Smartphone-controlled biosensor for viral respiratory infectious diseases: Screening and response

Yaxing Ma,Yaoyu Luo,Xinrui Feng,Chuixiu Huang,Xiantao Shen

Outbreaks of emerging viral respiratory infectious diseases (VRIDs) including coronavirus disease 2019 (COVID-19) seriously endanger people's health. However, the traditional nucleic acid detection required professionals and larger instruments and antigen-antibody detection suffered a long window period of target generation. To facilitate the VRIDs detection in time for common populations, a smartphone-controlled biosensor, which integrated sample preparation (electromembrane extraction), biomarker detection (red-green-blue model) and remote response technology (a built-in APP), was developed in this work. With the intelligent biosensor, VRIDs could be recognized in the early stage by using endogenous hydrogen sulfide as the biomarker. Importantly, it only took 15 min to accomplish the whole process of screening and response to VRIDs. Moreover, the experimental data showed that this smartphone-controlled biosensor was suitable for ordinary residents and could successfully differentiate non-communicable respiratory diseases from VRIDs. To the best of our knowledge, this is the first time that a smartphone-controlled biosensor for screening and response to VRIDs was reported. We believe that the present biosensor will help ordinary residents jointly deal with the challenges brought by COVID-19 or other VRIDs in the future.

Inverse opal microneedles arrays for fluorescence enhanced screening skin interstitial fluid biomarkers

Kexin Yi,Yunru Yu,Yuetong Wang,Yuanjin Zhao

Infectious diseases caused by bacteria have become serious health concerns. Efficient and sensitive early detections are of pivotal significance in the diagnosis and treatment of bacterial infection. Here, we develop novel inverse opal microneedles (MNs) arrays for in situ extraction and fluorescence-enhanced detection of target biomarkers in skin interstitial fluid (ISF). As the MNs array is fabricated by applying a resin to replicate the colloidal crystal-filled negative mold , it possesses the exceptional arranged porous structure and unique photonic band gap (PBG) properties. Benefitting from these features, the inverse opal MNs array enables non-invasive skin ISF sampling and provides enhanced fluorescence signal intensity towards specific bio-probes, facilitating improved detection sensitivity. On the basis of the inverse opal MNs array, we have further verified its value of in situ extraction and fluorescence-enhanced detection of lipopolysaccharide (LPS) with high efficiency and sensitivity in skin ISF from infected rats. These results indicate the great potential value of the inverse opal MNs array in non-invasive clinical diagnosis and other related biomedical applications .

Photonic Hydrogels for Synergistic Visual Bacterial Detection and On-Site Photothermal Disinfection

Ge Xie,Shuo Du,Qiuyi Huang,Min Mo,Yujie Gao,Miaomiao Li,Juan Tao,Lianbin Zhang,Jintao Zhu

Rapid and sensitive diagnostics in the early stage of bacterial infection and immediate treatment play critical roles in the control of infectious diseases. However, it remains challenging to develop integrated systems with both rapid detection of bacterial infection and timely on-demand disinfection ability. Herein, we demonstrate a photonic hydrogel platform integrating visual diagnosis and on-site photothermal disinfection by incorporating Fe3O4@C nanoparticles into a poly(hydroxyethyl methacrylate)-co-polyacrylamide (PHEMA-co-PAAm) matrix. In vitro experiments demonstrate that such a hydrogel can respond to pH variation caused by bacterial metabolism and generate the corresponding color changes to realize naked-eye observation. Meanwhile, its excellent photothermal conversion ability enables it to effectively kill bacteria by destroying cell membranes under near-infrared irradiation. Moreover, the pigskin infection wound model also verifies the bacterial detection performance and disinfection ability of the hydrogel in vivo. Our strategy demonstrates a new approach for visual diagnosis and treatment of bacterial infections.

