Protein glycosylation is of great significance in various physiological processes. Nevertheless, it remains a huge challenge to identify glycopeptides in complex biosamples by the direct mass spectrometry analysis due to the low ion efficiency and low abundance of glycopeptides. In this study, a novel hydrogel (denoted as ZIF-8/SAP) with a stable three-dimensional (3D) network structure and excellent hydrophilicity was successfully fabricated to capture glycopeptides with high efficiency. Owing to the unique characteristics, ZIF-8/SAP exhibited great selectivity (1 : 1000), great sensitivity (1 fmol μL−1), large binding capacity (300 mg g−1) and satisfactory reusability (seven cycles). Inspired by the great enrichment performance of the as-prepared material toward glycopeptides, ZIF-8/SAP was further applied to capture glycopeptides from a real human serum sample. The experimental results demonstrated that 217 N-glycosylation sites were identified in 283 N-glycopeptides, corresponding to 95 glycoproteins identified from 10 μL human serum by the nano-LC-MS/MS analysis, revealing the great potential of the novel ZIF-8/SAP hydrogel for glycopeptide enrichment and glycoproteomic research.

In this work, the binding interaction of an artificial sweetener, acesulfame (ACS) with human serum albumin (HSA) are investigated at the molecular level by using spectral methods and molecular modeling. ACS has the ability to induce static quenching of the intrinsic fluorescence of HSA by a complex formed between HSA and ACS through weak multi-noncovalent forces including hydrophobic, hydrogen bond and van der Waals forces . ACS enters subdomain IIA of HSA to induce the tertiary structure changes of HSA and decreased the hydrophobicity of protein. In addition, ACS binding does not obviously alter the secondary structure of HSA. This study is hoped to provide some crucial information for further investigations of the biosafety of sweetener.

Development of rapid and accurate detection of miRNAs in complex samples is of great significance for potential early diagnosis of disease. Herein, we report a magnetic relaxation switching (MRS)-based strategy for direct detection of miRNAs in complex samples via the assistance of signal amplification system of CRISPR/Cas13a which has the ability to specifically recognize target RNA. In the designed strategy, 30 nm-magnetic nanoparticles (MB 30 ) and 1000 nm-magnetic particles (MM 1000 ) linked by single-strand RNA 1 complexes (MB 30 -RNA 1 - MM 1000 ) were employed as signal probe. After the target miRNAs (taking miR-21 as model) recognition by CRISPR/Cas13a system, the resulted trans -cleavage degrades the MB 30 -RNA 1 -MM 1000 , releasing MB 30 which caused transverse relaxation time (T 2 ) signal change. The combination of CRISPR/Cas13a assisted signal amplification and the MRS assay achieved direct detection of miR-21 in the serum sample without extracting within 60min, with a detection limit of 0.22 pM. Moreover, the detection accuracy is confirmed by performing the detection of miR-21 using qRT-PCR. The CRISPR/Cas13a system assisted MRS assay successfully achieved accurate, simple, and rapid detection of miRNAs in complex samples, showing great potential for detection miRNAs in potential clinical applications.

Spinal muscular atrophy (SMA) is a rare autosomal recessive neuromuscular disease. Nusinersen sodium (NS) is the world's first antisense oligonucleotide (ASO) drug for SMA precise targeted therapy. However, the limited half-life of oligonucleotides and their tendency to accumulate in hepatic and renal tissues presented significant challenges for clinical investigation and therapeutic drug monitoring. In this study, we proposed an analytical strategy based on the specific capture of oligonucleotide functionalized fluorescent probes by single stranded binding proteins (SSB) for ultra-sensitive and high-throughput detection of nusinersen sodium in human serum. The magnetic nanoparticles modified with single-strand binding protein (MNPs-SSB) selectively bonded to the red fluorescent quantum dots functionalized with oligonucleotides (RQDs-ssDNA) that were complementary to nusinersen sodium. Upon interaction with nusinersen sodium, RQDs-ssDNA formed a double-stranded complex (RQDs-ssDNA-NS), resulting in enhanced red fluorescence after magnetic separation as it was no longer captured by MNPs-SSB but remained in the supernatant. A quantitative analysis of nusinersen sodium in biological samples was successfully achieved by establishing a relationship between fluorescence intensity and its concentration. The detection signal F/F0 exhibited a linear correlation (R 2 = 0.9871) over a wide range from 0.1 nM to 200 nM, with a limit of detection (LOD) of 0.03 nM, demonstrating the high specificity and rapid analysis time (only 30 min). This method provided a novel approach for sensitive, high-throughput, and specific analysis of nusinersen sodium and similar ASO drugs.

