Integrated physiological, transcriptomics, and metabolite analysis reveal photosynthetic toxicity related to the bisphenol A stress response mechanism in pepper (Capsicum annuum L.)

Chao Huang,Yilin Liu,Jing Lu,Xunzhi Deng,Zhoubin Liu,David P. Molloy,Langtao Xiao,Zhoufei Luo

Bisphenol A (BPA), an emerging endocrine-disrupting chemical found in irrigation water, soil, and agricultural plastic mulch, presents potential exposure risks to pepper plants ( Capsicum annuum L.). This study investigated BPA impact, observing that BPA accumulates in a concentration-dependent manner, with highest levels in roots and lowest in fruit. High BPA exposure decreased photosynthetic pigment content, reduced stomatal size, caused chloroplast dilation, and increased reactive oxygen species and malondialdehyde accumulation, disrupting hormone balance in leaves with a 2.3-fold increase in abscisic acid content under 100 mg/kg BPA treatment. Integrated multi-omics analyses revealed that BPA toxicity operates through interference with galactose metabolism, starch-sucrose metabolism, plant hormone signaling, and glutathione metabolism pathways. Key molecular responses included significant downregulation of photosynthesis-related genes and substantial metabolic perturbations, notably an 8.1-fold reduction in uridine diphosphate fructose and 10.6-fold accumulation of indole-3-ethanol. By employing an integrated multi-omics approach, this study provides a comprehensive mechanistic understanding of BPA phytotoxicity that extends beyond conventional physiological observations. The findings offer novel insights into the molecular regulatory networks underlying plant stress responses to endocrine-disrupting chemicals.

UPLC-Q-TOF-MS/MS-based urine metabolomics studies on the toxicity and detoxication of Tripterygium wilfordii Hook. f. after roasting

Shu Chen,Li Liu,Hong-Xia Jiang,Qun Sun,Liang Zhang,Jian-Qun Liu,Li-Fang Liu

Tripterygium wilfordii (TW), a well-known traditional Chinese medicine , was widely used in the treatment of autoimmune disorders and inflammatory diseases . However, the clinical use of TW was limited by severe toxicities, such as hepatotoxicity and nephrotoxicity . Our previous studies indicated that roasting was an effective approach for reducing TW-induced toxicity. After roasting, celastrol was completely decomposed, partially converted into 1-hydroxy-2,5,8-trimethyl-9-fluorenone and the total alkaloids content were significantly reduced. However, the detoxication mechanisms of roasting on TW were poorly unknown. This study aimed to explore the toxicity and detoxification mechanisms of TW after roasting based on urine metabolomics. Promising biomarkers were evaluated by multiple comparison analyses. Sixteen toxicity biomarkers were identified between control group and total extract group. Twelve toxicity biomarkers were identified between control group and total alkaloids group. Eight toxicity biomarkers were identified between control group and celastrol group. These metabolites were mainly involved in seven metabolic pathways, summarized as pentose and glucuronate interconversions, lipid metabolism (sphingolipid metabolism, glycerophospholipid metabolisms, fatty acid biosynthesis and steroid hormone biosynthesis) and amino acid metabolism (taurine and hypotaurine metabolism, tryptophan metabolism). After roasting, the toxicities of total extract, total alkaloids and celastrol were relieved by ameliorative serum parameters and pathological changes in hepatic and renal tissues which revealed that the reduction of celastrol and total alkaloids played important roles in the detoxification of roasting on TW. Furthermore, roasting regulated the levels of fourteen potential biomarkers in the total extract group, ten potential biomarkers in the total alkaloids group and seven candidate biomarkers in the celastrol group to normal levels. Biological pathway analysis revealed that roasting may ameliorate TW-induced metabolic disorders in pentose and glucuronate interconversions, lipid metabolism and amino acid metabolism. This study provided evidence for the application of roasting in TW.

