucleotide variants

Liu Qiong,Jiang Zhou,Li Sheng,Li Yinfeng,Wan Yingfei,Hu Zhenyu,Ma Shimeng,Zou Zhen,Yang Ronghua

Accurate identification of single-nucleotide variants (SNVs) is critical in clinical diagnostics but remains challenging due to subtle free energy variations, particularly for hard-to-detect SNVs such as wobble base pairs and those in high guanine–cytosine (GC) regions. Here we report a high-energetic-penalty SNV detection (HEPSD) platform that redesigns the hybridization regions of CRISPR RNA (crRNA) in the CRISPR/Cas12a system. This system employs a binary crRNA architecture design that enables the activation of the cleavage activity of Cas12a while amplifying the energetic penalty for single-nucleotide mismatches through nonequilibrium hybridization-driven regulation. Consequently, the entire targeting region of CRISPR/Cas exhibits a marked preference for mutations in genomic DNA, while preventing false activation induced by sequences containing a single mismatched nucleotide. Moreover, HEPSD exhibits exceptional differentiation performance for hard-to-detect SNVs including wobble mutations at extreme GC contents. As proof of principle, profiling of BRAF V600E and EGFR L858R tumor mutations down to a 0.01% variant allele frequency was achieved, enabling accurate discrimination of 132 clinical sample pairs, which showed high consistency with quantitative polymerase chain reaction-based techniques and next-generation sequencing. The proven effectiveness of this platform showcases its potential for clinical molecular diagnostics and expands the fundamental scope of hybridization-based protocols.

Functionalized MXene Fiber Electrode for the Electrochemical Sensing of Urinary Ammonia

Yu Liu,Xu Zeng,Yulong Wu,Habib Ullah Khan,Fei Xiao,Bao Yu Xia,Yimin Sun

The development of technologically advanced fiber-based flexible microelectrodes has been of extensive research interest in healthcare systems because of their unique construction and synergistic effect on multifunctional properties. In this work, we constructed functional MXene fiber (MXF) by a simple and versatile wet spinning method, and then, a well-aligned ZIF-67 nanoarray was grown in situ on the surface. Using the chemical vapor deposition (CVD) method, carbon nanotubes (CNTs) were produced on MXF via the pyrolysis of ZIF-67 and melamine. Finally, Pt nanoparticles were electrodeposited on the CNTs forest, and a Pt@CNTs/MXF electrode was obtained. Owing to the plethora of surface active sites and the synergistic effects between MXene, CNTs, and Pt nanoparticles, the as-fabricated fiber electrode enabled the precise detection of ammonia under alkaline conditions via differential pulse voltammetry (DPV), which exhibited a linear range of 0.1 μM-10 mM and a detection limit of 73.2 nM. Due to their good performance in ammonia detection, Pt@CNTs/MXF electrodes could be adopted to determine the ammonia concentration in urine for clinical estimation, which provides a practical approach for the diagnosis of urinary ammonia-associated diseases.

Fluorescence turn-on detection of human serum albumin based on the assembly of gold nanoclusters and bromocresol green

Xiao Wenxiang,Li Yaoxin,Xiong Yinan,Chen ZhenCheng,Li Hua

As the most abundant protein in plasma, human serum albumin plays a vital role in physiological processes, such as maintaining blood osmotic pressure and carrying small-molecule ligands. Since the content of albumin in the human serum can reflect the status of liver and renal function, albumin quantitation is significant in clinical diagnosis. In this work, fluorescence turn-on detection of human serum albumin (HSA) had been performed based on the assembly of gold nanoclusters and bromocresol green. Gold nanoclusters (AuNCs) capped by reduced glutathione (GSH) were assembled with bromocresol green (BCG), and the assembly was used as a fluorescent probe for HSA. After BCG assembling, the fluorescence of gold nanoclusters was nearly quenched. In acidic solution, HSA can selectively bind to BCG on the assembly and recover the fluorescence of the solution. Based on this turn-on fluorescence, ratiometric HSA quantification was realized. Under optimal conditions, HSA detection by the probe possessed a good linear relationship in the range of 0.40–22.50 mg·mL −1 , and the detection limit was 0.27 ± 0.04 mg·mL −1 (3σ, n  = 3). Common coexisting components in serum and blood proteins did not interfere with the detection of HSA. This method has the advantages of easy manipulation and high sensitivity, and the fluorescent response is insensitive to reaction time. Graphical abstract

Defect-rich CoOOH nanorings: A biocompatible and cost-efficient material for clinical diagnosis of children heart failure

Yi Wang,Aiping Tu,Xinwen Zhang,Mengyao Li,Laicheng Liu,Yongfei Li,Jiangling Wu,Pu Zhang

