This study investigated the in vitro fermentation of polysaccharides from Morus alba L., the contribution of its carbohydrates to the fermentation, and the effect on the composition of gut microbiota. Over 48 h of fermentation, the pH value in the fecal culture decreased from 7.12 to 6.14, and the total short chain fatty acids (SCFA) and acetic, propionic, and butyric acids all significantly increased. After 48 h of fermentation, 45.36 ± 1.36% of the total carbohydrates in the polysaccharide, including 35.72 ± 1.51% of arabinose, 23.1 ± 1.19% of galactose, 41.43 ± 1.52% of glucose, 26.36 ± 1.93% of rhamnose and 65.57 ± 1.07% of galacturic acid, were consumed. The increase in acetic and butyric acids was primarily due to the fermentation of galactose and galacturonic acid in the polysaccharide, while the increase in propionic acid resulted mainly from the fermentation of arabinose and glucose. In addition, the polysaccharide could modulate the gut microbiota composition by increasing the Bacteroidetes population and decreasing the Firmicutes population. The results may facilitate the development of food products known as prebiotics, aimed at improving gastrointestinal health.

Inulin was fermented by adult faecal microbiota and 10 exogenous strains for 24 or 48 h. The contents of acetate, propionate, butyrate and lactate were quantified in the fermented products, and the growth-inhibitory and apoptosis-inducing effects on a human colon cell line (HCT-116 cells) were assessed. Most of these strains increased contents of acetate, propionate and butyrate, and promoted lactate conversion. Correlation analysis suggested that butyrate and lactate in the fermentation products were positively and negatively correlated with the measured inhibition ratios (p < .05). The results were mostly consistent with the verification trial results using standard acid solutions. The fermentation products could cause apoptosis via inducing DNA fragmentation and increasing total apoptotic populations in the treated cells. Moreover, the fermentation products with higher growth-inhibitory activities demonstrated the increased apoptosis-inducing properties. In conclusion, these strains could cooperate with adult faecal microbiota to confer inulin fermentation products with higher anti-colon cancer activity.

Fucoidan is widely applied in food and pharmaceutical industry for the promising bioactivities. Low-molecular weight hydrolyzed fucoidan has gained attention for its beneficial health effects. Here, the modulation on microbiome and metabolome features of fucoidan and its acidolyzed derivatives (HMAF, 1.5–20 kDa; LMAF, <1.5 kDa) were investigated through human fecal cultures. Fucose is the main monosaccharide component in fucoidan and LMAF, while HMAF contains abundant glucuronic acid. LMAF fermentation resulted in the highest production of short-chain fatty acids, with acetate and propionate reaching maximum levels of 13.46 mmol/L and 11.57 mmol/L, respectively. Conversely, HMAF exhibited a maximum butyrate production of 9.28 mmol/L. Both fucoidan and acidolyzed derivatives decreased the abundance of Escherichia-Shigella and Klebsiella in human fecal cultures. Fucoidan and HMAF prefer to improve the abundance of Bacteroides . However, LMAF showed positive influence on Bifidobacterium , Lactobacillus , and Megamonas . Untargeted metabolome indicated that fucoidan and its derivatives mainly altered the metabolic level of lipids, indole, and their derivatives, with fucoidan and HMAF promoting higher level of indole-3-propionic acid and indole-3-carboxaldehyde compared to LMAF. Considering the chemical structural differences, this study suggested that hydrolyzed fucoidan can provide potential therapeutic applications for targeted regulation of microbial communities.

This study explores the effect of ultrasound-assisted cellulase (UE) processing on the structure, functional performance, and prebiotic properties of guava insoluble dietary fiber (RIDF). As the ultrasound exposure time increased (0–40 min), the particle size (from 126.3 μm to 38.7 μm) and crystallinity (from 30.72% to 14.42%) of RIDF significantly decreased, while its soluble carbohydrate content (from 1.40% to 6.17%) and water solubility (from 8.6% to 16.4%) notably increased. These crucial changes suggested the degradation and disorganization of RIDF. Meanwhile, UE-RIDF (modified RIDF) demonstrated a honeycomb network microstructure with a larger surface area, indicating distinct structural and morphological properties compared to RIDF. Consequently, UE-RIDF showed stronger retention/adsorption capacity and improved in vitro prebiotic activity. Interestingly, UE treatment increased RIDF fermentability, leading to additional shifts in its modulation of human fecal microbiota, including a higher abundance of Proteobacteria and Phascolarctobacterium . Furthermore, compared to pure cow/goat yogurt, yogurt fermented with UE-RIDF displayed a lower pH, better stability, and improved texture and rheological properties during a 21-day refrigeration period. These findings highlight the potential application of RIDF as a novel prebiotic active ingredient in fermented foods for the first time, underscoring the critical role of UE treatment in enhancing its performance.

