Chitosan-Derived Carbon Quantum Dots with Dual ROS Scavenging and Anti-inflammatory Functionalities for Accelerated Wound Repair

Miaomiao Yao,Qi He,Yameng Tao,Xiaoyang Kang,Xinru Wu,Fangkai Shi,Yujia Wei,Jingjing Liu,Zhiyun Meng,Ruolan Gu,Hui Gan,Guifang Dou,Shuchen Liu,Yunbo Sun

Blue fluorescent carbon quantum dots (CDs) were synthesized by a one-step hydrothermal method using chitosan as the only raw material. This material overcomes multifunctional integration limitations in traditional dressings, achieving coordinated wound repair across all phases. During inflammation (0–3 days), CDs scavenge reactive oxygen species and suppress inflammatory factors (TNF-α↓49.63%, IL-6↓70.8%). In proliferation (3–7 days), they promote angiogenesis (CD31↑56.89%) and cell proliferation (Ki67↑74.3%). During remodeling (7–14 days), functional parallel collagen deposition occurs (collagen deposition 1.26× Celox group). The seamless “anti-inflammation/prohealing/remodeling” mechanism yielded 59% wound closure by day 3 postirradiation (vs Celox 37.34%). CDs synergistically activate Nrf2 while inhibiting NF-κB pathways, enabling combined anti-inflammatory/antioxidant regulation in irradiated environments. Nanoscale reconstruction retains chitosan’s hemostasis and avoids gene modification/vector loading via one-step synthesis (cost-reduced), with biosafety confirmed by acute toxicity tests. This study provides a prohealing nanomaterial and establishes a paradigm for traditional biomaterial redevelopment.

Study on the Application of Zeolitic Imidazolate Framework-8 Loaded With Artemisia Argyi Essential Oil in the Treatment of Bacterial Infected Wounds

Yiyang Xu,Ying Li,Zixuan Ye,Cheng Wang,Pengfei Cui,Shuwen Zhou,Lin Qiu,Jianhao Wang

Wound bacterial infection deteriorates with antibiotic misuse, boosting bacterial drug resistance, threatening human health. Therefore, combining natural antibacterials with efficient broad-spectrum materials offers new solutions. Artemisia argyi is a kind of medicinal plant distributed in Asia, known for its rich biological active compounds. With the advancement of modern analytical technology, Artemisia argyi has shown excellent antibacterial and anti-inflammatory potential, particularly Artemisia argyi essential oil (AAEO) which is considered to be the most active substance in Artemisia argyi. However, its practical application is limited due to its poor water solubility, strong volatility, high sensitivity to light and heat, and its irritating odor. Zeolitic imidazolate framework-8 (ZIF-8) is a type of metal–organic framework material (MOF) with good biocompatibility, simple synthesis, high porosity, and antimicrobial activity. ZIF-8 can be used as an ideal vehicle for the preparation of antibacterial materials. In this study, we loaded AAEO onto ZIF-8 to successfully develop AAEO@ZIF-8 antibacterial nanomaterial. The bactericidal ability and antibacterial mechanism of AAEO@ZIF-8 were evaluated through in vitro experiments such as bacterial coating, Live/Dead staining, and crystal violet staining. It was confirmed that AAEO@ZIF-8 had significant antibacterial activity against both Staphylococcus aureus and Escherichia coli . In in vivo experiments, the wound model using C57BL/6 male mice was established, and wound tissue staining with Gram, CD31 + immunohistochemistry, H&E, and Masson staining was performed to evaluate the antibacterial activity of AAEO@ZIF-8. The results showed that AAEO@ZIF-8 exhibited the best wound repair performance in the model. These findings suggested that AAEO@ZIF-8 is a novel and effective antimicrobial nanomaterial targeting pathogenic bacteria. This study successfully transformed the storage mode of AAEO from liquid to solid, providing a promising strategy for the application of AAEO in the treatment of wound bacterial infections.

An antibacterial bilayer hydrogel modified by tannic acid with oxidation resistance and adhesiveness to accelerate wound repair

Yang Li,Rongzhan Fu,Chenhui Zhu,Daidi Fan

Bacterial infection and oxidative stress remain critical problems for wound closure because they frequently trigger severe complications and delay wound healing. In addition, maintaining a moist microenvironment can promote skin regeneration. In this study, a bilayer hydrogel modified with tannic acid (TA) was constructed to accelerate wound repair. The bilayer hydrogel, composed of a layer with large pores to absorb the fluids and allow gas exchange and small pores to maintain the wound moist and prevent bacterial invasion, was initially developed. Thereafter, TA was introduced into the hydrogel to form a dual crosslinked network and endowed the hydrogel with adhesiveness, antibacterial, and oxidation resistance. In addition, the TA@bilayer hydrogel exhibited shape memory behaviour and self-healing ability due to the hydrogen bonds formed between TA and the bilayer hydrogel. As a result, the TA@bilayer hydrogel significantly promoted wound closure by accelerating collagen deposition, reducing tumour necrosis factor-α (TNF-α) levels, and facilitating the expression of vascular endothelial growth factor (VEGF).