Nanomaterials have been widely used in the detection of biomarkers, but only a few of them can be really applied in clinical disease diagnosis in hospital. Herein, we fabricate a kind of defect-rich CoOOH nanorings (NRs) and employ them as probes to improve the clinical diagnosis performance for children heart failure (HF). This diagnosis platform is dependent on ultrasensitive and specific detection of the “gold standard biomarker” of HF, B-type natriuretic peptide (BNP), which is realized based on the prevention of fluorescence resonance energy transfer. The detection range for BNP is 30–800 fg/mL with a limit of detection as low as 30 fg/mL. For diagnosis of a large number of clinical cases of children who behave sign or symptom of HF (respiratory distress, edema, or tachycardia), a very close correlation ( r  = 0.990) is obtained between this method and commercially available chemiluminescence immunoassay (CLIA, Siemens) that commonly adopted by hospital. It is worth noting that the present method has better diagnostic performance for children HF than CLIA including specificity (84.6% vs 61.5%), accuracy (82.2% vs 66.7%) and positive predictive value (40.0% vs 20.0%). Moreover, this method is more cost-effective than CLIA for BNP detection. The defect-rich, biocompatible, and low-cost CoOOH nanomaterial is promising to be applied in early clinical diagnosis of HF in hospital.

Enzyme-activated anchoring of peptide probes onto plasma membranes for selectively lighting up target cells

Julan Xia,Sujuan Sun,Xianhua Wu,Yan Huang,Chunyang Lei,Zhou Nie

In a cellular microenvironment, numerous biomolecules are involved in various physiological and pathological processes. However, for the in-depth and comprehensive understanding of their roles at the molecular level, there is still a lack of detection techniques for the in situ tracking of these biomolecules in a local environment. Herein, we engineered a membrane insertion peptide (MIP) as an enzyme-activated membrane insertion peptide probe (eaMIP) that allowed the in situ tracking of the activity of target enzymes in living cells. In this strategy, the membrane insertion capacity of the MIP motif in each eaMIP was caged by appending a chemical moiety. In the presence of target enzymes, the caging moiety in each eaMIP was removed by enzymatic decaging, leading to the generation of active MIPs. The versatility of this design was demonstrated by lighting up different tumor cells with distinct fluorescence signal patterns, affording an alternative tool for clinical diagnostics, biochemical research and membrane engineering.

Green-emissive carbon dots for multitarget staining of cells, fungi, and bacteria: Insights into mechanisms and clinical applications

Jingbo Sun,Liushunhao Chen,Hongchen Li,Chen Xu,Dong Xu,Jiali Ren

Biological staining is essential for imaging, functional investigation, and disease diagnostics. In this research, innovative green-emissive carbon dots (G-CDs) were hydrothermally prepared from 1,2,4-triaminobenzene and ascorbic acid (AA) and further explored for versatile imaging of mammalian cells, plant cells, and microorganisms. The G-CDs, exhibiting fluorescence enhancement, outperform conventional dyes in terms of staining efficiency and specificity across various biological systems. For mammalian cells (SH-SY5Y, Hep3B, and HEK293), the G-CDs can stain both the nuclei and nucleoli in 10 min via a micropinocytic endocytosis approach, with the latter brighter. Fluorescence enhancement was greater for RNA than for DNA. Interestingly, for plant cells, only cell walls were stained, because positive charges on G-CDs enable strong adsorption onto the negatively charged cell walls, thereby hindering their further entry. G-CDs can stain cell walls and nuclei for fungi (including Aspergillus flavus and Aspergillus niger ), and cell walls and nucleoids for bacteria ( Bacillus subtilis and Escherichia coli ). The targeted staining arises from both the binding of G-CDs to DNA, RNA, pectin, peptidoglycan, and cellulose, and fluorescence enhancement due to surface passivation inhibiting rotation and vibration of functional groups. Electrostatic interactions, coordination, hydrogen bonding, and amidation collectively account for the interactions between G-CDs and these biomolecules . Finally, their practical applications in leucorrhea examinations demonstrated their capability to differentiate between bacterial and fungal vaginosis, highlighting their potential in clinical diagnostics.

