Drosophila suzukii (spotted wing drosophila) has become a major invasive insect pest of soft fruits in the America and Europe, causing severe yield losses every year. The female D. suzukii shows the oviposition preference for ripening or ripe fruit by cutting the hard skin with its serrated ovipositor. A recent study reported that mechanosensation is involved in the texture discrimination during egg-laying behaviour in D. suzukii . However, the underlying mechanism and molecular entity that control this behaviour are not known. The transient receptor potential (TRP) channels and degenerin/epithelial sodium channels (DEG/ENaC) are two candidate gene families of mechanically activated ion channels. Thus, we first identified TRP and DEG/ENaC genes in D. suzukii by bioinformatic analysis. Using transcriptome sequencing, we found that many TRP genes were expressed in the ovipositor in both D. suzukii and D. melanogaster , while some DEG/ENaCs showed species-specific expression patterns. Exposure to drugs targeting TRP and DEG/ENaC channels abolished the oviposition preference for harder texture in female D. suzukii . Therefore, mechanosensitive ion channels may play significant roles in the texture assessment of egg-laying behaviour in D. suzukii , which has promising implications to further research on the development of novel control measures.

Simple SummaryThe Solanaceae crops are the main hosts ofZeugodacus tau, a pest insect that causes serious damage to economically important crops of Solanaceae. In the winter–spring period, female adults ofZ. tauhave been found to oviposit on the pepino melonSolanum muricatum. However, the differences in the ability of the fruits ofS. muricatumand other Solanaceae crops to attract gravidZ. taufemales have not been clarified. In this study, oviposition and trapping bioassays were performed to clarify whether such differences existed. GC-MS was used to identify the compounds and relative contents of the volatile odors ofS. muricatum. The Y-tube olfactometer system was used to determine the compounds that induced behavioral responses in gravidZ. taufemales. The results show thatSolanum muricatumodor was attractive forZ. taufemales. The nine compounds from the volatiles ofS. muricatuminduce behavioral responses in gravidZ. taufemales. These results are beneficial for preventingZ. taufemales from harmingS. muricatum, developing attractants and repellents forZ. taufemales, and establishing a system of ecological control forZ. taufemales.AbstractZeugodacus tau(Walker) (Diptera: Tephritidae) is a pest seriously harmful to Solanaceae crops and was found to oviposit on the pepino melonSolanum muricatum(Aiton). To date, the differences in the ability of the fruits ofS. muricatumand other Solanaceae crops to attract gravidZ. taufemales have seldom been reported. Oviposition and trapping bioassays were performed to clarify whether such differences existed. A combination of GC-MS and the Y-tube olfactometer system was used to identify and determine the compounds inducing behavioral responses in gravidZ. taufemales to the volatile odors ofS. muricatum. The results show thatS. muricatumodors play a role in attracting gravidZ. taufemales. The odors of Solanaceae crops influence their ability to attract these organisms. The nine compounds from the volatiles ofS. muricatuminduce tendency or repellency responses in gravidZ. taufemales. Hexyl acetate, butyl acetate, amyl actate, and isoamyl acetate caused tendency behavior in gravidZ. taufemales, while hexyl hexanoate, butyl isovalerate, butyl valerate, and isoamyl hexanoate caused repellency behavior. Heptyl acetate caused repellency behavior in gravidZ. taufemales at higher concentrations (5 mg/mL) but caused tendency behavior at a low concentration (0.5 mg/mL). These results suggest that vigilance against the harm caused byZ. tauis required during the cultivation ofS. muricatum. The nine compounds of the volatile odors ofS. muricatumcould help to develop attractants and repellents for gravidZ. taufemales. These results are beneficial for preventingZ. taufemales from harmingS. muricatum, developing attractants and repellents forZ. taufemales, and establishing a system of ecological control forZ. taufemales. Keywords:Zeugodacus tau;Solanum muricatum;induced oviposition;attraction rate;volatile

In a heterogeneous and changing environment, oviposition site selection strongly affects the survival and fitness of the offspring. 1 , 2 Similarly, competition between larvae affects their prospects. 3 However, little is known about the involvement of pheromones in regulating these processes. 4 , 5 , 6 , 7 , 8 Here, we show that mated females of Drosophila melanogaster prefer to lay eggs on substrates containing extracts of conspecific larvae. After analyzing these extracts chemically, we test each compound in an oviposition assay and find that mated females display a dose-dependent preference to lay eggs on substrates spiked with (Z)-9-octadecenoic acid ethyl ester (OE). This egg-laying preference relies on gustatory receptor Gr32a and tarsal sensory neurons expressing this receptor. The concentration of OE also regulates larval place choice in a dose-dependent manner. Physiologically, OE activates female tarsal Gr32a + neurons. In conclusion, our results reveal a cross-generation communication strategy essential for oviposition site selection and regulation of larval density.