Systematic impacts of fluoride exposure on the metabolomics of rats

Shiyuan Zhao,Jinxiu Guo,Hongjia Xue,Junjun Meng,Dadi Xie,Xi Liu,Qingqing Yu,Haitao Zhong,Pei Jiang

Fluoride is widely present in the environment. Excessive fluoride exposure leads to fluorosis, which has become a global public health problem and will cause damage to various organs and tissues. Only a few studies focus on serum metabolomics, and there is still a lack of systematic metabolomics associated with fluorosis within the main organs. Therefore, in the current study, a non-targeted metabolomics method using gas chromatography-mass spectrometry (GC-MS) was used to research the effects of fluoride exposure on metabolites in different organs, to uncover potential biomarkers and study whether the affected metabolic pathways are related to the mechanism of fluorosis. Male Sprague-Dawley rats were randomly divided into two groups: a control group and a fluoride exposure group. GC-MS technology was used to identify metabolites. Multivariate statistical analysis identified 16, 24, 20, 20, 24, 13, 7, and 13 differential metabolites in the serum, liver, kidney, heart, hippocampus, cortex, kidney fat, and brown fat, respectively, in the two groups of rats. Fifteen metabolic pathways were affected, involving toxic mechanisms such as oxidative stress, mitochondrial damage, inflammation, and fatty acid, amino acid and energy metabolism disorders. This study provides a new perspective on the understanding of the mechanism of toxicity associated with sodium fluoride, contributing to the prevention and treatment of fluorosis.

Metabolic profiling of rats poisoned with paraquat and treated with Xuebijing using a UPLC-QTOF-MS/MS metabolomics approach

Wen Liu,Sha Li,Yang Ke Wu,Xiao Yan,Yi Ming Zhu,Fei Ya Jiang,Yu Jiang,Liang Hong Zou,Tong Tong Wang

Xuebijing (XBJ) is a compound Chinese medicine that contains Paeoniae Radix Rubra, ChuanXiong Rhizoma, Salvia Miltiorrhiza Radix et Rhizoma, Carthami Flos, and Angelicae Sinensis Radix. It is widely used in China to treat sepsis. Previous studies have demonstrated that XBJ can decrease mortality in patients with moderate paraquat poisoning. However, the mechanism by which it exerts this effect is not completely clear. In this study, an ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry (UPLC-QTOF-MS/MS)-based metabolomics approach was used to perform a metabolic profiling analysis. Principal component analysis (PCA), random forest (RF), and partial least squares discriminant analysis (PLS-DA) were used to identify metabolites to clarify the mechanism of XBJ's activity. XBJ clearly alleviated lung injury in a Sprague Dawley (SD) rat model of paraquat (PQ) poisoning. Seven metabolites related to four pathways, including those involved in sphingolipid and phospholipid metabolism, amino acid metabolism, unsaturated fatty acid metabolism, and pantothenic acid and CoA biosynthesis, were present at different levels in PQ-poisoned rats treated with XBJ compared with untreated rats. XBJ can ameliorate the effects of PQ poisoning in SD rats. Using a metabolomics approach enabled us to gain new insight into the mechanism underlying this effect.

Liquid Chromatograph-Mass Spectrometry-Based Non-targeted Metabolomics Discovery of Potential Endogenous Biomarkers Associated With Prostatitis Rats to Reveal the Effects of Magnoflorine

Yuan Yin,Dong Fei-Xue,Liu Xu,Xiao Hong-Bin,Zhou Zhong-Guang

Magnoflorine (Mag) has multiple pharmacological activities for the prevention and treatment of prostatitis. However, its molecular mechanisms andpharmacological targets are not clear. In this study, the ultra-performance liquid tandem mass spectrometry-based metabolomics method was used to clarify the intervention of Mag against prostatitis and the biological mechanism. A total of 25 biomarkers associated with the prostatitis model were identified by metabolomics, and a number of metabolic pathways closely related to the model were obtained by MetPA analysis. After given Mag treatment, the results of each indicator were shown that Mag alkaloid could inhibit the development of prostatitis effectively. We found that Mag had regulative effects on potential biomarkers of prostatitis model, which can regulate them to the control group. Our results indicated that alkaloids have an effective intervention therapy for prostatitis, and five types of metabolic pathways closely related to prostatitis model were obtained, including phenylalanine, tyrosine and tryptophan biosynthesis, phenylalanine metabolism, tyrosine metabolism, arginine and proline metabolism, glycine, serine and threonine metabolism, alanine, aspartate and glutamate metabolism. This study has provided the basic experimental data for the development of Mag in the prevention and treatment of prostatitis.

