Totally biodegradable poly(trimethylene carbonate/glycolide-block-L-lactide/glycolide) copolymers: synthesis, characterization and enzyme-catalyzed degradation behavior

Wu Xiaomeng,Chen Xiaoyu,Fan Zhongyong

The macroinitiator of poly(1,3-trimethylene carbonate- ran -glycolide) (P(TMC- ran -GA)) copolymers terminated with hydroxyl groups were synthesized by ring-opening polymerization (ROP) at 120 °C for 12 h. Then, the novel block copolymers consisting of P(TMC- ran -GA) segments bonding with L -lactide ( L LA) and glycolide (GA) random copolymerization segments were prepared. The poly(TMC/GA- block - L LA/GA)) block copolymers were synthesized by ROP in bulk at 130 °C with high M n ¯ (above 2.0 × 10 5  Da). Such kind of block copolymers composed of P(TMC- ran -GA) and P( L LA- ran -GA) segments show good mechanical properties and adjustable degradation rate. The molecular structures of block copolymers were characterized by GPC, FTIR, and 1 H NMR. Chain microstructure analysis was performed in detail with 13 C NMR spectroscopy. The effect of GA units content on thermal behaviors, mechanical properties as well as biodegradability of copolymers was investigated by DSC, XRD, stress-strain measurements and enzymatic degradation tested in comparison with corresponding block copolymers P(TMC- block - L LA/GA). The results shown that amorphous P(TMC- ran -GA) segments significantly improve the flexibility of block copolymers. The incorporation of GA units strongly decreases the crystallization ability of L LA segment within copolymers due to more random L LA-GA sequence and shorter average L LA block length. Meanwhile, the tensile strength of material and flexibility are significantly improved with the raising GA units content. The enzymatic degradation rate is also accelerated with the increase GA unit content. Thus, the biodegradable block copolymers with unique molecular structure and modifiable properties are promising new materials for biomedical applications.

Degradable polyurethane for marine anti-biofouling

Chunfeng Ma,Liguo Xu,Wentao Xu,Guangzhao Zhang

Degradable polyurethane (PU) with copolyester oligomer consisting of ε-caprolactone (CL) and glycolide (GA) as the soft segments has been prepared by a combination of ring-opening polymerization and condensation reaction. Enzymatic and hydrolytic degradation experiments demonstrate that the PU can degrade in seawater. Such a polyurethane exhibit a more rapid degradation in comparison with that with poly(ε-caprolactone) (PCL) soft segments because the introduction of GA can reduce the crystallinity, as revealed by differential scanning calorimetry (DSC) and polarizing optical microscope (POM). Marine field tests show that the degradable polyurethane has good antifouling ability due to its self-renewal property. Besides, such polyurethane can serve as a carrier and controlled release system for an antifoulant, and the incorporation of an antifoulant in the polyurethane can significantly improve the antifouling ability and duration.

Drug release and biocompatibility of self-assembled micelles prepared from poly (ɛ-caprolactone/glycolide)-poly (ethylene glycol) block copolymers

Feng Su,Xiangke Sun,Rongye Li,Yuandou Wang,Laishun Xi,Yangsheng Chen,Suming Li

A series of poly(ɛ-caprolactone/glycolide)-poly(ethylene glycol) (P(CL/GA)-PEG) diblock copolymers were prepared by ring opening polymerization of a mixture of ɛ-caprolactone and glycolide using mPEG as macro-initiator and stannous octoate as catalyst. Self-assembled micelles were prepared from the copolymers using nanoprecipitation method. The micelles were spherical in shape. The micelle size was larger for copolymers with longer PEG blocks. In contrast, the critical micelle concentration of copolymers increased with decreasing the overall hydrophobic block length. Drug loading and drug release studies were performed under in vitro conditions, using paclitaxel as a hydrophobic model drug. Higher drug loading was obtained for micelles with longer poly(ε-caprolactone) blocks. Faster drug release was obtained for micelles of mPEG2000 initiated copolymers than those of mPEG5000 initiated ones. Higher GA content in the copolymers led to faster drug release. Moreover, drug release rate was enhanced in the presence of lipase from Pseudomonas sp., indicating that drug release is facilitated by copolymer degradation. The biocompatibility of copolymers was evaluated from hemolysis, dynamic clotting time, and plasma recalcification time tests, as well as MTT assay and agar diffusion test. Data showed that copolymer micelles present outstanding hemocompatibility and cytocompatibility, thus suggesting that P(CL/GA)-PEG micelles are promising for prolonged release of hydrophobic drugs.

