View large Download slide View large Download slide Close modal The environmental health risks of acetochlor (ACE) have received widespread attention. The removal of ACE via the ultraviolet/persulfate (UV/PS) process was investigated in this study. The degradation of ACE in the UV/PS process fitted pseudo-first-order kinetics. ACE can be effectively removed in the pH range of 5.0–9.0. In addition, the removal efficiency of ACE increases with the increase in PS concentration. The experimental and calculation results prove that the contributions of UV irradiation, sulfate radicals (SO 4 •- ) and hydroxyl radicals (HO·) were 40, 52.1, and 6.43%, respectively, and the second-order rate constants between SO 4 •- and with ACE are 3.5 × 10 9 and 5.6 × 10 9 M −1 s −1 , respectively. The influences of different water matrices were explored. Both chloride ions and bicarbonate can inhibit the degradation of ACE, while the effects of sulfate ions and nitrate can be negligible. Humic acid (HA) has an inhibitory effect of ACE degradation. The intermediates were detected and the possible degradation pathway of ACE was proposed. The results of this study can provide technical support for the control of ACE in the environment. The degradation of acetochlor in the UV/PS process was investigated. The kinetic model of acetochlor degradation in the UV/PS process was proposed. Both UV irradiation, SO 4 •- , and are involved in the degradation of acetochlor. The degradation pathway of acetochlor was proposed in the UV/PS process.

The hyporheic zone is a key area for water circulation. However, hyporheic zone pollution is widespread all over the world and remediations of hyporheic zones, when polluted by both heavy metals and organics, are challenging. Thus, the mechanisms and application of Calcium peroxide (CaO 2 ) coupled with Zero-valent iron (ZVI) to simultaneously remove cadmium (Cd), lead (Pb), and acetochlor were investigated in this work. The effects of mass ratio of ZVI and CaO 2 , initial pH, and water matrices were evaluated. At initial pH 3, with the dosage of 1.5 g·L −1 ZVI and 0.5 g·L −1 CaO 2 , the removal efficiencies of Cd, Pb, and acetochlor were 96.7%, 99.8%, and 100%, respectively. Acetochlor was degraded into four main byproducts, and the toxicity evaluation indicated that the ecotoxicity of acetochlor was reduced during the degradation by CaO 2 /ZVI. In column transport experiments, with the mass ratios of ZVI and CaO 2 to quartz sand of only 0.13% and 0.05%, the mass recovery efficiencies of Cd, Pb, and acetochlor from the effluent were 42.5%, 10.4%, and 0%, respectively. Cd was removed mainly by the adsorption of ZVI and iron(hydr)oxides (represented Fe 3 O 4 and lepidocrocite); removal of Pb chiefly resulted from ZVI reduction, and hydroxyl radicals derived from the Fenton reaction accounted for acetochlor degradation. This work provides new insights and a facile strategy for in-situ remediations of hyporheic zones polluted simultaneously by heavy metals and organics.

The aim of this work was to develop a fast, simple, and reliable UPLC-MS 3 method for the sensitive detection of acetochlor in biological samples. In MS 3 mode, the ion transition m/z 270.1 → 224.1→148.1 was chosen for quantification with butachlor as the internal standard. In the UPLC system, separation was performed on a UPLC column (2.1 × 50 mm ID, 1.7 μm) with 0.1 % FA in water and acetonitrile as mobile phases . After simple protein precipitation via acetonitrile, the method was well validated with good linearity (0.5–20 ng/mL, r > 0.995), accuracy (−3.70 %-2.98 %), and precision (<15 %). The selectivity and sensitivity were improved obviously in MS 3 mode than that in MRM mode. The developed UPLC-MS 3 method was successfully applied to the cellular pharmacokinetics study of acetochlor in MCF-7 cells.

