To promote the bone repair ability of drug-loaded scaffolds, poly(lactic acid) (PLA)/graphene oxide (GO)/Salvianolic acid B (Sal-B)/aspirin (ASA) dual drug-loaded biomimetic composite scaffolds were prepared. The results showed that the addition of these two drugs delayed the gel formation of the composite system, but a biomimetic nanofiber structure could still be obtained by extending the gel time. The addition of Sal-B increased the hydrophilicity of the scaffold, while an increase in ASA reduced the porosity. Dual drug-loaded scaffolds had good haemocompatibility and synergically promoted the proliferation of MC3T3-E1 cells and enhanced alkaline phosphatase activity. Sustained-release experiments of the two drugs showed that the presence of ASA slowed the cumulative release of Sal-B, while Sal-B promoted the release of ASA. Kinetic modeling showed that the release of both drugs conforms to the Korsmeyer–Peppas model, but Sal-B conforms to the Fick diffusion mechanism and ASA follows Fick diffusion and carrier swelling/dissolution.

Biomimetic scaffolds loaded with drugs can improve the osteogenesis and neovascularisation of scaffolds. A series of PLA/GO/Sal-B drug-loaded scaffolds was prepared by thermally induced phase separation. The addition of Sal-B increased the diameter of the fibres, but the scaffold showed a porous nanofibrous structure after drug release. X-ray diffraction results showed that the addition of Sal-B did not affect the formation of the nanofibre biomimetic structure of the scaffold. FTIR results indicated a certain interaction between Sal-B and PLA/GO. Water absorption and porosity test results revealed that the scaffolds had good hydrophilicity and appropriate porosity. The addition of Sal-B was also conducive to the formation of sediments possibly due to the good water solubility of Sal-B itself. The prepared scaffolds had good blood compatibility and cytocompatibility, and a small additional amount of Sal-B could significantly promote cell proliferation and alkaline phosphatase activity. Their sustained release performance indicated that the biomimetic scaffolds had controlled the release of Sal-B. The kinetic model showed that the PLA/GO/Sal-B drug-loaded biomimetic scaffolds followed the diffusion mechanism.

Scarring poses a significant challenge in wound healing, especially for chronic wounds like those linked to diabetes. This study developed a dual drug-loaded core-shell fiber scaffold (PSPC) containing curcumin (Cur) in the shell and salvianolic acid B (SAB) in the core via electrospinning to address this challenging issue through synergistic anti-inflammatory and anti-fibrosis efficacy. The PSPC scaffold presented favorable tensile properties (3.67 ± 0.32 MPa of tensile strength and 337.64% ± 32.07% of elongation at break) and suture retention (2.71 ± 0.17 N). In vitro experiments confirmed that this scaffold was biocompatible, supporting cell adhesion and proliferation. Additionally, it also scavenged excessive intracellular reactive oxygen species (ROS) and promoted macrophage polarization towards the regenerative M2 phenotype. In vivo studies using a mouse diabetic wound model showed that the PSPC scaffold effectively mitigated inflammatory responses, promoted neoangiogenesis, and facilitated epidermal formation and re-epithelialization. More importantly, the scaffold also modulated collagen synthesis, decreased the collagen type I/III ratio, supported the functional regeneration of hair follicles, sebaceous glands, and other skin appendages, then ultimately achieved the goal of chronic diabetic wound healing and scar reduction. The developed dual drug-loaded fiber scaffold integrating anti-inflammatory and anti-fibrosis presents a promising strategy for the anti-scarring treatment of chronic wounds.

Salvianolic acid B (SAB) is a widely used cardioprotective agent, while its clinical application was limited by poor intestinal absorption and low oral bioavailability. In this study, SAB phospholipid (SP) complex was first prepared to improve the lipophilicity of SAB and then combined with d-glucose to further enhance intestinal absorption. Compared with free SAB, SP or the mixture of SAB and d-glucose, combination of SP and d-glucose showed higher intestinal absorption evidenced by increased effective permeation coefficient (Peff) in the in situ single-pass intestinal perfusion (SPIP) assay. Subsequently, SP and d-glucose at mass ratio of 1:6, with the highest Peff of SAB, were chosen for the preparation of complexed pellets to improve oral absorption efficiency of SAB. As expected, the obtained pellets significantly enhanced oral bioavailability of SAB in the pharmacokinetic study characterized by increasing Cmax and AUC0–t of SAB by 14.88-fold and 5.02-fold than free SAB, respectively. In conclusion, combination of d-glucose in SP pellets can effectively improve the intestinal absorption and oral bioavailability of SAB.

