Metabolic responses to ethanol and butanol in Chlamydomonas reinhardtii

Jiang Yongguang,Xiao Peng,Shao Qing,Qin Huan,Hu Zhangli,Lei Anping,Wang Jiangxin

Background Microalgae have been demonstrated to be among the most promising phototrophic species for producing renewable biofuels and chemicals. Ethanol and butanol are clean energy sources with good chemical and physical properties as alternatives to gasoline. However, biosynthesis of these two biofuels has not been achieved due to low tolerance of algal cells to ethanol or butanol. Results With an eye to circumventing these problems in the future and engineering the robust alcohol-producing microalgal hosts, we investigated the metabolic responses of the model green alga Chlamydomonas reinhardtii to ethanol and butanol. Using a quantitative proteomics approach with iTRAQ-LC–MS/MS technologies, we detected the levels of 3077 proteins; 827 and 730 of which were differentially regulated by ethanol and butanol, respectively, at three time points. In particular, 41 and 59 proteins were consistently regulated during at least two sampling times. Multiple metabolic processes were affected by ethanol or butanol, and various stress-related proteins, transporters, cytoskeletal proteins, and regulators were induced as the major protection mechanisms against toxicity of the organic solvents. The most highly upregulated butanol response protein was Cre.770 peroxidase. Conclusions The study is the first comprehensive view of the metabolic mechanisms employed by C. reinhardtii to defend against ethanol or butanol toxicity. Moreover, the proteomic analysis provides a resource for investigating potential gene targets for engineering microalgae to achieve efficient biofuel production.

Assessing the Effects of Cadmium Stress on the Growth, Physiological Characteristics, and Metabolic Profiling of Rice (Oryza sativa L.) Using HPLC-QTOF/MS

Zhenni Lan,Qing He,Mingxia Zhang,Huahong Liu,Liusen Fang,Jinfang Nie

Cadmium (Cd) pollution is an important environmental problem, as it is easily absorbed by plants and gradually accumulates in the human body through the food chain. This study aimed to elucidate the changes in the metabolic response of the rice cultivar “TanLiangYou215” under Cd stress. Rice was grown in soil culture at 0 (Control), 2 (Low group), and 10 (High group) mg/kg CdCl2for 90 days. The ultrastructural, Cd content, antioxidant activity, and metabolic changes to the rice in different tissues were analyzed. Phenotypic characterization and ultrastructure showed that the rice roots and leaves were significantly damaged and plant growth was inhibited in the High group, while plant growth was promoted in the Low group. Overall, Cd showed a regularity of “low promotion and high inhibition”. Physiological indices revealed that rice was significantly affected by Cd stress. Compared to the Control, Cd stress resulted in higher antioxidant enzyme activities, and the Low group suffered less oxidative damage than the High group. Metabolomic studies revealed that Cd stress significantly altered the metabolic profiles of rice plants. Rice responded to Cd stress by upregulating amino acids and regulating related pathways, including alanine, aspartate and glutamate metabolism, and arginine and proline metabolism. The significant expression of flavonoids with antioxidant properties helped rice resist the oxidative damage caused by Cd accumulation in the root tissue; Cd stress significantly downregulated glycerophospholipid metabolism in the stem and leaf tissues, which affected the cellular activities in rice stem and leaf tissues. We investigated the effects of Cd stress on ultrastructure, antioxidant activity, and metabolic changes in different tissues of the rice variety TLY215. Moreover, the different tissues of TLY215 can regulate these metabolic pathways to resist Cd stress, which provides valuable insights into the response of TLY215 to different concentrations of Cd.Keywords:cadmium (Cd);rice (Oryza sativaL.);phenotypic characterization;physiological activity;metabolomics

Self-detoxification behaviors and tissue-specific metabolic responses of fishes growing in erythromycin-contaminated water

