Green extraction of the active lignans from Schisandra chinensis has received considerable attention due to their promising applications in the biomedical field. In this study, microwave-assisted deep eutectic solvent method was used to simultaneously extract five lignans: Schizandrol A, Schizandrol B, Schisantherin A, Schisandrin A and Schizandrin B. The extraction parameters were optimized according to the calculated comprehensive score obtained from the analytic hierarchy process combined with the entropy weight method. The optimal extraction conditions were that the molar ratio of choline chloride and glycolic acid is 4:1, water content is 30% (V/V), solid–liquid ratio is 1:20 (W g :V mL ), extraction time is 20 min, and extraction temperature is 70°C, under which the extraction rate of the five lignans were 10.89, 8.616, 4.019, 4.893, and 5.318 mg/g. These results confirmed that our extraction method is efficient and feasible for green extraction of lignans from S. chinensis .

Schisandra sphenanthera and Schisandra chinensis are widely consumed either as food or for medicinal purposes. Nevertheless, no detailed comparative assessments of their physicochemical properties and biological activity have been reported. In this paper, using hot-water extraction, alcohol precipitation, and deproteinization, we obtained polysaccharidic extracts from Schisandra sphenanthera and Schisandra chinensis (denoted as SSP and SCP, respectively) and investigated their antioxidant and immunological activities. The extracts were different from each other with regard to sugar, protein, and uronic acid contents. Both extracts were mainly composed of arabinose, glucose, and galactose, but their contents varied greatly; SSP had more galacturonic acid. Compared with SCP, SSP had stronger free radical scavenging ability, protective effects on biomolecules, cellular antioxidant activity, owing to its higher protein (35.35 ± 1.73%) and uronic acid (12.81 ± 1.15%) contents. With respect to cell viability, neutral red phagocytosis, NO production, and acid phosphatase activity, SCP had stronger effects than SSP; this was largely due to its high levels of mannose, galactose, arabinose, and glucose. These results provide evidence to support the use Schisandra -derived polysaccharides for several purposes, including clinical, agricultural, and industrial applications.

A significant quantity of residues will produce when volatile oil in Schisandra chinensis fruit is extracted by steam distillation. It is important to investigate the residues for potential use. Currently, there is an increasing demand for anti-photoaging products due to the skin damage caused by UV radiation. In this paper, the ingredients of the residues were investigated, and the anti-photoaging activities of residues extracts were studied. The results showed that the residues still contained notable amounts of lignans, flavonoids, and organic acids. In the extracted fraction of dichloromethane (DE), the residues contained higher amounts of schizandrol B and schisantherin A compared to the fresh fruit. The levels were particularly high at 17.39 ± 0.02 and 52.58 ± 0.04 mg/ml, respectively. By lowering the amount of matrix metalloproteinase-1 (MMP-1), reactive oxygen species (ROS) accumulation, and type I collagen (COL-I) loss in fibroblasts, the residues showed targeted benefits in the fight against skin photoaging. In particular, the DE-R phase of residues was very successful in raising the level of COL-I, matching or even exceeding that of retinol (ROL) and undistilled fresh fruits. Protocatechuic acid, schizandrol A, schizandrol B, schisantherinA, schisandrin A and schizandrin B were found to have a significant correlation with the anti-photoaging ability of residues. Particularly, there was a strong and significant correlation between an increase in COL-I and schizandrol B (|ρ| = 0.829*) and schisantherin A (|ρ| = 0.928**), and the distillation procedure has the potential to increase the amount of schizandrol B and schisantherin A in the residues. ABSTRACT Cholestasis is caused by the obstacle of bile formation or secretion and can develop into severe liver diseases. We previously reported the ethanol extract of Schisandra sphenanthera (Wuzhi tablet, WZ) can significantly protect against lithocholic acid (LCA)-induced intrahepatic cholestasis in mice, partially due to the activation of PXR pathway and promotion of liver regeneration. However, the effect of WZ on the bile acids profile and gut microbiome in cholestastic mice remain unknown. In this study, the effect of WZ against LCA-induced liver injury was evaluated and its effect on the bile acids metabolome and gut microbiome profiles in cholestastic mice was further investigated. Targeted metabolomics analysis was performed to examine the change of bile acids in the serum, liver, intestine and feces. The change of intestinal flora were detected by the genomics method. Targeted metabolomics analysis revealed that WZ enhanced the excretion of bile acids from serum and liver to intestine and feces. Genomics analysis of gut microbiome showed that WZ can reverse LCA-induced gut microbiome disorder to the normal level. In conclusion, WZ protects against LCA-induced cholestastic liver injury by reversing abnormal bile acids profiles and alteration of gut microbiome.

