Two pectin-polysaccharides, ASP-B 2 (M w , 5.32 kDa) and B 3 (M w , 30.51 kDa), were obtained from Acanthopanax senticosus leaves. An HILIC-UPLC-ESI − -HCD-MS/MS technique was introduced for structural elucidations of skeletal structure of pectin-polysaccharides in addition to empolying SEC-MALLS, GC–MS, UPLC-ESI + -MS, methylation analysis, FT-IR and NMR methods . Skeletal structure of ASP-B 2 was characterized by combining HG smooth region and RG hairy region with a 2:1 chain length ratio, wheres ASP-B 3 had a distinguishing 3:1 chain length ratio (HG:RG). HG were defined as repeated → 4 GalA 1 →, and RG were featured by repeated → 4 GalA 1 → 2 Rha p 1 → with branched points at C-3, C-4 and C-3,4 of rhamnosyl. Side chains mainly consisted of galactans , arabinans and arabinogalactans. Both showed a random coil conformation and no triple-helix conformation. ASP-B 2 and B 3 possess stronger DPPH and hydroxyl radical scavenging activities, with a relatively low reducing power. Thus, two polysaccharides can be further explored as novel natural antioxidant.

Acanthopanax senticosus has been used to extract active products. However, abundant Acanthopanax senticosus residues (ASR), which contain plenty of lignin are discarded after extraction. An appropriate extraction method should be chosen to obtain the lignin with such desirable properties. Thus, this study investigated the effect of alkali, milled wood, deep eutectic solvent and ethanol methods on the lignin. Lignin obtained from different extraction methods were characterized, yields, chemical structure, thermal behavior, molecular weight and phenolic content were evaluated. The results show that the process of lignin acquisition has a great influence on the properties of lignin. Moreover, the multifarious functional groups exist in lignin macromolecules , such as phenolic, ether groups and other chromophores, conferred good UV resistance to lignin. Among them, the lignin from alkali method has the most phenolic-OH groups and smallest molecular weight result in a good UV-resistant, the SPF value achieves 2.39 at 1% AL content, the alkali method was the best way to make sunscreen blended with cream take various factors into consideration. This study used lignin as a bioactive ingredient to provide UV-resistant property to sunscreen formulations. Furthermore, lignin extracted from Acanthopanax senticosus residue provides a new application for the treatment of herb residue waste.

Objectives: To evaluate the Spectrum-Effect relationships between the High-Performance Liquid Chromatography (HPLC) Fingerprints of fractions with different polarities and the anti-inflammatory and antioxidant effects of A. trifoliatus and to identify its pharmacodynamic material basis.Materials and Methods: Chemical Fingerprints of A. trifoliatus fractions with different polarities (all fractions and ethyl acetate, n-butanol and water fractions) were obtained by HPLC, and Principal Component Analysis (PCA) was performed. The foot swelling and prostaglandin E2 (PGE2) content in mice induced by carrageenan were used as anti-inflammatory markers, and 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) methods were used as antioxidant indicators. Moreover, the Spectrum-Effect relationship between common peaks and efficacy value was conducted by Grey Relational Analysis (GRA) and Partial Least Squares analysis (PLS).Results: PCA identified three principal components, and all fractions of A. trifoliatus had good anti-inflammatory and antioxidant effects. Based on GRA and PLS analysis, peaks 1 and 2 (neochlorogenic acid), 4 and 5 (chlorogenic acid), 8, 9, 10, 11 (isochlorogenic acid B), and 12 were involved in anti-inflammatory efficacy, and peaks 1 and 2 (neochlorogenic acid), 3, 4, and 5 (chlorogenic acid), and 6, 7, 9, 10, and 13 (isochlorogenic acid A) were involved in antioxidant efficacy. Conclusion: This study on the Spectrum-Effect relationship of different polar fractions of A. trifoliatus provides data to support the discovery of the pharmacodynamic material basis of the anti-inflammatory and antioxidant properties of A. trifoliatus.

Acanthopanacis cortex (the dried root bark of Acanthopanax gracilistylus W. W. Smith) has been used for the treatment of rheumatic diseases in China for over 2000 years. Four previously undescribed lignans ( 1 – 4 ) and 12 known lignans ( 5 – 16 ) were isolated from Acanthopanacis cortex. In this study, the inhibitory activities of compounds 1 – 16 against neutrophil elastase (NE), cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2) are reported. The results show that compounds 1 – 16 exhibit weak inhibitory activities against NE and COX-1. However, compounds 2 , 6 – 8 and 13 – 16 demonstrate better COX-2 inhibitory effects with IC 50 values from 0.75 to 8.17 μ Μ. These findings provide useful information for the search for natural selective COX-2 inhibitors.

