A dual-functional polyaniline film-based flexible electrochemical sensor for the detection of pH and lactate in sweat of the human body

Chonghui Zhu,Hua Xue,Hongran Zhao,Teng Fei,Sen Liu,Qidai Chen,Bingrong Gao,Tong Zhang

A flexible dual functional electrochemical sensor based on macroscopic polyaniline (PANI) film is reported. The PANI film is prepared by interfacial synthesis without any additional templates and surfactants, and is easily transferred from the water surface to any substrate. The surface of PANI film is flat and has a certain degree of crystallization. The PANI film exhibits good electrochemical properties, which is attributed to the order structure of PANI. The flexible sensor based on PANI film exhibits good electrochemical performances to pH and lactate. And the flexible PANI sensor has good reproducibility, selectivity and long-term stability. Meanwhile, the PANI sensor is also applied to detect the actual sample (such as food and human sweat), and the results are in accordance with the commercial pH meter, indicating the reliability of the PANI sensor.

Self-adhesive wearable poly (vinyl alcohol)-based hybrid biofuel cell powered by human bio-fluids

Yaqin Yuan,Zhen Zhang,Jinlong Cao,Xiaowen Zhao,Lin Ye,Guanglin Wang

Advancement of wearable microelectronics demands their power source with continuous energy supply, skin-integration and miniaturization. In light of poly (vinyl alcohol) (PVA) hydrogel with nontoxicity, good biocompatibility and low cost, an advanced wearable PVA-based hybrid biofuel cells (HBFCs) with high self-adhesiveness was developed. Through the reaction between PVA molecules and succinic anhydride (SAA), the carboxylated PVA (PVA/SAA) was obtained, and by incorporation with PDA as crosslinker , the self-adhesive PVA/SAA-DA hydrogel electrolytes formed by dual covalent and hydrogen bonding . With increasing SAA and PDA content, the pore size decreased, and a uniform and dense network formed, endowing the hydrogel with a relatively high absorption capacity of PBS solution of lactate as cell fuel. Meanwhile the various functional groups of hydrogel, including catechol , quinone , amino and hydroxyl groups , contributed to impressive tissue adhesion strength against pigskin under dry and wet conditions. The PVA/SAA-DA hydrogel displayed high conductive property, and the integrated PVA-based HBFC generated open circuit voltage of 0.50 V and maximum power density of 128.76 μW/cm 2 in 20 mM lactate solution, which was optimized to be 0.57 V/224.85 μW/cm 2 when the pore size was enlarged. The power retention reached above 70% in one week, showing long-term stability of HBFC. The PVA-based HBFC was further adhered to human skin without extra adhesive tapes to scavenge human sweat as biofuel, and the maximum power density reached 85.34 μW/cm 2 , while by connected with a DC-DC converter, the HBFC could power watch, exhibiting promising application potentials as wearable electronic device to provide bioelectricity .

Porous carbon-based robust, durable, and flexible electrochemical device for K+ detection in sweat

Tingting Jiang,Bing Yin,Xiaobo Liu,Lihui Yang,Hongyu Pang,Jie Song,Shuo Wu

Ion sensors are attracting attention for real-time ion monitoring in biological fluids, which requires the development of sensitive, stable, flexible, robust and durable ion-selective electrodes (ISEs) and reference electrodes (REs). In this paper, a highly robust and durable ion sensor was prepared by coating polymer membranes on porous carbon electrodes. A high sensitivity of 58.6 mV per decade with a rapid response time of 0.8 s, and a negligible potential drift less than 1.4 mV h−1 were obtained simultaneously. In addition, after six washing cycles, the K+ ion sensors still have an average sensitivity of 53.2 mV per decade. Importantly, the polymer membrane permeated and packed the porous structure tightly, and thus the ion sensors presented outstanding robustness and durability. The Nernst slope of the K+ ion response fluctuated from 60.2 to 57.9 mV per decade between 0° and 60° bending angles. Repeated bending for 8000 cycles did not result in the delamination of the sensing and reference membranes or reduction of the sensitivity (57.4 mV per decade). Furthermore, five kinds of flexible reference electrodes (LEC, bare Ag, bare Ag/AgCl, PVB + NaCl on Ag/AgCl, PVC/agarose + NaCl on Ag/AgCl) were fabricated and evaluated in terms of the sensitivity for Cl− and long-term stability. Finally, the flexible K+ ion sensor was integrated with microfluidic channels and connected to a portable electrochemical workstation to realize the real-time analysis of human sweat.