Fast Self-Healing Hyaluronic Acid Hydrogel with a Double-Dynamic Network for Skin Wound Repair

Kaidan Yang,Junfeng Yang,Ruina Chen,Qi Dong,Yingshan Zhou

Developing extracellular matrix-derived hydrogel with a fast self-healing capacity to provide a sustainable moist environment able to accelerate wound healing is highly desired for full-thickness skin wound repair. In this study, a fast self-healing hyaluronic acid hydrogel with a dual dynamic network was constructed through a primary reversible acylhydrazone bond formed between aldehyde-modified hyaluronic acid, 3,3′-dithiobis (propionyl hydrazide) (DTP), and secondary dynamic ionic interactions between κ-carrageenan (KC) and K+. Because of the presence of various dynamic covalent bonds such as the acylhydrazone bond, disulfide bond, and noncovalent bonds including hydrogen bonding and ionic interactions, as well as the notable thermoreversible nature of KC, the resultant hydrogel could be self-healed rapidly within 30 min under physiological temperature with a self-healing efficiency of 100%, which was significantly better than other hyaluronic acid hydrogels, as reported previously. Besides, the hydrogel displayed excellent cytocompatibility. According to this study, the hydrogel was administered into the wounds and achieved a superior performance of promoting full-thickness skin wound healing by increasing granulation tissue formation, deposition of collagen as well as the acceleration of re-epithelialization and neovascularization, compared to commercial products, e.g., gauze and 3 M hydrocolloid. We also anticipate that this strategy of double-dynamic network cross-linking can be adopted to fabricate self-healing materials for multiple applications.

Prussian Blue-Assisted NIR Triggered in-Situ Polymerized Nanocomposite Hydrogel for Wound Repair

Qimeng Wu,Zhixue Zhang,Yao Li,Yuan Gu,Xiaokang Ding,Hao Hu,Fu-Jian Xu

Hydrogels are commonly used as wound dressings to help maintain a moist environment around the wound and isolate contaminants, thus promoting healing. For irregular wounds, the slow healing process and even infection may occur due to the inability of dressings to adhere well to the wound. Prussian blue (PB) is a metal-organic framework (MOF) material with excellent photothermal conversion and superior stability. In this paper, a kind of near-infrared (NIR) light triggered in-situ polymerized antimicrobial hydrogel was prepared. The free radical initiator was encapsulated in the hollow PB by a phase change material (PCM) to maintain stability. The raised temperature triggered by NIR induced the release and decomposition of the initiator. The matrix was formed by the cross-linking of double bonds on modified chitosan. The quaternary amine groups of modified chitosan and the photothermal properties of PB enhanced the antimicrobial properties of the hydrogel. High-quality wound healing was demonstrated in the whole skin defect model. This study provides a new reference for the preparation of in-situ polymerized hydrogel dressings for irregular wounds.

Eggshell membrane powder reinforces adhesive polysaccharide hydrogels for wound repair

Zhuojun Zhang,Nianci Li,Li Sun,Zihao Liu,Yan Jin,Ye Xue,Biyun Li,Hongyun Xuan,Huihua Yuan

Multifunctional polysaccharide hydrogels with strong tissue adhesion, and antimicrobial and hemostatic properties are attractive wound healing materials. In this study, a chitosan-based hydrogel (HCS) was designed, and its properties were enhanced by incorporating oxidized eggshell membrane (OEM). Hydrogel characterization and testing results showed that the hydrogel had excellent antimicrobial properties, cytocompatibility, satisfactory adhesion properties on common substrates, and wet-state adhesion capacity. A rat liver injury model confirmed the significant hemostatic effect of the hydrogel. Finally, the ability of the hydrogel to promote wound healing was verified using rat skin wound repair experiments. Our findings indicate that HCS/OEM hydrogels with added eggshell membrane fibers have better self-healing properties, mechanical strength, adhesion, hemostatic properties, and biocompatibility than HCS hydrogels, in addition to having superior repair performance in wound repair experiments. Overall, the multifunctional polysaccharide hydrogels fabricated in this study are ideal for wound repair.