Xiaosheng prescription (XSP) has been used for dry eye disease (DED) for more than 10 years in Eye Hospital of China Academy of Chinese Medicine Sciences. However, the effective ingredients involved have remained unclear, which was investigated in this study by the correlation of ingredient and therapeutic activity. Human corneal epithelial cells (HCEC) cultured with 110 mM NaCl solution in vitro and C57BL/6 mice injected subcutaneously with scopolamine hydrobromide were used to establish dry eye models, and the therapeutic effect of XSP extract 1 was better than that of XSP extract 2 significantly. Then, UPLC-Q-TOF/MS and data analysis program Progenesis QI and Makerlynx XS were used to analyze the potential effective ingredients of XSP, and 4 compounds were speculated and identified, in which Schisandrin and 1 μM of Schisantherin A could obviously increase the cell survival rate of injured cells on the cell model. It can be indicated that Schisandrin and Schisantherin A are probably the potential effective ingredients in XSP for DED.

Schisandra chinensis (Turcz.) Baill (S. chinensis), a functional food, is used as a tonic and sedative agent in traditional Chinese medicine. Modern pharmacological research has proved that S. chinensis could prevent and treat age-related neurodegenerative diseases. The presence of bioactive lignans in S. chinensis is the main reason for its neuroprotective and cognitive enhancement effects. This study aimed to clarify the mechanism of lignans in S. chinensis in ameliorating learning and memory deficits in Alzheimer's disease (AD) animals. The step-down test and Morris water maze (MWM) test were used to verify the effects of lignans in S. chinensis on learning and memory in AD animals. Then, metabolomics approaches based on ultraperformance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS) were used to clarify the mechanism of lignans in S. chinensis in treating AD. Finally, quantitative analysis of AD-related neurotransmitters in the brain was conducted after treatment with lignans in S. chinensis. In the MWM and step-down tests, lignans in S. chinensis showed a clear ability to ameliorate the impaired learning and memory of AD animals. A total of 31 endogenous metabolites were identified after treatment with lignans in S. chinensis, which were associated with lignans ameliorating learning and memory. These biomarkers were mainly associated with polyunsaturated fatty acid metabolism and amino acid and vitamin metabolism. Moreover, lignans in S. chinensis upregulated the levels of γ-aminobutyric acid (GABA), 5-hydroxytryptamine (5-HT), acetylcholine (Ach), norepinephrine (NE) and glycine (Gly) and downregulate the level of aspartic acid (Asp). Lignans in S. chinensis might alleviate the neurotoxic effects of neurological inflammation and oxidative stress, Aβ deposition, and tau phosphorylation via the regulation of multiple endogenous metabolic pathways during pathological AD. The research might provide useful support for the further study of pharmacology and new drug development of lignans in S. chinensis.

Diabetic nephropathy (DN) is a progressive kidney disease due to glomerular capillary damage in diabetic patients, with inflammation and oxidative stress implicated as crucial pathogenic factors. There is an urgent need to develop effective therapeutic drug. Natural medicines are rich resources for active lead compounds. They would provide new opportunities for the treatment of DN. The present study was designed to investigate the protective effects of Schisandrin B ( SchB ) on DN and to delineate the underlying mechanism. Oral administration of SchB in the diabetic mouse model significantly alleviated hyperglycemia-induced renal injury, which was accompanied by maintenance of urine creatinine and albumin levels at similar to those of control non-diabetic mice. Histological examination of renal tissue indicated that both development of fibrosis and renal cell apoptosis were dramatically inhibited by SchB . The protective effect of SchB on DN associated with suppression of inflammatory response and oxidative stress. These results strongly suggested that SchB could be a potential therapeutic agent for treatment of DN. Moreover, our findings provided a fuller understanding of the regulatory role of NF-κB and Nrf2 in DN, indicating that they could be important therapeutic targets.

