Differential metabolic profiles of pigment constituents affecting leaf color in different albino tea resources

Yingjuan Chen,Yuxin Han,Shiqiang He,Qinghua Cheng,Huarong Tong

Albino tea cultivars with distinct leaf colors have rarely been used to explore the metabolic mechanism of leaf color formation. Carotenoids and chlorophyll a re primary pigments affecting leaf color with differential metabolic mechanisms in albino tea resources. TEM analysis showed that in albino stage, much less chloroplasts with abnormal structure were observed in white and yellow tea leaves, which corresponded to the significant reduction of total chlorophyll, Chl a and Chl b, and increase of Chl a/b, resulting in tea leaves to lose their green color. Compared to green tea ‘Fudingdabaicha’ (FD), three albino tea cultivars had higher number of carotenoids, but much lower contents. For albino mutants, significant lower α-Carotene, α-cryptoxanthin and lutein involved in lutein biosynthesis and remarkable higher zeaxanthin ( P  < 0.05) were responsible for the leaf yellowing of ‘Huangjinya’ (HJY), while zeaxanthin degradation and accumulation of antheraxanthin, violaxanthin and neoxanthin contribute to the white appearance of ‘Jibai’ (JB).

Characterization of metabolic fate of phellodendrine and its potential pharmacological mechanism against diabetes mellitus by ultra-high-performance liquid chromatography-coupled time-of-flight mass spectrometry and network pharmacology

Feng-Xiang Zhang,Yu-Lin-Lan Yuan,Shuang-Shuang Cui,Min Li,Rui-Man Li

Rationale Characterizing the functional mechanism of quality control marker (Q-marker) was of great importance in revealing the primary pharmacological mechanism of herbs or the other complex system, and drug-related metabolites always contribute to the pharmacological functions. Cortex Phellodendri was used as a core herb in the treatment of diabetes mellitus (DM). As a Q-marker of Cortex Phellodendri, the role of phellodendrine in DM was still unclear. Thus, the characterization of phellodendrine-related metabolites in vivo and the subsequent induced functional mechanism exerted great importance in elucidating the anti-DM mechanism of Cortex Phellodendri. Methods An ultra-high-performance liquid chromatography-coupled time-of-flight mass spectrometry (UHPLC/Q-TOF MS) method was developed to profile metabolites of phellodendrine in rats. The potential pharmacological mechanism against DM was predicted by network pharmacology. Results A total of 19 phellodendrine-related metabolites were screened out in rats for the first time. Among them, M4 , M5 , M9 , and M12 were regarded as the primary metabolites. Meanwhile, phase I metabolic reactions of hydroxylation, demethylation, and isomerization and phase II reactions of glucuronidation and sulfation occurred to phellodendrine; glucuronidation and hydroxylation were the two main metabolic reactions. Moreover, the potential targets of phellodendrine and three main metabolites ( M4 , M5 , and M12 ) were predicted by a network pharmacological method, and they mainly shared 52 targets, including PDE5A, CHRNA3, SIGMAR1, F3, ESR1, DRD1, DRD2, DRD3, and DRD4. Furthermore, Kyoto Encyclopedia of Genes and Genomes pathway analysis showed that calcium signaling pathway, cGMP-PKG signaling pathway, and cAMP signaling pathway were regarded as the core mechanism of phellodendrine to treat DM. Conclusion The metabolic feature of phellodendrine in vivo was revealed for the first time, and its anti-DM mechanism information for further pharmacological validations was also supplied. It also gave a direction to further elucidation of pharmacological mechanism of Cortex Phellodendri in treating DM.

Landscape of circulating metabolic fingerprinting for keloid

Hu Yu,Zhou Xuyue,Chen Lihao,Li Rong,Jin Shuang,Liu Lingxi,Ju Mei,Luan Chao,Chen Hongying,Wang Ziwei,Huang Dan,Chen Kun,Zhang Jiaan

Background: Keloids are a fibroproliferative disease characterized by unsatisfactory therapeutic effects and a high recurrence rate. Objective: This study aimed to investigate keloid-related circulating metabolic signatures. Methods: Untargeted metabolomic analysis was performed to compare the metabolic features of 15 keloid patients with those of paired healthy volunteers in the discovery cohort. The circulating metabolic signatures were selected using the least absolute shrinkage. Furthermore, the selection operators were quantified using multiple reaction monitoring-based target metabolite detection methods in the training and test cohorts. Results: More than ten thousand metabolic features were consistently observed in all the plasma samples from the discovery cohort, and 30 significantly different metabolites were identified. Four differentially expressed metabolites including palmitoylcarnitine, sphingosine, phosphocholine, and phenylalanylisoleucine, were discovered to be related to keloid risk in the training and test cohorts. In addition, using linear and logistic regression models, the respective risk scores for keloids based on a 4-metabolite fingerprint classifier were established to distinguish keloids from healthy volunteers. Conclusions: In summary, our findings show that the characteristics of circulating metabolic fingerprinting manifest phenotypic variation in keloid onset.

