Biomimetic Platelet Membrane-Based Polyphenol–CeO2 Nanozyme Complex: A Broad-Spectrum Antioxidative System for Comprehensive Atherosclerosis Treatment

Yao Chen,Qingfa Tang,Qinglan Zhu,Meiting Lu,Errong Chang,Ziting Gao,Zidong Wang,Lu Zhang,Qiang Liu,Junju Zou,Zhong Zuo,Chunyan Shen,Cuiping Jiang

Atherosclerosis (AS) is a chronic cardiovascular disease characterized by the accumulation of excess lipids within arterial walls. Since oxidative stress is one of the key drivers in its pathogenesis, antioxidative therapy serves as a promising strategy for AS treatment. Despite the potent antioxidant properties of polyphenolic salvianic acid A (SAA) and salvianolic acid B (SAB), clinical application is limited by their short half-life, insufficient target selectivity, and poor chemical stability. To overcome these limitations, we developed a biomimetic platelet membrane-coated polyphenol–cerium dioxide nanozyme complex (SS-CeO2@PLTM) as a broad-spectrum antioxidative system for comprehensive AS treatment. The SS-CeO2@PLTM was constructed by conjugating SAA and SAB on the surface of cerium dioxide via a one-pot method, followed by surface coating with a biomimetic platelet membrane. Systematic characterization using UV–vis spectroscopy, Fourier transform infrared (FT-IR), X-ray photoelectron spectroscopy (XPS), transmission electron microscopy-energy dispersive X-ray spectroscopy (TEM-EDS) elemental mapping analysis, Forster resonance energy transfer (FRET), molecular simulations, and pH-responsive release assays validated the successful synthesis and pH-responsive drug release behavior of SS-CeO2@PLTM. It was also found that SS-CeO2@PLTM exhibited broad-spectrum antioxidant activities, effectively scavenging superoxide anions, DPPH, H2O2, and intracellular reactive oxygen species (ROS) while enhancing hepatic SOD and GSH activities. Further cellular uptake and in vivo targeting assays confirmed their specific targeting for atherosclerotic plaques. SS-CeO2@PLTM demonstrated great efficiency in attenuating oxidative stress, inhibiting lipid uptake and lipid accumulation, mediating cholesterol efflux, and inhibiting inflammatory factor (IL-1β, IL-6, and TNF-α) secretion in RAW264.7 cells. We also found that SS-CeO2@PLTM exhibited improved lipid metabolism and diminished plaque area in apoE–/– mice via attenuating oxidative stress and NF-κB-mediated inflammation. Furthermore, biosafety was verified via hemolysis and in vivo safety tests. This study is the first to reveal the superior efficacy of the PLTM-based nanozyme complex in targeted AS therapy, offering a paradigm shift for multifactorial cardiovascular disease management.

Improving the antioxidant activity and enriching salvianolic acids by the fermentation of Salvia miltiorrhizae with Geomyces luteus

Yun Xing,Le Cai,Tian-peng Yin,Yang Chen,Jing Yu,Ya-rong Wang,Zhong-tao Ding

The antioxidant activities and total phenolic content of fermented Salvia miltiorrhiza with fungus Geomyces luteus were investigated. The results revealed that G. luteus fermentation could significantly improve the antioxidant activity and total phenolic content of S. miltiorrhiza . The main antioxidant constituents were characterized by spectroscopic analysis as salvianolic acids. High-performance liquid chromatography (HPLC) quantification also showed the enhanced content of salvianolic acid B after fermentation. The present study suggests that G. luteus fermentations are effective in the S. miltiorrhiza salvianolic acids’ enrichment process.

