A simple and rapid chemiluminescence aptasensor for acetamiprid in contaminated samples: Sensitivity, selectivity and mechanism

Yingying Qi,Fu-Rong Xiu,Minfeng Zheng,Baoxin Li

Ultralow concentration and selective detection of pesticide residue is important to evaluate the environmental and biological pollution and the threat to human health which single component pesticide can bring. Herein, we report an amplified chemiluminescence (CL) sensing platform for ultrasensitive and selective acetamiprid (widely used pesticide) detection. It is based on aptamer's high binding affinity to target and the relevance between AuNPs' morphology and its catalytic effect to stimulate the generation of CL in the presence of H 2 O 2 and luminol. Moreover, AuNPs morphological slight change induced by aptamers’ conformation during targets binding could lead to the significant change of catalytic properties . Therefore, the proposed sensing platform for pesticide residue exhibited a high sensitivity toward acetamiprid with a detection limit of 62   pM, which was about 100-fold lower than that of other aptamer-based sensor for acetamiprid detection. Because of the intrinsic specificity of aptamer's recognization, this sensing platform has high selectivity . So, this sensing platform provides a label-free and cost-effective approach for sensitive and selective detection of single component pesticide residue. More importantly, this CL method was successfully used to determine acetamiprid in real contaminated samples.

Aptamer-based Resonance Light Scattering for Sensitive Detection of Acetamiprid

Wang Chengke,Chen Dan,Wang Qingqing,Wang Qiuxia

In this work, an aptasensor-based resonance light-scattering (RLS) method was developed for the sensitive and selective detection of acetamiprid. The ABA (acetamiprid binding aptamer)-stabilized gold nanoparticles (ABA-AuNPs) were used as a probe. Highly specific single-strand DNA (ssDNA, i.e , aptamers) that bind to acetamiprid with high affinity were employed to discriminate other pesticides, such as edifenphos, kanamycin, metribuzin et. al. The sensing approach is based on a specific interaction between acetamiprid and ABA. Aggregation of AuNPs was specifically induced by the desorption of the ABA from the surface of AuNPs, which caused the RLS signal intensity to be enhanced at 700 nm. The alteration of AuNPs’ aggregation has been successfully optimized by controlling several conditions. Under the optimal conditions, the RLS intensity changes ( I/I0 ) of AuNPs were linearly correlated with the acetamiprid concentration in the range of 0–100 nM. The detection limit is 1.2 nM (3σ). This method had also been used for acetamiprid detection in lake water samples.

Quantum Dots-Based Immunochromatographic Strip for Rapid and Sensitive Detection of Acetamiprid in Agricultural Products

Liu Ying,Zhao Ying,Zhang Tianyi,Chang Yunyun,Wang Shuangjie,Zou Rubing,Zhu Guonian,Shen Lirong,Guo Yirong

In this study, a rapid and sensitive immunochromatographic strip (ICS) assay, based on quantum dots (QDs), was developed for the qualitative and quantitative detection of acetamiprid in agricultural samples. Acetamiprid-ovalbumin conjugates (ACE-OVA) and goat anti-mouse IgG were sprayed onto a nitrocellulose membrane as a test and control line. Two kinds of anti-acetamiprid monoclonal antibodies (mAb) obtained in our lab were characterized by the ELISA and surface plasmon resonance assay. The competitive immunoassay was established using a QDs-mAb conjugate probe. The visual detection limit of acetamiprid for a qualitative threshold was set as 1 ng/mL to the naked eye. In the quantitative test, the fluorescence intensity was measured by a portable strip reader and a standard curve was obtained with a linear range from 0.098 to 25 ng/mL, and the half maximal inhibitory concentration of 1.12 ng/mL. The developed method showed no evident cross-reactivities with other neonicotinoid insecticides except for thiacloprid (36.68%). The accuracy and precision of the developed QDs-ICS were further evaluated. Results showed that the average recoveries ranged from 78.38 to 126.97% in agricultural samples. Moreover, to test blind tea samples, the QDs-ICS showed comparable reliability and a high correlation with ultra-performance liquid chromatography-tandem mass spectrometry. The whole sample detection could be accomplished within 1 h. In brief, our data clearly manifested that QDs-ICS was quite qualified for the rapid and sensitive screening of acetamiprid residues in an agricultural product analysis and paves the way to point-of-care testing for other analytes.

