Pulsed Electric Field-Assisted Ethanolic Extraction of Date Palm Fruits: Bioactive Compounds, Antioxidant Activity and Physicochemical Properties

Azhari Siddeeg,Muhammad Faisal Manzoor,Muhammad Haseeb Ahmad,Nazir Ahmad,Zahoor Ahmed,Muhammad Kashif Iqbal Khan,Abid Aslam Maan,Xin-An Zeng,Al-Farga Ammar

The current study was aimed to observe the influence of pulsed electric field (PEF) on the extraction of bioactive components; antioxidant activity and physicochemical properties of date palm fruit extract (DPFE) as compared to the extract untreated by PEF. The fruit was treated with PEF (frequency: 10 Hz, time: 100 µs, pulses number: 30, electric field strength (EFS): 1, 2, and 3 kV/cm. The results show that PEF has a positive impact on the total content of carotenoids, anthocyanins, flavonoids, and phenolics by increasing the EFS. DPFE treated with PEF exhibit a strong antioxidant activity as compared to untreated extract, while electrical conductivity, pH values, and titratable acidity were not affected by PEF. The results indicate a notable increase in the volatile components in DPFE treated with PEF at 3 kV/cm. Thus, PEF treatment can assist the ethanolic extraction of DPFE to improve the bioactivity and antioxidative activity. These findings suggest that PEF can be a more suitable technique to enhance solvent extraction on a commercial basis. Keywords:dates;PEF;bioactive compounds;antioxidant activity;ethanolic extraction

Ethanol extract of Sophora japonica flower bud, an effective potential dietary supplement for the treatment of hyperuricemia

Danni Song,Honghui Zhao,Lingling Wang,Fuqi Wang,Lina Fang,Xu Zhao

Hyperuricemia (HUA) is a common metabolic condition associated with a variety of diseases, mainly manifested by the increase in uric acid (UA) levels. As xanthine oxidase (XOD) is the key enzyme in UA production, it has become a target for treating HUA. In this study, an extract named EESJ was extracted from Sophora japonica using 80% ethanol solution and administered to HUA mice to verify its UA-lowering effect. The results confirmed that administering EESJ could reduce the UA levels in vivo by inhibiting XOD activity, and ameliorating renal injury in a dose-dependent manner. To further study the active components in EESJ, 243 chemical components and 32 blood components were identified by the UHPLC-Q-TOF-MS/MS technology, of which flavonoids were the main constituents. The molecular docking results showed that the following six compounds, namely hydroxygenkwanin, genistein , tectorigenin, kaempferol , sophoricoside, and quercetin , had strong binding properties with XOD; among which hydroxygenkwanin had the best binding capacity. In conclusion, this study confirmed that EESJ could effectively reduce UA levels, and speculated the active compounds playing a role in lowering UA levels, which has certain guiding significance for Sophora japonica and natural compounds as potentially effective treatment options for HUA.

Fuqin He,Yongjun Liu,Ruiying Yu,Tianli Zhang,Fang Guo,Xiayu Wu,Yi Fan,Xuedong Zhu,Qianyi Zhou,Dan Shu

Objective Wound repair remains a significant challenge in the field of regenerative medicine. Notably, while numerous studies have investigated the effects of isolated nicotine or chlorogenic acid, the systematic evaluation of tobacco ethanol extract (TEE) on cutaneous wound healing has remained uncharted territory. Therefore, this study aims to systematically explore TEE’s multifaceted effect on wound healing. Methods The hepatorenal toxicity of 5% TEE was evaluated in 6-7-week-old ICR mice. Subsequently, full-thickness 6 mm wounds were created, and images were captured on days 0, 3, 6, 9, and 12. Tissues were collected for H&E staining, Masson trichrome staining, and immunohistochemistry to assess wound healing. Results The TEE we prepared showed little hepatorenal toxicity (p > 0.05). By day 12, the wound closure rate in the TEE group reached 95.26%, significantly higher than that in the vehicle group (89.20%, p < 0.05). Histopathological analysis demonstrated that TEE reduced neutrophil infiltration in the wound tissues, accelerated granulation tissue formation, and increased collagen content by 20.99% compared to the vehicle group (p < 0.05). Immunohistochemical staining revealed that on day 6, the positive areas of IL-6 and TNF-α in the TEE group were 5.91% and 4.87% lower than those in the vehicle group (p < 0.05), respectively. On day 9, CD31 expression in the TEE group (8.58%) was significantly higher than that in the vehicle group (5.54%) (p < 0.05).Conclusion This study reveals that TEE accelerates wound healing, providing new insights into effective topical therapies for skin wounds while highlighting the potential for tobacco waste valorization.