Toxicity Effects and Mechanisms of Graphdiyne towards Freshwater Microalgae Scenedesmus obliquus

Wei Hong,Fan Zhang,Zhuang Wang

Graphdiyne (GDY), an emerging 2D carbon nanomaterial, holds promise yet remains unexplored for environmental hazards. Herein, we investigated the toxicity effects and mechanisms of GDY towards the freshwater microalga Scenedesmus obliquus at environmentally relevant concentrations. The results revealed that 1 mg/L of GDY inhibited the growth of the algae and significantly induced a decrease in photosynthetic pigments. Exposure to 0.1 and 1 mg/L of GDY led to a reduction in cell membrane permeability and induced intracellular oxidative stress in the algae. Further, 1 mg/L of GDY caused oxidative damage to the algal cells. Molecular modeling indicated that GDY could directly affect the stability of dsDNA fragment. Transcriptome analysis showed that GDY at 1 mg/L influenced the expression of 2216 genes, with the glycerolipid metabolism pathway being enriched significantly. Metabolome analysis identified 129 differentially expressed metabolites (DEMs) in the algae exposed to 1 mg/L of GDY, revealing three significantly disrupted pathways: glycerophospholipid metabolism, pentose phosphate pathway, and flavonoid biosynthesis. The integrated transcriptome and metabolome analysis suggested that GDY exposure downregulated the level of a lysophosphatidylcholine species (LPC 18:3) by inhibiting the expression of genes related to FMN binding molecular functions. Simultaneously, the algae responded to GDY exposure by upregulating genes involved in the biological process of carbohydrate metabolic process and the molecular function of hydrolase activity, as well as increasing the levels of DEMs, specifically a lysophosphatidic acid species (LPA 16:4). This study elucidates the stress mechanisms induced by GDY in aquatic organisms and emphasizes the importance of monitoring its acute biological effects.

Temporal metabolomics analysis reveals the metabolic patterns in goat cumulus cells during oocyte maturation

Zhiyuan Shen,Yixin Ma,Ming Gao,Ling Gu

Cumulus cells play a crucial role in the oocyte growth and maturation processes through providing necessary nutrients and growth signals by gap junction communication. However, a global overview of metabolic events in goat cumulus cells is still lacking. In the present study, we collected cumulus cells from goat cumulus-oocyte complexes (COCs) at different developmental stages. Metabolomics analysis was performed to investigate the global metabolic patterns in cumulus cells during oocyte in vitro maturation. In particular, we revealed the several significantly altered metabolic pathways and metabolic   characteristics in goat cumulus cells, including the accumulation of fatty acids, steroid hormones metabolism, active catabolism of arginine during meiotic resumption, and a progressive decline in nucleotide metabolism. In conclusion, the dataset generated by our metabolomic profiling will provide valuable information to understand the key metabolic pathways and metabolites involved in COCs development.

Investigation of Dioscorea bulbifera Rhizome-Induced Hepatotoxicity in Rats by a Multisample Integrated Metabolomics Approach

Dong-Sheng Zhao,Li-Long Jiang,Ya-Xi Fan,Ling-Li Wang,Zhuo-Qing Li,Wei Shi,Ping Li,Hui-Jun Li

The use of herbal medicines continues to expand globally, meanwhile, herb-associated hepatotoxicity is becoming a safety issue. As a conventional Chinese medicinal herb, Dioscorea bulbifera rhizome (DBR) has been documented to cause hepatic toxicity. However, the exact underlying mechanism remains largely unexplored. In the present study, we aimed to profile entire endogenous metabolites in a biological system using a multisample integrated metabolomics strategy. Our findings offered additional insights into the molecular mechanism of the DBR-induced hepatotoxicity. We identified different metabolites from rat plasma, urine, and feces by employing gas chromatography-mass spectrometry in combination with multivariate analysis. In total, 55 metabolites distributed in 33 metabolic pathways were identified as being significantly altered in DBR-treated rats. Correlation network analysis revealed that the hub metabolites of hepatotoxicity were mainly associated with amino acid, bile acid, purine, pyrimidine, lipid, and energy metabolism. As such, DBR affected the physiological and biological functions of liver via the regulation of multiple metabolic pathways to an abnormal state. Notably, our findings also demonstrated that the multisample integrated metabolomics strategy has a great potential to identify more biomarkers and pathways in order to elucidate the mechanistic complexity of toxicity of traditional Chinese medicine.