MicroRNA-21 (miRNA-21) is a class of small non-coding RNAs that play a crucial role in the regulation of post-transcriptional gene expression. By leveraging the principle of base complementary pairing, the detection of miRNA-21 can be achieved through its analogue, MicroDNA-21 (miDNA-21), which possesses an identical sequence. Utilizing screen-printed carbon electrodes (SPCE), a conducting polymer (polypyrrole, Ppy), and metal nanocomposites (gold-platinum nanoparticles, Au@Pt NPs) were sequentially electrodeposited to create an electrochemical biosensor for the detection of miDNA-21. This biosensor employed ferrocyanide/ferricyanide (Fe(CN) 6 3−/4− ) as the electrochemical signaling probe, and its structure was elucidated via field emission scanning electron microscopy (FESEM) and Energy Dispersive Spectrometer (EDS). Quantitative detection was facilitated through differential pulse voltammetry (DPV), yielding a concentration range and linear fitting equation of 10.00–7.00 × 10.00 −14 mol/L: ΔI = 0.5143 lg c + 9.191, R 2 = 0.9986. The limit of detection (LOD) was determined to be 2.90 × 10 −15 mol/L, with a sensitivity of 9.42 μA/μmol/L·cm 2 . Subsequent performance assessments confirmed the sensor’s excellent selectivity, reproducibility, and stability. Evaluation using the standard addition method, with normal human serum as the matrix, yielded recoveries ranging from 98.97 % to 102.70 %, and relative standard deviation (RSD) values below 1.00 %. These results demonstrate the method’s viability for practical application in detecting miRNA in human serum.

Elevated levels of cysteine (Cys) have been associated with many diseases and it is of vital importance to discriminate and quantify Cys in human serum. However, analogues of Cys especially homocysteine (Hcy) and glutathione (GSH) possess similar structure, which lead to a big challenge for specific detection of Cys without interference from other coexist biothiols. In this work, we developed a novel sensing approach for highly selective and sensitive determination of Cys by using fluorescence carbon dots (CDs) and cobalt-derived specific recognition. To construct such a sensing platform, cobalt-doped CDs (Co-CDs) were synthesized directly by using folic acid (FA) as precursor and CoCl 2 as dopant. The fluorescence of the Co-CDs can be specifically and intensively suppressed by the addition of Cys due to the combination with cobalt, while Hcy and GSH cannot induce any obvious fluorescence changes. Besides the robust selectivity, this new developed sensing strategy displayed a good correlation with Cys from 0.1 to 100 μM with a limit of detection (LOD) of 80 nM. This sensing platform was successfully applied to detect Cys in human serum samples with some advantages including highly selective and sensitive, convenient and rapid, as well as cost-effective, suggesting its great potential to be used in specific Cys analyzing in biological and bioanalytical applications.

Gold nanoparticles (AuNPs) catalyze the mild reaction between the weak reducing agent kojic acid (KA) and silver ions (Ag + ) to form Au@Ag bimetallic NPs by the combination of the intrinsic catalysis with plasmonic properties This is proposed as a novel optical assay to determine the tyrosinase (TYRase) concentration. The nanoparticles have been characterized by UV-vis spectroscopy, transmission electron microscope (TEM) images, and X-ray photoelectron spectroscopy (XPS). The sensing mechanism is based on the fact that KA binds to TYRase by chelating with dicopper active site of TYRase and the introduction of TYRase restrains the Au@Ag bimetallic NP formation by the precedent binding with KA. A clear color variation from yellow to pink and UV-vis spectral changes are observed at the optimal wavelength of 410 nm. The assay works in the range 0.13~0.73 U mL −1 with a detection limit (LOD) of 0.019 U mL −1 . The impact from matrix interfering substances including glucose, uric acid, common oxidases, and amino acids is negligible. The applicability is demonstrated by quantitative determination of TYRase in human serum samples with 74 to 89% recovery and RSD less than 4.0%, which accords with the level for bio-sample analysis.

l -Kynurenine (KYN) is a metabolite of the Kynurenine pathway and is a known potential marker of immune suppressant disorders and cancer. Here, we present a molecularly imprinted two dimensional (2D) Photonic crystal (PC) hydrogel sensor for the detection of L-KYN in human serum. The sensor utilizes polystyrene-based 2D PC colloidal arrays (2D PCCA) hydrogel with methacrylic acid as the functional monomer which can imprint the L-KYN template by hydrogen bonding . After removal of the template, the resulting nanocavities in the hydrogel can selectively bind and recognize L-KYN in the serum samples. The binding is selective for L-KYN, which is revealed by shrinkage of the hydrogel volume and decrease in the particle spacing that can be easily monitored through changes in the Debye diffraction ring diameter using a LASER pointer. The sensor demonstrates visible red to green color shift upon binding to L-KYN. The 2D PC sensor demonstrates the limit of detection (LOD) of 50 nM, linear relationship of particle spacing versus L-KYN concentration range (50–1000 nM) with the analytical recovery of up to 92 % in the spiked serum samples. The sensor can distinguish between L-KYN and D-KYN and is re-usable up to five times. The sensor is available for the rapid and quantitative detection of L-KYN in the human serum.

