Simple SummaryThe fall armyworm (FAW),Spodoptera frugiperda(J.E. Smith), is a voracious insect pest native to the Western Hemisphere, particularly in South America. The polyphagous fall armyworm feeds on more than 350 plants in such families, including Poaceae, Asteraceae, and Fabaceae. Transgenic plants that expressBacillus thuringiensisproteins (Bt plants) and synthetic insecticides are the main tactics to controlSpodoptera frugiperda, although widespread usage of synthetic chemicals has resulted in the emergence of resistance. We assessed cross-resistance, resistance mechanism, and fitness costs based on the life history traits ofSpodoptera frugiperda. Results indicated that after 24 generations of selection, the resistance to indoxacarb was increased by 472.67-fold as compared to the Ind-UNSEL. Significantly longer developmental time of larvae extended pupal duration, shorter adult longevity, and lower fecundity were observed in Ind-SEL as compared with the Ind-UNSEL population. Butoxide synergist increased susceptibility to indoxacarb, indicating that P450 enzymes may be involved in indoxacarb resistance. Therefore, it is crucial that we comprehend how insecticides work and how resistance develops in order to develop techniques for managing resistance. These data are valuable to understand the indoxacarb resistance mechanism and provides vital information for scientific-based guidance of pest management decisions.AbstractThe fall armyworm,Spodoptera frugiperda(J.E. Smith), is a voracious insect pest that is difficult to control due to resistance to insecticides and Bt proteins. We assessed cross-resistance, resistance mechanism, and fitness costs based on the life history traits ofS. frugiperda. We established anS. frugiperdastrain selected for resistance to indoxacarb (Ind-SEL) from a field-collected population and an unselected strain, Ind-UNSEL. Results indicated that after 24 generations of selection, the resistance to indoxacarb was increased by 472.67-fold as compared to the Ind-UNSEL. There was high cross-resistance to deltamethrin (31.23-fold) with very low or negligible cross-resistance to chlorantraniliprole, emamectin benzoate, and/or methoxyfenozide in the Ind-SEL population. Butoxide synergist increased susceptibility to indoxacarb, indicating that P450 enzymes may be involved in indoxacarb resistance. Significantly longer developmental time of larvae extended pupal duration, shorter adult longevity, and lower fecundity were observed in Ind-SEL as compared with the Ind-UNSEL population. The Net reproductive rate (R0) was the only growth parameter that differs between crosses of Ind-SEL♂ × Ind-UNSEL♀ (176 ± 46) and Ind-SEL♀ × Ind-UNSEL♂ (328 ± 57). On the other hand, all population growth parameters differ between Ind-SEL and Ind-UNSEL strains. Our work contributes to the growing body of research that demonstrates the importance of strain genetics in fitness cost experiments and helps resistance management programs make decisions. Keywords:Spodoptera frugiperda;indoxacarb selection;cross-resistance;inheritance of resistance;fitness cost

BACKGROUND Laodelphax striatellus (Fallén) (Hemiptera: Delphacidae) is one of the most important insect pests of rice, and it has been able to develop strong resistance to many insecticides. Triflumezopyrim, a new type of mesoionic insecticide developed by Corteva Agriscience, showed high biological activity in controlling piercing-sucking insect pests such as planthopper and leafhopper. RESULTS In this study, we continuously selected a susceptible laboratory stain (Unsel) of L. striatellus for 16 generations by exposing it to triflumezopyrim in the laboratory. A 45.1-fold triflumezopyrim-resistant strain (Tri-sel) was established, in which cross-resistance to nitenpyram and acetamiprid was not detected. The realized heritability ( h 2 ) of the Tri-sel strain was estimated at 0.13. The mortalities of the testing F 1 (the offspring of a cross between Unsel and Tri-sel strains) suggested that the resistance of L. striatellus to triflumezopyrim was autosomal and incompletely dominant, as well as a polygenic inheritance. In addition, the results of synergist experiment showed that P450s potentially contributed to the triflumezopyrim resistance. The activities of detoxification enzymes in the Unsel and Tri-sel strains indicated that the activity of P450s in the Tri-sel strain was significantly increased, consistent with the results of synergist experiments. Furthermore, 12 P450 genes demonstrated significant up-regulation. CONCLUSIONS L. striatellus has a certain risk of resistance to triflumezopyrim after continuous selection. Triflumezopyrim resistance did not result in cross-resistance to neonicotinoid insecticides. The up-regulation of multiple P450 genes may mediate triflumezopyrim resistance in L. striatellus . © 2022 Society of Chemical Industry.

Cantharidin (CTD) is a natural toxin with effective toxicity to lepidopteran pests. Nevertheless, little information is available on whether pests develop resistance to CTD. After being exposed to CTD (50 mg/L to 90 mg/L) or 10 generations, the resistance ratio of laboratory selected cantharidin-resistant Mythimna separata (Cantharidin-SEL) strain was only elevated 1.95-fold. Meanwhile, the developmental time for M. separata was prolonged (delayed1.65 in males and 1.84 days in females). The reported CTD target, the serine/threonine phosphatases (PSPs), have not been shown significant activity variation during the whole process of CTD-treatment. The activity of detoxification enzymes (cytochrome monooxygenase P450, glutathione-S-transferase (GST) and carboxylesterase) were affected by CTD selection, but this change was not mathematically significant. More importantly, no obvious cross-resistance with other commonly used insecticides was observed in the M. separata population treated with CTD for 10 generations (resistance ratios were all lower 2.5). Overall, M. separata is unlikely to produce target-site insensitivity resistance, metabolic resistance to CTD. Meanwhile, cantharidin-SEL is not prone to develop cross-resistance with other insecticides. These results indicate that CTD is a promising biogenetic lead compound which can be applied in the resistance management on M. separata .

