3D Bioprinting of Artificial Skin Substitute with Improved Mechanical Property and Regulated Cell Behavior through Integrating Patterned Nanofibrous Films

Shaoquan Bian,Xiaohua Hu,Hao Zhu,Weili Du,Chenmin Wang,Liangliang Wang,Liuzhi Hao,Yuming Xiang,Fengzhen Meng,Chengwei Hu,Zhiyun Wu,Jing Wang,Xiaohua Pan,Min Guan,William Weijia Lu,Xiaoli Zhao

Three-dimensional (3D) bioprinting has advantages for constructing artificial skin tissues in replicating the structures and functions of native skin. Although many studies have presented improved effect of printing skin substitutes in wound healing, using hydrogel inks to fabricate 3D bioprinting architectures with complicated structures, mimicking mechanical properties, and appropriate cellular environments is still challenging. Inspired by collagen nanofibers withstanding stress and regulating cell behavior, a patterned nanofibrous film was introduced to the printed hydrogel scaffold to fabricate a composite artificial skin substitute (CASS). The artificial dermis was printed using gelatin–hyaluronan hybrid hydrogels containing human dermal fibroblasts with gradient porosity and integrated with patterned nanofibrous films simultaneously, while the artificial epidermis was formed by seeding human keratinocytes upon the dermis. The collagen-mimicking nanofibrous film effectively improved the tensile strength and fracture resistance of the CASS, making it sewable for firm implantation into skin defects. Meanwhile, the patterned nanofibrous film also provided the biological cues to guide cell behavior. Consequently, CASS could effectively accelerate the regeneration of large-area skin defects in mouse and pig models by promoting re-epithelialization and collagen deposition. This research developed an effective strategy to prepare composite bioprinting architectures for enhancing mechanical property and regulating cell behavior, and CASS could be a promising skin substitute for treating large-area skin defects.

Bimodal Tactile Sensor without Signal Fusion for User-Interactive Applications

Xiaole Ma,Chunfeng Wang,Ruilai Wei,Jiaqi He,Jing Li,Xianhu Liu,Fengchang Huang,Shuaipeng Ge,Juan Tao,Zuqing Yuan,Ping Chen,Dengfeng Peng,Caofeng Pan

Tactile sensors with multimode sensing ability are cornerstones of artificial skin for applications in humanoid robotics and smart prosthetics. However, the intuitive and interference-free reading of multiple tactile signals without involving complex algorithms and calculations remains a challenge. Herein a pressure–temperature bimodal tactile sensor without any interference is demonstrated by combining the fundamentally different sensing mechanisms of optics and electronics, enabling the simultaneous and independent sensing of pressure and temperature with the elimination of signal separation algorithms and calculations. The bimodal sensor comprises a mechanoluminescent hybrid of ZnS–CaZnOS and a poly(3,4-ethylenedioxythiophene):poly(styrene sulfonate) (PEDOT:PSS) thermoresistant material, endowing the unambiguous transduction of pressure and temperature into optical and electrical signals, respectively. This device exhibits the highest temperature sensitivity of −0.6% °C–1 in the range of 21–60 °C and visual sensing of the applied forces at a low limitation of 2 N. The interference-free and light-emitting characteristics of this device permit user-interactive applications in robotics for encrypted communication as well as temperature and pressure monitoring, along with wireless signal transmission. This work provides an unexplored solution to signal interference of multimodal tactile sensors, which can be extended to other multifunctional sensing devices.