Bimetallic FeCo phosphide-enabled electrochemical sensor for rapid tanshinol quantification in Salvia miltiorrhiza herb at near-neutral condition

Mao Huang,Rui Wang,Fan Yu,Qi Ding,Yanyan Zhu

Tanshinol, an active ingredient extracted from the Salvia miltiorrhiza herb, is widely used in Chinese medicine or health supplements. Accurately and rapidly quantifying of tanshinol under near-neutral or neutral conditions is of great significance but still a significant challenge. Herein, a novel electrochemical sensor based on bimetallic FeCo phosphides (Fe x Co 1-x P) was developed for rapid and sensitive detection of tanshinol in near-neutral environments. Fe x Co 1-x P nanocrystals supported on porous graphene (Fe x Co 1-x P/GFs) were synthesized, with Fe 0.25 Co 0.75 P/GFs demonstrating an enhanced electrochemical response for tanshinol detection at pH 6.5. The Fe 0.25 Co 0.75 P/GFs-based tanshinol electrochemical sensor exhibited linear detection ranges of 0.20–11.65 μM and 11.65–39.98 μM, which could accurately quantify tanshinol in Salvia miltiorrhiza herb. This study highlights the potential of earth‐abundant FeCo phosphides for the electrochemical detection of tanshinol under benign conditions, offering a promising approach for the standardized analysis of Salvia miltiorrhiza herb.

pH and glucose dual-responsive hydrogels for IL-4 and salvianolic acid A delivery to enhance diabetic wound healing

Lifang Xie,Tianxiang Yuan,Su Hu,Chenmin Zhao,WenLi Huang,Wei Ding,Ye Zhang,Xianwen Hu

Chronic diabetic wounds exhibit impaired healing due to a complex interplay of pathological mechanisms, including persistent hyperglycemia, excessive matrix metalloproteinase-9 (MMP9) activity, and chronic inflammation, which disrupt tissue repair and regeneration. A central driver of this dysregulated healing is macrophage polarization imbalance, with excessive pro-inflammatory M1 and insufficient reparative M2 macrophages, prolonging inflammation and delaying tissue remodeling. To address these challenges, we developed a dual-responsive hydrogel composed of phenylboronic acid-functionalized polyethyleneimine (PEI-PBA), polyvinyl alcohol (PVA), and Benzaldehyde-PEG-Benzaldehyde (DF-PEG-DF) for controlled co-delivery of interleukin-4 (IL-4) and salvianolic acid A (SalA). This dynamically crosslinked hydrogel responds to the diabetic wound microenvironment, enabling pH- and glucose-triggered release. IL-4 promotes M2 macrophage repolarization, resolving inflammation and enhancing tissue repair, while SalA scavenges reactive oxygen species (ROS) and inhibits MMP9 activity, preserving extracellular matrix (ECM) integrity and supporting regeneration. In vivo, the hydrogel significantly accelerated wound closure in diabetic mice by promoting M2 polarization and reducing MMP9-mediated matrix degradation. This coordinated modulation of the inflammatory and proteolytic microenvironment attenuated inflammation, enhanced angiogenesis, and increased collagen deposition, collectively facilitating tissue regeneration. These findings suggest this intelligent hydrogel is a promising therapeutic strategy for chronic diabetic wound healing with translational potential.

Salvianolic acid B and Senkyunolide I synergistically alleviate cardiac hypertrophy and regulate MAP3K1 signaling

Liu Changtong,Guo Rui,Zhou Yue,Zhu Miao,Shao Li,Wang Yingchao,Zhao Lu

Background Cardiac hypertrophy, characterized by the thickening of the heart muscle, arises from factors such as hypertension and genetic mutations, often leading to adverse outcomes like heart failure and arrhythmias. Guanxinning tablets (GXNT), a botanical drug composed of the blood-activating herbs Salvia miltiorrhiza Bunge. and Ligusticum striatum DC., are widely used in the treatment of cardiovascular diseases. However, the active ingredients and their molecular mechanisms are yet to be fully understood. Methods We evaluated the anti-hypertrophic effects of GXNT and screened its active substances via cardiac function live-imaging on an aristolochic acid A-stimulated zebrafish cardiac hypertrophy model, and through F-actin immunostaining on a phenylephrine-induced hypertrophic NRCMs model. Additionally, the protective effects of GXNT’s active substances were analyzed in a mouse model of cardiac hypertrophy using echocardiography, histopathology analysis, and western blotting. Results The anti-hypertrophic effects of GXNT were assessed using an aristolochic acid A-stimulated zebrafish model and phenylephrine-induced hypertrophic NRCMs. GXNT demonstrated significant anti-hypertrophic effects in both models. Phenotypic screening identified Senkyunolide I (Sen I) from Ligusticum striatum as the active component in the zebrafish model, while Salvianolic acid B (Sal B) and Rosmarinic acid from Salvia miltiorrhiza emerged as the key anti-hypertrophic compound in NRCMs. In a mouse model of isoproterenol-induced cardiac hypertrophy, Sal B and Sen I showed synergistic effects, improving cardiac function, reducing oxidative stress, and suppressing inflammation. Mechanistically, transcriptomic sequencing highlighted cooperative modulation of MAP3K1 signaling by the two compounds. Notably, siRNA-mediated knockdown of MAP3K1 in cardiomyocytes attenuated the hypertrophic phenotype, supporting its essential role in the pathological process. Molecular docking and dynamic simulations further supported their binding potential to MAP3K1. Conclusion These findings underscore GXNT’s potent anti-hypertrophic effects, possibly driven by the synergistic actions of Sal B and Sen I, and offer insights into its therapeutic potential through MAP3K1 signaling regulation.