Black-grass (Alopecurus myosuroidesHuds.) is a common weed in Chinese wheat fields, and has become troublesome due to its evolution of herbicide resistance. One black-grass population (HN-14) collected from a wheat field where herbicides were applied was suspected to be resistant (R) to mesosulfuron-methyl. This study aims to establish a cross-resistance pattern and explore potential resistance mechanisms. The results of a whole-plant dose response assay showed that the resistant (R) population had a high of resistance to mesosulfuron-methyl (33-fold); meanwhile, no synergism of P450s activity inhibitor malathion was observed. The sequencing results revealed that ALS resistance mutation Trp-574-Leu occurred in R plants. The results of in vitro ALS enzyme activity assays also supported that the extractable ALS from R plants were 23.22-fold resistant to mesosulfuron-methyl. In the light of the “R” resistance rating system, HN-14 has evolved RRR and RR resistance to fenoxaprop-P-ethyl, clodinafop-propargyl, haloxyfop-methyl, and fluazifop-P-butyl and R? (resistance may be developing) to pinoxaden, however remains sensitive to imazethapyr, quizalofop-P-ethyl, tralkoxydim, and isoproturon. These results indicated that the mesosulfuron-methyl resistance in the black-grass population HN-14 was driven by a target-site mechanism rather than a nontarget (at least P450s-mediated) mechanism. Keywords:herbicide resistance mechanism;cross resistance;acetolactate synthase (ALS) inhibitor;Alopecurus myosuroideshuds

Rhopalosiphum padi is an important global wheat pest. The pyrethroid insecticide bifenthrin is widely used in the control R. padi. We explored the resistance potential, cross-resistance, adaptive costs, and resistance mechanism of R. padi to bifenthrin using a bifenthrin-resistant strain (Rp-BIF) established in laboratory. The Rp-BIF strain developed extremely high resistance against bifenthrin (1033.036-fold). Cross-resistance analyses showed that the Rp-BIF strain had an extremely high level of cross-resistance to deltamethrin (974.483-fold), moderate levels of cross-resistance to chlorfenapyr (34.051-fold), isoprocarb (27.415-fold), imidacloprid (14.819-fold), and thiamethoxam (11.228-fold), whereas negative cross-resistance was observed to chlorpyrifos (0.379-fold). The enzymatic activity results suggested that P450 played an important role in bifenthrin resistance. A super-kdr mutation (M918L) of voltage-gated sodium channel (VGSC) was found in the bifenthrin-resistant individuals. When compared with the susceptible strain (Rp-SS), the Rp-BIF strain was significantly inferior in multiple life table parameters, exhibiting a relative fitness of 0.69. Our toxicological and biochemical studies indicated that multiple mechanisms of resistance might be involved in the resistance trait. Our results provide insight into the bifenthrin resistance of R. padi and can contribute to improve management of bifenthrin-resistant R. padi in the field.

Cucumber target spot, a major disease that threatens cucumber production, is caused by Corynespora cassiicola . Cyclobutrifluram, a novel succinate dehydrogenase inhibitor (SDHI) developed by Syngenta, has demonstrated strong inhibitory activity against various plant pathogenic fungi and nematodes. However, its antifungal spectrum, resistance risk as well as underlying mechanisms of resistance in C. cassiicola remain poorly understood. In this study, cyclobutrifluram exhibited potent inhibitory activity against anamorphic fungi and selected ascomycetes, with the mean sensitivity of C. cassiicola isolates to the fungicide being 0.98 ± 1.26 μg/mL. Additionally, five laboratory-derived cyclobutrifluram-resistant mutants showed comparable or lower biological fitness than their respective parental isolates. The resistant mutants and field isolates were also found to possess nine distinct point mutations in the CcSdhB , CcSdhC or CcSdhD genes. Finally, cyclobutrifluram exhibited positive cross-resistance with other SDHIs, with the resistance levels varying depending on the specific mutations present. In conclusion, cyclobutrifluram was found to be effective against anamorphic fungi and selected ascomycetes. C. cassiicola ’s risk of resistance development to cyclobutrifluram was assessed as moderate to high and was primarily associated with mutations in CcSdh genes.

Digitaria sanguinalis is a competitive and annual grass weed that commonly infests crops across the world. In recent years, the control of D. sanguinalis by nicosulfuron has declined in Hebei Province, China. To determine the resistance mechanisms of D. sanguinalis to nicosulfuron, a population of D. sanguinalis where nicosulfuron had failed was collected from a maize field of Hebei Province, China. Whole-plant dose–response experiments demonstrated that the resistant population (HBMT-15) displayed 6.9-fold resistance to nicosulfuron compared with the susceptible population (HBMT-5). Addition of the glutathione S-transferase (GSTs) inhibitor 4-chloro-7-nitrobenzoxadiazole (NBD-Cl) significantly reduced the resistance level of the HBMT-15 population to nicosulfuron, and the GSTs activity of the HBMT-15 population was higher than the HBMT-5 population after nicosulfuron treatment. In vitro acetolactate synthase (ALS) enzyme experiments revealed that the nicosulfuron I 50 value for the HBMT-15 population was 41 times higher than that of the HBMT-5 population. An Asp 376 to Glu substitution in the ALS gene was identified in the HBMT-15 population. The HBMT-15 population had a moderate (2- to 4-fold) level of cross-resistance to three other ALS inhibitors (imazethapyr, pyroxsulam, and flucarbazone‑sodium), but was susceptible to pyrithiobac‑sodium. This study demonstrated that both an Asp 376 to Glu substitution in the ALS gene and GSTs-involved metabolic resistance to ALS inhibitors coexisted in a D. sanguinalis population .