Mechanically robust, stretchable and environmentally adaptable organohydrogels with cross-linked fibrous structure for sensory artificial skins

Kexuan Wang,Bohui Zheng,Ping Wu,Jialiang Lai,Hongli Zhang,Weifeng Zhao,Xilang Jin,Aijie Ma,Weixing Chen,Hanbin Liu,Hongwei Zhou

Hydrogels are promising materials for fabricating sensory artificial skins (SASs), but their mechanical properties and environment adaptability are often limited, leading to restricted performances of SASs. In this work, mechanically robust, stretchable and environment adaptable organohydrogels with cross-linked fibrous structure are designed and constructed. The organohydrogels are prepared by introducing doped polyaniline (PANI) into cross-linked fibrous mats of poly(acrylic acid) (PAA) and poly(vinyl alcohol) (PVA). Cross-linked PAA-PVA mat works as skeleton to account for the mechanical robustness while PANI serves as the conductive component to achieve mechanosensing functionality. Due to the unique structure, the resultant organohydrogels, being denoted as PAA-PVA/PANI organohydrogels, exhibit high tensile strength (5.06 MPa), stretchability, fatigue resistance and excellent environment adaptability (anti-freezing, anti-drying and swelling resistance). SASs of such organohydrogels have achieved a gauge factor of 1.81, a sensing range of 0–70 %, a response time of 0.28 s and a reversible sensing in more than 2,000 cycles. In addition, SASs are further attempted in monitoring diverse human motions and physiological activities, such as joint bending, muscle motion and pulse. Overall, this work has provided promising soft materials for future SASs that work properly under complicated environmental conditions.

3D-bioprinted multifunctional artificial skin patches with synergistic antimicrobial-mechanoadaptive functions for accelerating infected wound healing

Ting Dong,Rui Li,Zhaojing Lu,Jian Zhou,Mutao Xu,Shasha Feng,Tingting Cui,Ning Gu

This study employs advanced 3D bioprinting to molecularly integrate polycaprolactone (PCL), carboxymethyl chitosan (CMCS), and curcumin (CCM), formulating an artificial skin patch with dual antibacterial and regenerative functionalities. The patch employs directional hydrogen bonding between CMCS and bioactive CCM to reduce the contact angle of the PCL matrix from 107.5° to 57.3°, significantly enhancing antibacterial efficacy ( S. aureus : 92.89 %; E. coli : 99.31 %), promoting cell migration, and achieving an optimal balance between tensile strength (11 MPa) and elongation (30 %). The material demonstrates exceptional mechanical resilience with a storage modulus (G′ = 3.1 kPa) greater than the loss modulus (G″ = 1.6 kPa), and over 95 % modulus recovery within 30 s after 10 % strain, ensuring structural integrity under dynamic wound conditions. In vivo animal studies confirmed that the artificial skin patch significantly promoted the healing of infected wounds, with only approximately 5.51 ± 1.56 % of the wound area remaining unhealed after 12 days of treatment. Histological analysis further revealed that the patch effectively suppressed IL-6 expression and facilitated epithelialization, collagen deposition, angiogenesis, and macrophage polarization. Both in vitro and in vivo assessments validate its superior wound-healing capacity, offering a highly efficient, customizable, and transformative solution for clinical wound management.

Layer-by-Layer Assembly of Nanofiber/Nanoparticle Artificial Skin for Strain-Insensitive UV Shielding and Visualized UV Detection

Fei Liang,You Jun Fan,Shuang Yang Kuang,Hai Lu Wang,Ying Wang,Peng Xu,Zhong Lin Wang,Guang Zhu

Here, a strain-insensitive, durable, and visualized ultraviolet (UV) detectible multilayered artificial UV shielding skin (MUVS-skin) is demonstrated. The electrospinning of polyurethane nanofibers and electrospraying of TiO 2 nanoparticles are alternatingly conducted to assemble the MUVS-skin. The results show that the stability of UV shielding property under different strains (50%, 100%, 150%, and 200%) is enhanced as more layers are assembled. The ultraviolet protection factor of the strain-free MUVS-skin (119 µm) is 10 810 and reaches to 5685 at 200% strain. Moreover, the effective transmittance (at 365 nm) of MUVS-skin (119 µm) after being stretched for 10 000 cycles at 200% strain is only 0.125%. This stable UV shielding property is ascribed to the low-level variation in the porosity of the multilayered structure, which originates from the enhanced slippage between adjacent layers. Furthermore, ultraviolet A (UVA) and ultraviolet B (UVB) can be precisely detected as the luminescence of blue and red for the MUVS-skin integrates with a phosphor layer (MUVS-skin/P). Owing to the excellent stable UV shielding property, visualized UV detection and durability against cyclic deformation, this MUVS-skin/P might be extensively applied to the smart wearable artificial skin and protective clothing.