Untargeted metabolomics of the intestinal tract of DEV-infected ducks

Cai Haiqing,Yang Xia,Yang Yunyun,Feng Yi,Wen Anlin,Yang Ying,Wen Ming,Ou Deyuan

IntroductionDuck enteritis virus (DEV) mainly causes infectious diseases characterized by intestinal haemorrhage, inflammation and parenchymal organ degeneration in ducks and other poultry. However, the mechanism by which it causes intestinal damage in ducks is not well understood. Metabolomics can provide an in-depth understanding of the full complexity of the disease. Methods In this study, 24 clinically healthy green-shell ducks (weight 1.5 kg ± 20 g) were randomly divided into 2 groups (experimental group, 18; control group, 6). The experimental group was intramuscularly injected with 0.2 mL of DEV virus in solution (TCID50 3.16 × 108 PFU/mL), and the control group was injected with 0.2 mL of sterile normal saline. Duck duodenum and ileum tissue samples were collected at 66 h, 90 h and 114 h post-injection (12 h of fasting before killing), and metabolomics analysis of duck duodenum and ileum tissues at the three time points (66, 90, 114 h) was performed by liquid chromatography–mass spectrometry (LC–MS) to screen for and analyse the potential differentiated metabolites and related signalling pathways. Results Screening was performed in the positive/negative mode (Pos: Positive ion mode; the ionization of substances at the ion source with positive ions such as H+, NH4+, Na+ and K+; Neg: Negative ion mode; the ionization of substances at the ion source with negative ions such as Cl−, OAc−), and compound abundance was compared to that in the control group. The total number of differentially abundant compounds in the duodenum at 66 h, 90 h and 114 h of DEV infection gradually increased, and metabolites such as cytidine, 2′-deoxyriboside and 4-guanidinobutyric acid were differentially abundant metabolites common to all three time periods. The metabolic pathways related to inflammatory response and immune response were tryptophan acid metabolism, cysteine-methionine metabolism, histidine metabolism and other amino acid metabolism and fat metabolism. Among them, the metabolic pathways with more differentially abundant metabolites were amino acid biosynthesis, cysteine and methionine metabolism, tryptophan metabolism, unsaturated fatty acid biosynthesis and purine metabolism, and the metabolic pathways with more enrichment factors were the IgA-related intestinal immune network pathway and lysosome pathway. Compared with the control group, there were 16 differentially abundant metabolites in the ileum tissue of DEV-infected ducks at 66 h of infection, 52 at 90 h of infection, and 40 at 14 h of infection with TD114. The metabolic pathways with more enriched differentially abundant metabolites were pyrimidine metabolism, tyrosine metabolism, phenylalanine metabolism and tryptophan biosynthesis. The metabolic pathways with the most enrichment factors were the mTOR signalling pathway, ferroptosis pathway, tryptophan metabolism pathway and caffeine metabolism pathway. Conclusion Comparative analysis showed that the number of differentially abundant metabolites in the duodenum and ileum differed to some extent after DEV infection, with significantly more differentially abundant metabolites in duodenal tissues and fewer in ileal tissues; after DEV infection, the highest number of differentially abundant metabolites was obtained at 114 h of DEV infection, followed by the second highest at 90 h of infection and the lowest at 66 h of infection. The common differentially abundant metabolites in duodenal and ileal tissues were prostaglandins, arachidonic acid, and arachidonic ethanolamine. The main metabolic pathways in the duodenum were the IgA-associated intestinal immune network pathway and the lysosomal pathway, and the metabolic pathways with more enriched factors in the ileum were the mTOR signalling pathway, the ferroptosis pathway, and the tryptophan metabolism pathway.

Effects of steam processing on the flavor formation mechanism of male and female mussels

Nana Zhao,Xinxing Xu,Shiyuan Dong,Yuanhui Zhao,Tianhong Liu,Li Liu,Yue Zhao,Mingyong Zeng,Kang Liu

In this study, the flavor quality of male and female mussel meat was investigated by sensory evaluation, gas chromatography-ion mobility spectrometry (GC‒IMS), gas chromatography‒mass spectrometry (GC‒MS) and metabolomics. The sensory evaluation results indicated that the flavor quality of male and female mussel meat was optimal when it was steamed for 8 min and that “fresh” was the most prominent odour attribute. The GC‒IMS and GC‒MS results revealed that aldehydes, ketones and alcohols were the characteristic flavor compounds in male and female mussel meat during different steaming times. Glycerol phospholipid metabolism was the main metabolic enrichment pathway shared by male and female mussel meat, and glycine (Gly), serine (Ser), and threonine (Thr) metabolism were the main metabolic pathways of female mussel meat. Phosphatidylethanolamine, phosphatidylcholine, phosphatidyl-L-serine, phosphatidylglycerol, and 1-acyl-n-glycerin-3-phosphocholine are the precursors of key odour compounds in male and female mussel meat at different steaming times.