Determination of acetamiprid using electrochemiluminescent aptasensor modified by MoS2QDs-PATP/PTCA and NH2-UiO-66

Li Jingxian,Jiang Ding,Shan Xueling,Wang Wenchang,Ou Guofu,Jin Haozhe,Chen Zhidong

A novel aptasensor has been fabricated based on the resonance energy transform (RET) system from MoS 2 QDs-PATP/PTCA (donor) to NH 2 -UiO-66 (acceptor). The electrochemiluminescence (ECL) signal of PTCA was greatly amplified due to the decoration of MoS 2 QDs-PATP, and the NH 2 -UiO-66 was utilized to label the signal probe DNA (pDNA), which hybridizes with the exposed aptamer anchored on the surface of MoS 2 QDs-PATP/PTCA. With the target acetamiprid, the specific binding of acetamiprid to aptamer causes the connection between the donor and the acceptor to be interrupted and produce an “on” ECL signal. Thus, an “off-on” ECL sensing platform for sensitive and selective acetamiprid assay was designed. Under the optimal condition, the ECL signal of the aptasensor was found to be linearly related to the logarithm of the acetamiprid concentration ranging from 0.1 fM to 0.1 μM with a detection limit of 0.064 fM. More importantly, the recovery rate of the ECL aptasensor was calculated to be 98.7 ~ 106% with a RSD lower 5.1% for the residual acetamiprid assay in real food samples, which indicated that the aptasensor has high potential for practical applications. Anti-Agglomeration Behavior and Sensing Assay of Chlorsulfuron Based on Acetamiprid-Gold Nanoparticles

Guangyang Liu,Ruonan Zhang,Lingyun Li,Xiaodong Huang,Tengfei Li,Meng Lu,Donghui Xu,Jing Wang

Monitoring of low levels of chlorsulfuron in environmental water samples is important. Although several detection methods have been developed, they all have some drawbacks, such as being time-consuming, requiring expensive instruments and experienced operators, and consuming large volumes of organic solvents. There is an urgent need for a simple, rapid, and inexpensive detection method for chlorsulfuron. Herein, such a method was developed using anti-aggregation of gold nanoparticles (AuNPs) in the presence of acetamiprid in agricultural irrigation water samples. Aggregation of the AuNPs was induced by acetamiprid, and this produced a distinct color change from Bordeaux red to blue. However, the strong hydrogen bonding interaction between chlorsulfuron and acetamiprid could inhibit AuNP aggregation. The effect of chlorsulfuron on the anti-agglomeration behavior of AuNPs was monitored by ultraviolet–visiblespectroscopy (UV-Vis) and the naked eye over a concentration range 0.1–100 mg/L. The detection limit for chlorsulfuron was 0.025 mg/L (signal-to-noise ratio of three). This colorimetric method was successfully applied to the determination of chlorsulfuron in spiked tap water and agricultural irrigation water with satisfactory recoveries (76.3%–94.2%).

Impedimetric aptasensor based on highly porous gold for sensitive detection of acetamiprid in fruits and vegetables

Yiwei Xu,Wen Zhang,Jiyong Shi,Zhihua Li,Xiaowei Huang,Xiaobo Zou,Weilong Tan,Xinai Zhang,Xuetao Hu,Xin Wang,Chao Liu

A novel electrochemical aptasensor modified with highly porous gold and aptamer was prepared for the determination of acetamiprid in fruits and vegetables. Highly porous gold was synthesized by electroreduction at −4 V in an electrolyte containing 2.5 mol/L NH 4 Cl and 10 mmol/L HAuCl 4 . Acetamiprid-binding aptamer was immobilized on highly porous gold by self-assembly. Acetamiprid could be captured by aptamer on the sensing interface, resulting in an increment of electron transfer resistance. Thanks to the large specific surface area of highly porous gold and the high affinity of aptamer, the aptasensor exhibited a highly sensitive impedance response for acetamiprid. Under optimal condition, the aptasensor displayed a linear response for acetamiprid in the concentration range of 0.5–300 nmol/L, and the detection limit was 0.34 nmol/L. Furthermore, the aptasensor showed high selectivity, good reproducibility and stability. Finally, the aptasensor was applied for the determination of acetamiprid in fruits and vegetables with satisfactory results.

