Elevating enzymatic hydrolysis efficiency and ethanol yield of poplar by hydrogen peroxide assisted p-toluenesulfonic acid pretreatment

Wei Tang,Xinyue Wang,Ziqin Hua,Li Zhang,Yucai He

Utilizing hydrogen peroxide (H 2 O 2 ) and acid medium for oxidation-reduction reaction to facilitate the efficient biorefining of lignocellulose is the ultimate goal of oxidative pretreatment . Here, the p-toluenesulfonic acid (TsOH) was employed in the H 2 O 2 pretreatment of poplar, which has achieved excellent depolymerizing effect of lignin and xylan . The results displayed that 99.9% xylan and 68.4% lignin were dramatically removed when 2% H 2 O 2 and 40 g/L TsOH were introduced to the pretreatment (HTP) at 160 °C. After enzymatic hydrolysis and simultaneous saccharification and fermentation, the enzymatic hydrolysis efficiency and ethanol yield of poplar were advanced by 59.0% and 59.4%, respectively. The structural characterization indicated that the incremental efficiency was attributed to the advantageous accessibility, surface area , crystallinity, and crystal size of poplar by HTP’s augmentative removal of non-cellulosic components. This work expanded the application of acidic H 2 O 2 pretreatment and suggested that HTP was a synergistic and feasible way in biorefinery.

Enhancement of bio-jet fuel production from corncob by iron ion-catalyzed hydrogen peroxide pretreatment

Rui Zhang,Yuting He,Rui Zhu,Can Zhu,Yu Jiang,Yuming Ni,Minghui Fan,Quanxin Li

Developing efficient and green pretreatment pathways is of great significance for the production of biofuels. The purpose of this work is to evaluate that iron-ion catalyzed hydrogen peroxide (Fe-HP) pretreatment can be used for the enhancement of bio-jet fuel production using corncob as a typical lignocellulosic biomass. Typically, the Fe-HP pretreatment was conducted under the following pretreatment conditions: H 2 O 2 concentration of 5.0 wt%, iron ion concentration of 400 ppm, biomass loading of 10 wt%, 70 °C and 60 min. The Fe-HP pretreated corncob shows a high sugar yield of 39.2 % (w/w) under the optimal enzymolysis conditions (50 ℃, pH 4.8, 72 h). Ethanol yield of 35.2 % (w/w) was obtained under the optimal fermentation conditions (30 ℃, pH 5.5, 72 h). Especially, the directional transformation of corncob to bio-jet fuels was achieved by integrating biomass   fermentation and catalytic synthesis. High ethanol conversion of 95.9C-mol % and good jet fuel selectivity of 70.9C-mol % were obtained by using the bi-functional catalyst (HSAPO-34/NiHβ) under the optimal synthesis conditions (200 °C, 5 MPa, 3 h). Based on the biomass characterization and reactive oxygen specie’s analysis, the possible role and mechanism of the iron-ion catalyzed hydrogen peroxide pretreatment was addressed. The iron ion catalyst can accelerate the decomposition of hydrogen peroxide to form hydroxyl radicals, and thus promoting biomass depolymerization and lignin removal from lignocellulose. The pretreatment increased sugar yield and concentration during enzymatic hydrolysis, which indirectly enhanced ethanol fermentation and bio-jet fuel production.

Development of a Robust Saccharomyces cerevisiae Strain for Efficient Co-Fermentation of Mixed Sugars and Enhanced Inhibitor Tolerance through Protoplast Fusion

Jianzhi Zhao,Yuping Zhao,Longhao Wu,Ning Yan,Shuo Yang,Lili Xu,Deyun He,Hongxing Li,Xiaoming Bao