Zhi Wu,Jin Zhao,Shuang Zhu,Mengxing Chen,Dan Wu,Yiyou Wu,Junbin Lin,Renyun Miao,Rencai Feng,Xiang Li,Bingcheng Gan,Tao Wang

Oudemansiella raphanipesethanolic extract (ORE) was prepared via ultrasonication-assisted ethanolic extraction and enriched through silica gel and macroporous adsorption resin chromatography to afford a non-/weakly polar fraction (ORE-S) and a polar fraction (ORE-N), respectively. This study aimed to (1) quantify major bioactive components (e.g., polyphenols, alkaloids, and terpenes) in ORE-S and ORE-N, (2) assess their antioxidant activities, (3) correlate compositional differences with antioxidant function, and (4) identify key antioxidant compounds along with their potential mechanisms of action. By integrating widely targeted metabolomics with network pharmacology, we not only elucidated how enrichment methods influence the antioxidant properties of ORE but also demonstrated the potential of ORE-N as a valuable source of bioactive compounds and natural antioxidants.

The purpose of this study was to evaluate the efficacy of ethanol extracts fromTorreya grandisseed (EST) as a functional food in hyperuricemia mice. We investigated EST by analyzing its chemical composition. Using a mouse model of hyperuricemia induced by potassium oxonate (PO), we evaluated the effects of EST on uric acid (UA) production, inflammation-related cytokines, and gut microbiota diversity. The primary constituents of EST consist of various flavonoids and phenolic compounds known for their antioxidant and anti-inflammatory properties in vitro. Notably, our findings demonstrate that EST significantly reduced UA levels in hyperuricemia mice by 71.9%, which is comparable to the effects observed with xanthine treatment. Moreover, EST exhibited an inhibitory effect on xanthine oxidase activity in mouse liver, with an IC50value of 20.90 μg/mL (36%). EST also provided protective effects to the mouse kidneys by modulating oxidative stress and inflammation in damaged tissues, while also enhancing UA excretion. Finally, EST influenced the composition of the intestinal microbiota, increasing the relative abundance of beneficial bacteria such asAkkermansia muciniphila,Corynebacterium parvum,Enterorhabdus, Muribaculaceae,Marvinbryantia, andBlautia. In summary, our research unveils additional functions ofTorreya grandisand offers new insights into the future of managing hyperuricemia.

Evaluation on the traditional safe use of Kochiae Fructus oriented by antioxidant properties and oral safety of its ethanolic extract

Xuelin Chen,Kun Zhang,Hanlei Wang,Xia Zhao,Yumei Zhang

Kochiae Fructus (KF) is a traditional Chinese medicine, which has been used to delay aging and treat inflammation, such as rubella, eczema, cutaneous pruritus, etc . In order to fully understand the traditional medicinal value of KF, we evaluated the antioxidant properties and oral safety of its ethanolic extract. Considering flavonoids and phenolics in medicinal plants generally have strong antioxidant activity, we firstly detected the total flavonoids and phenolics contents of KFEE and its fractions. Secondly, we evaluated the antioxidant activities of KFEE and its fractions. Finally, we evaluated the oral safety of KFEE by the acute and 28-day subacute toxicities. The n-butanol fraction (ENBF) possessed the highest phenolics and flavonoids with values of 77.30 ± 3.17 mg gallic acid equivalents/g and 228.81 ± 7.56 mg rutin equivalents/g, respectively. The results of antioxidant tests showed that ENBF possessed potent antioxidant ability. Among them, the high antioxidation capacity observed in ENBF could be attributed to its rich content of flavonoids and phenolics. The results of toxicological studies showed that the LD 50 value of KFEE was 6000 mg/kg BW, and the no observed adverse effect level (NOAEL) of KFEE was 600 mg/kg BW. According to the standards of the American Academy of Sciences for the classification of toxic substances, KFEE can be classified as practically non-toxic substance, which provided valuable evidence for the oral safety of KF as a natural aging delay medicine.