Biodegradable poly(trimethylene carbonate-b-(L-lactide-ran-glycolide)) terpolymers with tailored molecular structure and advanced performance

Xiaoyu Chen,Xiaomeng Wu,Zhongyong Fan,Qinghua Zhao,Qing Liu

The macroinitiator of poly(1,3-trimethylene carbonate) (PTMC) with number-average molecular weight ( ) of 9.6 × 10 3 g mol −1 was synthesized by ring-opening polymerization at 120°C. Then, the novel terpolymer P(TMC- b -( L LA- ran -GA)) consisting of PTMC homopolymer segment attached with various monomer molar ratios of L -lactide ( L LA) and glycolide (GA) random copolymerization block was prepared with about 5.0 × 10 4 g mol −1 by ring-opening polymerization in bulk at 140°C. The tailored molecular structures of P(TMC- b -( L LA- ran -GA)) were characterized by 1 H nuclear magnetic resonance, 13 C NMR, FTIR, and gel permeation chromatography, and chain microstructure analysis was performed in detail with 13 C NMR spectroscopy. The effect of GA units on the thermal and crystallization behaviors, mechanical properties, as well as biodegradability of terpolymers was investigated by differential scanning calorimetry, wide-angle X-ray diffraction, stress-strain measurements, and in vitro tests in comparison with corresponding poly(trimethylene carbonate- block - L -lactide) copolymer P(TMC- b - L LA). The results show that amorphous PTMC segments have a significant effect on condensed state behavior of the terpolymers, and the incorporation of GA units strongly decreases the crystallinity and crystallization ability of L LA segment within terpolymers because of more random L LA-GA sequence and shorter average L LA block length. Meanwhile, the toughness of materials is greatly improved, and in vitro degradation is also accelerated. Peripheral vascular stents were 3D printed for the first time and met the requirements for application. The results show totally biodegradable terpolymers with unique molecular structure, and modifiable properties are promising new biomaterials with advanced performance for biomedical application.

MBG/ PGA-PCL composite scaffolds provide highly tunable degradation and osteogenic features

Jiangfeng Li,Chunyi Wang,Guoxing Gao,Xing Yin,Ximing Pu,Bing Shi,Yang Liu,Zhongbing Huang,Juan Wang,Jingtao Li,Guangfu Yin

It remains a challenge to achieve satisfactory balance between biodegradability and osteogenic capacity in biosynthetic bone grafts. In this study, we aimed to address this challenge by incorporating mesoporous bioactive glass (MBG) into poly(caprolactone-co-glycolide) (PGA-PCL) at gradient ratios. MBG/PGA-PCL (PGC/M) scaffolds with MBG incorporation ratio at 0, 10%, 25% and 40% (PGC/M0-40) were synthesized using a modified solvent casting-particulate leaching method, and their physiochemical and biological properties were comprehensively evaluated. PGC/M scaffolds exhibited highly perforated porous structure with a large-pore size of 300–450 μm, with ordered MBGs of around 6.0 nm mesopores size uniformly dispersed. The increase in MBG incorporation ratio significantly improved the scaffold surface hydrophilicity, apatite-formation ability and pH stability, increased the weight loss rate while insignificantly influenced the molecular chains degradation of PGA-PCL component, and facilitated the attachment, spreading, viability and proliferation of rat bone marrow stromal cells (rBMSCs) on scaffolds. Moreover, rBMSCs cultured on PGC/M10-40 scaffolds demonstrated enhanced ALP activity and osteogenesis-related gene expression in a MBG dose-dependent manner as compared with those cultured on PGC/M0 scaffolds. When implanted to the rat cranial bone defect, PGC/M25 and PGC/M40 scaffolds induced significantly better bone repair as compared to PGC/M0 and PGC/M10 scaffolds. Besides, the biodegradability of PGC/M scaffolds correlated with the MBG incorporation ratio. These data suggested this novel PGC/M scaffolds as promising bone repair biomaterial with highly tunable hydrophilicity, bioactivity, cytocompatibility, osteogenic activity as well as biodegradability.