Combating Traumatic Brain Injury: A Dual-Mechanism Hydrogel Delivering Salvianolic Acid A and Hydroxysafflor Yellow A to Block TLR4/NF-κB and Boost Angiogenesis

Guoying Zhou,Yujia Yan,Linh Nguyen,Jiangkai Fan,Xiao Zhang,Li Gan,Tingzi Yan,Haitong Wan

Traumatic brain injury (TBI) leads to severe neurological dysfunction, disability, and even death. Surgical intervention and neurorehabilitation represent the current clinical management methods, yet there remains no effective treatment for recovery after TBI. Post-traumatic hyperinflammation and vascular injury are the key therapeutic challenges. Therefore, a novel-designed multifunctional HT/SAA/HSYA hydrogel based on hyaluronic acid (HA) co-loaded with salvianolic acid A (SAA) and hydroxysafflor yellow A (HSYA) was developed in order to simultaneously target inflammation and vascular injury, addressing key pathological processes in TBI. The HT hydrogel was formed through covalent cross-linking of tyramine-modified HA catalyzed by horseradish peroxidase (HRP). Results demonstrated that the HT hydrogel possesses a porous structure, sustained release capabilities of loaded drugs, suitable biodegradability, and excellent biocompatibility both in vitro and in vivo. WB, immunofluorescence staining, and PCR results revealed that SAA and HSYA significantly reduced the expression level of pro-inflammatory cytokines (IL-1β and TNF-α) and inhibited M1 macrophage polarization through the suppression of the TLR4/NF-κB inflammatory pathway. In vivo experiments confirmed that the HT/SAA/HSYA hydrogel exhibited remarkable pro-angiogenic effects, as evidenced by increased expression of CD31 and α-SMA. Finally, H&E staining showed that the HT/SAA/HSYA hydrogel effectively reduced the lesion volume in a mouse TBI model, and demonstrated more pronounced effects in promoting brain repair at the injury site, compared to the control and single-drug-loaded hydrogel groups. In conclusion, the HT hydrogel co-loaded with SAA and HSYA demonstrates excellent anti-inflammatory and pro-angiogenic effects, offering a promising therapeutic approach for brain repair following TBI.

Tanshinone and salvianolic acid biosynthesis are regulated by SmMYB98 in Salvia miltiorrhiza hairy roots

Xiaolong Hao,Zhongqiang Pu,Gang Cao,Dawei You,Yang Zhou,Changping Deng,Min Shi,Shivraj Hariram Nile,Yao Wang,Wei Zhou,Guoyin Kai

Salvia miltiorrhiza Bunge is an herb rich in bioactive tanshinone and salvianolic acid compounds. It is primarily used as an effective medicine for treating cardiovascular and cerebrovascular diseases. Liposoluble tanshinones and water-soluble phenolic acids are a series of terpenoids and phenolic compounds, respectively. However, the regulation mechanism for the simultaneous promotion of tanshinone and salvianolic acid biosynthesis remains unclear. This study identified a R2R3-MYB subgroup 20 transcription factor (TF), SmMYB98 , which was predominantly expressed in S. miltiorrhiza lateral roots. The accumulation of major bioactive metabolites, tanshinones, and salvianolic acids, was improved in SmMYB98 overexpression (OE) hairy root lines, but reduced in SmMYB98 knockout (KO) lines. The qRT-PCR analysis revealed that the transcriptional expression levels of tanshinone and salvianolic acid biosynthesis genes were upregulated by SmMYB98-OE and downregulated by SmMYB98-KO . Dual-Luciferase (Dual-LUC) assays demonstrated that SmMYB98 significantly activated the transcription of SmGGPPS1 , SmPAL1 , and SmRAS1 . These results suggest that SmMYB98-OE can promote tanshinone and salvianolic acid production. The present findings illustrate the exploitation of R2R3-MYB in terpenoid and phenolic biosynthesis, as well as provide a feasible strategy for improving tanshinone and salvianolic acid contents by MYB proteins in S. miltiorrhiza .