The excessive use of herbicide acetochlor (ACT) threatens crop health and the environment, necessitating effective remediation strategies. This study focused on a consortium named AT1, enriched from ACT-contaminated soil. Under optimized conditions (25 °C, pH 7, 1 % inoculum), AT1 almost completely degraded ACT (50–1000 mg/L) within 6–12 days. High-throughput sequencing of 16S rRNA gene revealed a reduction in community diversity over time, with Sphingomonas (58.6 %) and Diaphorobacter (26.43 %) as dominant taxa. A structure model and network analysis indicated strong microbial competition during the peak degradation. Predicted functions and liquid chromatography-mass spectrometry based metabolomics data identified benzene ring intermediates during ACT degradation, including 2,6-dimethylaniline, resorcinol, phenol, 3-ethyl-1,2-benzenediol, 1,2,3-trihydroxybenzene, phloroglucinol, and benzene-1,2,4-triol. Joint omics analysis revealed that AT1 likely degrades ACT via N -dealkylation by Pseudomonas , amide bond hydrolysis by Diaphorobacter , and carboxylation and hydroxylation by Sphingomonas , leading to the formation of these intermediate metabolites. Moreover, AT1 efficiently degraded key intermediates, particularly 2,6-dimethylaniline, phenol, and resorcinol, further enhancing ACT mineralization. Notably, AT1 efficiently degraded ACT in soil, resulting in a significant decrease in ACT environmental residues. These findings provide valuable insights for the discovery and identification of herbicide-degrading bacterial resources and the metabolic transformation of herbicides, and developing approaches for pollution control and biodegradation of amide herbicides.

Acetochlor is a high-volume herbicide used on a global scale and toxicity assessments are needed to define its potential for adverse effects in wildlife and humans. This study was conducted to determine the effects of acetochlor on human liver carcinoma cells (HepG2), a cell model widely used to assess the potential for chemical hepatotoxicity. Experiments were conducted at concentrations ranging 0–800 μM acetochlor over a 12 to 48h period to quantify underlying mechanisms of toxicity. Our data indicate that acetochlor suppressed HepG2 cell proliferation in both a concentration- and time-dependent manner. Acetochlor induced reactive oxygen species (ROS) generation more than 700% with exposure to 400 μM acetochlor, and acetochlor decreased the activities and levels of anti-oxidant responses (superoxide dismutase, glutathione) following exposure to 100 μM, 200 μM and 400 μM acetochlor. Acetochlor also (1) induced HepG2 cell damage through apoptotic-signaling pathways; (2) enhanced intracellular free Ca 2+ concentration (>400%); (3) decreased mitochondrial transmembrane potential (∼77%), and reduced ATP levels (∼65%) following exposure to 400 μM acetochlor compared to untreated cells. Notably, cell cycle progression was blocked at G0/G1 phase in HepG2 cells when treated for 24 h with 400 μM acetochlor. Taken together, acetochlor induced significant cytotoxicity toward HepG2 cells, and the underlying toxicity mechanisms appear to be related to ROS generation, mitochondrial dysfunction and disruption in the cell cycle regulation. These data contribute to toxicity assessments for acetochlor, a high-use herbicide, to quantify risk to wildlife and human health.

Herbicide acetochlor (ACT) is widely used in agricultural production, but it remains unclear whether microplastics (MPs) from agricultural films exacerbate its harmful effects on soil health. Here, a microcosm cultivation experiment was conducted to investigate the impacts of PE (Polyethylene)-MPs and PBAT (polybutylene adipate terephthalate)-MPs with ACT (MPs/ACT) on soil health. Results showed that MPs/ACT treatment caused soil acidification, depletion of organic carbon and ammonium, altered enzyme activities, and influenced greenhouse gas emissions compared to ACT alone. MPs/ACT co-exposure also restructured microbial communities, reducing alpha diversity, increasing co-occurrence network complexity, enriching putative contaminant degraders, and enhancing the relative abundance of key nitrogen-cycling genes, enzymes, and carbon-cycling genes. Notably, PBAT/ACT stimulated nitrification-denitrification processes more than PE/ACT, indicating polymer-specific effects on soil nutrient cycling. Overall, MPs/ACT decreased soil health by 8.89 % - 14.63 %, greater than 8.39 % caused by ACT alone. These results demonstrate that MPs exacerbate the toxic effects of ACT on soil health. Mitigation strategies, such as reducing MP inputs, optimizing herbicide use, and incorporating biochar or microbial amendments, may help to restore soil health and maintain ecosystem stability. This study provides critical insights into the compounded impacts of MPs and herbicides on soil ecosystems.

