Cuttlebone as a hemostatic traditional Chinese medicine is usually applied in powder directly to the wound for hemostasis. In order to improve the hemostatic utility of cuttlebone, gelatin/cuttlebone (Gel/Cb) membranes with different Gel/Cb ratios were fabricated through electrospinning and characterized by scanning electron microscopy, Fourier transform infrared spectroscopy, and X-ray diffraction. The hemostatic effect and coagulation process of Gel/Cb electrospun membranes were evaluated in vitro by dynamic whole-blood coagulation test, in vitro hemolysis test, determination of clotting time, and platelet adhesion test and in vivo by liver and tail vein hemostasis models. It was noted that the addition of cuttlebone could significantly decrease the hemolysis rates of Gel/Cb electrospun membranes, which was only about one-sixth of the Gel electrospun membrane. The animal experiments demonstrated effective reduction in bleeding time and volume in Gel/Cb-treated mice. Specially, compared with the commercially available gauze group. Gel/Cb (7:3) electrospun membrane reduced the clotting time by 80.92% and the average bleeding volume by 50.83%. The results further indicated that Gel/Cb electrospun membranes could accelerate coagulation, possibly via the coagulation pathway activation to stimulate platelet contraction and rapid aggregation at the wound surface, providing a new method for rapid hemostasis of acute trauma. Effective wound management remains a major clinical challenge, especially in preventing infections and promoting rapid healing. However, traditional wound dressings often lack multifunctionality, limiting their ability to support hemostasis and antibacterial protection. To address this challenge, in this work, a novel wound dressing was developed utilizing nano-cuttlebone particles, which are rich in calcium carbonate and chitin, to enhance hemostatic and antimicrobial functionality. The dressing, composed of polyvinyl alcohol, sodium alginate, and nano-cuttlebone, addresses the need for advanced wound care solutions. The particle size distribution of the powder was evaluated using a laser particle size analyzer, and the particle size of the nano-cuttlebone was around 75 nm. The drug release experiment showed that the drug release of nano cuttlebone hydrogel was 1.4 times that of micro cuttlebone hydrogel in 48 h. The antibacterial activity of the nano-cuttlebone-doped dressing against S. aureus was 97.32% and against E. coli was 99.99%. The cell scratch test showed that the nano-cuttlebone hydrogel gradually migrated to the central area, and its migration rate was 61.43% after 12 hours. The hydrogel also demonstrated excellent hemostatic ability in a mouse liver injury model and could rapidly stop bleeding within 1 min. In a full-thickness wound model, the nano-cuttlebone hydrogel enhanced collagen deposition and promoted faster wound closure. This study highlights the potential of marine biomaterials for developing multifunctional wound dressings to improve patient outcomes.

The harmful effects of antibiotics on aquatic environments have become a growing concern of modern society. Developing high-performance photocatalysts capable of degrading antibiotics under solar light is, therefore, crucial. In this study, TiO2-cuttlebone composites are prepared via the sol–gel method, to produce carbonate radicals (•CO3−) under solar light irradiation. The •CO3−radicals exhibit high selectivity for the degradation of tetracycline hydrochloride (TC). Compared to TiO2alone, the TiO2-cuttlebone composite demonstrates excellent solar-driven photocatalytic activity for TC degradation in both freshwater and seawater. The reaction pathways of TC degradation in seawater are elucidated using HPLC-MS/MS analysis. Moreover, a TiO2-cuttlebone self-suspending photocatalyst device is fabricated using 3D printing technology and low-temperature deposition methods, with aluminum–plastic (AP) as a substrate. This innovative device is easily recyclable from photocatalytic solutions while maintaining high stability, making it highly desirable for practical applications.

While the regulatory roles of biomaterial topology in bone regeneration are recognized, the specific osteogenic mechanisms driven by distinct morphological features, particularly without biochemical cues, remain poorly understood. This study aimed to independently elucidate the osteogenic potential and underlying mechanisms solely attributable to the distinctive “S”-grooved topology derived from cuttlebone. Inspired by cuttlebone, we successfully fabricated a biomimetic polycaprolactone (PCL) membranes sheet with “S”-grooved topology using liquid silicone mold replication combined with PCL recasting techniques. Comprehensive characterization (SEM, CLSM) confirmed precise replication of the native “S”-grooved morphology (width: 100 ± 30 μm, depth: 20 ± 6 μm). In vitro studies revealed that these grooves promoted directional adhesion and proliferation of rat bone marrow mesenchymal stem cells (rBMSCs) via contact guidance, as evidenced by cytoskeleton alignment. Furthermore, the “S”-grooved topology significantly enhanced osteogenic differentiation of mouse pre-osteoblasts (MC3T3-E1), demonstrated by increased alkaline phosphatase (ALP) activity and mineralized nodule formation. Mechanistic investigations using RT-qPCR, Western blot, and dimethyl labeling-based quantitative proteomics on rBMSCs identified key involvement of the Rap1-ERK signaling pathway. Specifically, the “S”-grooved topology upregulated Rap1 expression and enhanced ERK phosphorylation, leading to increased expression of osteogenic markers (e.g., Runx2, OPN). These findings demonstrate that the biomimetic “S”-grooved topology alone, through contact guidance and activation of the Rap1-ERK mechanotransduction pathway, significantly enhances osteogenic differentiation, providing a foundation for designing topologically optimized biomaterials for bone regeneration biomimetic PCL membranes sheet with the “S”-grooved topology.