Comprehensive Analysis of the R2R3-MYB Transcription Factor Gene Family in Salvia miltiorrhiza and the Regulatory Role of SmMYB50 in Salvianolic Acid Metabolism

Fan Huiyan,Zhou Jingzhi,Ying Yiling,Tang Kaijie,Han Jiayi,Wang Qiuying,Kai Guoyin

Background (TF) R2R3-MYB transcription factors (TFs) play crucial roles in regulating plant secondary metabolism, which is vital for growth, development, and stress responses. Salvia miltiorrhiza , a medicinal plant, is known for its bioactive compounds such as salvianolic acids. Understanding the R2R3-MYB TF family in S. miltiorrhiza could provide insights into the regulation of secondary metabolism and contribute to quality improvement in this important medicinal plant. Methods and Results We conducted a comprehensive analysis of the R2R3-MYB TF family in S. miltiorrhiza , identifying 431 MYB-like TFs, including 149 R2R3-MYB TFs. Phylogenetic analysis classified these TFs into 27 distinct groups. Chromosomal localization analysis revealed their widespread distribution across all chromosomes. Structural analysis showed that most R2R3-SmMYB genes contain untranslated regions (UTRs) and conserved motifs, with motif5 potentially playing a key role in differentiation. Interspecific collinearity analysis demonstrated the highest collinearity between S. miltiorrhiza and Salvia baicalensis , with 12 intraspecific collinear genes exhibiting interspecific collinearity with multiple species. Promoter analysis identified abundant light- and hormone-responsive elements, particularly MeJA-responsive elements. Expression profiling highlighted SmMYB50 as highly responsive to MeJA, ABA, and YE induction, promoting salvianolic acid accumulation. Transgenic validation confirmed that SmMYB50 activates key biosynthetic genes such as SmCYP98A14 , crucial for SalB synthesis. Conclusions Our findings provide valuable insights into the functional roles of R2R3-SmMYB-TFs in regulating secondary metabolism in S. miltiorrhiza . SmMYB50 was identified as a key regulator, offering new directions for future research on metabolic pathways and quality improvement in this important medicinal plant.

Treating Acute Respiratory Distress Syndrome with a Multifaceted Nanomedicine: Inhibition of PANoptosis and Enhancement of Lung Barrier Integrity

Yanhui Cui,Xueqin Wang,Caiyang Lu,Liling Ran,Zirui Guo,Jiayu Yao,Tian Yu,Xuanxi Liu,Fang Li,Changqi Li,Yingcai Meng,Wenhu Zhou

Acute lung injury (ALI)/Acute Respiratory Distress Syndrome (ARDS) is a life-threatening condition marked by severe inflammatory responses and disruption of the alveolar-capillary barrier, leading to high mortality rates and lack of effective treatments. Recent research has underscored the crucial role of programmed cell death pathways-pyroptosis, apoptosis, and necroptosis-in exacerbating inflammation and barrier dysfunction in ALI/ARDS. However, effective therapeutic agents targeting this process remain scarce. In this study, we reveal that PANoptosis, which integrates these cell death pathways through the PANoptosome complex, plays a central role in the pathogenesis and progression of ALI/ ARDS. The levels of PANoptosis-related molecules were significantly elevated in both the ALI mice and the clinical ARDS patient samples, highlighting it as a novel therapeutic target. Building on this insight, we developed a multifunctional nanomedicine, TPNs/Sal B, which integrates tea polyphenol-based nanoparticles (TPNs), a bioactive nanomaterial with anti-pyroptotic and anti-necroptotic properties, with salvianolic acid B (Sal B), known for its anti-apoptotic effects. Our results demonstrate that the nanomedcine TPNs/Sal B effectively inhibit PANoptosis, thereby attenuating lung tissue pathological damage, reducing inflammation, and improving lung epithelial barrier function in ALI models. Moreover, we identified LIM and SH3 protein 1 (LASP1) which may play an critical role in modulating alveolar epithelial barrier function during ALI/ARDS progression, and treatment with TPNs/Sal B effectively restored LASP1 levels. Our findings underscore the therapeutic potential of TPNs/Sal B as a targeted treatment for ALI/ARDS, offering a promising strategy for modulating PANoptosis and preserving lung function.