Boosting humoral and cellular immunity with enhanced STING activation by hierarchical mesoporous metal-organic framework adjuvants

Chengjie Huang,Xiao Zhi,Tianbao Ye,Xiuyuan Wang,Ke Li,Yiyang Li,Qiang Zhang,Lai Jiang,Xianting Ding

Vaccination is essential for preventing and controlling infectious diseases, along with reducing mortality. Developing safe and versatile adjuvants to enhance humoral and cellular immune responses to vaccines remains a key challenge in vaccine development. Here, we designed hierarchical mesoporous MOF-801 (HM801) using a Cocamidopropyl betaine (CAPB) and a Pluronics F127 in an aqueous phase system. Meanwhile, we synthesized a novel SARS-CoV-2 nanovaccine (R@M@HM801) with a high loading capacity for both the STING agonist (MSA-2) and the Delta receptor binding domain (Delta-RBD) antigen. R@M@HM801 enhanced MSA-2 and RBD utilization and effectively co-delivered MSA-2 and RBD antigens to antigen-presenting cells in the draining lymph nodes, thereby promoting the activation of both T and B cells. Lymphocyte single-cell analysis showed that R@M@HM801 stimulated robust CD11b + CD4 + T cells, CXCR5 + CD4 + T follicular helper (T fh ), and durable CD4 + CD44 + CD62L − , CD8 + CD44 + CD62L − effector memory T cell (T EM ) immune responses, and promoted the proliferative activation of CD26 + B cells in vivo. Meanwhile, R@M@HM801 induced stronger specific antibodies and neutralization of pseudovirus against Delta compared to the RBD + MAS-2 and RBD + MAS-2 + Alum vaccines. Our study demonstrated the efficacy of a hierarchical mesoporous HM801 and its potential immune activation mechanism in enhancing adaptive immune responses against viruses and other diseases.

Multi-walled carbon nanotubes decorated with silver nanoparticles for antimicrobial applications

H.I. Hamouda,H.M. Abdel-Ghafar,M.H.H. Mahmoud

Great attention has been paid to antimicrobial agents' development to face and control infection transmission of infectious diseases spreading. Decorated multi-walled carbon nanotubes (MWCNTs) with silver nanoparticles (Ag NPs) are useful substances with antibacterial potency that might be used for a wide range of medical applications and water disinfection to limit the spreading of infectious diseases. In this work, silver nanoparticles were loaded on the surface of MWCNTs via the wet-impregnation technique followed by thermal treatment at 450 °C for 4 h in an inert atmosphere . Different ratios of Ag NPs (0, 3, 6, 12 and 24) were loaded on the surface of MWCNTs. The synthesized Ag-MWCNTs nanocomposites were characterized by XRD, XPS, and TEM . The antibacterial efficacy of Ag-MWCNTs nanocomposites was tested towards different microbial strains (bacteria and fungus); P. aeruginosa , E. coli , B. subtilis , S. aureus , C. albicans , and A. niger, which proved the wide spectrum of Ag-MWCNTs. The synthesized nanocomposite showed a higher antimicrobial capability against gram-positive than gram-negative bacteria. Biological studies indicated a large inhibition zone for each bacterial strain that reaches 18.5 mm. SEM and fluorescent microscopy were tested to examine the structural disruption in the outer membranes after incubation with the antibacterial agent. The nanocomposite firmly agglutinated to the surface of the bacterial cells, leading to cells' wrapping, morphology deterioration, and cell death. The inhibition ability of Ag-MWCNTs nanocomposite with a broad spectrum of microorganisms provides exciting opportunities to enhance and maximize potable water disinfection and medical applications to control infectious diseases spreading.