Acute alkalinity stress induces functional damage and alters immune metabolic pathways in the gill tissue of spotted scat (Scatophagus argus)

Lei Yang,Adili Abudu,Kecheng Zhu,Tong Han,Cunyu Duan,Yu Chen,Xiaolong Li,Gang Shi,Chunhua Zhu,Guangli Li,Changxu Tian

The spotted scat ( Scatophagus argus ), known for its robust osmoregulatory abilities as a euryhaline fish, represents a promising candidate for large-scale aquaculture in saline-alkaline waters. In this study, spotted scat were exposed to control conditions and a treatment group with alkalinity at 30 mmol/L for varying durations (0 h, 12 h, 24 h, 48 h, 72 h, and 96 h). Histopathological analysis, biochemical assays, and transcriptomic evaluation were employed to examine the impact of alkalinity stress on gill tissue. The results indicated that gill tissues exhibited varying degrees of damage when compared to the control group across different time points. Prolonged alkalinity stress resulted in an increased number of chlorine-secreting cells and caused the gill lamellae to curve, display inconsistent orientation, and swell at the base. Biochemical analyses revealed that acute alkalinity stress disrupted oxidative balance, leading to decreased activities of superoxide dismutase (SOD) and catalase (CAT), with a notable minimum observed at 96 h. In contrast, the activities of glutathione peroxidase (GSH-Px), acid phosphatase (ACP), alkaline phosphatase (ALP), and malondialdehyde (MDA) concentration gradually increased with extended alkaline exposure. Transcriptomic analysis of gill tissues from the control and 24 h acute alkalinity stress groups revealed significant changes in gene expression, identifying 513 differentially expressed genes (DEGs), with 292 up-regulated and 221 down-regulated. These DEGs were significantly enriched in pathways related to drug metabolism-cytochrome P450 and drug metabolism-other enzymes. Additionally, pathways involved in arachidonic acid metabolism, as well as arginine and proline metabolism, were found to play critical roles in adapting to alkalinity stress. This study contributes to a valuable insight into the molecular mechanisms underlying alkali resistance in spotted scat.

Metabolomics approach in lung tissue of septic rats and the interventional effects of Xuebijing injection using UHPLC-Q-Orbitrap-HRMS

Xu Tanye,Zhou Lin,Shi Yingying,Liu Liwei,Zuo Lihua,Jia Qingquan,Du Shuzhang,Kang Jian,Zhang Xiaojian,Sun Zhi

Sepsis is the dysregulated host response to an infection which leads to life-threatening organ dysfunction. Metabolomic profiling in bio-fluid or tissue is vital for elucidating the pathogenesis of sepsis and evaluating therapeutic effects of medication. In this study, an untargeted metabolomics approach was applied to study the metabolic changes in lung tissue of septic rats induced by cecal ligation and puncture (CLP) and investigate the treatment effects of Xubijing injection (XBJ). Metabolomics analyses were performed on ultra-high performance liquid chromatography-Q Exactive hybrid quadrupole-orbitrap high-resolution accurate mass spectrometry (UHPLC-Q-Orbitrap-HRMS) together with multivariate statistical analysis. A total of 26 differential metabolites between CLP and sham-operated group were identified. The altered metabolic pathways included energy metabolism, amino metabolism, lipid metabolism, fatty acid metabolism and hormone metabolism. Among the 26-varied metabolites, 15 were significantly regulated after XBJ treatment. The metabolic pathway network of sepsis was drawn to interpret the pathological feature of lung damage caused by sepsis and the underlying regulating mechanism of XBJ on the molecular levels. Our findings display that LC-MS-based metabolomics is a useful tool for uncovering the underlying molecular mechanism of sepsis, and XBJ may exert therapeutic effect by regulating multiple metabolic pathways.