A high-performance liquid chromatography method for detecting acylation impurities in acetate triptorelin microspheres for injection

Yan-nan Zan,Yu Chen,Fei Xie,Shao-hua Shang,Ning Chen,Yi-mei Ding

An efficient high-performance liquid chromatography (HPLC) method was established to detect acylated impurities in triptorelin acetate microspheres for injection. Additionally, HPLC coupled with quadrupole time-of-flight tandem mass spectrometry (HPLC-QTOF-MS) was employed to identify the types of acylated impurities. In HPLC, the separation of triptorelin and its acylated impurities was achieved on a Welch Ultimate XB-C18 column (250 mm × 4 mm, 5 μm). The mobile phase consisted of a 20 mmol·L -1 aqueous solution of ammonium acetate (pH 6.9) and acetonitrile, delivered in a gradient elution mode with a flow rate of 0.5 mL min -1 . The detection wavelength was set at 210 nm. For the HPLC-QTOF-MS method, the aforementioned HPLC conditions were utilized, and the mass spectrometer employed an electrospray ionization (ESI) ion source in positive ion mode. The scanning range spanned from 100 to 2000 m/z . The carrier gas was N 2 , with an atomization gas flow rate of 3 L·min -1 . The interface voltage was maintained at 4.5 kV, the desolvation temperature at 250 °C, and the drying gas flow rate at 15 L·min -1 . Additionally, the heating block temperature was set at 400 °C, and the interface temperature at 350 °C. Results Using this method, 14 types of acylated impurities in triptorelin were identified, further confirming that the acylation site of triptorelin is the serine alcohol hydroxyl group. Conclusion Both the HPLC and HPLC-QTOF-MS methods can be effectively utilized for routine quality control analysis of triptorelin acetate microspheres for injection.

Collagen-Inspired 3D Printing Electrospinning Biomimetic Patch for Abdominal Wall Defect Regeneration

Tao Yinghua,Luo Peiyu,Jing Fengya,Liu Tao,Tan Xin,Lyu Zhiyang,VeerleBernaerts Katrien,Zhang Tianzhu,Jia Ruipeng

Repairing abdominal wall defects presents significant challenges, due to the high infection risk, poor biocompatibility, and insufficient mechanical strength associated with synthetic materials. To overcome these limitations, we developed a bioinspired multifunctional 3DPF patch by integrating 3D printing and electrospinning technologies. The core material of the patch is 4arm-PLGA-GPO (4A-GPO), synthesized by conjugating the Gly-Pro-Hyp (GPO) peptide sequence with 4arm-PLGA(4A), which significantly enhances bioactivity and mechanical properties. Additionally, the patch encapsulates basic fibroblast growth factor (bFGF) to stimulate cell proliferation and migration, while an antibacterial layer composes of emodin (EMO) and tobramycin to prevent infection. In vivo studies demonstrate the 3DPF patch effectively accelerates tissue repair by reducing fibrosis and adhesions, promoting angiogenesis and collagen deposition, and modulating the immune response. Transcriptomic analysis reveals that the patch downregulates IL-17 mediated inflammatory pathways while upregulating cell adhesion molecule-related pathways, synergistically facilitating microenvironment reconstruction. Furthermore, molecular docking studies suggest the patch interacts with key molecules such as VEGF and COL3, enhancing angiogenesis and matrix remodeling. In summary, this biomimetic patch, composed of bioactive materials with well-defined chemical compositions, integrates mechanical support, immune modulation, and antibacterial protection. by offering a comprehensive solution for abdominal wall repair, it holds significant potential for clinical translation in complex tissue engineering applications.

