Photodynamic therapy (PDT) has been widely used in cancer treatment. However, hypoxia in most solid tumors seriously restricts the efficacy of PDT. To improve the hypoxic microenvironment, we designed a novel mesoporous platinum (mPt) nanoplatform to catalyze hydrogen peroxide (H 2 O 2 ) within the tumor cells in situ without an extra enzyme. During the fabrication, the carboxy terminus of the photosensitizer chlorin e6 (Ce6) was connected to the amino terminus of the bifunctional mercaptoaminopolyglycol (SH-PEG-NH 2 ) by a condensation reaction, and then PEG-Ce6 was modified onto the mPt moiety via the mercapto terminal of SH-PEG-NH 2 . Material, cellular and animal experiments demonstrated that [email protected] catalyzed H 2 O 2 to produce oxygen (O 2 ) and that Ce6 transformed O 2 to generate reactive oxygen species (ROS) upon laser irradiation. The [email protected] nanoplatform with uniform diameter presented good biocompatibility and efficient tumor accumulation. Due to the high atomic number and good near-infrared absorption for Pt, this [email protected] nanoplatform showed computed tomography (CT) and photoacoustic (PA) dual-mode imaging ability, thus providing an important tool for monitoring the tumor hypoxic microenvironment. Moreover, the [email protected] nanoplatform reduced the expression of hypoxia-inducible factor-1 α (HIF-1 α ) and programmed death-1 (PD-1) in tumors, discussing the relationship between hypoxia, PD-1, and PDT for the first time.

Elevating photothermal conversion efficiency (PCE) is the key to enhance curative efficacy during photothermal therapy. Nevertheless, current functionalized or synthetic methods for preparing photothermal agents with high PCE are too complicated, greatly hindering the development of photothermal therapy. In this study, we developed a tumor-targeting flower-like nanocomposite termed mesoporous platinum nanoparticles@Cu 2- x Se-AS1411 (MPNPs@Cu 2- x Se-AS1411, denoted as MPCSA) with remarkable PCE for high-resolution photoacoustic (PA) imaging guided photothermal/chemodynamic therapy for tumor ablation. We discovered that MPNPs served as carriers for delivering PA contrast agents. Cu 2- x Se nanosheets emerged as promising near-infrared-II (NIR-II) PA imaging and photothermal/chemodynamic therapy agents owing to their strong localized surface plasmon resonance property in the NIR-II region. The amine-modified AS1411 aptamer, which is connected to the surface of the flower-like MPNPs@Cu 2- x Se can target the nucleolar protein on the surface of 4T1 cells, achieving the accumulation of MPCSA at the tumor site in mice. Notably, these MPCSA nano-flowers exhibited an excellent PCE up to 82.33 % under 1064 nm laser irradiation, resulting in highly efficient cancer cell ablation and greatly hindering in vivo tumor growth. This study has paved the way for imaging-guided accurate cancer diagnosis and targeted cancer therapy.

Combination cancer therapy could maximize the benefits of immunotherapy by remodeling the tumor microenvironment. Platinum has been used in cancer therapy due to its unique antitumor, photothermal and catalytic properties. Herein, we successfully developed a mesoporous platinum nanoparticle (mPt)-based trinity strategy nanoplatform for enabling synergistic photodynamic therapy (PDT), photothermal therapy (PTT) and immunotherapy. Specifically, platinum nanoparticles were first fabricated and decorated with a silica shell for photosensitizer hematoporphyrin monomethyl ether (HMME) conjugation and further coated with a thermosensitive lipid bilayer containing an immune adjuvant resiquimod (R848), resulting in a multifunctional nanoplatform (RH@PSN@Lip). The visible red laser activation not only directly killed tumor cells but also induced tumor immunogenic cell death (ICD). mPt also served as a nano-enzyme to alleviate the hypoxic microenvironment of tumors and generated oxygen for PDT. Furthermore, the visible red laser served as a “switch” to trigger the release of R848 from the lipid bilayer of RH@PSN@Lip, thereby strengthening anti-tumor immune responses and remodeling the tumor microenvironment. Collectively, this trinity strategy nanoplatform successfully increased tumor-infiltrating lymphocytes and reversed the suppressive condition in the tumor microenvironment. Consequently, melanoma growth was remarkably inhibited, and mice were also protected from tumor rechallenging. Overall, this work presents an innovative and promising synergistic therapeutic strategy for clinical cancer treatment.

Mesoporous platinum (mPt) nanozyme possessed enzyme-like property of catalase (CAT) and peroxidase (POD), but the insufficient hydrogen peroxide (H 2 O 2 ) concentration severely restricted its application in photodynamic therapy (PDT) and catalytic therapy. Herein, by depositing ultrasmall gold nanoparticles (AuNPs) and modifying photosensitizer IR808, a multifunctional nanozyme (mPt@Au-IR808) was designed to promote PDT/catalytic therapy through cascade enzyme-like reactions of glucose oxidase (GOx) and CAT/POD. In tumor microenvironment, the CAT-like oxygen (O 2 ) generation improved the PDT efficacy, and the POD-like hydroxyl radical (·OH) generation achieved endogenous catalytic therapy. Using the GOx/CAT-like activities and endogenous H 2 O 2 , the yields of singlet oxygen and ·OH were significantly promoted. Furthermore, mPt@Au-IR808 showed higher photothermal conversion efficiency (41.2%) than mPt (36.1%). By combining the photothermal therapy and enhanced PDT/catalytic therapy, the developed mPt@Au-IR808 nanozyme showed excellent anti-tumor efficacy, which will be promising as cascade nanozyme to promote photo/catalytic therapy.
