Capacity, stability and energy requirement of divalent mercury uptake by non-methylating/non-demethylating bacteria

Fuyu Xie,Qingke Yuan,Yukang Qie,Ying Meng,Fubo Luan

Methylmercury (MeHg) uptake by demethylating bacteria and inorganic divalent mercury [Hg(II)] uptake by methylating bacteria have been extensively investigated because uptake is the initial step of the intracellular Hg transformation. However, MeHg and Hg(II) uptake by non-methylating/non-demethylating bacteria is overlooked, which may play an important role in the biogeochemical cycling of mercury concerning their ubiquitous presence in the environment. Here we report that Shewanella oneidensis MR-1, a model strain of non-methylating/non-demethylating bacteria, can take up and immobilize MeHg and Hg(II) rapidly without intracellular transformation. In addition, when taken up into MR-1 cells, the intracellular MeHg and Hg(II) were proved to be hardly exported over time. In contrast, adsorbed mercury on cell surface was observed to be easily desorbed or remobilized. Moreover, inactivated MR-1 cells (starved and CCCP-treated) were still capable of taking up nonnegligible amounts of MeHg and Hg(II) over an extended period in the absence and presence of cysteine, suggesting that active metabolism may be not required for both MeHg and Hg(II) uptake. Our results provide an improved understanding of divalent mercury uptake by non-methylating/non-demethylating bacteria and highlight the possible broader involvement of these bacteria in mercury cycling in natural environments.

Effects of divalent mercury on myosin structure of large yellow croaker and its binding mechanism: Multi-spectroscopies and molecular docking

Yingnan Liu,Ying Bu,Wenhui Zhu,Jianrong Li,Xuepeng Li

Heavy metals have long biological half-lives and are therefore a major threat to aquatic organisms, especially fish. Divalent mercury (Hg(II)) is an important form from a toxicological viewpoint. In this paper, we studied the interaction mechanism between large yellow croaker myosin and Hg(II) by multi-spectroscopies and molecular docking. Hg(II) had a positive effect on improving the elasticity of myosin gel, and the constant increase of charge would destroy the gel. Hg(II) caused myosin to aggregate, and the protein's apparent structure rapidly increased in length. The content of α-helix obviously decreased, β-turns and β-sheet increased. The myosin and Hg(II) quenching type was static quenching. Thermodynamic analysis suggested hydrogen bonding and van der Waals forces were the main forces for the combination. The molecular docking further confirmed the mechanism of action. This study provides a theoretical guidance for the preventions and control of marine heavy metals.

Mercury Re-Emission in Flue Gas Multipollutants Simultaneous Absorption System

Yue Liu,Qingfeng Wang,Rongjun Mei,Haiqiang Wang,Xiaole Weng,Zhongbiao Wu

Recently, simultaneous removal of SO2, NOx and oxidized mercury in wet flue gas desulfurization (WFGD) scrubber has become a research focus. Mercury re-emission in traditional WFGD system has been widely reported due to the reduction of oxidized mercury by sulfite ions. However, in multipollutants simultaneous absorption system, the formation of a large quantity of nitrate and nitrite ions as NOx absorption might also affect the reduction of oxidized mercury in the aqueous absorbent. As such, this paper studied the effects of nitrate and nitrite ions on mercury re-emission and its related mechanism. Experimental results revealed that the nitrate ions had neglected effect on mercury re-emission while the nitrite ions could greatly change the mercury re-emission behaviors. The nitrite ions could initially improve the Hg0-emission through the decomposition of HgSO3NO2–, but with a further increase in the concentration, they would then inhibit the reduction of bivalent mercury owing to the formation of Hg-nitrite complex [Hg(NO2)x2-x]. In addition, the subsequent addition of Cl– could further suppress the Hg0 emission, where the formation of a stable Hg-SO3–NO2–Cl complex was assumed to be the main reason for such strong inhibition effect.

Development of a novel composite resin for dissolved divalent mercury measurement using diffusive gradients in thin films

Heng Yao,Yujie Zhao,Che-Jen Lin,Fengjiao Yi,Xuefeng Liang,Xinbin Feng

In this work, a composite resin gel incorporating thiol-modified metal double hydroxide ( TM-MDH) nanoparticles is developed for application in diffusive gradients in thin films (DGT) devices to sample and concentrate divalent Hg (Hg(II)) in water and sediment samples. The DGT device uses the TM-MDH resin as a sorption layer and an agarose gel as a diffusive layer. Complete digestion of the TM-MDH resin after sampling can be achieved in 5 mL of 12 N HCl solution for 30 min for direct aqueous Hg(II) analysis. The recovery of Hg(II) uptake onto the resin in aqueous solution reaches 95.4 ± 1.9%. The effect of ionic strength and pH on the performance of DGT device for Hg(II) is assessed. It is found that there is no significant difference on Hg(II) uptake over a pH range of 3.5–8.5 and an ionic strength range of 1–500 mM NaCl. The diffusion coefficient of Hg(II) at 25 °C was estimated to be 9.48 × 10 −6  cm 2 /s at 50 μg/L solution. The sorption capacity of TM-MDH-DGT for Hg(II) reaches 41.0 μg/cm 2 . Field validations performed in reservoir water and in contaminated paddy soil demonstrate that the developed TM-MDH DGT device can accurately determine Hg(II) concentrations in these samples and outperform traditional sampling methods for both high and low Hg(II) concentrations.

Acute effects of mercury on brine shrimp: First evidence highlighting the molecular mechanism through integrated transcriptome and metabolomic analysis

Hangyu Lin,Yanlin Wei,Junjie Huang,Qun Lu,Song Zhu,Jianguang Qin,Shengqi Su,Tao He

Mercury poses significant hazards to aquatic organisms, causing neurotoxicity, and metabolic disorders. Research on the dynamic distribution of mercury within zooplankton is limited, and the metabolic mechanisms of mercury in zooplankton remain unclear. In this study, we employed a mercury-specific fluorescent probe (aggregation-induced emission luminogen, AIEgen) to visualize the in vivo distribution of Hg 2 + and MeHg in brine shrimp. Both Hg 2+ and MeHg accumulated predominantly in the head, ocellus, midgut, and midgut–hindgut transition, with Hg 2+ further affecting the hindgut and anal opening. Transcriptomics and metabolomics analyses revealed that Hg 2+ exposure in the midgut likely induced the efflux activity of ABC transporters. The linoleic acid metabolism, α-linolenic acid metabolism, tryptophan metabolism, and retinol metabolism pathways were potentially associated with MeHg accumulation in the head and ocellus. Additionally, the cytochrome P450 pathway participated in detoxification. Enzyme assays and multi-omics data further demonstrated that both Hg 2+ and MeHg elicited oxidative stress and impaired growth, development, and energy metabolism in brine shrimp. Integrated omics analysis revealed that glutathione metabolism is highly associated with Hg 2+ , while glycerophospholipid metabolism is highly associated with MeHg. Our study revealed the toxicity effects of mercury in zooplankton brine shrimp and promoted the understanding of the underlying mechanism and coping strategies for mercury toxicity in the zooplankton of the aquatic food chain.