Hypertrophic scar (HS) represents a natural consequence of abnormal fibroproliferation occurring during wound healing, which not only leads to tissue dysfunction but also causes psychological issues for patients. Up to now, no completely effective therapy for HS has been found. Salvianolic acid B (Sal B), one of the natural plant-derived compounds, has the capacity to suppress the fibrosis process in scarring treatment. Nevertheless, due to its poor skin-penetration ability and unsatisfactory stability in existing formulations, its current clinical applicability is restricted. In this study, inspired by the separation behavior of the thorns on the trunks of the silk floss tree, we developed a hyaluronic acid (HA) dissolving microneedle (MN) platform loaded with Sal B. This platform can enhance the sustainable delivery of Sal-B directly into scar tissue in a minimally invasive manner, thereby enhancing the efficacy of HS therapy and improving the stability of Sal B. To evaluate the therapeutic efficacy of Sal-B MN, we established a rabbit ear HS model. After being administered five times, Sal-B MN significantly reduced the scar elevation index (SEI) and down-regulated the expressions of collagen I, transforming growth factor beta 1 (TGF-β1), Signal Transducer and Activator of Transcription 3 (STAT3), and alpha-smooth muscle actin (α-SMA). Notably, these effects were still evident even after six months of storage at room temperature. This indicates that tree-inspired Sal-B MN holds great potential as a highly effective tool for HS treatment with a long shelf-life. This makes the system suitable for clinical application and storage under various conditions.

Kidney-specific delivery is critically important for the treatment of renal fibrosis with drugs such as salvianolic acid B (Sal B). Here we report a kidney-specific nanocomplex formed by the coordination-driven assembly of catechol-modified low molecular weight chitosan (HCA-Chi), calcium ions and Sal B. The prepared HCA-Chi-Ca-Sal B (HChi-Ca-Sal B) nanocomplex reversed the TGF-β1-induced epithelial–mesenchymal transition (EMT) in HK-2 cells. In vivo imaging demonstrated a kidney-specific biodistribution of the nanocomplex. The anti-fibrosis effect of HChi-Ca-Sal B was tested in a mouse model of unilateral ureteral obstruction (UUO). Significant attenuation of the morphological lesions and the levels of extracellular matrix (ECM) proteins in the tubulointerstitium was observed in mice treated with HChi-Ca-Sal B, suggesting that the nanocomplex was able to prevent fibrosis better than the treatment with free Sal B. It was concluded that the HChi-Ca-Sal B nanocomplex showed a specific renal targeting capacity and could be utilized to enhance Sal B delivery for treating renal fibrosis.

An in-capillary 2,2′-azinobis-(3-ethylbenzthiazoline-6-sulphonate) as oxyradicals combining field-enhanced sample injection with micellar electrokinetic chromatography was developed for screening and determination of the major antioxidants in Yangxinshi Tablet. To obtain simultaneous separation and detection of radicals and analytes, relevant factors were optimized separately. Under the optimum conditions, four compounds including salvianolic acid B, hyperoside, puerarin, and caffeic acid were identified as the major antioxidants. All validation results covering recovery, precision, and stability demonstrated good applicability of the method. On this basis, the total antioxidant activity was successfully evaluated in terms of the decreased peak area of radicals. There was a correlation coefficient of 0.8974 between the total contents of major antioxidants and the total antioxidative activity of the sample. Therefore, these four compounds were selected as combinatorial markers for the quality evaluation of Yangxinshi Tablet. It was concluded that the established method presented a powerful potential to screen and quantify active ingredients in the complex preparation of Chinese medicine.

Traumatic brain injury (TBI) leads to structural damage in the brain, and is one of the major causes of disability and death in the world. Herein, we developed a composite injectable hydrogel (HA/Gel) composed of hyaluronic acid (HA) and gelatin (Gel), loaded with vascular endothelial growth factor (VEGF) and salvianolic acid B (SAB) for treatment of TBI. The HA/Gel hydrogels were formed by the coupling of phenol-rich tyramine-modified HA (HA-TA) and tyramine-modified Gel (Gel-TA) catalyzed by horseradish peroxidase (HRP) in the presence of hydrogen peroxide (H2O2). SEM results showed that HA/Gel hydrogel had a porous structure. Rheological test results showed that the hydrogel possessed appropriate rheological properties, and UV spectrophotometry results showed that the hydrogel exhibited excellent SAB release performance. The results of LIVE/DEAD staining, CCK-8 and Phalloidin/DAPI fluorescence staining showed that the HA/Gel hydrogel possessed good cell biocompatibility. Moreover, the hydrogels loaded with SAB and VEGF (HA/Gel/SAB/VEGF) could effectively promote the proliferation of bone marrow mesenchymal stem cells (BMSCs). In addition, the results of H&E staining, CD31 and α-SMA immunofluorescence staining showed that the HA/Gel/SAB/VEGF hydrogel possessed good in vivo biocompatibility and pro-angiogenic ability. Furthermore, immunohistochemical results showed that the injection of HA/Gel/SAB/VEGF hydrogel to the injury site could effectively reduce the volume of defective tissues in traumatic brain injured mice. Our results suggest that the injection of HA/Gel hydrogel loaded with SAB and VEGF might provide a new approach for therapeutic brain tissue repair after traumatic brain injury.