Heme and lithospermic acid synergistically inhibit aggregation of human islet amyloid polypeptide: A novel hIAPP inhibitor for the potential therapy of type 2 diabetes

Bin Xiao,Junhao Xiao,Xiaoying Xiao,Sisi Liu,Hongmei Chen,Shengping Dai,Yan Sui,Huixian Ye

Amyloid deposition of human islet amyloid polypeptide (hIAPP) is closely linked to the pathogenesis and progression of type 2 diabetes mellitus (T2DM). Developing effective inhibitors to suppress hIAPP aggregation holds significant therapeutic potential for the prevention and treatment of T2DM. Recent researches indicate that both heme and lithospermic acid (LPA) can inhibit hIAPP aggregation. However, heme is prone to induce protein damage under oxidative stress, while LPA exhibits limited inhibitory efficacy despite its antioxidant properties. To overcome these limitations, we aimed to develop a dual-component inhibitor comprising heme and LPA. thioflavin T (ThT) fluorescence, transmission electron microscopy (TEM), circular dichroism (CD) and gel electrophoresis were combined to observe the inhibitory efficacy of heme-LPA co-formulation on hIAPP aggregation. The results demonstrate that LPA and heme can synergistically inhibit hIAPP aggregation. The inhibitory effect of heme-LPA co-formulation on hIAPP aggregation is significantly stronger than that of either component alone. The heme-LPA not only prevents the complete conversion of hIAPP into β-sheet fibrillar structures but also maintains its active monomeric conformation for extended periods. Furthermore, peroxidase activity assays revealed that the presence of LPA significantly reduces the peroxidase activity of heme in a concentration-dependent manner and attenuates peptide nitration damage under H₂O₂-NO₂ − oxidative stress. At a heme-to-LPA ratio of 1:4, peptide nitration bands were virtually undetectable. These findings indicate that the dual-component inhibitor heme-LPA represents an efficient and safe strategy for inhibiting hIAPP aggregation. This research provides an important avenue for developing novel anti-hIAPP aggregation inhibitors for the prevention and treatment of T2DM.

Rapid Endothelialization Enabled by Integrating Bi-oriented Topography and Salvianolic Acid B Modification on a Small-Diameter Fibrous Graft

Jixia Deng,Changlin Zhai,Lei Zhan,Kuihua Zhang,Tao Yang,Qinfei Ke,Chen Huang,Anlin Yin

The long-term reliability of small-diameter vascular grafts (SDVGs) is still largely confined by acute thrombosis, intimal hyperplasia and delayed endothelialization. In this study, a fibrous SDVG containing both physical and biochemical cues was developed to promote rapid endothelialization. The success of our method depends on the integration of a bi-oriented luminal surface with the conjugation of salvianolic acid B (Sal B) via aminolysis and subsequent amidation reaction. Specifically, longitudinally aligned poly (lactide acid-co-ε-caprolactone) (PLCL) microfibers having nano-grooved surface texture provided bi-oriented topographical cues to guide the elongation and migration of endothelial cells (ECs), whilst the incorporation of Sal B served as a biochemical signal to inhibit thrombosis, enhance EC growth and increase von Willebrand factor (vWF) expression in ECs. When implanted into a rat abdominal aorta defect model, the graft was able to facilitate luminal EC coverage, smooth muscle cells infiltration and collagen deposition without causing stenosis or thrombosis. The combination of bi-oriented topography with surface functionalization may become a platform design to maintain patency and accelerate in situ tissue regeneration of SDVG.