Sweat lactate biosensor based on lactate oxidase immobilized with flower-like NiCo2O4 and carbon nanotubes

Liang Tian,Lei Cai,Zhengyuan Ding,Yaqiu Zhou,Yan Zhang,Quan Liu,Xin Ge,Chunmei Yu

We design a portable and non-invasive lactate biosensor that can conveniently monitor lactate levels in real human sweat. The proposed biosensor is constructed through the immobilization of lactate oxidase (LOx) and horseradish peroxidase (HRP) on flower-like NiCo 2 O 4 microspheres coupled with single-walled carbon nanotubes (SWCNTs). The synergistic effect between NiCo 2 O 4 microspheres and SWCNTs can increase the conductivity and the active surface area of the electrode. Using ferrocene–methanol (FcMeOH) as the electron transfer mediator in buffer, the developed biosensor can detect lactate in a wide linear range of 0.1 mM to 30 mM with a detection limit of 39.9 µM under optimal conditions. Additionally, the biosensor exhibits excellent selectivity and stability. As proof of concept, the biosensor was applied to monitor lactate level in real human sweat samples, and the results were in good agreement with those obtained by high-performance liquid chromatography (HPLC) analysis. The lactate biosensor is cost effective and can be as a non-invasive alternative to blood analysis, which is suitable for rapid analysis of lactate levels in the fields of fitness, health monitoring and clinical diagnosis.

Evaporation-induced ion enrichment for high-performance glucose detection in a microfluidic thread/paper-based analytical system

Xiong Jing,Wu Liang,Xiao Gang,Ma Yan,Ran Ruilong,Sun Wei,Wang Wei,Mao Cuiping,Qiao Yan,Lu Zhisong

A microfluidic thread/paper-based analytical device (µTPAD) is of great significance in biomedical analysis and point-of-care diagnostics. Nevertheless, the research has not been done on the ion distribution on the thread and how it affects sensing so far. Inspired by the sweat stains on the clothes, a high-performance µTPAD based on the evaporation-induced ion enrichment is presented for glucose monitoring in salty solutions. In comparison to the multi-ply cotton thread, the single-ply cotton thread exhibits the most significant ion-enrichment capability at the rear segment, which could be attributed to the balance between the water evaporation and the solution replenishment in the thread. The salts accumulated at the rear segment could increase the ionic strength of the solution passed through the thread, promoting the colorimetric reaction on the paper-based sensing unit. The single-ply thread with strong ion-enrichment capability was integrated with a paper-based glucose sensor to prepare a µTPAD for highly sensitive glucose detection in both PBS and artificial sweat (-0.673µM −1 in PBS and − 0.253 µM −1 in artificial sweat). Moreover, the system could detect glucose in human sweat samples with high accuracy and precision. This work may provide a strategy to prepare a highly sensitive µTPAD for precise analysis of substances in bio-fluids, particularly human sweat. Graphical Abstract

NH2-MIL-101(Fe) anchored onto nanoporous gold microelectrode: Highly sensitive electrochemical platform for simultaneously sensing of ascorbic acid and uric acid

Yan Chen,Xiaojuan Yang,Zhonggang Liu,Guohua Liu,Zheng Guo

Resolving the issue of poor conductivity and easy aggregation of MOFs is recognized as one important factor for their successful applications in electrochemical sensors and electrocatalysis. In this work, we propose an assembly of NH 2 -MIL-101(Fe) onto a nanoporous gold (NPG) microelectrode surface. The nanoporous gold could greatly improve the conductivity and reduce the aggregation of NH 2 -MIL-101(Fe). The proposed NH 2 -MIL-101(Fe)/NPG microelectrode was developed for the electroanalysis of ascorbic acid (AA) and uric acid (UA). It exhibited excellent electrochemical performance for the individual and simultaneous sensing of AA and UA in a 0.1 M PBS solution (pH 7.4). High sensitivities (2.03 μA μM −1 cm −2 for AA; 29.30 μA μM −1 cm −2 for UA) were achieved in the simultaneous analysis process. The improved electrochemical performance was attributed to the electrocatalytic properties and large specific surface area of NH 2 -MIL-101(Fe)/NPG, as well as the electrostatic interaction between NH 2 -MIL-101(Fe) and analytes. Moreover, the NH 2 -MIL-101(Fe)/NPG microelectrode was successfully used for the simultaneous detection of AA and UA in complicated environment (human sweat and advanced DMEM). This work shows great promise for the simultaneous detection of AA and UA and extends the applications of MOFs in electrochemical devices.