The evolution of pest resistance to insecticidal proteins from Bacillus thuringiensis (Bt) restricts the sustainable application of transgenic Bt crops, and exploring other ecofriendly insecticides to efficiently suppress Bt-resistant populations is thus urgently needed. The plant secondary metabolite rutin possesses pest resistance effects and plays an important role in plant resistance to pests, but the effect of rutin on pink bollworms is unclear. We investigated the effects of rutin on two types (Bt-susceptible and Bt-resistant) of pink bollworms ( Pectinophora gossypiella ). Rutin had significant inhibitory effects on the growth, development, and oviposition of both Bt-susceptible and Bt-resistant strains. For both strains, rutin ingestion by larvae did not significantly affect larval survival at 7 days after treatment or egg hatching rate but significantly decreased pupation rate, eclosion rate, neonate-to-adult survival, pupal weight and eggs per female and significantly prolonged larval developmental periods, and the effect increased with increasing rutin content. The response to rutin ingestion was basically consistent between Bt-resistant and Bt-susceptible strains, indicating that the Bt-resistant strain did not exhibit cross-resistance to rutin. These results provide a reference basis for green control and resistance management of pink bollworms and may be used as a component of comprehensive management for this pest.

Bactrocera dorsalis is a major pest causing economic losses in fruit and vegetable production. Current control methods, such as synthetic pesticides and male annihilation techniques, have drawbacks, including health risks, resistance development, and ineffectiveness against females. This study investigates the attractions and oviposition stimulant activities of five common volatiles—[isobutyl acetate (A), isoamyl acetate (B), isobutyl butyrate (C), isoamyl isovalerate (D), and isoamyl butyrate (E)]—from the most preferred hosts: mango, guava, orange, and banana. These volatiles were tested at three different concentrations, both individually and in complex blends. Choice tests were performed using a Y-tube olfactometer and Petri plate with agarose assays, and antennal responses were determined using electroantennogram (EAG) recordings. Among them, a blend comprising 1.25% isoamyl acerate (10.95 µg/µl), 1.25% isoamyl isovalerate (10.75 µg/µl), and 20% isoamyl butyrate (172.40 µg/µl) (B1D1E3) emerged as the most potent attractant for both sexes and oviposition stimulant for females. Moreover, combining B1D1E3 with mango, banana, or orange juice significantly enhanced egg-laying behavior. Electrophysiological recordings supported these findings, with B1D1E3 eliciting the strongest antennal responses. Results suggest B1D1E3 could be used to elicit strong oviposition responses in mass rearing of B. dorsalis for the Sterile Insect Technique.

The area planted with insect-resistant genetically engineered crops expressing Bacillus thuringiensis (Bt) genes has greatly increased in many areas of the world. Given the nearby presence of non-Bt crops (including those planted as refuges) and non-crop habitats, pests targeted by the Bt trait have a choice between Bt and non-Bt crops or weeds, and their host preference may greatly affect insect management and management of pest resistance to Bt proteins. In this study, we examined the oviposition preference of the target pest of Bt rice, Chilo suppressalis, for Bt versus non-Bt rice plants as influenced by previous damage caused by C. suppressalis larvae. The results showed that C. suppressalis females had no oviposition preference for undamaged Bt or non-Bt plants but were repelled by conspecific-damaged plants whether Bt or non-Bt. Consequently, C. suppressalis egg masses were more numerous on Bt plants than on neighbouring non-Bt plants both in greenhouse and in field experiments due to the significantly greater caterpillar damage on non-Bt plants. We also found evidence of poorer performance of C. suppressalis larvae on conspecific-damaged rice plants when compared with undamaged plants. GC-MS analyses showed that larval damage induced the release of volatiles that repelled mated C. suppressalis females in wind tunnel experiments. These findings suggest that Bt rice could act as a dead-end trap crop for C. suppressalis and thereby protect adjacent non-Bt rice plants. The results also indicate that the oviposition behaviour of target pest females should be considered in the development of Bt resistance management strategies.