A novel flower-like film composed of uniform hollow TiO 2 -NH 2 nano-shell (HTNs), luminol and aniline was synthesized by electro-copolymerization method in acidic ionic liquid EmimCl (IL, 1-ethyl-3-methylimidazolium chloride). The surficial morphology of the film was characterized by scanning electron microscope (SEM), transmission electron microscope (TEM), selected-area electron diffraction (SAED) and atomic force microscope (AFM), which showed the polycrystalline property. Its copolymerization mechanism was discussed in detail, which presented symmetric arrangement. The most obvious multiple increase of peak current in CV indicated the conjugate of HTNs in luminol structure and in aromatic ring skeleton. This film with excellent electrocatalysis performance used for nonelectroactive acetochlor (ACT) detection showed the lowest catalytic potential. Some important parameters were optimized for a homogeneous film and for a sensitive detection. On the optimum film, the electrocatalysis sensor exhibited a linear response within the concentration range of ACT from 2.0×10 −7 M to 1.5×10 −5 M, and an absolute detection limit of 0.15×10 -9 M at a signal-to-noise ratio of 3. Then the proposed method was used for the detection of ACT residue in the real strawberry sample with good results. The promising sensor presents good reproducibility and stability, providing a new fast and sensitive method for the analysis of real samples in environment.

Imazethapyr (IM) is an acetolactate synthase (ALS)-inhibiting herbicide that has been widely used in recent years. However, IM spraying can lead to the accumulation of herbicide residues in leaves. Here, we determined the effects of IM spraying on the plant growth and leaf surface microbial communities of Arabidopsis thaliana after 7 and 14 days of exposure. The results suggested that IM spraying inhibited plant growth. Fresh weight decreased to 48% and 26% of the control value after 7 and 14 days, respectively, of 0.035 kg/ha IM exposure. In addition, anthocyanin content increased 9.2-fold and 37.2-fold relative to the control content after 7 and 14 days of treatment, respectively. Furthermore, IM spraying destroyed the cell structures of the leaves, as evidenced by increases in the number of starch granules and the stomatal closure rate. Reductions in photosynthetic efficiency and antioxidant enzyme activity were observed after IM spraying, especially after 14 days of exposure. The diversity and evenness of the leaf microbiota were not affected by IM treatment, but the composition of community structure at the genus level was altered by IM spraying. Imazethapyr application increased the abundance of Pseudomonas , a genus that includes species pathogenic to plants and humans, indicating that IM potentially increased the abundance of pathogenic bacteria on leaves. Our findings increase our understanding of the relationships between herbicide application and the microbial community structures on plant leaves, and they provide a new perspective for studying the ecological safety of herbicide usage.

In this study, a weakly polar pesticide acetochlor in water was tried to degrade by BiOBr catalysts under visible light irradiation. Its degradation reaction rate increased exponentially with increasing catalyst dosage, which was significantly different from conventional photocatalytic patterns established for polar or water-soluble pollutants. Thus, the rate-dosage relationship of BiOBr/acetochlor was compared to that of BiOBr/Rhodamine B (RhB) (a typical strong polar dye pollutant in water) at different BiOBr dosages. The degradation rate of RhB increased with the BiOBr dosage and then plateaued due to more catalyst particles blocking the light into the inner solution, which is a typical behavior for heterogeneous photocatalysis of polar organic compounds in aqueous solution. While acetochlor behaved differently, its degradation rate increased exponentially with the BiOBr dosage, and no plateau was observed even up to 9.0 g/L BiOBr dosage. This special rate-dose relationship for weakly polar acetochlor was correlated with a unique adsorption property of BiOBr to acetochlor in water solution. There was a re-distribution effect that always exclude excessive acetochlor adsorbed on BiOBr surface regardless either substrates or BiOBr overdose. This finally resulted in every BiOBr particle having enough adsorption sites available for dissolved dioxygen to function thereby accelerate the degradation of weakly polar organic compounds in water.