Xu-Hui Huang,Rong Liu,Jia-Nan Chen,Xiu-Ping Dong,Bei-Wei Zhu,Lei Qin

Erythromycin has caused severe risk on the aquatic ecological environment and food chain propagation due to its extensive use, environmental persistence, and low efficiency of sewage removal. Our study monitored the erythromycin accumulation, self-detoxification behavior, and metabolic response of turbot in erythromycin-contaminated environments using a multi-omics method with the aim to improve the safety of turbot consumption and provide guidance for turbot culture. Erythromycin was found to rapidly accumulated and reached its peak in turbot in the first two days of exposure to erythromycin. Residual levels in the gill, liver, kidney, muscle, and blood were detected at 95.19, 72.80, 68.74, 51.3, and 12.33 ng/g, respectively. The risk of erythromycin residues in turbot can be effectively reduced by a withdrawal period longer than 12 d. Erythromycin-contaminated turbot muscles showed a significant decrease in flavor properties, particularly umami, sweet, and aroma precursors. A total of 232 lipids in four main categories, glycerides, phospholipids , fatty acyl, and sphingolipids, were found to show significant changes in the turbot muscles. The interacting metabolic network formed by taurine, glycerophospholipid, arginine, alanine , and glutamate metabolism, wherein turbot responded to erythromycin was verified through 14 significantly altered metabolites. A withdrawal period of at least half a month and the corresponding product labels for antibiotics should be formulated before the fish exposed to antibiotics during intensive culture are sold to protect the interests of consumers. This study could serve as a reference for the quality and safety risk assessment of turbot and other highly economical fishes affected by antibiotics exposure.

Metabolic responses of tea (Camellia sinensis L.) to the insecticide thiamethoxam

Weiting Jiao,Ping Zhang,Chuanjian Cui,Min Yan,Qing X. Li,Yongfeng Tang,Nan Zhang,Xinyi Wang,Ruyan Hou,Rimao Hua

Background Thiamethoxam (TMX) is insecticidal, but also can trigger physiological and metabolic reactions of plant cycles. The objective of this work was to evaluate the physiological and metabolic effect of TMX on tea plants and its potential benefits. Results In this study, dose of TMX (0.09, 0.135 and 0.18 kg a.i./ha) were tested. Except for peroxidase (POD) and glutathione S-transferase (GST), chlorophyll, carotenoid, catalase (CAT) and malondialdehyde (MDA) were significantly affected compared with the controls. The CAT activity was increased by 3.38, 1.71, 2.91 times, respectively, under three doses of TMX treatment. The metabolic response between TMX treatment and control groups on the third day was compared using a widely targeted metabolomics. A total of 97 different metabolites were identified, including benzenoids, flavonoids, lipids and lipid-like molecules, organic acids and derivatives, organic nitrogen compounds, organic oxygen compounds, organoheterocyclic compounds, phenylpropanoids and polyketides, and others. Those metabolites were mapped on the perturbed metabolic pathways. The results demonstrated that the most perturbation occurred in flavone and flavonol biosynthesis. The beneficial secondary metabolites luteolin and kaempferol were upregulated 1.46 and 1.31 times respectively, which protect plants from biotic and abiotic stresses. Molecular docking models suggest interactions between TMX and flavonoid 3-O-glucosyltransferase. Conclusion Thiamethoxam spray positively promoted the physiological and metabolic response of tea plants. And this work also provided the useful information of TMX metabolism in tea plants as well as rational application of insecticides. © 2023 Society of Chemical Industry.

Deciphering Alternaria metabolic responses in microbial confrontation via an integrated mass spectrometric targeted and non-targeted strategy

Ye Tian,Mohamed F. Abdallah,Marthe De Boevre,Kris Audenaert,Cheng Wang,Sarah De Saeger,Aibo Wu

Microbial confrontation is ubiquitously present in nature, such as mycoparasitism, competition and antibiosis between biocontrol agents and microbial pathogens. However, the internal metabolic responses of fungal pathogens under microbial interaction scenario have been scarcely investigated. In this study, we set up an integrated mycotoxin analysis and non-targeted metabolomics workflow to decipher metabolic changes of Alternaria pathogens when confronted with selected Trichoderma strains , as well as mycotoxin metabolization in the Trichoderma spp. Results demonstrated that Trichoderma spp. significantly influenced mycotoxin production and whole metabolome of Alternaria pathogens when in cocultivation, and one Trichoderma strain could metabolize alternariol into its hydroxylated form. These differential metabolites revealed fungal physiological alternations in various confrontation conditions . In all, a MS-based strategy was proposed to investigate microbial metabolic profiles under fungal/fungal and fungal/mycotoxin cocultivation, and this generic methodology would be significant for understanding the occurrence and change of food contaminants in microbial interactions.