Artemisia sphaerocephala Krasch polysaccharide (ASKP) and its two fractions—60P (branched xylan) and 60S (branched glucomannan), were subjected to simulated gastrointestinal digestion and in vitro fermentation by human fecal microbiota. The results showed that all polysaccharide fractions could transit through gastrointestinal tract without dramatic degradation and be utilized by gut microbiota. ASKP exhibited the highest depletion rate and highest capability to decrease the pH than its fractions. Meanwhile, 60S showed the stronger capability to increase the production of propionic acid and reduce the ratio of acetic acid to propionic acid. At the phylum level, all polysaccharides efficiently reduced the Firmicutes/Bacteroidetes ratio and relative abundance of Proteobacteria, with ASKP being the most capable to suppress the proliferation of Proteobacteria. At the genus level, ASKP and 60P markedly promoted the growth of Bacteroidetes , and 60S promoted the growth of Parabacteroides and Collinsella . Prediction on metabolic function revealed that polysaccharide administration could dramatically change the metabolic profile of bacteria compared with fructooligosaccharides. Besides, all the polysaccharides dramatically promoted the bile acid metabolism. Compared with 60S, ASKP and 60P showed stronger ability to suppress the metabolisms on carbohydrate and amino acid. In summary, both ASKP and its two fractions showed the prebiotic potentials.

Acrolein (ACR) derives from the external environment and the endogenous metabolism of organisms. It has super-reactivity and can induce various diseases. We investigated the capacity of cyanidin-3-O-glucoside (C3G) and its degradants/metabolites to capture ACR during thermal processing or in vivo. Our results indicated that both C3G and its degradants, including phloroglucinaldehyde (PGA) and protocatechuic acid (PCA), could efficiently trap ACR to form adducts, such as C3G–ACR, C3G–2ACR, PGA–ACR, PGA–2ACR, PCA–ACR, and PCA–2ACR. Additionally, these adducts were detected in commercial canned red bayberry products. The adducts of C3G and its metabolites conjugated with ACR, such as C3G–ACR, C3G–2ACR, PGA–ACR, and 4-hydroxybenzoic-acid–ACR (4-HBA–ACR), were also detected in mice feces treated with C3G by oral gavage, where the adduct level was dose-dependent. A similar pattern was observed in tests on human consumption of red bayberry. In human urine, only PGA–2ACR and 4-HBA–ACR, were found, whereas C3G–ACR, C3G–2ACR, myricetin-3-O-rhamnoside–ACR (M3R–ACR), PGA–2ACR, 4-HBA–ACR and ferulic acid–ACR (FA–ACR) were detected in human feces following administration of red bayberry. Our results are the first demonstration that C3G and its metabolites can capture ACR in vitro and in vivo (mice and humans) and present a novel strategy, the development of C3G as a promising ACR inhibitor.

Chitooligosaccharides (COS), with a specific degree of polymerization, were evaluated for their effect on human fecal microbiota, as well as their metabolite, using an in vitro anaerobic batch fermentation model. COS increased the richness and diversity of the gut microbial community. More importantly, the microbial community structure of the COS groups was significantly different from the control and fructooligosaccharides (FOS) groups. COS specifically enriched the butyric and propionic acid-producing microbiota and targeted Faecalibacterium , Clostridium_sensu_stricto_1 , C._sensu_stricto_13 , and Fusicatenibacter . Furthermore, COS increased the acetic, propionic, and butyric acid contents. The production of propionic acid was greater than that obtained using FOS. Metabolome analysis suggested that COS could upregulate allocholic acid, 1,3-butanediol, 3,4-dihydroxybutanotic acid, and 2-hydroxybutyric acid production and downregulate putrescine, ethanamine, hexanoic acid, and branched chain amino acid ( l -valine and l -leucine) production in the fermentation broth. These differential metabolites were closely related to lipid, glucose, and amino acid metabolism. These results should inform further development of COS as potential raw materials for functional foods and health products.
