Background Peptides have diverse and important physiological roles in plants and are ideal markers for species identification. It is unclear whether there are specific peptides in Panax quinquefolius L. (PQ). The aims of this study were to identify Quinetides, a series of diverse posttranslational modified native peptides of the ribonuclease-like storage protein (ginseng major protein), from PQ to explore novel peptide markers and develop a new method to distinguish PQ from Panax ginseng. Methods We used different fragmentation modes in the LTQ Orbitrap analysis to identify the enriched Quinetide targets of PQ, and we discovered Quinetide markers of PQ and P. ginseng using ultrahigh-performance liquid chromatography–quadrupole time-of-flight mass spectrometry analysis. These “peptide markers” were validated by simultaneously monitoring Rf and F11 as standard ginsenosides. Results We discovered 100 Quinetides of PQ with various post-translational modifications (PTMs), including a series of glycopeptides, all of which originated from the protein ginseng major protein. We effectively distinguished PQ from P. ginseng using new “peptide markers.” Four unique peptides (Quinetides TP6 and TP7 as markers of PQ and Quinetides TP8 and TP9 as markers of P. ginseng ) and their associated glycosylation products were discovered in PQ and P. ginseng . Conclusion We provide specific information on PQ peptides and propose the clinical application of peptide markers to distinguish PQ from P. ginseng .

A new triterpenoid glycoside( 1 ) and seven known triterpenoid glycosides, pseudoginsenoside RT 2 ( 2 ), yesanchinoside R 2 ( 3 ), vinaginsenoside R13( 4 ), vinaginsenoside R8( 5 ), notoginsenoside E( 6 ), 6′′′- O -acetylginsenoside Re( 7 ), 6″- O -acetylginsenoside Rb 1 ( 8 ), were isolated from the rhizomes of Panacis majoris . The new triterpenoid glycoside was elucidated as 3- O -[ β - D -glucopyranosyl-(1→2)- β - D -(6′- O -ethyl)-glucuronopyranosyl]-oleanolic acid-28- O - β - D -glucopyranoside by extensive spectroscopic and phytochemical methods. Compounds 2 ― 8 were obtained from the plant for the first time. Compounds 3 and 4 displayed good activities against adenosine diphosphate (ADP)-induced platelet aggregation, and compounds 1, 5, 6 and 8 showed moderate activities. Compound 6 exhibited moderate antiplatelet aggregation activity induced by arachidonic acid(AA).

Central composite design and response surface methodology are adopted to explore the extraction process of polyphenols from Panax quinquefolius L ., including the extraction time, extraction temperature, ethanol concentration, and the influence of interaction effects, and establish the multiple regression model. The results show that the optimal extraction conditions for polyphenols of P. quinquefolius L . are 61 °C of extraction temperature, 38 min of extraction time, 48% of ethanol concentration and the polyphenol yield is 79.53 ± 0.43 gallic acid equivalent (mg GAE g −1 ). The actual polyphenols extraction yield of P. quinquefolius L . is close to the predicted value, which is 99.21% of the predicted value. The extraction method is simple, low cost and can be used to extract polyphenols from P. quinquefolium L .

Lipidomics currently is still confronted with challenges from chromatographic separation and lipids identification. Here we report a lipidomics platform by integrating ultrahigh performance supercritical fluid chromatography/quadrupole time-of-flight mass spectrometry (UHPSFC/QTOF-MS) and collision cross section (CCS) measurement using ion mobility spectroscopy/time-of-flight mass spectrometry (IMS/QTOF-MS), aiming to enhance the profiling performance and identification reliability of lipids. The lipidomes extracted from three congeneric Panax species ( P. ginseng , P. quinquefolius , and P. notoginseng ) by methyl tert -butyl ether are comprehensively profiled and compared by use of this platform. A potent UHPSFC/QTOF-MS approach was developed on a 1.7-μm particles packed Torus 2-PIC column using CH 3 OH (in CO 2 ) as a modifier and CH 3 OH/0.2 mM ammonium acetate as the makeup liquid, enabling well resolution of six lipid subclasses by both positive and negative MS E modes. In contrast to the reversed-phase chromatography, “normal-phase” like elution order and better resolution of polar lipids and some lipid isomers were achieved by UHPSFC separation. Pattern recognition chemometric analysis of 60 batches of Ginseng samples ultimately unveiled 24 lipid markers, of which triacylglycerols were the most important. Aside from the automated MS database searching against HMDB and LIPID MAPS, the application of CCS retrieval or CCS prediction improved lipid identification by reducing the possible hits. In conclusion, this integral platform can significantly improve the chromatographic separation and the reliability of lipids identification in lipidomics studies. It is the first report that systematically compares the lipidomic difference of three reputable Panax species, providing useful information for their quality control in addition to ginsenoside analysis.

The flower buds of threePanaxspecies (PGF:P. ginseng; PQF:P. quinquefolius; PNF:P. notoginseng) widely consumed as health tea are easily confused in market circulation. We aimed to develop a green, fast, and easy analysis strategy to distinguish PGF, PQF, and PNF. In this work, fast gas chromatography electronic nose (fast GC e-nose), headspace-gas chromatography-ion mobility spectrometry (HS-GC-IMS), and headspace solid phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS) were utilized to comprehensively analyze the volatile organic components (VOCs) of three flowers. Meanwhile, a principal component analysis (PCA) and heatmap were applied to distinguish the VOCs identified in PGF, PQF, and PNF. A random forest (RF) analysis was used to screen key factors affecting the discrimination. As a result, 39, 68, and 78 VOCs were identified in three flowers using fast GC e-nose, HS-GC-IMS, and HS-SPME-GC-MS. Nine VOCs were selected as potential chemical markers based on a model of RF for distinguishing these three species. Conclusively, a complete VOC analysis strategy was created to provide a methodological reference for the rapid, simple, and environmentally friendly detection and identification of food products (tea, oil, honey, etc.) and herbs with flavor characteristics and to provide a basis for further specification of their quality and base sources. Keywords:Panax;flower bud;fast GC e-nose;HS-GC-IMS;HS-SPME-GC-MS;multivariate statistical analysis
