Aggregation of micron-particles in microfluidic texture of artificial joint to improve tribological properties

Qi Liu,Hengdi Yuan,Daolong Yang,Lanfeng Zhang,Nsilani Kouediatouka Ange,Guangneng Dong

Artificial joint lubricants injected in vitro are easily absorbed and diluted by human body, resulting in a loss of lubrication effect. In this study, the lubricating substance used was micron-sized gelatin particles containing sodium hyaluronate (GSH particles), and an attempt was made to aggregate the dilute particle solution in the microfluidic texture of Ti6Al4V surface to improve the lubrication. The COMSOL simulation results demonstrated that micron-sized particles were enriched in the microfluidic texture, where the concentration was up to 2.8 times higher than released particles. Finally, the results of particle aggregation were observed by preparing transparent microfluidic texture on PDMS surface. Aggregation of lubricant can effectively improve the lubrication environment and has potential to prolong the injection cycle of lubricant.

Tribological properties of hierarchical structure artificial joints with poly acrylic acid (AA) - poly acrylamide (AAm) hydrogel and Ti6Al4V substrate

Deng Yaling,Sun Jianjun,Ni Xingya,Yu Bo

Natural joints have the hierarchical structure of bone and cartilage, providing excellent lubrication and load-carrying capacity. In our research, the hierarchical structure artificial joints of Poly Acrylic Acid (PAA)- Poly Acrylamide (PAAm) hydrogel coating and Ti6Al4V substrate were formed using a simple yet versatile method by mimicking the hierarchical structure of natural joints. The efficient lubrication at friction interfaces was achieved. The composition and microstructure were confirmed by Fourier-transform infrared (FTIR), X-ray photoelectron spectroscopy (XPS), Scanning electron microscopy (SEM). The hydrogel coatings were successfully prepared on the surface of Ti6Al4V substrate through the adsorption of the charged group. This hierarchical structure material exhibits the lower dynamic friction, approximately 0.085, which is much lower than bare Ti6Al4V, about 0.429. The wear on the surfaces after the tribological experiment is super-shallow and has no significant fracture area around the scratch. The PAA-PAAm hydrogel has a more uniform and compact cross-linked network porous structure, and the porous size is much smaller than PAA hydrogel. The cross-linked network porous structure is the main factor accounting for the low dynamic friction. This hierarchical structure of soft and hard improve the load-carrying ability and has the potential uses in artificial joints.

The Low Friction Coefficient and High Wear Resistance UHMWPE: The Effect of Pores on Properties of Artificial Joint Materials

Chunmin Yang,Junhua Zhang,Honglin Yue,Xueqin Kang

Ultrahigh-molecular-weight-polyethylene (UHMWPE) is extensively applied to make bone and cartilage implants in the field of biomaterial application. UHMWPE matched with a metal or ceramic component withstands the long-term effect of cyclic stress, which induces UHMWPE serious wear, and affects the service life of the artificial joint. This investigation focuses on the influence of pores on the mechanical and tribological property of UHMWPE. The porosity, crystallinity, yield strength, tensile strength, hardness, compression yield strength, creep resistance, wettability, friction performance, and wear mechanism of solid and porous UHMWPE were evaluated and compared. The research results indicated that the pore had a remarkable influence on the mechanical, friction, and wear property of UHMWPE. The porosity of porous UHMWPE was 29.7% when 50 wt. % sodium chloride (NaCl) was added and the pore size was about 200 μm. The crystallinity, hardness, creep resistance, strength, and elongation decreased after NaCl was added and dissolved. However, the yield strength in the tensile and compression test was closer to that of the natural cartilage. The friction coefficient and wear loss of porous UHMWPE were higher than that of solid UHMWPE in dry conditions, but these values of porous UHMWPE were lower than that of solid UHMWPE in the calf serum lubrication condition. The main wear mechanism of porous and solid UHMWPE was abrasive. The lubricity of calf serum reduced wear surface scratches and furrows, especially for porous UHMWPE.