Phytochemical analysis and anticancer effect of Camellia oleifera bud ethanol extract in non-small cell lung cancer A549 cells

Niu Jingming,Jia Xiaoyan,Yang Nian,Ran Yuanquan,Wu Xia,Ding Furong,Tang Dongxin,Tian Minyi

Camellia oleifera is a medicine food homology plant widely cultivated in the Yangtze River Basin and southern China due to its camellia oil. Camellia oleifera bud and fruit exist simultaneously, and its bud is largely discarded as waste. However, C. oleifera bud has been used in traditional Chinese medicine to treat a variety of ailments. Thus, the purpose of this study was to identify the chemical components of C. oleifera bud ethanol extract (EE) and first evaluate its anticancer effects in non-small cell lung cancer A549 cells. Based on UHPLC-Q-Orbitrap-MS analysis, seventy components were identified. For anticancer activity, C. oleifera bud EE had remarkable cytotoxic effect on non-small cell lung cancer A549 (IC50: 57.53 ± 1.54 μg/mL) and NCI-H1299 (IC50: 131.67 ± 4.32 μg/mL) cells, while showed lower cytotoxicity on non-cancerous MRC-5 (IC50 > 320 μg/mL) and L929 (IC50: 179.84 ± 1.08 μg/mL) cells. It dramatically inhibited the proliferation of A549 cells by inducing cell cycle arrest at the G1 phase. Additionally, it induced apoptosis in A549 cells through a mitochondria-mediated pathway, which decreased mitochondrial membrane potential, upregulated Bax, activated caspase 9 and caspase 3, and resulted in PARP cleavage. Wound healing and transwell invasion assays demonstrated that C. oleifera bud EE inhibited the migration and invasion of A549 cells in a dose-dependent manner. The above findings indicated that C. oleifera bud EE revealed notable anticancer effects by inhibiting proliferation, inducing apoptosis, and suppressing migration and invasion of A549 cells. Hence, C. oleifera bud ethanol extract could serve as a new source of natural anticancer drugs.

The Ethanol Extract of Syringa pubescens Turcz. Flower Ameliorates Inflammation Responses by Inhibiting the Expression of Cyclooxygenase-2/Inducible Nitric Oxide Synthase and the Release of Inflammatory Factors

Yanfang Wu,Jiameng Zhang,Ting Zhang,Chenyu Wang,Wanting Bi,Yanxia Wu,Pu Liu,Xinsheng Wang

The flower of Syringa pubescens Turcz., frequently prepared as herbal tea, exhibits various biological properties, including anti-inflammatory, antioxidant, and hepatoprotective activities. Herein, we identify the chemical constituents from ethanol extract of S. pubescens flower (SPFE) and explore its antioxidant, anti-xanthine oxidase (anti-XOD), and 5-lipoxygenase (5-LOX) enzyme activities, and anti-inflammatory activities and mechanisms. SPFE contained a variety of glycosides and showed good antioxidant capacity and effectively inhibited the activity of 5-LOX and XOD enzymes in vitro. In the model of RAW264.7 cell inflammation generated by lipopolysaccharide, SPFE not only impeded the expression of inflammatory mediators and the accumulation of malondialdehyde, but also restored the capacity of antioxidant enzymes. Moreover, SPFE could notably relieve or treat inflammation by suppressing the expression of inducible nitric oxide synthase and cyclooxygenase-2 proteins. In vivo inflammation models, oral SPFE remarkably reduced the swelling of the ear, paw tissues in mice and had a good anti-inflammatory activity, reducing cell infiltration in the claw tissue, and restoring the degree of oxidative damage in the organism. Our findings demonstrate that SPFE exhibits promising potential as a novel therapeutic agent or nutraceutical for anti-inflammatory applications. Graphical

Mengjia Xu,Huixia Feng,Mingquan Guo

Cyclocarya paliurus has been widely applied for therapeutic feeding as a traditional Chinese medicine homologous food with high nutritional and medical value. Nevertheless, the specific bioactive compounds liable for hypoglycemic and hypolipidemic properties and underlying mechanisms remain unexplored. In the present study, the in vitro hypoglycemic and hypolipidemic effects of C. paliurus were assessed by α-glucosidase and pancreatic lipase inhibition assays, and the results indicated that 70% ethanol extract exhibited remarkable α-glucosidase and pancreatic lipase inhibitory activity with the IC 50 value of 3.14 μg/mL and 0.98 μg/mL, respectively. A total of 18 and 15 potential ligand candidates were screened, and the chemical structures were characterized. Furthermore, the docking result showed that chlorogenic acid, quercetin, kaempferol 3- O -rhamnoside, and pterocaryoside A contributed to the underlying hypoglycemic and hypolipidemic effects of C. paliurus , further strengthened the bindings of the enzyme-ligand complex and suppressed the enzyme's activity with the binding energies ranged from −4.00 to −6.31 kcal/mol. The interrelations between the specific bioactive constituents, α-glucosidase or lipase, and hypoglycemic or hypolipidemic activities were also carified. The present study reveals substantial prospects of C. paliurus to be developed as a natural dietotherapy, disease-prevention, and healthcare for diabetes and obesity in the near future.