Gold nanorod-mediated multicolor biosensor for visual detection of Streptococcus agalactiae during pregnancy in actual clinical samples

Xiaozhuang Zhang,Yubin Zhang,Fukang Li,Huaiyu Jia,Min Wang,Guozhen Tian

Background Streptococcus agalactiae ( S. agalactiae ) has attracted much attention due to its potential threat to the health of mothers and infants. Especially during the perinatal period, its infection may lead to a series of serious complications, thus conducting tests for S. agalactiae serves as a highly effective preventive measure during pregnancy. Currently, available S. agalactiae detection methods used in clinical trials suffer from several notable limitations. These include prolonged testing durations, the necessity for sophisticated and costly equipment, and an inability to provide real-time visualization of test results. Results In this study, we successfully devised multicolor visualization assay using gold nanorods (GNRs) as mediators for a simple, rapid, sensitive, and reliable of S. agalactiae detection. This method innovatively utilizes recombinase polymerase amplification (RPA) to efficiently amplify cfb gene, while employing streptavidin-modified magnetic beads (MBs-SA) for rapid separation of amplification products. This approach substantially shortens reaction time, allowing entire detection process to be completed within 30 min. The localized surface plasmon resonance (LSPR) of GNRs enable multicolor changes in results and naked-eye identification, eliminating the reliance on expensive detection instruments, while achieving an impressive sensitivity of 1 copy/μL. Significance The proposed method demonstrates satisfactory accuracy and specificity, requires no complex instrumentation, and enables visual detection of results. It holds significant potential and value for application in resource-limited settings such as home care facilities, and primary hospitals for preventing S. agalactiae infection.

Novel antibacterial peptide HfAMP against methicillin-resistant Staphylococcus aureus

Lin Fei,Jin Weiwei,Yang Longbing,Tian Chunren,Zhang Jin,Jiao Zhenglong,Peng Jian,Guo Guo

Objectives The urgent development of novel antibacterial agents to combat antibiotic-resistant Staphylococcus aureus is imperative. Antimicrobial peptides are considered promising alternatives to traditional antibiotics for addressing this pressing issue. This study investigates the antibacterial activities and potential mechanisms of the peptide HfAMP against methicillin-resistant S. aureus (MRSA). Methods The antibacterial activity of HfAMP against MRSA was evaluated using the broth microdilution method, growth curve, and time-kill assay. Its potential mechanism of action was investigated through propidium iodide, laurdan, DiSC 3 (5), 2’,7’-dichlorodihydrofluorescein diacetate, and ATP quantification assays. To assess the safety and stability of HfAMP, cytotoxicity, hemolysis, and antibacterial activity were examined under different conditions (i.e., temperatures, ions, serum, pH, and enzymes). Furthermore, a mouse skin infection model was employed to evaluate the in vivo efficacy of HfAMP. Results The results demonstrate that HfAMP exhibits bacteriostatic and bactericidal activity against MRSA, with a minimum inhibitory concentration of 4 µg/mL. It remains stable under a range of pH, temperatures, and serum, although it is susceptible to degradation by pepsin. Mechanistic investigations show that HfAMP compromises bacterial membranes by interacting with membrane components and disrupting the proton motive force, leading to metabolic disturbances. In a mouse model of MRSA-induced skin infection, HfAMP significantly reduced bacterial burden and inflammation in the affected skin tissues. Conclusion Collectively, these findings indicate that HfAMP possesses potent antibacterial activity against MRSA by compromising bacterial viability and inhibiting virulence, highlighting its potential as a promising candidate for the treatment of MRSA skin-infection.

Rapid diagnosis of tuberculosis using a three-dimensional nanofiber paper-based electrochemical sensor

Manman Du,Junting Zhou,Zhiwei Zhang,Tianteng Hao,Junfeng Han,Jin Wei,Xinwu Xie,Can Wang