Combination of Metabolite Analysis and Network Pharmacology to Explore the Potential Anticough Mechanism of Protopine—A Marker in Zhi-Ke-Bao Tablets

Qi-Feng Zou,De-Jian Chen,Cheng-Jun Liu,Zi-Hao Chen,Xia Yang,Rong-Huang Xu,Zhen-Hui Zhou,Jian-Xin Chen,Wei Shi,Feng-Xiang Zhang

Rationale Protopine, an active alkaloid in Papaver somniferum L., was abundant in a well-known anticough traditional Chinese medicine preparation—Zhi-Ke-Bao tablets. Till now, the metabolism feature and anticough mechanism of protopine have not been fully elucidated, restricting its further development. Methods The metabolites of protopine in rats were profiled by using ultra-high performance liquid chromatography coupled with time-of-flight mass spectrometry, and its anticough targets and mechanism were predicted by network pharmacology. Results In rats, a total of 19 metabolites were identified following ingestion of protopine (21 mg/kg/day, i.g.), including 4 in plasma, 6 in urine, 5 in feces, 10 in liver, 2 in spleen, 4 in lung, 3 in kidney, 3 in heart, and 3 in brain. The main metabolic features were ring-opening, methylation, demethylation, glucuronidation, sulfation, and hydroxylation. Among them, methylation, sulfation, and hydroxylation of protopine in vivo were revealed for the first time. The network pharmacology results show that protopine and its metabolites regulate physiological activities by acting on STAT3, SRC, CASP3, MTOR, MMP9, ESR1, and other targets, involving PI3K-Akt signaling pathway, FoxO signaling pathway, and TNF signaling pathway, etc. Conclusions The metabolic features of protopine and its potential mechanisms for anticough effects were outlined, providing data for further anticough pharmacological validation of protopine.

Antibiotic-induced gut microbiota dysbiosis altered host metabolism

Mengxue He,Jiachen Shi,Aiyang Liu,Yong-Jiang Xu,Yuanfa Liu

Antibiotics are useful for treating infections caused by bacteria, but they have negative effects on the host body. The goal of this study was to determine whether antibiotics alter the metabolic phenotype of the host. We found that taking antibiotics reduced the diversity and richness of gut microbiota and affected the composition of the microbiome, which in turn altered the metabolic profiles of plasma and fecal samples. Additionally, plasma and fecal metabolites and gut microbiota genera showed a significant association. The most significant pathways related to the gut dysbiosis induced by antibiotics including purine, pentose, and glucuronate metabolism, histidine, ascorbate and alternate, lysine degradation, and fatty acid biosynthesis. The relationship between gut microbiota and altered metabolites of plasma and feces provides information about bacterial action, which is useful for designing new microbiota-based disease prevention and treatment interventions.

Tissue distribution, excretion characteristics and metabolic profiling studies of oxypeucedanin in rats using liquid chromatography tandem mass spectrometry

Huixiao Duo,Liyun Wang,Pei Lu,Xiaodan Zhang,Mingcong Zheng,Hanying Song,Juan Zhou,Wenzhuo Lei,Shushu Ding,Jie Jia Li,Junxu Li,Qing Zhu

Oxypeucedanin (OPD) is a linear furanocoumarin extracted from Chinese herbs, which has been reported to exhibit various pharmacological effects such as analgesia, antitussive, anticancer and anti-inflammatory activities. In this paper, the tissue distribution, excretion and metabolism characteristics of OPD in rat was conducted in order to better understand this compound and for potential development of OPD as a medicine. We first used the established HPLC/MS/MS method to examine the plasma protein binding rate and the tissue distribution of OPD in rats after oral administration. The excretion characteristics of OPD in urine, feces and bile, as well as its metabolic profiles in plasma, urine and bile were also investigated. The results demonstrated that after OPD administration, extensive metabolism occurred in rats, involving first-pass effect and enterohepatic circulation, with significant sex differences observed in the metabolic process. The study identified eight metabolites in rats, indicating that OPD metabolism primarily occurs through oxidation, reduction, and glucuronidation. In summary, this study systematically examined the basic process of OPD in vivo, providing a robust foundation for accurately predicting the behavior, efficacy and safety of OPD in humans.