A needle-like reusable surface-enhanced Raman scattering substrate, and its application to the determination of acetamiprid by combining SERS and thin-layer chromatography

Kang Yan,Wu Ting,Han Xiaozhen,Gu Haixin,Zhang Xin

A micro surface-enhanced Raman scattering (SERS) substrate has been fabricated by electrochemical deposition of dendrite-like gold on carbon fiber needles (Au-CFNs). Scanning electron microscopy and energy dispersive spectroscopy were used to confirm the presence of the gold nanostructure on the CFNs. This substrate has a Raman scattering enhancement factor as high as 3.3 × 10^7 when using rhodamine 6G as the reporter molecule. The high SERS sensitivity is attributed to the massive hotspots on gold bulges that enhance the local surface plasmon resonance. The Au-CFN substrate was reproduced 10 times after electrochemically wiping off the analytes from the needle-like electrode. The substrate has attractive features such as convenient sampling, low sample dosage, and minimal invasion. It was applied, in combination with thin-layer chromatography, for the determination of acetamiprid on vegetables. The result was more accurate because the sample information of both the surface and the bulk can be obtained at the same time after inserting the tip of this needle substrate into the TLC plate. The limit of detection for acetamiprid is 0.05 μg⋅mL -1 and the linear range is 0.1–10 μg⋅mL -1 . A reusable micro needle-like SERS substrate was fabricated and applied for pesticide residue analysis. The SERS signal of acetamiprid can be acquired on the tip of this micro needle-like Au-CFN substrate. This substrate can be reused for 10 times.

Mechanochemical modification of kaolin surfaces for immobilization and delivery of pesticides in alginate-chitosan composite beads

Yan Huiqiong,Chen Xiuqiong,Wu Tiantian,Feng Yuhong,Wang Chunxiu,Li Jiacheng,Lin Qiang

The surface modification with poly(methyl hydrosiloxane) (PMHS) using a grinding method as a pesticide carrier had been successfully achieved. The characteristics of the modified kaolin were represented by Fourier transform infrared, X-Ray diffraction, BET specific surface area measurements, laser particle size analysis, transmission electron microscopy and contact angle measurement. Subsequently, immobilization of acetamiprid onto clay’s surface was carried out by means of freeze-drying technology. And the drug-loaded clays were encapsulated in alginate-chitosan composite beads by extrusion into calcium chloride solution. The composite beads showed regularly spherical shapes and uniform sizes with diameters of 1.44 ± 0.24–1.75 ± 0.19 mm. The immobilization and release of acetamiprid were carried out to examine the effect of the modified kaolin on the properties of the composite bead. The results showed that the intensive force of superfine pulverization during the grinding process was effective to increase the specific surface areas of the modified kaolin with particle size reduction. The modified kaolins as promising carrier materials had improved the drug loading rate and encapsulation efficiency from 4.15 and 42.01 % for alginate-chitosan composite beads to 6.02 and 82.95 %, which were higher than 4.33 and 54.29 % for SA-CS-kaolin composite beads, respectively. The controlled-release formulation containing the modified kaolin had shown slow and sustained release properties to promote the efficient use of organic pesticide to reduce pollution effect on the environment.

Highly efficient analysis of fipronil and ethiprole in milk using ternary deep eutectic solvent-functionalized UiO-66-based tip-SPE − HPLC

Yehong Han,Ruiyang Ma,Juntao Wang,Hongyuan Yan,Yufei Shan,Zihan Liu,Zhi Wang

In this study, for the effective determination of fipronil and ethiprole residues in milk, a new analytical approach, which was based on a tip solid-phase extraction (TSPE) by using a newly prepared ternary deep eutectic solvent-functionalized UiO-66 (TDES@UiO-66) as the adsorbent and HPLC-UV detection, was developed. The method integrated the advantages of both the high adsorption capability of the TDES@UiO-66 and the high efficiency of the TSPE, and enabled a rapid extraction of fipronil and ethiprole from milk samples within 8 min by utilizing only 3.0 mg of TDES@UiO-66 adsorbent. The developed method demonstrated an excellent analytical performance, including good linearity (linear rang: 0.02–10 μg/mL; r : 0.9990–0.9997), high precision (relative standard deviations: ≤ 9.9 %), good accuracy (recoveries: 91.1 %–109.8 %), and good sensitivity (limits of quantification: 0.011–0.018 μg/mL). This study provides a rapid and practical approach for the detection of fipronil and ethiprole in different milk samples.