Tuberculosis (TB) is the leading cause of death caused by a single infectious pathogen. Currently, a variety of conventional methods are widely used for detecting Mycobacterium tuberculosis ( Mtb ) or diagnosing TB, which have made significant contributions to controlling the TB epidemic. However, these existing TB diagnostic methods still face some difficulties and cannot provide rapid, low-cost, high-sensitivity, and high-accuracy diagnostic results simultaneously. Therefore, it is imperative to develop new TB diagnostic methods that are rapid, cost-effective, and sensitive through rigorous evaluation of potential biomarkers and new detection methods. In this study, a three-dimensional nanofiber (3D NFs) paper-based electrochemical sensor was prepared using electrospinning technology, and Mtb secretory protein Ag85B was utilized as a biomarker to achieve point-of-care detection of TB clinical samples. The 3D NFs paper-based electrochemical sensor can achieve a wide range of Bacillus Calmette–Guérin detection of 10 1 -10 8 CFU/mL with good specificity. When the clinical bronchoalveolar lavage fluid samples were diluted 10 3 or 10 4 times, the 3D NFs paper-based electrochemical sensor could detect TB clinical samples with a sensitivity of up to 92.9 % and a specificity of 60 % within 5 min. 3D NFs paper-based electrochemical sensor has the advantages of low cost, high sensitivity, and fast response speed, and can realize rapid preliminary screening of TB patients.

An activatable type I photosensitizer for NIR fluorescence imaging and photodynamic therapy of drug-resistant bacterial infections

Zhongkun Wang,Rui Zhu,Chenrui Li,Weiwei Xu,Liangyou Zhao,Guangjin Gao,Weihao An,Xiang Dong,Qingqing Miao,Qing Li

Bacterial infections pose a significant global health challenge, further exacerbated by the emergence of antibiotic-resistant strains, thereby necessitating the development of alternative therapeutic strategies. Type I photosensitizers, as an emerging phototheranostic modality, hold great promise for antibacterial treatment but are limited by poor specificity. Herein, we report a hydrogen sulfide (H 2 S)-activatable theranostic probe (CyIN) that integrates near-infrared (NIR) fluorescence imaging and type I photodynamic therapy (PDT) functionalities into a single platform for the precise treatment of drug-resistant bacterial infections. CyIN is rationally constructed by masking the hydroxyl group of iodine-substituted hemicyanine with a H 2 S-cleavable 2,4-dinitrophenyl group, which efficiently quenches NIR fluorescence and suppresses reactive oxygen species generation. Such molecular configuration allows CyIN to turn on its NIR fluorescence signal and type I photodynamic action upon interaction with bacterial-derived H 2 S, achieving distinguishable detection of drug-resistant bacterial infection, potent bacterial clearance (>99 %), and accelerated healing of bacteria-infected wounds. Furthermore, histological analyses confirm that the CyIN treatment can reduce inflammation, enhance collagen deposition, and promote tissue regeneration without systemic toxicity. Thus, this work presents a promising H 2 S-responsive phototheranostic platform for precise bacterial detection and efficient photodynamic eradication of drug-resistant bacterial infections.

SU3327: A Multi-Target Compound Targeting Bacterial Menaquinone and DNA

Shuge Wang,Ke Zhao,Ziqi Chen,Chengtao Sun,Hongliang Chen,Yang Wang,Congming Wu,Dejun Liu

Background and Purpose The rapid emergence of antibiotic resistance (AMR) threatens global health by rendering existing antibiotics ineffective, demanding novel antimicrobial agents with unique mechanisms of action. SU3327 (also known as Halicin), identified through deep learning-based drug screening, has demonstrated broad-spectrum antibacterial activity and high safety in vivo. However, its precise mechanism of action remains unclear. This study aimed to elucidate the antibacterial mechanism of SU3327 and to explore its potential as a multi-target therapeutic. Experimental Approach We employed microbiological, biochemical/biophysical, mass spectrometry, electrochemical, and transcriptomic analyses to study SU3327′s effects. We examined its impact on proton motive force (PMF), intracellular ATP synthesis, and electron transport chain (ETC) function, including Complex I inhibition and menaquinone interaction. We also evaluated nitroreductase bioactivation, identified the hydroxylamine metabolite, and quantified resulting oxidative DNA damage. Key Results SU3327 disrupts bacterial energy metabolism by targeting menaquinone (MK) in the ETC. It inhibits Complex I, impeding electron transfer, disrupting the PMF, and impairing ATP synthesis. Additionally, SU3327 enters Escherichia coli ATCC 25922, undergoing nitroreductase bioactivation, forming hydroxylamine compounds (R-NHOH) that react with DNA, generating hydroxyl radicals (•OH) and inducing oxidative DNA damage. These results indicate that SU3327 operates via a dual-target mechanism: disrupting bacterial respiration and causing DNA damage. Conclusions and Implications SU3327 is a first-in-class multi-target antibacterial agent acting via two synergistic mechanisms. Its dual action enhances efficacy and may reduce resistance likelihood by requiring mutations in distinct pathways. This study advances understanding of SU3327 and supports its development as a novel antibiotic for veterinary medicine.