Acetochlor is a widely used and highly effective herbicide. Its overuse poses significant threats to biosecurity and ecological integrity, particularly affecting free-ranging birds. Data on its impact, especially mechanisms of liver toxicity in chickens, are lacking. Thus, we established an animal-cell-animal model to explore intrinsic mechanisms at multiple levels. We found that acetochlor exposure caused liver cell swelling, inflammatory cell accumulation, and lipid deposition. Transcriptomic analyses revealed that differential gene were mainly enriched in hepatic immune, inflammatory, and programmed cell death pathways. We next focused on the gene GPR120, conducting transfection and agonism experiments in LMH, HD11, and co-cultured cells. Acetochlor significantly increased ROS accumulation, activated the NLRP3 inflammasome, and which induced PANoptosis. HD11 cells exhibited M1 polarization with upregulated pro-inflammatory factors. Silencing GPR120 exacerbated cellular damage and immune responses, whereas its agonist, GSK7A, dramatically reduced macrophage M1 polarization and mitigated immune damage to LMH cells. Finally, we returned to animal studies, adding Omega-3-a known GPR120 agonist-to the diet. Omega-3 effectively reversed acetochlor-induced hepatitis and PANoptosis. Given that acetochlor residues pose potential threats to ecosystems and avian health, it is crucial to strengthen residue control, conduct risk assessments, and explore targeted pathways and nutritional supplementation to counteract these negative impacts.

Acetochlor, a commonly used herbicide, poses significant risks to ecosystem and organism health through contamination of the food chain. Despite its widespread use, there is a lack of comprehensive studies on its toxicological effects on avian species. This study investigates the impact of environmental acetochlor exposure on chicken liver health using metabolomics analysis and histopathological techniques. Microscopic examination revealed autophagy-like structures and endoplasmic reticulum (ER) expansion, with significant effects observed at higher exposure levels. Biochemical analysis and metabolomics also demonstrated acetochlor-induced ferroptosis, highlighting disruptions in liver function. Further, in vitro studies revealed that acetochlor stimulates autophagy, which regulates ferroptosis via ferritin degradation, mediated through the ER-CaMKII pathway. These findings emphasize the importance of understanding the molecular mechanisms involved in acetochlor toxicity, particularly the role of the Ca 2+ /CaMKII pathway, ER stress, and autophagy in ferroptosis. The study contributes to a deeper understanding of how environmental contaminants affect avian species, providing critical insights for better herbicide risk assessment, pollution control, and sustainable agricultural practices.

Acetochlor is a high-volume herbicide whose widespread use threatens ecosystems and affects aquaculture. Apoptosis and autophagy are important causes of hepatotoxicity caused by toxicants, which can be mediated by oxidative stress and the inhibition of PPAR/RXR pathway. However, the mechanism of acetochlor on fish hepatocyte damage still needs to be further investigated. Therefore, we treated the Ctenopharyngodon idella hepatic cell line (L8824 cells) with different concentrations (10, 20, and 40 μM) of acetochlor and/or ROS scavenger NAC (1 mM) for 24 h. The results showed that acetochlor decreased the cell viability in a dose-dependent manner. AO/EB staining and flow cytometry verified the increased apoptotic rates . Quantitative analysis of gene expression levels or protein expression levels displayed that the expression levels of Beclin1, P62, LC3B, BAX, and cleaved Casp3 were increased, and the expression of BCL2 was reduced. Besides, we detected the increased ROS contents and decreased PPAR/RXR pathway expressions after acetochlor treatment. The clearance of ROS alleviated the inhibition of the PPAR/RXR pathway and lightened apoptosis and autophagy under acetochlor stress. Overall, these results revealed that acetochlor exposure triggered BCL2/BAX/Casp3-cascaded apoptosis and Beclin1-dependent autophagy through ROS-mediated PPAR/RXR inhibition. The results partially explain the toxicological mechanism of acetochlor and provide targets for the development of its antidote.