Bioactive Metal Ion-Coordinated Dynamic Hydrogel with Antibacterial, Immunomodulatory, and Angiogenic Activities for Infected Wound Repair

Xinrui Li,Xu Chen,Lian Guan,Wenbo He,Weiling Yin,Dongcheng Ye,Jia Gao,Miao Wang,Guoqing Pan

The repair of infected wounds is a complex physiopathologic process. Current studies on infected wound treatment have predominantly focused on infection treatment, while the factors related to delayed healing caused by vascular damage and immune imbalance are commonly overlooked. In this study, an extracellular matrix (ECM)-like dynamic and multifunctional hyaluronic acid (HA) hydrogel with antimicrobial, immunomodulatory, and angiogenic capabilities was designed as wound dressing for the treatment of infected skin wounds. The dynamic network in the hydrogel dressing was based on reversible metal–ligand coordination formed between sulfhydryl groups and bioactive metal ions. In our design, antibacterial silver and immunomodulatory zinc ions were employed to coordinate with sulfhydrylated HA and a vasculogenic peptide. In addition to the desired bioactivities for infected wounds, the hydrogel could also exhibit self-healing and injectable abilities. Animal experiments with infected skin wound models indicated that the hydrogel dressings enabled minimally invasive injection and seamless skin wound covering and then facilitated wound healing by efficient bacterial killing, continuous inflammation inhibition, and improved blood vessel formation. In conclusion, the metal ion-coordinated hydrogels with wound-infection-desired bioactivities and ECM-like dynamic structures represent a class of tissue bionic wound dressings for the treatment of infected and chronic inflammation wounds.

Normal-pressure-prepared chitosan carbonate liquid dressing: Spontaneous transformation into a pure chitosan water-resistant film and enhanced wound repair

Yuanzhu Long,Xingxing Liu,Fengxiang Huang,Lu Peng,Yingqian Duan,Guohui Bai,Zhaoqiong Zhu,Diansan Su

Liquid dressings are essential in wound care due to their excellent conformability and spontaneous film-forming properties. However, their application is hindered by two key challenges: strict biocompatibility requirements and the need for water-resistant film formation upon drying. Additionally, incorporating active repair functions is extremely difficult. Chitosan, a natural biopolymer with proven wound-healing properties, could address these limitations. However, conventional solubilization methods using acetic acid compromise biocompatibility and impede water-resistant film formation. Herein, we report the first normal-pressure synthesis of purified chitosan carbonate liquid dressing using an ammonium bicarbonate-mediated acetate replacement strategy. This solution spontaneously forms a water-resistant film of pure chitosan upon drying (water contact angle: 99.4°). In full-thickness wound models, it demonstrated remarkable reparative efficacy, including hair follicle regeneration within 10 days, a feature absent in commercial benchmarks like 3 M Cavilon. Importantly, this chitosan carbonate liquid dressing not only overcomes the limitations of conventional chitosan-based solutions in direct biomedical applications but also provides a new repair-facilitating option for the limited pool of liquid dressings. Its unique transformation into a pure chitosan film upon drying resolves long-standing processing limitations of chitosan, unlocking broader applications in biomedical fields such as drug delivery and tissue engineering.

Polyglutamic acid grafted dopamine modified collagen-polyvinyl alcohol hydrogel for a potential wound dressing

Guofei Yu,Changkai Yang,Nianhua Dan,Weihua Dan,Yining Chen

Natural collagen has good biocompatibility and ability to promote tissue regeneration and repair, but the poor mechanical properties and intolerance of degradation of natural collagen limit its applications in the biomedical field. In this research, we synthesized a skin wound repair hydrogel with good biological activity, high strength and excellent water absorption properties. Inspired by the theory of wet healing, dopamine was introduced into the side chain of the water-absorbing polymer polyglutamic acid to synthesize a cross-linking agent (PGAD) with both water absorption and cell adhesion ablities, and then it was introduced into collagen/polyvinyl alcohol (PVA-COL) system to form a double network hydrogel. Scanning electron microscope observation of the morphological characteristics of the hydrogel showed that after the introduction of PGAD, the hydrogel formed an obvious pore structure, and the swelling rate showed that the introduction of PGAD significantly improved the water absorption rate of the hydrogel. In addition, PVA-COL-PGAD hydrogel has good mechanical properties and water absorption behavior. In vitro experimental results revealed that the hydrogel has good biocompatibility. In vivo wound healing experiments showed that hydrogel can promote wound healing process. These results indicated that our hydrogel has great potential as a medical wound dressing.