A tannic acid-reinforced PEEK-hydrogel composite material with good biotribological and self-healing properties for artificial joints

Yuntong Liu,Dangsheng Xiong

Polyetheretherketone (PEEK) is widely considered as a promising material for joint implants but it still has limitations involving high friction and wear. To mimic the cartilage-subchondral bone structure in natural joints, a polyvinyl alcohol (PVA) hydrogel layer was fabricated on the PEEK substrate to provide a lubrication mechanism. In addition, tannic acid was applied to form dynamic hydrogen bonds with PVA molecules, for the purpose of strengthening the hydrogel layer and endowing it with self-healing ability. Our experimental results demonstrated that the prepared PEEK-hydrogel composite exhibited good biotribological performance with a low average friction coefficient around 0.06 and little wear after the friction test. It also could repair the scratch made by a blade spontaneously at room temperature taking advantage of the reversibility of the hydrogen bonds. The influence of the properties of the PVA hydrogel and the concentration of tannic acid on the frictional and self-healing behavior of the composite structure was investigated and the internal mechanism was discussed. This work presents a facile method to fabricate a PEEK-hydrogel composite possessing outstanding tribological properties and self-healing capacity simultaneously, hopefully promoting its potential in producing artificial joints.

Macroscale superlubricity achieved between zwitterionic copolymer hydrogel and sapphire in water

Zhongnan Wang,Jinjin Li,Yuhong Liu,Jianbin Luo

Achievement of macroscale superlubricity from the hydration effect could provide a potential application for artificial articular cartilage. In this work, two zwitterionic polymers, 2-methacryloyloxyethyl phosphorylcholine (MPC) and sulfobetaine methacrylate (SBMA), were used to synthesize a P(MPC-co-SBMA) copolymer hydrogel through the additional polymerization of the alkane carbon-carbon double bond. The incorporation of MPC promoted the water-binding property of copolymer hydrogels as well as improved their anti-compression capability. A superlubricity state with a friction coefficient of approximately 0.002 was achieved when a P(MPC-co-SBMA) copolymer hydrogel hemisphere slid on a sapphire in water, which demonstrated its dependence on sliding velocity, load, and aqueous lubricants. The superlubricity mechanism was mainly attributed to the hydration effect from zwitterionic MPC and SBMA polymer chains, which led to the formation of a uniform hydration layer on the hydrogel surface and the strong adsorption of water molecules on the sapphire surface, which provided additional stabilized hydration layers. These findings might provide insight into the superlubricity mechanism of zwitterionic hydrogels based on the hydration effect, thereby broadening their extensive applications.

Quantitative analysis and degradation mechanisms of different protein degradation methods

Tao Zhang,Dekun Zhang,Hongtao Liu,Kai Chen

The abrasive debris produced by wear test of artificial joints in vitro is encapsulated by proteins in serum lubricants, which hinder the characterization of debris analysis. One of the key issues of isolating wear debris from serum is degrading the proteins wrapping the wear debris. In this article, the proteins in calf serum were degraded by a strong alkali, a strong acid, and an enzyme. The residual concentration of proteins in calf serum was detected by UV absorption. Quantitative analysis of protein degradation and the protein degradation rate was proposed, following treatment with different degradation reagents and different incubation times. The results showed that when 10 mL of 25% volume calf serum was added with 40 mL of NaOH and incubated at 65°C for 24 h, the protein degradation rate reached a maximum of 95.52%. The protein degradation rate in the solution ranged from 31.86% to 71.64% when a different volume of 37% HCl was added and incubated at 60°C. The highest protein degradation rate was 94.98% in the protease degradation solution. When the protein degradation rate is less than 70%, the particles were coated by protein. When the protein degradation rate was more than 95%, there was no protein coating around the particles. The three protein degradation methods have different processes and protein degradation rates. A suitable method for protein degradation can be selected according to these practical applications.