Enhancing biocompatibility and degradation control of biodegradable polymers through amino acid grafting: A study on 4arm-PLGA-Amino acid copolymers

Yinghua Tao,Xin Tan,Tianzhu Zhang

Non-degradable polymers, when retained in the body for long periods, can cause chronic inflammation and immune responses. This issue increases the costs related to secondary surgeries and overall healthcare. Conversely, while biodegradable polymers such as PLGA offer an alternative, their degradation rate is challenging to precisely control. Additionally, their mechanical properties deteriorate during the degradation process, and the degradation by-products may lead to localized acidification or other adverse reactions. In this study, we introduce a new biodegradable copolymer called 4arm-PLGA-Amino acid (4aPLGA-Amino acid). This copolymer is synthesized by grafting amino acids onto 4arm-PLGA (4aPLGA) using triphosgene, which is both environmentally friendly and cost-effective. The incorporation of amino acids aims to enhance the biocompatibility and functionality of the copolymer. By varying the types of amino acids grafted, the degradation rate of 4aPLGA can be modulated, thereby improving its potential for applications in drug delivery systems. This modification also promotes cell attachment and proliferation, making it suitable for tissue engineering and regenerative medicine. The study demonstrated that 4aPLGA-Amino acid significantly improved biocompatibility, degradation control, and mechanical properties, broadening its applicability in various biomedical engineering fields.

Assembled granular hydrogels loaded with growth factors for enhanced mesenchymal stem cell therapy in abdominal wall defect repair

Liuxin Yang,Fengya Jing,Dandan Wei,Xiaocong Zhao,Yinghua Tao,Tao Liu,Tianzhu Zhang

Abdominal wall defects caused by trauma, congenital rupture, and intra-abdominal infection remain challenging due to the large wound area and complex complications. Herein, an assembled mesenchymal stem cell (MSCs)-laden granular hydrogel (termed assembled GSD@FPs), loaded with basic fibroblast growth factor (bFGF) and platelet-derived growth factor (PDGF), is developed. This hydrogel is constructed through dynamic covalent cross-linking ( via borate ester bonds) among dopamine-grafted gelatin methacrylamide (GelMA-DA), phenylborate-modified hyaluronic acid (HA-PBA), and epigallocatechin-3-gallate (EGCG), serving as multifunctional bulk building blocks for cell delivery and abdominal wall repair. The designed assembled granular hydrogels possess good rheological properties, self-healing, injectability, and tissue-adhesion properties. Detailed in vitro cell experiments are conducted, revealing that the GSD@FPs granular hydrogels can effectively promote cell proliferation, cell migration and angiogenesis. Furthermore, in abdominal wall defects, assembled GSD@FPs significantly accelerates the tissue healing process by simultaneously inhibiting the inflammatory response, promoting collagen deposition, and promoting cell proliferation and angiogenesis. Importantly, the assembled GSD@FPs granular hydrogels can also provide mechanical support and increase the thickness of regenerated tissue (1727.8 ± 169.6 μm for the control group, 3204.2 ± 278.5 μm for the assembled GSD@FPs group at 14 d). Eventually, the GSD granular hydrogels biodegraded, facilitating tissue remodeling and generating new muscle tissues. Therefore, this study provides a promising strategy with great potential for application in abdominal wall repair.