High-precision Helicobacter pylori infection diagnosis using a dual-element multimodal gas sensor array

Jiaying Wu,Shiyuan Xu,Xuemei Liu,Jingwen Zhao,Zhengfu He,Aiwu Pan,Jianmin Wu

Helicobacter pylori (H. pylori) is a globally widespread bacterial infection. Early diagnosis of this infection is vital for public and individual health. Prevalent diagnosis methods like the isotope 13C or 14C labelled urea breath test (UBT) are not convenient and may do harm to the human body. The use of cross-response gas sensor arrays (GSAs) is an alternative way for label-free detection of metabolite changes in exhaled breath (EB). However, conventional GSAs are complex to prepare, lack reliability, and fail to discriminate subtle changes in EB due to the use of numerous sensing elements and single dimensional signal. This work presents a dual-element multimodal GSA empowered with multimodal sensing signals including conductance (G), capacitance (C), and dissipation factor (DF) to improve the ability for gas recognition and H. pylori-infection diagnosis. Sensitized by poly(diallyldimethylammonium chloride) (PDDA) and the metal–organic framework material NH2-UiO66, the dual-element graphene oxide (GO)-composite GSAs exhibited a high specific surface area and abundant adsorption sites, resulting in high sensitivity, repeatability, and fast response/recovery speed in all three signals. The multimodal sensing signals with rich sensing features allowed the GSA to detect various physicochemical properties of gas analytes, such as charge transfer and polarization ability, enhancing the sensing capabilities for gas discrimination. The dual-element GSA could differentiate different typical standard gases and non-dehumidified EB samples, demonstrating the advantages in EB analysis. In a case–control clinical study on 52 clinical EB samples, the diagnosis model based on the multimodal GSA achieved an accuracy of 94.1%, a sensitivity of 100%, and a specificity of 90.9% for diagnosing H. pylori infection, offering a promising strategy for developing an accurate, non-invasive and label-free method for disease diagnosis.

Detection of the level of DNMT1 based on self-assembled probe signal amplification technique in plasma

Zhenzhen Wan,Fangfang Gong,Mimi Zhang,Leiliang He,Yilin Wang,Songcheng Yu,Jie Liu,Yuming Wu,Li'e Liu,Yongjun Wu,Lingbo Qu,Jiaqi Sun,Fei Yu

DNA (cytosine-5)-methyltransferase1 (DNMT1) is the most abundant DNA methyltransferase in somatic cells, and it plays an important role in the initiation, occurrence, and rehabilitation of tumors. Herein, we developed a novel strategy for the detection of the level of DNMT1 in human plasma using the self-assembled nucleic acid probe signal amplification technology. In this method, the DNMT1 monoclonal antibody (McAb DNMT1 ) was immobilized on carboxyl magnetic beads to form immunomagnetic beads and then captured DNMT1 specifically. After that, DNMT1 polyclonal antibody (PcAb DNMT1 ) and biotinylated sheep anti-rabbit IgG (sheep anti rabbit IgG-Biotin) were sequentially added into the system to react with DNMT1 and form biotinylated double antibody sandwich immunomagnetic beads. In the presence of the bridging medium streptavidin, the biotinylated double antibody sandwich immunomagnetic beads would form a complex with biotinylated poly-fluorescein (Biotin-poly FAM), and the fluorescence intensity of the complex was proportional to the concentration of DNMT1. Immunomagnetic beads can capture the target DNMT1 in the sample, and Biotin-poly FAM can realize signal amplification. Using these strategies, we got a linear range of the system for DNMT1 level detection was from 2 nmol/L to 200 nmol/L, and the limit of detection (LOD) was 0.05 nmol/L. The method was successfully applied for the determination of DNMT1 in human plasma with the recovery of 101.3–106.0%. Therefore, this method has the potential for the detection of DNMT1 level in clinical diagnosis.

Diagnosis of prosthetic joint infection based on Cas12a-catalytic aggregation mediated by supramolecular chemistry

Yining Zhao,Ziye Liu,Ziting Liu,Juan Zhang,Jinghui Peng,Genxi Li

Prosthetic joint infection (PJI), which is still difficult to be diagnosing, is caused by bacterial infection. Rapid detection for trace amounts of bacteria should be a solution; however, the existing assays cannot serve for PJI diagnosis due to the inherent limitations. Herein, a strategy based on supramolecular chemistry mediated Cas12a catalytic aggregation (CCA) is explored in this work for ultrasensitive detection of universal bacteria. In the design, supramolecular chemistry is introduced to impede the cis-cleavage of activated Cas12a, resulting in its aggregation in the surrounding of bacteria with the linkage of CCA probe. Since Cas12a aggregation owns excellently enhanced catalytic efficiency, the CCA-based strategy can be used for bacterial analysis with a limit of detection as low as 1 CFU/mL. With high sensitivity, reproducibility, generalizability, and stability, the CCA-based strategy can be applied for the detection of clinical effusion samples. The remarkable performance of CCA-based strategy has also demonstrated its robust potential to be a simple, rapid, and highly sensitive bacteria diagnostic tool, paving the way for prompt diagnosis and management of PJI.