Distinguish Esophageal Cancer Cells through VOCs Induced by Methionine Regulation

Jijuan Zhou,Dianlong Ge,Yajing Chu,Yue Liu,Yan Lu,Yannan Chu

Detection of exhaled volatile organic compounds (VOCs) is promising for noninvasive screening of esophageal cancer (EC). Cellular VOC analysis can be used to investigate potential biomarkers. Considering the crucial role of methionine (Met) during cancer development, exploring associated abnormal metabolic phenotypes becomes imperative. In this work, we employed headspace solid-phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS) to investigate the volatile metabolic profiles of EC cells (KYSE150) and normal esophageal epithelial cells (HEECs) under a Met regulation strategy. Using untargeted approaches, we analyzed the metabolic VOCs of the two cell types and explored the differential VOCs between them. Subsequently, we utilized targeted approaches to analyze the differential VOCs in both cell types under gradient Met culture conditions. The results revealed that there were five/six differential VOCs between cells under Met-containing/Met-free culture conditions. And the difference in levels of two characteristic VOCs (1-butanol and ethyl 2-methylbutyrate) between the two cell types intensified with the increase of the Met concentration. Notably, this is the first report on VOC analysis of EC cells and the first to consider the effect of Met on volatile metabolic profiles. The present work indicates that EC cells can be distinguished through VOCs induced by Met regulation, which holds promise for providing novel insights into diagnostic strategies.

Untargeted metabolomics combined with chemometrics reveals distinct metabolic profiles across two sparkling cider fermentation stages

Wenting Li,Qi Guo,Yuning Zhao,Tianli Yue,Yahong Yuan

This study aimed to examine the metabolic profiles of Saccharomyces cerevisiae yeasts (WLS21 and Y41) in two phases of sparkling cider making (normal and pressure fermentation) by combining untargeted metabolomic with chemometrics. The results showed that of the 634 nonvolatile metabolites identified using LC-MS and 83 volatile metabolites identified by GC–MS, the differential metabolites were 226 and 54, respectively. Metabolic pathway and correlation analyses showed that aspartic acid, phenylalanine and tyrosine, glutamic acid and purine metabolism were associated with flavor formation. The pressure fermentation process increased apigenin, naringenin, toxifolin, pyridoxine and thiamine contents in the final cider. These findings provide useful information and new research ideas for the formation of flavor in sparkling cider and the regulation of phenolic and vitamin production by microbial stress fermentation.

Distinct effects of nano-polystyrene, erythromycin, and their mixtures on the composition and metabolic profile of intestinal microbiota in Nereis succinea

Dali Wang,Yuheng Zhong,Qi Ding,Xiaofeng Han,Kai Chen,Bo Pan,Jing You

Nanoplastics (NPs) and antibiotics are emerging contaminants that co-occur ubiquitously in coastal environments and pose risks to ecosystems. Here, we report the single and combined effects of nano-sized polystyrene (PS) and erythromycin on a marine benthic invertebrate N. succinea at both individual and intestinal microbial levels. It was found that PS at 10 and 1000 μg L−1 and erythromycin at 249 and 7170 ng L−1, individually and in combination, did not cause mortality of N. succinea, but altered organism weight and behaviors and led to morphological changes and oxidative stresses in the intestines. Synergistic effects on glutathione-S-transferase activity and burrowing behavior were observed for mixtures containing high levels of PS. The results of 16S rRNA sequencing showed that erythromycin at both concentrations did not cause changes in microbial composition at the phylum level, while PS and the mixtures significantly increased the relative abundance of Proteobacteria, and reduced Actinobacteriota, Bacteroidota and Firmicutes. At the genus level, erythromycin decreased some potential pathogens such as Vibrio, Nautella, and Pseudomonas, whereas PS and the mixtures reduced beneficial genera such as Megasphaera and Megamonas. Community-level physiological profiles showed that erythromycin and PS had distinct influences on the metabolism of the cultivable intestinal microbiota, with erythromycin enhancing while PS and the mixtures inhibiting the metabolic capacity and diversity. Collectively, our results illustrated that PS NPs and erythromycin exerted distinct influences on the intestinal microbiota of N. succinea, and their mixture effects were dominated by the PS effect.