Silver-Nanoparticle-Modified Polyimide for Multiple Artificial Skin-Sensing Applications

Peng Bi,Xiaowei Liu,Yang Yang,Ziyu Wang,Jing Shi,Gaomin Liu,Fangfang Kong,Benpeng Zhu,Rui Xiong

Wearable devices have become very important for collecting health information and monitoring the condition of seriously ill patients. However, most recently developed wearable devices can only monitor limited signals within a very small range of values. A novel sensor is fabricated by modifying the surface of polyimide nanowires with silver nanoparticles. These nanowire sensors can sense both pressure and temperature changes and have antimicrobial properties. The fabricated sensor can detect a wide range of pressure variation from 5 KPa to 100 KPa. A theoretical model is developed to explain the sensing mechanism of the silver-nanoparticle-modified polyimide nanowires. The experimental and theoretical results for the nanowires are in good agreement. By attaching these sensors to human joints and fingers, real-time information can be detected for human motion, such as walking or holding a hot cup of water. The excellent performance of this device shows that it has great potential for use in artificial skins for detecting temperature and human motion.

Robust Gradient Hydrogel-Loaded Nanofiber Fleshy Artificial Skin via A Coupled Microfluidic Electrospinning-Reactive Coating Strategy

Qing Li,Rong Chen,Tingting Cui,Yuting Bai,Jie Hu,Jiafei Yu,Gefei Wang,Su Chen

Skin regeneration has attracted tremendous interest due to the important role of skin for human protection and beauty. Thus, methods allowing artificial skin to be carried out in a controllable fashion are potentially important for wound healing, which involves an intersection of materials, medicine, biology and other disciplines. Herein, aiming at a new general methodology for fleshy materials, we designed and fabricated a new hydrogel-loaded hydrophobic-hydrophilic nanofiber fleshy artificial skin. The gradient hydrogel-loaded nanofiber artificial skin integrates both advantages of nanofiber and hydrogel, exhibiting fleshy feature (comparability to real skin in terms of appearance, texture and function), excellent air permeability, compatibility, good mechanical and antibacterial property. Interestingly, the efficient transport channels are formed throughout the hydrogel-loaded nanofiber structure, which is beneficial for the water absorption and transfer. These advantages enable the establishment of a moist and favorable microenvironment, thus greatly accelerating wound healing process. This work couples microfluidic electrospinning with reactive coating technique, which is in favor of material design and fabrication with controllable and uniform structures. The hydrogel-loaded nanofiber fleshy artificial skin shows comparability to real skin in terms of beauty, texture and function, which would definitely provide new opportunities for the further optimization and upgrading of artificial skin. Advanced In-Situ functionalized conductive hydrogels with high mechanical strength for hypersensitive soft strain sensing applications

Yihao Hou,Yanjiao Chang,Zhiyong Zhao,Mingxi Zhang,Qian Zhao,Mingzhuo Guo,Jiale Zhao,Siyang Wu,Yunhai Ma