Finding ideal oviposition sites is a task of vital importance for all female insects. To ensure optimal conditions for their progeny, females of herbivorous insects detect not only the odors of a relevant host plant but also chemicals released by eggs, named oviposition-deterring pheromones (ODPs). It is reported that such chemicals play critical roles in suppressing female oviposition behavior; however, the molecular mechanism underlying the detection of egg-derived ODPs remains elusive. Here, we have identified three specific fatty acid methyl esters from the surface of eggs of Helicoverpa armigera serving as ODPs—methyl oleate (C18:1ME), methyl palmitate (C16:0ME), and methyl stearate (C18:0ME). We demonstrated that these ODPs are detected by the receptor, HarmOR56 , exclusively expressed in sensilla trichodea on female antennae. To assess the significance of this receptor, we disrupted HarmOR56 in H. armigera using CRISPR-Cas9 and found that mutant females did not respond to the ODPs, neither in behavioral nor in electrophysiological tests. We therefore conclude that HarmOR56 is indispensable for identifying the ODPs. This study explores, for the first time, how a female-specific odorant receptor detects chemicals from conspecific eggs. Our data elucidate the intriguing biological phenomenon of repulsion to conspecific eggs during oviposition and contribute new insight into a female-specific olfactory pathway linked to reproduction.

Citrus fruit fly Bactrocera minax (Enderlein) (Diptera:Tephritidae) is a devastating pest of citrus plants in China. In order to characterize the behavioral mechanisms and possible chemical cues involved in host selection by female citrus fruit fly, the host plant volatiles was analyzed and Y-tube olfactometer assays were used. Host odor from citrus fruits peels elicited upwind orientation flights followed by landing and egg-laying, demonstrating the essential role of host plant volatiles in B. minax host-finding and oviposition behavior. GC–MS analysis of volatiles from the peels of three varieties citrus fruits, that were Bingtang sweet oranges ( Citrus sinensis cv. Bingtang), Satsumas mandarins ( Citrus reticulata cv. Satsuma) and Sour oranges ( Citrus aurantium ), which adult and larval of B. minax preferred to, showed that the major compounds extracted from the basal region were different from those of the distal region to some extent. Furthermore, in Y-tube olfactometer attraction assays, female flies were significantly attracted ( P < 0.05) to four out of the thirteen tested compounds from the citrus volatiles, including nonanal , citral , limonene and linalool . Conversely, furfuryl alcohol and guaiastil significantly repelled females ( P < 0.05). The percentage composition of nonanal, citral, limonene and linalool from different regions of the fruits displayed a positive correlation with the numbers of egg marks made by females, whereas furfuryl alcohol and guaiastil displayed a correspondingly negative correlation. These results suggest that the oviposition preference of B. minax females is highly related to the chemical compounds emitted from host fruit peels.

Simple SummaryThe excessive utilization of chemical pesticides has resulted in the emergence of pesticide resistance, the disturbance of ecosystems, environmental degradation, and health hazards. Botanical pesticides offer a promising alternative to chemical pesticides, as they have minimal impacts on human health and the environment. Osthole, which has shown pesticidal activities, is the primary bioactive compound inCnidium monnieri(L.) Cusson. In this study, we assessed osthole for its contact toxicity, antifeedant activity, and oviposition preference against three agricultural pests: theTetranychus urticae,Myzus persicae, andBactrocera dorsalis, using laboratory bioassays. Osthole exhibited pesticidal effects against theT. urticaeandM. persicae, impeding their fecundity and development. Additionally, it demonstrated significant antifeedant activity against theT. urticaeand acted as a potent attractant for oviposition by theB. dorsalis. These findings highlight and expand the potential of osthole as a botanical pesticide for the control of these agricultural pests.AbstractOsthole, the dominant bioactive constituent in theCnidium monnieri, has shown acute pesticidal activities. However, its detailed toxicity, antifeedant, and oviposition preference effects against agricultural pests have not been fully understood, limiting its practical use. This study aimed to investigate the contact toxicity, antifeedant activity, and oviposition preference of osthole against three agricultural pests (Tetranychus urticae,Myzus persicae, andBactrocera dorsalis). Our results showed that theCnidium monnieri(L.) Cusson (CMC) has a high osthole content of 11.4 mg/g. Osthole exhibited a higher level of acute toxicity against theT. urticaeto four other coumarins found in CMC. It showed significant pesticidal activity againstT. urticaeandM. persicaefirst-instar nymphs and adults in a dose-dependent manner but not againstB. dorsalisadults. Osthole exposure reduced the fecundity and prolonged the developmental time of theT. urticaeandM. persicae. Leaf choice bioassays revealed potent antifeedant activity in theT. urticaeandM. persicae. Furthermore, the femaleB. dorsalisshowed a distinct preference for laying eggs in mango juice with 0.02 mg/mL osthole at 48 h, a preference that persisted at 96 h. These results provide valuable insights into the toxicity, repellent activity, and attractant activity of osthole, thereby providing valuable insights into its potential efficacy in pest control. Keywords:botanical pesticide;osthole;antifeedant;oviposition preference;Tetranychus urticae;Myzus persicae;Bactrocera dorsalis