Transcriptome-aligned metabolic profiling by SERSome reflects biological changes following mesenchymal stem cells expansion

Xinyuan Bi,Bin Ma,Wei Liu,Wei-Qiang Gao,Jian Ye,Hanyu Rao

Background Mesenchymal stem cells (MSCs) are widely applied in the treatment of various clinical diseases and in the field of medical aesthetics. However, MSCs exhibit greater heterogeneity limited stability, and more complex molecular and mechanistic characteristics compared to conventional drugs, making rapid and precise monitoring more challenging. Methods Surface-enhanced Raman spectroscopy (SERS) is an ultrasensitive, tractable and low-cost fingerprinting technique capable of identifying a wide range of molecules related to biological processes. Here, we employed SERS for reproducible quantification of ultralow concentrations of molecules and utilized spectral sets, termed SERSomes, for robust and comprehensive intracellular multi-metabolite profiling. Results We revealed that with increasing passage number, there is a gradual decline in cell expansion efficiency, accompanied by significant changes in intracellular amino acids, purines, and pyrimidines. By integrating these metabolic features detected by SERS with transcriptomic data, we established a correlation between SERS signals and biological changes, as well as differentially expressed genes. Conclusion In this study, we explore the application of SERS technique to provide robust metabolic characteristics of MSCs across different passages and donors. These results demonstrate the effectiveness of SERSome in reflecting biological characteristics. Due to its sensitivity, adaptability, low cost, and feasibility for miniaturized instrumentation throughout pretreatment, measurement, and analysis, the label-free SERSome technique is suitable for monitoring MSC expansion and offers significant advantages for large-scale MSC manufacturing. Lactiplantibacillus plantarum LZU-J-Q21 enhanced the functional metabolic profile and bioactivity of Cistanche deserticola

Kangkang Liu,Junxiang Li,Wenting Hao,Jingjing Li,Israr Khan,Yibo Liang,Haijuan Wang,Xiaofeng Li,Chunjiang Zhang

Microbial fermentation is an effective method to enhance the bioavailability of herbs. This study utilized Lactiplantibacillus plantarum LZU-J-Q21 to ferment Cistanche deserticola and evaluated its metabolic properties and biological activity. Results showed that the contents of total acid and flavone, and the clearance rates of DPPH, ABTS and OH − in fermented Cistanche deserticola (FCD) were increased by 142.74 %, 56.45 %, 58.1 %, 62.3 %,51.2 %, compared with non-fermented Cistanche deserticola (NFCD). The metabolic profile of FCD had remarkable changes, especially elevated glucose and adenosine (97.31 % and 59.18 %). Further, FCD increased the weight-bearing swimming time of mice by 88.57 %, reduced fatigue markers BUN, BLA, and MDA (18.47 %, 12.92 %, and 15.16 %), and enhanced liver/muscle glycogen and SOD (28.99 %, 28.57 %, and 14.47 %). The investigation into its anti-fatigue mechanism suggested that FCD enhanced GS protein expression by activating PI3K/AKT/GSK3β signaling. These findings suggest that FCD enhances anti-fatigue effects by modifying its metabolic properties and biological activity.

Plasma Metabolic Profiles of Chronic and Recurrent Uveitis Treated by Artesunate in Lewis Rats

Xinyi Gong,Jingchuan Fan,Hui Huang,Fei Xu,Kaijiao Hu,Jianping Liu,Yi Tan,Feilan Chen

Background/Objectives: Identifying effective and safe treatment options for non-infectious uveitis remains challenging due to chronic and relapsing ocular inflammation. Previous studies have shown that artesunate (ART) plays an immunosuppressive role in several classic autoimmune diseases, including uveitis. However, its impact on the plasma metabolic profile of recurrent autoimmune uveitis remains unclear. This study aims to explore the effect of ART on the plasma metabolic features of recurrent experimental autoimmune uveitis (EAU) in a Lewis rat. Methods: Rats were clinically and pathologically evaluated for the development of recurrent EAU induced by inter-photoreceptor retinoid-binding protein (IRBP) R16 peptide-specific T-cells (tEAU). The disruptive effects of ART on tEAU were investigated to evaluate the potential role of rat recurrent EAU. Differentially expressed metabolites were identified in the plasma of rats by untargeted metabolomics analysis after ART treatment. The differential metabolites were applied to subsequent pathway analysis and biomarker analysis by MetaboAnalyst. Results: ART can significantly alleviate the severity of clinical signs and pathological injuries of eyeballs with tEAU. Both non-supervised principal component analysis and orthogonal partial least-squares discriminant analysis showed 84 differential metabolites enriched in 16 metabolic pathways in the tEAU group compared with heathy controls and 51 differential metabolites enriched in 17 metabolic pathways, including arginine and proline metabolism, alanine metabolism, and aminoacyl-tRNA biosynthesis, in the ART-treated group compared with the tEAU group. Particularly, upregulated L-alanine levels in both alanine metabolism and aminoacyl-tRNA biosynthesis were associated with T-cell activation, while elevated spermidine and N-acetyl putrescine levels in arginine and proline metabolism related to T-cell differentiation proved to be valuable biomarkers for ART treatment. Conclusions: Our study demonstrates that ART treatment can alleviate recurrent uveitis by altering the plasma metabolic characteristics associated with T-cell activation and differentiation, which might provide novel insights for potential therapeutic treatments.