Conductive hydrogels are used in wearable electronics, soft robotics and artificial skin due to their flexibility, biocompatibility and multifunctionality. However, combining high mechanical strength, conductivity and sensitivity in these hydrogels remains a challenge. This paper proposes a novel in-situ conductive functionalization technology for hydrogels with high mechanical strength, achieving high conductivity (3223.727 S·m −1 ) and low resistance (0.92 Ω) without sacrificing mechanical properties. Comprehensive experiments reveal the mechanisms and optimal parameters for conductive functionalization. AgNO 3 and ascorbic acid (VC) concentrations influence the distribution, reaction rate and aggregation of Ag particles on hydrogel surfaces. Sodium carboxymethylcellulose (CMC) enhances mechanical strength and Ag + adsorption capacity. Optimal parameters are 1.00 M AgNO 3 and 0.08 M VC. An in-situ reduction reaction of silver particles establishes strong interfacial bonding between the silver layer and hydrogel matrix, forming the basis for strain sensing. The hypersensitized sensing mechanism, involving the appearance and recovery of microcracks on the silver layer via the tunneling effect, enables high sensitivity (gauge factor = 36.65). The conductive hydrogels can monitor human physiological signals, tiny strain signals from water droplets (0.01 g) and airflow, with an average response time of 12.7 ms. The developed conductive hydrogels meet the technical requirements for combining high mechanical strength and sensitivity, offering an efficient new approach for conductive hydrogel-based soft strain sensors.

Construction of chitin nanocrystal/poly(deep eutectic solvent) elastomers via glycidyl methacrylate associated modification, polymerization and crosslinking for artificial skin applications

Huangjingyi Chen,Zicong Shi,Chong Tang,Xiaorui Liu,Shaoning Wang,Liang Liu,Juan Yu,Zhiguo Wang,Yimin Fan

A one-pot strategy was developed to fabricate a strong and ductile elastomer composed of chitin nanocrystals and poly(deep eutectic solvent) (ChNC/PDES), based on a dual-network structure formed through glycidyl methacrylate (GMA)associated modification, polymerization and crosslinking. This approach enables the integrated pretreatment, chemical modification, and nanodispersion of chitin within a lactic acid/choline chloride deep eutectic solvent (DES) system. Whereafter, the ultraviolet initiated polymerization of GMA with ChNC and DES components produced a homogeneous elastomer with a maximum tensile strength of 4.50 MPa and an elongation at break of 890 %. The mechanical performance was regulated by the degree of deacetylation (DDA) and aspect ratio of ChNC: higher DDA provided more amino groups for crosslinking and enhanced ductility, whereas it also reduced elastomer stiffness due to smaller nanocrystal size. Furthermore, a spin-coating and alcohol-water rinsing protocol enabled the fabrication of ultrathin elastomer films (~0.25 mm thick) that adhered conformably to human skin and accommodated complex deformations. These findings demonstrate the potential of bio-based ChNC/PDES elastomers as sustainable and flexible candidates for artificial skin applications.

Soft-tough polyacrylamide and polyacrylamide/polyvinyl alcohol/sodium alginate bilayer hydrogels for adaptive grasping and shape recognition

Ziwei Zhang,Wenjing Zhang,Lang Ding,Ming Liu,Bowen Zheng,Chenxin Yi,Jing Chen,Shuai Zhou

Human skin achieves high sensitivity and a wide range of tactile perception through its gradient structure and the synergistic function of various receptors. However, significant challenges remain in achieving multifunctional synergy enhancement for artificial skin. This study develops a soft-tough bilayer gradient hydrogel based on a salting-out and ion coordination toughening strategy, achieving high sensitivity (7.35 kPa −1 , 0–3 kPa) and a broad stress sensing range (0–75.44 kPa). The soft layer is designed with a low-density polyacrylamide (PAM) network, where weak chemical crosslinking improves both elasticity and sensitivity. Meanwhile, the tough layer is formed by a high-density dual crosslinked network of polyacrylamide /polyvinyl alcohol/sodium alginate, followed by post-treatment in ZnSO 4 solution. The combined effects of salting-out and ion coordination greatly enhance toughness, extend the stress detection range, and strengthen interlayer adhesion. Furthermore, this study integrates the hydrogel into a robotic arm and develops a gel-controller-computer tactile feedback system that simulates the conditioned reflex mechanism, enabling non-destructive recognition and precise grasping of objects with varying hardness. Moreover, the bilayer gradient hydrogel-based sensor facilitates comprehensive recognition of an object's weight and shape. This research offers a novel approach to advancing artificial skin, bioinspired robotics, and intelligent sensing technologies.