Honokiol-loaded nanoparticles for targeted bacterial eradication and treatment of MRSA infection

Shouli Yi,Hongjuan Zhang,Zhen Yang,Di Wu,Dan Shao,Jiongjie He,Yu Liu,Baocheng Hao,Shengyi Wang

Methicillin-resistant Staphylococcus aureus (MRSA) infections and the formation of associated biofilms often lead to refractory and recurrent bacterial infections, posing a serious threat to public health. Herein, a pH-responsive antibiotic-free nanoparticle (HNK@RHL-COS NPs) was prepared by encapsulating the natural antimicrobial agent honokiol (HNK) with rhamnolipid-modified chitosan, enabling targeted disassembling at the acidic infection site, destruction of the biofilm by rhamnolipid, thereby enhancing the penetration of honokiol and chitosan, thus achieving their synergistic bactericidal effect. HNK@RHL-COS NPs exhibited enhanced antimicrobial effects against MRSA, with a minimum inhibitory concentration (MIC) value of 2 μg/mL, significantly inhibited the formation of biofilm and eradicated the mature biofilm. Furthermore, HNK@RHL-COS NPs efficiently accelerated the healing of wounds in MRSA-infected mice, alleviated bacterial peritonitis in mice, and demonstrated excellent biocompatibility in vitro and in vivo . Collectively, this study provides a novel and safety solution for the clinical treatment of MRSA infections.

Rapid and sensitive detection of Candida albicans using nystatin-functionalized magnetic spheres

Zhen-Yu Zhao,Ya-Ning Che,Qian Huang,Han-Wen Yang,Xian-Hua Wang,Lin-Yi Dong

Candida albicans is the leading cause of invasive fungal disease; candidemia carries 30–40 % mortality. It commonly colonizes human mucosa as a commensal, but immunosuppression or disrupted microbiota enables opportunistic infection. Therefore, rapid, reliable diagnostics are vital to improve public health. In this study, a magnetic solid-phase microextraction (MSPE) technique based on nystatin-functionalized magnetic nanoparticles (Fe₃O₄@NYS) was developed to achieve the dual objectives of efficiently enriching C. albicans rapidly and extracting its deoxyribonucleic acid (DNA). Optimization of capture conditions enabled rapid enrichment of C. albicans with a maximum recovery rate of approximately 90 %. Subsequent to the capture of C. albicans, DNA extraction was performed on the Fe₃O₄@NYS- C. albicans complexes. Notably, Fe₃O₄@NYS facilitated the extraction of a substantial quantity of DNA, with the extraction yield reaching 1200 ng/mg. Therefore, the integration of DNA extraction with quantitative polymerase chain reaction (qPCR) technology enables sensitive and specific detection of C. albicans in real samples. The strategy demonstrated excellent linearity within the concentration range of 4.20 × 10 2 to 4.20 × 10 7  CFU mL −1 for C. albicans . The high sensitivity and accuracy of this method were confirmed in honey samples, showcasing a limit of detection (LOD) 420 CFU mL −1 , recoveries between 98.1 % and 99.8 %, and relative standard deviations (RSD) not exceeding 2.51 %. This method presents an innovative strategy for point-of-care clinical diagnosis, disease surveillance, and biosafety management, demonstrating substantial potential for the efficient isolation of DNA from complex biological matrices.