The economical and efficient commercial production of second-generation bioethanol requires fermentation microorganisms capable of entirely and rapidly utilizing all sugars in lignocellulosic hydrolysates. In this study, we developed a recombinantSaccharomyces cerevisiaestrain, BLH510, through protoplast fusion and metabolic engineering to enhance its ability to co-ferment glucose, xylose, cellobiose, and xylooligosaccharides while tolerating various inhibitors commonly found in lignocellulosic hydrolysates. The parental strains, LF1 and BLN26, were selected for their superior glucose/xylose co-fermentation capabilities and inhibitor tolerance, respectively. The fusion strain BLH510 demonstrated efficient utilization of mixed sugars and high ethanol yield under oxygen-limited conditions. Under low inoculum conditions, strain BLH510 could completely consume all four kinds of sugars in the medium within 84 h. The fermentation produced 33.96 g/L ethanol, achieving 84.3% of the theoretical ethanol yield. Despite the challenging presence of mixed inhibitors, BLH510 successfully metabolized all four sugars above after 120 h of fermentation, producing approximately 30 g/L ethanol and reaching 83% of the theoretical yield. Also, strain BLH510 exhibited increased intracellular trehalose content, particularly under conditions with mixed inhibitors, where the intracellular trehalose reached 239.3 mg/g yeast biomass. This elevated trehalose content contributes to the enhanced stress tolerance of BLH510. The study also optimized conditions for protoplast preparation and fusion, balancing high preparation efficiency and satisfactory regeneration efficiency. The results indicate that BLH510 is a promising candidate for industrial second-generation bioethanol production from lignocellulosic biomass, offering improved performance under challenging fermentation conditions. Our work demonstrates the potential of combining protoplast fusion and metabolic engineering to develop superiorS. cerevisiaestrains for lignocellulosic bioethanol production. This approach can also be extended to develop robust microbial platforms for producing a wide array of lignocellulosic biomass-based biochemicals.

The promoting effect of electron beam irradiation on enzymatic saccharification and alcohol fermentation of sorghum meal: Related mechanisms

Mengting Yan,Chenyang Sun,Jiangtao Yu,Junqing Bai,Huishan Shen,Xiuyun Zhang,Yifan Lu,Zhuangzhuang Sun,Xiangzhen Ge,Wei Liang,Jie Zeng,Haiyan Gao,Guoquan Zhang,Wenhao Li

In this study, sorghum meal underwent 2–10 kGy electron beam irradiation (EBI) to investigate alterations in its elemental composition, relevant physicochemical characteristics, and enzymatic hydrolysis fermentation properties. The results indicated a decrease in amylose content and an increase in reducing sugar content in sorghum meal after EBI, along with severe damage to the internal structure of starch granules, leading to increased solubility in water. Furthermore, irradiation enhanced the shear resistance and gelatinization stability of sorghum meal paste. Most importantly, EBI-induced degradation of sorghum starch molecules enhanced their susceptibility to interact with α-amylase, significantly improving the enzymatic hydrolysis and alcohol fermentation efficiency of sorghum meal. At an irradiation dose of 8 kGy, raw and cooked sorghum meal exhibited a 43.40% and 33.33% increase in ethanol concentration and a 43.44% and 30.29% increase in ethanol yield, respectively. This finding indicated that irradiation is an effective pretreatment method for starch-based ethanol fermentation.

Protecting Saccharomyces cerevisiae Cells with Nanozymes to Increase Alcohol Production

Cailin Qiao,Xinrou Wang,Yuwei Gao,Yi Ma,Huibo Luo,Suyi Zhang,Danqun Huo,Changjun Hou

A method was proposed to enhance the growth of Saccharomyces cerevisiae cells and increase the alcohol yield. This strategy utilized nanozymes, specifically FeCo-Prussian blue analogue nanoparticles (FeCo-PBA NPs), FeCo-N-C, CoSO porous nanocubes (CoSO PNCs), and FeN3P-single-atom nanozymes (FeN3P-SAzyme), to mimic the antioxidant enzymes. When subjected to H2O2 stress, the nanozymes significantly stimulated cell growth and effectively reduced the levels of reactive oxygen species (ROS), as evidenced by the decrease in the malondialdehyde (MDA) content. Additionally, the decline in lactate dehydrogenase (LDH) activity within the fermentation environment suggested that nanozymes provided protection against cell membrane damage caused by H2O2. This protective effect can be attributed to their inhibitory impact on ROS production. Further results demonstrated that 50 μg mL–1 of nanozymes led to an increase in the ethanol yield and total alcohol yield in S. cerevisiae. This approach held great potential for enhancing alcohol production efficiency in the S. cerevisiae industrial fermentation process.