Composite lubricating layer with enhanced wear-resistant properties between HXLPE and Cu/ZrO2 ceramic friction interface

Liu Sisi,Deng Yuxing,Jiang Shengqiang,Liu Jingang

Poly (2-methacryloyloxyethyl phosphorylcholine) (PMPC), as a highly hydrophilic as well as biocompatible material, has been widely used in biomedical applications. In this study, the friction reduction and wear resistance of HXLPE were improved by grafting PMPC on its surface. However, the PMPC will be sheared off during friction, which is difficult to meet the long-term service requirements of artificial joints. This can be achieved by improving the lubrication environment to minimize PMPC shedding. To simulate the human lubrication environment, free PMPC (lubrication factor) and Cu 2+ (transition group metal ion catalyst) were added to neonatal bovine serum (NBCS). When the friction pair consisting of grafted PMPC-modified HXLPE and Cu/ZrO 2 ceramics is rubbed therein, the grafted PMPC interacts with the free PMPC, while Cu 2+ catalyzes the oxidative hydrolysis of serum proteins into carbon-containing small molecules and adsorbs on the surface of the Cu/ZrO 2 ceramics to form a carbon-rich lubrication layer. The composite lubrication layer consisting of grafted-free PMPC and carbon-rich lubrication layer provides good load-bearing and lubrication properties at the friction interface. And the wear rate is reduced by 45% compared to rubbing in deionized water. This technique can reduce the shedding of grafted PMPC state and improve the load-bearing performance of the interfacial lubrication layer, thus reducing the wear of HXLPE and prolonging the service life of the artificial joint.

Improved biotribological performance of Ti6Al4V alloy through the synergetic effects of porous TiO2 layer and zwitterionic hydrogel coating

Lingling Cui,Hang Li,Jiewen Huang,Dangsheng Xiong

To improve the biotribological properties of Ti6Al4V alloy for artificial joints, motivated by the composite structure of ‘natural articular cartilage - subchondral bone’ and the hydration lubrication mechanism of articular cartilage, a soft/hard bilayer coating composed of a porous TiO 2 layer and a zwitterionic hydrogel coating was designed and fabricated on Ti6Al4V alloy through micro-arc oxidation (MAO) combined with ultraviolet (UV) radiation technology. Characterization results revealed that the bio-inspired bilayer coating could be successfully constructed on Ti6Al4V alloy, and such a coating displayed good hydrophilicity (WCA: 32°). Notably, by ball - on - disk friction test, this biomimetic bilayer coating exhibited an ultra-low friction coefficient (∼0.038) in PBS solution, which was reduced by an order of magnitude compared with that of the Ti6Al4V alloy substrate. This was due to the synergistic effects of soft/hard composite structure and the hydration lubrication of zwitterionic MPC polymer. This work provides a novel and effective strategy for surface modification of Ti6Al4V alloy for artificial joints.

Insights into synergic antioxidant effects for vitamin E and sugar alcohol stabilized ultrahigh molecular weight polyethylene composites for artificial joints

Rui Hong,Ri-Tong Lan,Yue Ren,Lu Xu,Yun-Xiang Xu,Nouman Ali Shah,Rizwan M. Gul,Shishu Huang,Lingli Li,Jia-Zhuang Xu,Zhong-Ming Li,Ka Li

Oxidation resistance is strongly necessary for the use of crosslinked ultrahigh molecular weight polyethylene (UHMWPE) in artificial joints. The hybrid antioxidant strategy emerged as a promising alternative to the single rival that relies on vitamin E (VE). The goal of this study was to investigate the universality of the antioxidant synergy between VE and sugar alcohol (SA). Three types of SA, including D-sorbitol, maltitol and erythritol, were employed to cooperate with VE. Compared to the use of VE alone, these hybrid antioxidants generally enabled the corresponding UHMWPE composites with prolonged oxidation induction time (OIT) and increased crosslink density. The correlation between OIT and crosslink density of VE/SA-stabilized UHMWPE composites showed low attenuation. The applicability of the hybrid antioxidants to protect UHMWPE against oxidation was further demonstrated using the in vitro clinically relevant accelerated aging. Among the used SA, D-sorbitol exhibited the strongest ability to enhance the synergetic efficiency. The possible mechanism was revealed from the following aspects: the number of secondary hydroxyls per molecular mass and the molecular polarity determined via polarity parameter calculation. These findings provide significant insights for fabricating UHMWPE bearing materials with superior comprehensive performance and extended service longevity.