Wearable electrochemical biosensor based on molecularly imprinted Ag nanowires for noninvasive monitoring lactate in human sweat

Qingwen Zhang,Danfeng Jiang,Changshun Xu,Yuancai Ge,Xiaohu Liu,Qingquan Wei,Liping Huang,Xueqian Ren,Chengde Wang,Yi Wang

We proposed a wearable electrochemical biosensor based on Ag Nanowires (AgNWs) and molecularly imprinted polymers (MIPs) prepared on the screen-printed electrode for the noninvasive monitoring of lactate in the human sweat. The MIPs were prepared by electropolymerization of 3-aminophenulboronic acid (3-APBA) with imprinted lactate molecule on the AgNWs-coated working electrode. The MIPs-AgNWs biosensor revealed high sensitivity and specificity for the detection of lactate from 10 −6 M to 0.1 M, with the detection limit of 0.22 μM. Furthermore, the sensors presented high stability and reproducibility with sensitivity recovery of 99.8% ± 1.7% during 7 months storage in a dark plastic box at room temperature. In addition, the flexible electrodes also showed stable electrochemical response after been bended and twisted for 200 times, respectively. Such MIPs-AgNWs biosensor was attached on volunteers’ skin for the noninvasive monitoring of the perspiration lactate in vivo . The wearable electrochemical biosensor provides feasibility in near future for the evaluation of human sweat in the military, sports and biomedical fields.

A Portable 3D Microfluidic Origami Biosensor for Cortisol Detection in Human Sweat

Xuan Weng,Zhuoyi Fu,Cheng Zhang,Wen Jiang,Hai Jiang

Analysis of cortisol levels in human sweat is increasingly important as it can be a “stress biomarker” in stress-related disorders, giving real-time information about human health status. In this study, a portable 3D microfluidic origami biosensor based on a smartphone was developed for cortisol-level detection in human sweat. Molybdenum disulfide (MoS2) nanosheet-mediated fluorescence resonance energy transfer (FRET) and fluorescently labeled aptamers were employed in the biosensing process. A multilayer-structured 3D origami microfluidic chip was fabricated and functionalized to facilitate low-volume perspired human sweat collection, transportation, and detection. The translatability of the biosensor was exhibited by the fluorescence analysis in a smartphone mounted in a custom-designed holder. The critical design parameters of the microfluidic origami biosensor, including the characterization of various paper substrates, the concentration of MoS2 nanosheets, and the incubation/reaction time, were adjusted to obtain an acceptable range for the assay dynamic range and limit of detection (LOD). Under optimum conditions, various doses of cortisol within the physiologically relevant range of 10–1000 ng/mL reported in human sweat were tested to evaluate the performance of the proposed biosensor. It displayed an LOD of 6.76 ng/mL at 3σ in artificial sweat, an analysis time of 25 min, and high selectivity. The performance of the proposed cortisol sensor was compared with an enzyme-linked immunosorbent assay (ELISA) for a spiked artificial sweat sample, and a correlation coefficient of 0.988 was found. The proposed biosensor also presented satisfactory results in the determination of the cortisol levels in a real human sweat sample. The resulting portable biosensor provides a rapid, low-cost, convenient, and non-invasive sensing solution for the point-of-care analysis of cortisol levels in sweat.

A conducting polymer PEDOT:PSS hydrogel based wearable sensor for accurate uric acid detection in human sweat

Zhenying Xu,Jingyao Song,Bingrui Liu,Shaoping Lv,Fengxian Gao,Xiliang Luo,Peipei Wang

Noninvasive wearable sensors present key information about targeting epidermally retrievable biofluids in human health monitoring. Tracking circulating uric acid (UA) in human sweat performs a simple and prompt way for reducing risk of gout and hyperuricemia. Herein, we propose a wearable microfluidic-based electrochemical sensor via incorporating conducting polymer PEDOT:PSS hydrogel towards accurate and sensitive detection of UA in sweat. The microfluidic device validly realizes the capture for sweat in real-time, and the conducting PEDOT:PSS hydrogel with large surface area plays multiple roles in this wearable sensor for UA detecting, electrolyte storing, and flexibility enhancing. The prepared flexible sensor shows an ultrahigh sensitivity of 0.875 µA µM −1 cm −2 and a low limit of detection (LOD) down to 1.2 μM (S/N = 3). Furthermore, it can detect UA levels in real human sweat samples collected from volunteers before and after purine-rich diets, and the results are highly correlated with those measured by the standard enzyme linked immunosorbent assay (ELISA). This PEDOT:PSS hydrogel based flexible sensor, together with the sweat-collecting microfluidic device, is believed to be promising for the construction of high performance wearable sensors to monitor biomarkers, metabolites and nutrients.