Herbivorous insects rely on olfactory cues to select oviposition sites on host plants, which is a process critical for their reproductive success. The rape stem weevil, Ceutorhynchus asper Roel. (Coleoptera: Curculionidae), is a major pest that causes significant damage to rapeseed and other cruciferous crop production, yet the chemical and molecular mechanisms underlying its oviposition preference remain unknown. Our behavioral assays revealed that C. asper females strongly avoided pre-infested rapeseed plants. Five monoterpenes (p-cymene, 1,8-cineole, ocimene, camphor, and α-pinene) were identified as herbivore-induced plant volatiles of C. asper -infested plants, with their emission levels significantly higher than those in healthy plants. Among these five compounds, subsequent electroantennogram (EAG) screening revealed that only 1,8-cineole triggered significant responses in C. asper and showed a concentration-dependent sensitivity. Behavioral assays showed that female C. asper exhibited significant behavioral and oviposition avoidance when exposed to 1,8-cineole. We identified CaspOR39 as the specific odorant receptor for 1,8-cineole, with molecular docking for prediction of the binding affinity and key residues. RNAi of CaspOR39 abolished female EAG responses, as well as behavioral and oviposition avoidance of 1,8-cineole. These results revealed a sex-specific chemical sensing mechanism: female C. asper can utilize CaspOR39 to detect 1,8-cineole as a key cue of plant infestation, driving adaptive oviposition behaviors of this beetle. Our results provide mechanistic and molecular insights into CaspOR39-mediated oviposition behaviors of C. asper , and a basis for the development of semiochemical-based strategies for management of this weevil.

Soy sauce, renowned for its distinctive taste and aroma, has become one of the most popular condiments worldwide. However, its odor attracts the flesh fly Parasarcophaga dux for larviposition, posing a threat to product quality and safety. Here, gas chromatography-ion mobility spectrometry (GC-IMS) was employed to analyze the differential volatile compounds in nine types of soy sauce with varying degrees of attractiveness to P. dux . Notably, 12 differential volatile compounds elicited antennal responses. Furthermore, olfactometer bioassays revealed 6 of them induced behavioral reaction. 3-hydroxy-2-butanone, 2-propanol, butanal, and 2-methylpyrazine were attractive to female flies, while β-myrcene and δ-octalactone exhibited repellent effects. These findings suggest that key volatile compounds with attractive or repellent properties may regulate the differential preference of P. dux for various soy sauces. This study provides a theoretical foundation for preventing P. dux larviposition in soy sauce through formula adjustment and supports the development of the soy sauce industry.

Simple SummaryThe Indian meal moth,Plodia interpunctella(Hübner, 1813) (Lepidoptera: Pyralidae), exhibited significant differences in oviposition preference between normal-oleic peanuts (NOPs) and high-oleic peanuts (HOPs). In this study, we found key volatile organic compounds (VOCs) from NOP and HOP peanuts that were attractive or repellent toP. interpunctellafemales by gas chromatography/mass spectrometry (GC-MS), gas chromatography/electroantennographic detection (GC-EAD), and electroantennogram (EAG) analysis, as well as behavioral responses in Y-tube olfactometer and wind tunnel bioassays. The identification of key VOCs may provide a foundation for developing food-based attractants and repellents for pest control. The Indian meal moth,Plodia interpunctella(Hübner, 1813) (Lepidoptera: Pyralidae), a primary stored peanut insect pest, exhibited a significant difference in oviposition preference among normal-oleic peanuts (NOPs) and high-oleic peanuts (HOPs). Identifying key volatile organic compounds (VOCs) that are attractive or repellent toP. interpunctellais of great significance for the ecological management of pests. The profiles and contents of VOCs among NOP and HOP varieties were measured and compared, and key bioactive VOCs were further confirmed via an electroantennogram (EAG) analysis, as well as behavioral responses in Y-tube olfactometer and wind tunnel bioassays. Females prefer to lay eggs on NOP varieties more than on HOP ones. Acetophenone, nonanal, decanal, dodecane, 2,5-dimethylbenzaldehyde, and 4-ethyl-benzaldehy derived from tested peanuts elicited stronger antennal EAG responses. The results of the Y-tube olfactometer and wind tunnel bioassay showed that the relative high levels of nonanal, dodecane, and unique VOC acetophenone in NOP varieties have a significant attraction toP. interpunctella. Conversely, 4-ethyl-benzaldehyde and the unique VOC 2,5-dimethyl-benzaldehyde commonly found in HOP varieties exhibit notable repellent effects onP. interpunctella. These VOCs could contribute to the development of attractants or repellents derived from special peanut varieties for pest management.