Natural plants with fascinating structures give inspirations to fabricate advanced affinity chromatography materials for glycoproteome and phosphoproteome analyses. In this study, inspired by the long-range ordered structure and rapid water transportation capability of lotus stem, a biomimetic hydrophilic material (PE/HAVP-Ti 4+ ) with parallel ordered channels is developed and applied to selectively enrich low-abundant glycopeptides and phosphopeptides in biological samples. The biomimetic PE/HAVP-Ti 4+ has honeycomb porous structure, robust mechanical property, excellent hydrophilicity, and abundant affinity sites. It demonstrates the excellent selectivity (molar ratio human IgG: BSA=1:1000, β-casein: BSA=1:2500), high sensitivity (0.2 fmol μL –1 for glycopeptides, 0.01 fmol μL –1 for phosphopeptides), satisfactory recovery rate (90.7 % for glycopeptides, 91.1 % for phosphopeptides), and good recyclability (at least 10 cycles). Moreover, after enrichment treatment by PE/HAVP-Ti 4+ , a total of 411 N - glycopeptides assigned to 212 N-glycoproteins and 63 phosphopeptides associated with 41 phosphoproteins in human serum are identified by mass spectrometry. The PE/HAVP-Ti 4+ has significant potential in applications of glycopeptide and phosphopeptide enrichment of real complex biological samples for the simultaneous study of glycoproteome and phosphoproteome.

Near−infrared (NIR) emitting Ag 2 S quantum dots (QDs) are excellent fluorescent nanoprobes for bioassays with low toxicity. A novel fluorescent sensing platform which employing NIR fluorescent Ag 2 S QDs and MnO 2 2D nanosheets as NIR emitters and quenchers is designed for rapid and selective determination of glutathione (GSH). A facile and efficient approach was demonstrated for the synthesis of NIR fluorescent Ag 2 S QDs with the emission of 845 nm. Then the NIR fluorescent nanoprobe of Ag 2 S QDs−MnO 2 nanosheets is obtained by adsorbing Ag 2 S QDs onto the surface of MnO 2 nanosheets which have atomically thick two−dimensional structure and high specific surface area. And the NIR fluorescence of Ag 2 S QDs is quenched by the MnO 2 nanosheets. The presence of GSH could reduce MnO 2 to Mn 2+ that results in the restoration of NIR fluorescence for Ag 2 S QDs. The NIR fluorescent nanoprobe could be used for highly selective detection of GSH. Also a low detection limit of 60 μmol/L was obtained. Because NIR fluorescence of the Ag 2 S QDs can efficiently reduce the interferences from background scattering and autofluorescence. The NIR fluorescent nanoprobe was directly applied to monitor the GSH level in human serum sample with high accuracy and precision.

Doxorubicin (Dox) is a DNA-targeting anthracycline antibiotic active against a wide spectrum of cancers. The interaction between Dox and double-stranded DNA (dsDNA) was used to load Dox using DNA duplexes as carriers. More importantly, the interesting DNA sequence-dependent fluorescence response of Dox could be exploited in the design of efficient Dox release systems and efficient fluorescence sensors. In this work, we demonstrated that separate introduction of G and C bases into T-rich single-stranded DNA (ssDNA) sequences afforded the best discrimination of Dox binding between dsDNA and ssDNA. For the first time, we successfully utilized this interesting DNA sequence-dependent fluorescence response of Dox as a signal transduction mechanism for the sensitive detection of biothiols in human serum. Cysteine, homocysteine, and glutathione were detected at as low as 26 nM, 37 nM, and 29 nM, respectively. The biosensors exhibited not only good selectivity, stability, and sensitivity in aqueous solutions but also a sensitive response in human serum, demonstrating their potential for diagnosis.

The development of electrochemical biosensors for the simultaneous detection of ascorbic acid (AA), dopamine (DA), uric acid (UA), tryptophan (Trp), and nitrite ( \({{\rm{NO}}}_{2}^{-}\) ) in human serum is reported in this work. Free-standing graphene nanosheets were fabricated on Ta wire using the chemical vapor deposition (CVD) method. CVD graphene, which here served as a sensing platform, provided a highly sensitive and selective option, with detection limits of AA, DA, UA, Trp, and \({{\rm{NO}}}_{2}^{-}\) of 1.58, 0.06, 0.09, 0.10, and 6.45 μM (S/N = 3), respectively. The high selectivity of the electrode is here explained by a relationship between the bandgap energy of analyte and the Fermi level of graphene. The high sensitivity in the oxidation current was determined by analyzing the influence of the high surface area and chemical structure of free-standing graphene nanosheets on analyte adsorption capacity. This finding strongly indicates that the CVD graphene electrode can be used as a biosensor to detect five analytes in human serum.