Biomimetic tri-layered artificial skin comprising silica gel-collagen membrane-collagen porous scaffold for enhanced full-thickness wound healing

Jing Tian,Caihong Fu,Wenhua Li,Na Li,Linyan Yao,Jianxi Xiao

Full-thickness wounds are severe cutaneous damages with destroyed self-healing function, which need efficient clinical interventions. Inspired by the hierarchical structure of natural skin, we have for the first time developed a biomimetic tri-layered artificial skin (TLAS) comprising silica gel-collagen membrane-collagen porous scaffold for enhanced full-thickness wound healing. The TLAS with the thickness of 3–7 mm displays a hierarchical nanostructure consisting of the top homogeneous silica gel film, the middle compact collagen membrane, and the bottom porous collagen scaffold, exquisitely mimicking the epidermis, basement membrane and dermis of natural skin, respectively. The 1-(3-Dimethylaminopropyl)-3-ethylcarbodiimide/N-Hydroxysuccinimide-dehydrothermal (EDC/NHS-DHT) dual-crosslinked collagen composite bilayer, with a crosslinking degree of 79.5 %, displays remarkable biocompatibility, bioactivity, and biosafety with no risk of hemolysis and pyrogen reactions. Notably, the extra collagen membrane layer provides a robust barrier to block the penetration of silica gel into the collagen porous scaffold, leading to the TLAS with enhanced biocompatibility and bioactivity. The full-thickness wound rat model studies have indicated the TLAS significantly facilitates the regeneration of full-thickness defects by accelerating re-epithelization, collagen deposition and migration of skin appendages. The highly biocompatible and bioactive tri-layered artificial skin provides an improved treatment for full-thickness wounds, which has great potential in tissue engineering.

3D bioprinting of prefabricated artificial skin with multicomponent hydrogel for skin and hair follicle regeneration

Xiaoxiao Ma,Xiaohui Zhu,Sheng Lv,Chunyan Yang,Zihao Wang,Meilan Liao,Bohao Zhou,Yiming Zhang,Shiyu Sun,Ping Chen,Zhonghua Liu,Haiyan Chen

Background: The timely management of large-scale wounds and the regeneration of skin appendages constitute major clinical issues. The production of high-precision and customizable artificial skin via 3D bioprinting offers a feasible means to surmount the predicament, within which the selection of bioactive materials and seed cells is critical. This study is aimed at employing skin stem cells and multicomponent hydrogels to prefabricate artificial skin through 3D bioprinting, which enables the regeneration of skin and its appendages. Methods and Results: We employed gelatin methacrylate (GelMA) and hyaluronic acid methacrylate (HAMA) as bioactive materials, in conjunction with epidermal stem cells (Epi-SCs) and skin-derived precursors (SKPs), to fabricate artificial skin utilizing 3D bioprinting. The photosensitive multicomponent hydrogel, comprising 5% GelMA and 0.5% HAMA, demonstrated excellent printability, suitable solubility and swelling rates, as well as stable mechanical properties. Moreover, this hydrogel exhibited exceptional biocompatibility, effectively facilitating the proliferation of SKPs while maintaining the cellular characteristics of both SKPs and Epi-SCs. The transplantation of this artificial skin into cutaneous wounds in nude mice led to complete wound healing and functional tissue regeneration. The regenerated tissue comprised epidermis, dermis, hair follicles, blood vessels, and sebaceous glands, closely resembling native skin. Remarkably, the artificial skin demonstrated sustained tissue regeneration capacity even after 12 h of in vitro culture, facilitating comprehensive functional skin regeneration. Conclusions: Our research presented a skin repair strategy for prefabricated cell-loaded artificial skin, thereby successfully facilitating the regeneration of the epidermis, dermis, hair follicles, blood vessels, and sebaceous glands within the wound.