Thermo-sensitive PLGA-PEG-PLGA hydrogel for sustained release of EGF to inhibit cervical cancer recurrence

Li Piao,Peng Xiang,Yangyang Zhou,Wei Zhao,Ting Yang,Shan Xia,Guanghui Gao,Kexin Chen,Dan Li

Overexpression of epidermal growth factor receptor (EGFR) in cancer is a key cause of recurrence of cervical cancer (CC). Although the EGF-EGFR pathway has been studied for decades, preventing tumor growth and recurrence caused by peripheral EGF remains a great challenge. In this work, a strategy is proposed to reduce the stimulation of high concentration EGF on tumor growth by using a thermo-sensitive hydrogel. The hydrogel is a triblock copolymer composed of polyethylene glycol (PEG) and poly (lactide glycolide) (PLGA). Based on the excellent temperature sensitivity, carrier capacity, swelling property and biocompatibility, the hydrogel can absorb the liquid around the tumor by injection and release EGF continuously at low concentration. The inhibitory effect of hydrogel on tumor growth is fully confirmed by an implanted tumor mouse model with human cervical cancer cell lines (HeLa) using triple-immunodeficient NCG mice. Compared with free EGF, the EGF-loaded hydrogel can hardly induce surface plasmon resonance (SPR) response, which proves that hydrogel can effectively weaken cytoskeleton rearrangement and inhibit cell migration by continuously releasing low concentration EGF. In addition, the EGF-loaded hydrogel can reduce cell proliferation by delaying the progress of cell cycle progression. Taken together, the hydrogel can effectively protect tumor microenvironment from the stimulation of high concentration EGF, delay cancer cellular processes and tumor growth, and thus providing an approach for inhibiting tumor recurrence of CC.

Spatiotemporal delivery of microenvironment responsive hydrogel incorporated with stem cells-loaded porous microspheres for abdominal wall repair

Liuxin Yang,Dandan Wei,Yinghua Tao,Fengya Jing,Tao Liu,Guanhua Jiao,Tianzhu Zhang

Healing the wounds of abdominal wall defects characterised by severe complications represents a significant clinical challenge. Mesenchymal stem cells (MSC)-based therapies have emerged as a promising strategy for tissue regeneration. However, the low engraftment and survival rates limit their application owing to the complicated wound microenvironment and large wound areas in abdominal wall defects. Inspired by focal biointerfaces, the present research engineered a spatiotemporal delivery microenvironment-responsive therapeutic platform, termed MSC-GSM@HG, for abdominal wall repair. The constructed platform proved capable of dissolving HA-PBA/Gel-DA (HG) hydrogels in response to in vivo inflammatory responses and subsequently releasing MSC-laden gelatine and silk fibroin microspheres (GSMs). Meanwhile, the HG hydrogels can not only protect the MSCs from excessive reactive oxygen species but also exhibit haemostatic and anti-inflammatory properties. Strikingly, this microsphere-releasing hydrogel platform actively enhanced the repair of abdominal wall defects in vivo , by reducing inflammatory response, facilitating angiogenesis and promoting collagen deposition. Therefore, the microenvironment-responsive platform represents a promising strategy for MSC-based therapies for abdominal wall defects through spatiotemporal delivery mechanisms.

TRAF-STOP alleviates osteoclastogenesis in periodontitis

Huang Yaxian,Wu Jinyan,Zhan Chi,Liu Rong,Zhou Zhaocai,Huang Xin,Tian Yaguang,Lin Zhengmei,Song Zhi

The enhanced osteoclastogenesis contributes to alveolar bone resorption in periodontitis, which increases the risk of tooth loss. To reduce bone destruction, the inhibition of osteoclast development is proposed as a feasible treatment. CD40L-CD40-TRAF6 signal transduction plays a crucial role in inflammation, but how it regulates osteoclast activity in periodontitis has not been elucidated. In this study, we showed the potential role of CD40L-CD40-TRAF6 signaling in periodontitis. CD40L obviously promoted osteoclast formation and bone resorption capacity in vitro. Mechanistically, we found that osteoclastogenesis was enhanced by the overexpression of NFATc1 and NF-κB activation. Importantly, osteoclast activity was effectively suppressed by TRAF-STOP, a small molecular inhibitor of TRAF6. Furthermore, local injection of TRAF-STOP-loaded injectable PLGA-PEG-PLGA hydrogel could alleviate ligation-induced periodontitis in vivo. Taken together, TRAF-STOP shows promising clinical efficacy in periodontitis through alleviating osteoclastogenesis.