Periplocymarin (PPM), a cardiac glycoside, has a narrow therapeutic index, poor tumor selectivity and severe cardiovascular toxicity which hinder its wide clinical applications in cancer treatment. Herein, we report novel redox-responsive prodrug-nanoparticles (MPSSV-NPs) self-assembled by co-nanoprecipitation of PPM-vitamin E conjugate and a PEG derivative of linoleate (mPEG2000-LA) in water. It was found that the characteristics of PPM-vitamin E nanoparticles (PSSV-NPs) were improved through co-nanoprecipitation with increased percentages of mPEG2000-LA. Moreover, the MPSSV-NPs were optimized according to the in vitro release and cytotoxicity study. Furthermore, the optimized MPSSV-NPs dramatically enhanced the circulation time and tumor distribution of PSSV-NPs after single intravenous injection. The in vivo studies in malignant H22-bearing mice revealed that MPSSV-NPs could effectively suppress tumor growth without causing obvious systemic toxicity. Altogether, these results suggested that MPSSV-NPs could offer a safe, multifunctional and viable nanoplatform for cardiac glycosides in cancer treatment.

Background Opioids are widely used in clinical practice. However, their long-term administration causes respiratory depression , addiction, tolerance, and severe immunosuppression . Traditional Chinese medicine (TCM) can alleviate opioid-induced adverse effects. Compound 511 is particularly developed for treating opioid addiction, based on Jiumi Liangfang, an ancient Chinese drug treatment and rehabilitation monograph completed in 1833 A.D. It is an herbal formula containing eight plants, each of them contributing to the overall pharmacological effect of the product: Panax ginseng C. A. Meyer (8.8%), Astragalus membranaceus (Fisch.) (18.2%), Datura metel Linn. (10.95%), Corydalis yanhusuo W. T. Wang (14.6%), Acanthopanar gracilistμlus W. W. Smith (10.95%), Ophiopogon japonicus (Linn. f.) Ker-Gawl. (10.95%), Gynostemma pentaphyllum (Thunb.) Makino (10.95%), Polygala arvensis Willd. (14.6%). This formula effectively ameliorates opioid-induced immunosuppression. However, the underlying mechanism remains unclear. Purpose To reveal the effects of Compound 511 on the immune response of morphine-induced immunosuppressive mice and their potential underlying molecular mechanism. This study provides information for a better clinical approach and scientific use of opioids. Methods Immunosuppression was induced in mice by repeated morphine administration. Th1/Th2/Th17/Treg cell levels were measured using flow cytometry. Splenic transcription factors of Th1/Th2/Th17/Treg and outputs of the regulatory PI3K/AKT/mTOR signaling pathway were determined. Subsequently, methicillin-resistant Staphylococcus aureus (MRSA) was administered intranasally to morphine-induced immunosuppressive mice pretreated with Compound 511. Their lung inflammatory status was assessed using micro-computer tomography (CT), hematoxylin and eosin (H&E) staining, and enzyme-linked immunosorbent assay (ELISA). Results Compared to morphine, Compound 511 significantly decreased the immune organ indexes of mice, corrected the Th1/Th2 and Treg/Th17 imbalance in the immune organs and peripheral blood, reduced the mRNA levels of FOXP3 and GATA3 , and increased those of STAT3 and T-bet in the spleen . It improved immune function and reduced MRSA-induced lung inflammation. Conclusion Compound 511 ameliorates opioid-induced immunosuppression by regulating the balance of Th1/Th2 and Th17/Treg via PI3K/AKT/mTOR signaling pathway. Thus, it effectively reduces susceptibility of morphine-induced immunosuppressive mice to MRSA infection .

Staphylococcus aureus is an opportunistic pathogen. Due to the widespread use and abuse of antibiotics, various drug-resistant strains of S. aureus have emerged, with methicillin-resistant Staphylococcus aureus (MRSA) being the most prevalent. Bacterial biofilm is a significant contributor to bacterial infection and drug resistance. Consequently, bacterial biofilm formation has emerged as a therapeutic strategy. In this study, the chemical constituents, antimicrobial and antibiofilm properties of tannins isolated from Penthorum chinense Pursh (TPCP) were investigated. In vitro , TPCP exhibited antimicrobial properties. The minimum inhibitory concentrations (MIC) and minimum bactericidal concentrations (MBC) for methicillin-sensitive Staphylococcus aureus (MSSA) and MRSA were 156.25 and 312.5 μg/mL, and 312.5 and 625 μg/mL, respectively. According to the growth curves, TPCP significantly inhibited the growth of MSSA and MRSA. The results of the crystal violet biofilm assay in conjunction with confocal laser scanning and scanning electron microscopy demonstrated that TPCP destroyed preformed MSSA and MRSA biofilms. TPCP significantly decreased the secretion of exopolysaccharides and extracellular DNA. Subsequently, the mechanism was investigated using RT-PCR. Examining the expression of ica A, cid A, sig B, agr A, and sar A genes in MRSA, we discovered that TPCP inhibited biofilm formation by affecting the quorum-sensing system in bacteria. Our study demonstrates that TPCP exerts antibacterial effects by disrupting the formation of bacterial biofilms, suggesting that TPCP has clinical potential as a novel antibacterial agent for the prevention and treatment of MSSA and MRSA infections.