Schisandra chinensis is a medicinal and edible plant that contains various bioactive compounds. Among these, lignans are the major functional compounds. Nevertheless, detailed information about lignans in Schisandra chinensis oil remains scarce. A powerful UPLC-Q/TOF-MS method was established for the rapid identification of the lignan constituents of Schisandra chinensis oils. The results showed that 21 lignans have been unambiguously identified, and four lignans have been tentatively identified in the Schisandra chinensis oils. In addition, semi-quantitative analysis indicated that the total lignan content in the Schisandra chinensis oils was distributed from 67.73 ± 0.06 to 87.61 ± 1.83 mg/g. Schisandrin and schisandrin B were the most abundant lignans in the Schisandra chinensis oils, their content ranging from 15.85 ± 0.09 to 20.57 ± 0.38 mg/g. Additionally, this study provided a systematic characterization of lignans in Schisandra chinensis oil and indicated that the oil might be used as lignan-related functional foods.

Schisandra chinensis is a functional fruit with tonic effect and was widely used by traditional Chinese medicine for treatment and health care. The quality of Schisandra chinensis fruit may vary by different storage condition. In this study, the influence of ambient temperature, humidity, packaging materials and period of storage on the quality of Schisandra chinensis fruits were investigated. The contents of main active components lignans and organic acids were simultaneously determined by ultra-performance liquid chromatography coupled to quadrupole electrostatic field orbitrap high resolution mass spectrometry (UPLC Orbitrap HRMS). The antioxidant activity was determined using DPPH radical scavenging capacity, ABTS + inhibition rate and FRAP value. The correlation of multicomponent and antioxidant activity was analyzed by grey relevance analysis. Taking the changes of multicomponent and antioxidant activity as investigation index, Schisandra chinensis fruits under different storage conditions was comprehensively evaluated. Schisandrol A, malic acid, sorbic acid, schizandrin A, schizandrin B, and schisandrol B were the main effective components of antioxidant activity. Ambient temperature at 5 °C and humidity at 40 % were more suitable for Schisandra chinensis fruits and kraft paper bag was better packaging material. Do not exceed 1 year was the effective storage period. For the safety evaluation, no aflatoxin was detected within the storage period of 2 years, demonstrated the storage was satisfactory. This study provided a reference for the high-quality storage and standardized operating procedures for storage of Schisandra chinensis fruits.

Schisandra chinensis fruit is a famous tonic traditional Chinese medicine and healthy food. The chemical components of dried fruits may vary with the initial processing methods of fresh fruits. Therefore, in this study, the influence of drying was evaluated based on targeted metabolomics analysis. The ultrahigh performance liquid chromatography-triple quadrupole mass spectrometry (UPLC-QQQ-MS) was applied in determination of multicomponent including lignans, organic acids, and oligosaccharides. The fresh fruits of Schisandra chinensis were processed by natural drying in the sun, hot air drying and vacuum drying at different temperatures, and vacuum freeze drying, respectively. The contents of lignans, organic acids, and oligosaccharides in different fruit samples were quantified and statistically analyzed by partial least squares discriminant analysis (PLS-DA). The variation of totally 10 lignans, 5 organic acids, and 9 oligosaccharides with different drying process were compared. The results showed that the drying time of natural drying method was much longer than the other methods using instrument. For hot air and vacuum drying samples, most of lignans, organic acids, and oligosaccharides gradually decreased with the increase of temperature. And the appearance of fruit drying at higher temperature was dark red. At the same processing temperature, vacuum drying takes longer time than hot air drying. In addition, organic acid and oligosaccharide contents of vacuum freeze drying treated samples were higher than the other processed samples significantly. According to the targeted metabolomics analysis, the lignans, organic acids, and oligosaccharides markers were screened out for holistic quality evaluation of Schisandra chinensis fruits. In comprehensive consideration of multicomponent, drying time, fruit appearance and storage, and production feasibility, hot air drying at 50°C is more suitable for the process of Schisandra chinensis. The result is expected to provide a scientific basis for the selection of proper method for drying process of fresh Schisandra chinensis fruit.