Enhancing soil aggregation and acetamiprid adsorption by ecofriendly polysaccharides hydrogel based on Ca2+- amphiphilic sodium alginate

Siqi Zhang,Furui He,Xiuqin Fang,Xinyu Zhao,Yuanyuan Liu,Gaobo Yu,Yang Zhou,Yuhong Feng,Jiacheng Li

Soil aggregation plays an important role in agricultural production activities. However, the structure of soil aggregation is destroyed by the natural environment and unreasonable farming management, resulting in the loss of water, fertilizers and pesticides in soil. At present, hydrogels have been widely reported to promote the formation of soil aggregation. In this paper, amphiphilic calcium alginate (ASA/Ca 2+ ) was applied to promote the formation of soil aggregation and enhance pesticide retention. Initially, an ASA was obtained through the one-pot Ugi condensation (a four-component green chemical reaction). Then, ASA/Ca 2+ hydrogel is prepared by Ca 2+ cross-linking. The formation of soil aggregation was determined through the Turbiscan Lab Expert stability analyzer, Confocal Laser Scanning Microscope (CLSM), and Transmission Electron Microscope (TEM). And the effect of soil aggregation on acetamiprid environmental behavior was investigated by adsorption kinetics, adsorption isotherms, and leaching. The results shown that the three-dimensional network structure of ASA/Ca 2+ hydrogel can promote the formation of soil aggregation. Aggregate durability index (ADI) was 0.55 in the presence of ASA/Ca 2+ hydrogel, indicating that amphiphilic hydrogel can enhance the stability of soil aggregation. The adsorbing capacity of acetamiprid was 1.58 times higher than pure soil, and the release of acetamiprid only about 20% in the presence of ASA/Ca 2+ hydrogel. These results would be helpful for the formation of soil aggregation and pesticides adsorption on soil aggregation. Thus, ASA/Ca 2+ hydrogel is likely to improve soil quality, simultaneously it can minimize the mobility of pesticides in the agricultural system.

Ultrasensitive molecularly imprinted electrochemiluminescence sensor based on enzyme-encapsulated liposome-linked signal amplification for trace analysis

Guangyan Liu,Jun Ling,Hanzhao Xie,Jianping Li

A molecularly imprinted polymer (MIP)-electrochemiluminescence (ECL) sensor was prepared and an ultrasensitive strategy was proposed for detecting an ultra-trace amount of ethiprole based on the enzymatic signal amplification of glucose oxidase (GOD). The ethiprole molecule on the MIP was marked with GOD-encapsulated liposomes via light-triggered click reaction after elution and readsorption. Many GOD particles were released from the liposomes and catalyzed the reaction to produce a large amount of H 2 O 2 in the presence of glucose, which considerably enhanced the ECL intensity of luminol. Therefore, the sensitivity for ethiprole determination was promoted remarkably. The ECL signal of the sensor was linearly related to the logarithm of ethiprole concentration from 5.00 × 10 −16 M to 5.00 × 10 −11 M, and the detection limit was 1.00 × 10 −16 M, which was the lowest of all the analytical methods used in ethiprole determination. Ethiprole in samples was determined directly without the tedious and complicated sample treatment procedures, such as separation and enrichment. The click chemistry labeling method using liposomes simplified the operation steps of the MIP-ECL sensor in ethiprole determination.

Detection of neonicotinoids in agricultural products using magnetic molecularly imprinted polymers-surface enhanced Raman spectroscopy

Xiaolin Cao,Yexuan Hu,Huimin Yu,Shuai Sun,Dan Xu,Ziping Zhang,Shuang Cong,Yongxin She

In this paper, magnetic molecularly imprinted polymers-surface-enhanced Raman spectroscopy (MMIPs-SERS) for rapidly analyzing acetamiprid and thiacloprid in agricultural products has been firstly developed. The magnetic imprinted polymers were obtained by polymerizing the imprinted layers on the surface of magnetic nanoparticles . The polymers were detailed characterized by using series of analytical techniques, and their adsorption and recognition performance were validated by adsorption tests. The results showed that the magnetic molecularly imprinted polymers possessed typically core-shell structure and exhibited class-specific recognition, fast adsorption saturation (only 1 min), and good magnetic separation performance towards targets. The adsorption and desorption conditions for MMIPs-SERS detection system were carefully investigated. Under optimum conditions, the good linear detection range of 1∼20 μg/g with LODs of 23.7–68.8 ng/g for acetamiprid and thiacloprid in peach and pear samples was obtained. Through the reusable and spiked experiments, the developed MMIPs-SERS method based on Au NPs as enhanced substrate was validated to be highly sensitive, accurate, efficient and applicable in analyzing neonicotinoids from pear and peach samples. This study provided a rapid and simple detection method for neonicotinoids with effective separation and detection properties based on the synergistic effect of imprinted polymers and SERS. More importantly, this developed method have good application potential in rapid analyzing field for neonicotinoids due to the amazing rapid adsorption time for extracting targets from complex food matrix (only 1 min).
