Dynamic changes of phenolic compounds during artificial aging of soybean seeds identified by high-performance liquid chromatography coupled with transcript analysis

Cui Meng,Wu Dong,Bao Kaixuan,Wen Zenglian,Hao Yingbin,Luo Liping

Phenolic compounds are important bioactive substances in plants, but study of their alteration during soybean seed aging is still limited. In this study, we conducted artificial aging on soybean seeds, detected the dynamic changes of phenolic compound concentrations using high-performance liquid chromatography, and analyzed the gene expression of key enzymes of phenolic metabolism. A detailed method for detection of 19 phenolic compounds during artificial aging of soybean seeds was constructed, and all of these phenols significantly changed in concentration. The content of protocatechuic acid, rutin, and morin decreased, whereas that of daidzein, glycitein, genistein, and baicalin increased. The concentration of caffeic acid, epicatechin, ferulic acid, daidzin, genistin, and resveratrol first rose and then declined, and the content of ferulic acid was highest after 2 days of artificial aging, with the other five phenolic compounds showing the highest content after 4 days of artificial aging. The total content of the 19 phenolic compounds reached a peak of 2357.43 μg g -1 dry weight at 2 days. Relative expression of PAL1 , PAL2 , PAL3 , CHS7 , CHS8 , IFS1 , IFS2 , CHR1 , 4CL2 , C4H , and CHI2 was mostly downregulated as the duration of artificial aging increased. This study provides novel insights into the storage and use of soybean seed resources. Graphical abstract

Molecular mechanism of high pressure shear grinding on Feng-flavour Chinese Baijiu ageing

Wei Jia,Zibian Fan,An Du,Lin Shi

Ultrahigh-pressure homogenization is a powerful technique for accelerating Baijiu ageing. In this study, the molecular mechanism of artificial ageing was thoroughly investigated utilizing foodomics combined with a mass spectrum information optimization program. Various characterizations of compounds in the ageing process were performed through ultra high performance liquid chromatography quadrupole-orbitrap high resolution mass spectrometry (UHPLC-Q-Orbitrap) based colloid theoretics. High-pressure shear grinding provides a high-pressure and high-temperature environment for Baijiu, accelerating the formation of flavour compounds and the decomposition of odour and harmful substances, thus improving the sensory characteristics of Feng-flavour Baijiu and shortening the ageing time. A total of 2069 substances were identified according to fragment spectra and Bayesian network model-based peak distribution. Molecules with VIP > 1, including 14 esters, 10 acids, 4 ketones, 3 aldehydes, 2 alcohols and 2 phenols, were quantified as potential markers of chemometric analysis. A novel prediction model for Baijiu ageing degree was furnished, and ageing years of Feng-flavour Chinese Baijiu were accurately predicted by factor analysis with KMO 0.784 and a polynomial regression model with r 0.94. The results demonstrated that in 400 bar, fresh distilled Baijiu could reach a natural ageing effect of 6.43 years, which was consistent with the sensory evaluation.

Thermocatalytic Degradation of Low Density Polyethylene Films at Artificial Aging Treatment under Lower Temperature

Si Zhao Zhang,Xue Guang Luo,Feng Ding,Ke Li,Xiao Yan Lin,Gao Long Zhu

Low density polyethylene (LDPE) films added thermal catalyst were investigated at artificial aging time of 0, 10, 20, and 30 days, respectively. The samples obtained were characterized by scanning electron microscopy (SEM), atomic force microscopy (AFM), gel permeation chromatograph (GPC) and fourier transform infrared spectroscopy (FT-IR). It shows that the surface of film was destroyed via thermocatalytic reactions at lower temperature. In addition, the changes of chemical groups were also observed in the thermocatalytic degradation process. Thus, the validation to the thermocatalytic route has been confirmed over lower temperature excitation. It is hoped that our work may provide a new insight into the degradation of polymeric films at lower temperature.

Low temperature catalytic oxidative aging of LDPE films in response to heat excitation

Xuegang Luo,Sizhao Zhang,Feng Ding,Xiaoyan Lin

The waste treatment of polymer materials is often conducted using the photocatalytic technique; however, complete decomposition is frequently inhibited owing to several shortcomings such as low quantum yield and the requirement of ultraviolet irradiation. Herein, we report a strategy to implement moderate management of polymeric films via thermocatalytic oxidative route, which is responsive to heat stimulus. Diverse LDPE-matrix films together with as-prepared thermal catalysts (TCs) or initiators were synthesized to further investigate heat-dependent-catalytic degradation effects. After artificial ageing, structural textures of the as-synthesized films could be chemically deteriorated, followed by a huge increase in surface roughness values, and appreciable loss was also found in the average molecular weights and mechanical parameters. We found an emergent phenomenon in which crystallization closely resembled two-dimensional (2D) growth, which displayed rod-like or disc-type crystal shapes. New chemical groups generated on film surfaces were monitored, and led to a higher limiting oxygen index because of strong catalytic oxidation, thus demonstrating the success of catalytic oxidative ageing by heat actuation. The underlying mechanism responsible for thermocatalytic oxidative pattern is also discussed. Accordingly, these findings may have important implications for better understanding the development of polymeric-matrix waste disposal.

Proteomics and Immunology Provide Insight into the Degradation Mechanism of Historic and Artificially Aged Silk

Ruru Chen,Mingzhou Hu,Hailing Zheng,Hui Yang,Lian Zhou,Yang Zhou,Zhiqin Peng,Zhiwen Hu,Bing Wang

The process and mechanism of silk degradation is still a bewildering mystery in the investigation and conservation of cultural relics, which rely on the development of accurate and tailored analysis technologies. Here, two advanced approaches, proteomics and immunology, were developed for determining the deterioration behavior of historic silk fabrics and artificially aged samples from the molecular to the holistic level. The surface morphology and secondary structure of silk were destroyed during degradation. Subsequently, the proteomics and immunology analysis demonstrated a new degradation model differing from previous reports. First, the amorphous region and the looser crystalline regions were destroyed together, and the macromolecular chains were broken randomly. Then, the tight β-sheet blocks in the crystalline region were exposed and deteriorated, which expedited the degradation of tight β-sheet blocks and relatively loose blocks in the crystalline domain as well as the amorphous domain, ultimately yielding small molecule polypeptides. However, the deterioration process of ancient fabrics could be accelerated by poor burial conditions, thus showing distinct destructive characteristics. Overall, the results gave us a more comprehensive and profound understanding of the degradation process of ancient silk.