A bionic PEEK composite structure with negatively charged surface adsorbing molecular brushes possessing improved biotribological properties for artificial joints

Yuntong Liu,Dangsheng Xiong,Xiaoduo Zhao

To improve the contact mode of artificial joints and their tribological performance , inspired by the ‘soft-on-hard’ structure of ‘cartilage-subchondral bone’ as well as negatively charged characteristic of the cartilage surface with brush-like molecules, a polyetheretherketone (PEEK)-hydrogel composite structure with negatively charged surface adsorbing cationic molecules through electrostatic interaction was designed and fabricated. Characterization results demonstrated that polyacrylic acid (PAA) hydrogel strengthened by polyvinyl alcohol (PVA) microcrystals was successfully fabricated on the PEEK substrate through ultra-violet (UV) radiation. Adsorption of cationic monomers was also confirmed. The PEEK-hydrogel composite structure had significantly reduced friction coefficient , ~80% lower than that of PEEK, owing to the synergistic effect of biphasic and boundary lubrication . This work proposes a novel route to develop the friction surface of artificial joints with advanced performance and contributes to the understanding of lubrication mechanism of artificial joints.

Antibacterial and anti-inflammatory ultrahigh molecular weight polyethylene/tea polyphenol blends for artificial joint applications

Yue Ren,Fei-Yu Wang,Zi-Jian Chen,Ri-Tong Lan,Ren-Huan Huang,Wan-Qun Fu,Rizwan M. Gul,Jing Wang,Jia-Zhuang Xu,Zhong-Ming Li

Periprosthetic joint infection (PJI) is one of the main causes for the failure of joint arthroplasty. In view of the limited clinical effect of oral/injectable antibiotics and the drug resistance problem, there is a pressing need to develop antibacterial implants with therapeutic antimicrobial properties. In this work, we prepared a highly antibacterial ultrahigh molecular weight polyethylene (UHMWPE) implant by incorporating tea polyphenols. The presence of tea polyphenols not only improved the oxidation stability of irradiated UHMWPE, but also gave it the desirable antibacterial property. The potent antibacterial activity was attributed to the tea polyphenols that produced excess intracellular reactive oxygen species and destroyed the bacterial membrane structure. The tea polyphenol-blended UHMWPE had no biological toxicity to human adipose-derived stem cells and effectively reduced bacteria-induced inflammation in vivo. These results indicate that tea polyphenol-blended UHMWPE is promising for joint replacement prostheses with multifunctionality to meet patient satisfaction.

A biomimetic bilayer coating on laser-textured Ti6Al4V alloy with excellent surface wettability and biotribological properties for artificial joints

Lingling Cui,Hang Li,Chenyang Gong,Jiewen Huang,Dangsheng Xiong

Titanium alloys with excellent mechanical properties and biocompatibility are widely used in surgical implants. However, due to its poor tribological properties, it is difficult to be used in joint bearing interfaces. Herein, mimicking the soft/hard hierarchical structure of articular cartilage/subchondral bone and the unique lubrication mechanisms of articular cartilage, a biomimetic bilayer coating consisting of a TiO 2 layer and a hydrogel layer was fabricated on laser-textured Ti6Al4V alloy by laser surface texturing, thermal oxidation and ultraviolet radiation techniques. Meanwhile, a zwitterionic polymer (SBMA) was introduced into the hydrogel layer to provide hydration lubrication. The experimental results demonstrated that the introduction of SBMA greatly improved the hydrophilicity and compressive modulus of the hydrogel layer. Compared with Ti6Al4V, the Ti6Al4V-hydrogel bearing interface exhibited a lower friction coefficient (0.06) and better wear resistance when lubricated in deionized water. More importantly, this Ti6Al4V-hydrogel bearing interface could maintain stable and low friction coefficients when lubricated in different physiological solutions for a long-term friction (4 h), and the lowest friction coefficient (0.039) was measured in phosphate buffer (PBS) solution. The outstanding biotribological performance was mainly attributed to the biphasic and hydration lubrication mechanisms of the hydrogel layer. This study provides new insights into the development of Ti6Al4V bearing interfaces for artificial joints.