Self-healing hydrogel based on oxidized pectin with grafted dopamine as gallic acid carrier for burn wound treatment

Junling Wang,Nannan Song,Liping Yin,Zhe Cui,Yong Wang,Chengyan Zhou,Jianheng Li,Jianglei Qin

When the barrier function of the skin tissue is destroyed, the burn wound is prone to bacterial infection and difficult to repair during the healing process. Therefore, there is an urgent need to design functional dressing materials to promote burn wound repairing. In this research, injectable hydrogel with mussel-inspired tissue adhesion was fabricated from dopamine grafted oxidized pectin (OP-DA) and hydrazide terminated polyethylene oxide (PEO-AH) as dressing material for burn wounds. The catechol moiety on the grafted dopamine (DA) enhanced the tissue adhesion of hydrogel and the wound sealing performance, which can well adhere onto the skin to cover irregular wounds for consistent sealing under body moving. On this basis, the hydrogel also showed hemostatic performance based on liver and tail hemostasis model. The hydrogel was further loaded with gallic acid (GA) to enhance the antibacterial property to E. coli and S. aureus to inhibit wound infection. Along with good biocompatibility and biodegradability, the hydrogel showed improved burn wound repairing performance to second-degree burn wound models on mice. As a result, the OP-DA/PEO-AH hydrogel showed great advantage as GA carrier in wound healing applications.

Five-In-One Hydrogel Integrating Bacteriostasis, Self-Healing Capability, Growth Factor Release, Electrical Stimulation, and Photothermal Stimulation Tailored for Complex Wound Repair

Simin Lai,Chenxi Shi,Liting Yuan,Kefeng Li,Xiaojing Wang,Xi Yu,Pengbi Liu,Huan Wang,Lihuan Wang,Hui Yu

Complex wound management remains a significant global challenge, and the development of multifunctional wound dressings that can effectively promote wound healing remains an urgent clinical need. Herein, a kind of multifunctional hydrogel wound dressing that combines bacteriostasis, self-healing capability, growth factor release, electrical stimulation, and photothermal stimulation is developed. This kind of wound dressing is generated by adding protocatechualdehyde (protocatechuic aldehyde (PA)), short core–shell fibers loading with platelet-rich-plasma (platelet-rich plasma fibers), and polydopamine-coated carbon nanotubes (PDA@CNTs) into quaternary ammonium chitosan (QCS) solution to form a shear-reversibly cross-linked QCS/PA/PDA@CNTs-PRP hydrogel. The obtained hydrogels possess impressive properties, including high swelling capacity (445–852%), strong adhesion ability (16.4–36.7 kPa), self-healing ability, injectability, conductivity (0.24–0.46 S/m), and photothermal properties. Notably, under near-infrared irradiation, the hydrogel exhibits a highly efficient bactericidal activity. In vitro experiments demonstrated that the hydrogel exhibits excellent biocompatibility and anti-inflammatory capability as well as its ability to stimulate cell proliferation, migration, and tubule formation. Moreover, the in vivo studies further confirmed that with the additional assistance of near-infrared light and electrical stimulation, the hydrogel further promotes wound epithelization, angiogenesis, and collagen deposition. Consequently, this hydrogel provides a promising therapeutic strategy for complex wound healing.

Inhibition and Rescue of Hyperglycemia-Induced Cellular Senescence by Mitochondrial Transfer from Enucleated Mesenchymal Stem Cell-Derived Microvesicles for Chronic Wound Healing

Zixuan Dong,Xiaobing Liu,Shichun Li,Xiaoling Fu

The aberrant cellular senescence in chronic wounds presents a significant barrier to healing. Mitochondrial dysfunction is critical in initiating and maintaining cellular senescence, underscoring therapeutic potential in restoring mitochondrial function by delivering healthy mitochondria to wound cells. However, approaches for delivering mitochondria to achieve optimized wound repair remain lacking. Herein, enucleated MSCs-derived microvesicles containing functional mitochondria (Mito@euMVs) via simple extrusion are developed. By controlling the size of microvesicles within a small micron-scale range, the mitochondrial encapsulation efficiency is optimized. Mito@euMVs effectively delivered mitochondria into fibroblasts and HUVECs, inhibiting and rejuvenating hyperglycemia-induced cellular senescence. To enhance the clinical applicability, soluble PVA microneedle patches for the transdermal Mito@euMVs delivery are utilized. In diabetic rats with pressure sores, the senescence-inhibiting and -rescuing properties of Mito@euMVs are further validated, along with their therapeutic efficacy, demonstrating their potential for chronic wound repair. Moreover, as a versatile delivery vehicle for mitochondria, Mito@euMVs hold promising for treating mitochondrial dysfunction and aging-related conditions.
