Facile preparation of fatigue-resistant Mxene-reinforced chitosan cryogel for accelerated hemostasis and wound healing

Zhengguo Wu,Shanshan Li,Xiaoqian Qin,Lu Zheng,Jiawei Fang,Lansheng Wei,Changliang Xu,Zhong Alan Li,Xiaoying Wang

The development of highly effective chitosan-based hemostatic materials that can be utilized for deep wound hemostasis remains a considerable challenge. In this study, a hemostatic antibacterial chitosan/N-hydroxyethyl acrylamide (NHEMAA)/Ti 3 C 2 T x (CSNT) composite cryogel was facilely prepared through the physical interactions between the three components and the spontaneous condensation of NHEMAA. Because of the formation of strong crosslinked network, the CSNT cryogel showed a developed pore structure (~ 99.07 %) and superfast water/blood-triggered shape recovery, enabling it to fill the wound after contacting the blood. Its capillary effect, amino groups, negative charges, and affinity with lipid collectively induced rapid hemostasis, which was confirmed by in vitro and in vivo analysis. In addition, CSNT cryogel showed excellent photothermal antibacterial activities, high biosafety, and in vivo wound healing ability. Furthermore, the presence of chitosan effectively prevented the oxidation of MXene, thus enabling the long-term storage of the MXene-reinforced cryogel. Thus, our hemostatic cryogel demonstrates promising potential for clinical application and commercialization, as it combines high resilience, rapid hemostasis, efficient sterilization, long-term storage, and easy mass production.

Biomimetic rhCOL17-P334 Conjugate for Enhanced Wound Healing

Yijie Lou,Chunyan Dai,Shuting Feng,Rijuan Jin,Shuangshuang Liu,Zhanyi Zhou,Minjun Wang,Yang Yang,Guoqiao Xu,Jue Hou,Jiayi Chen,Guping Tang,Nan Wang,Hongzhen Bai,Zhe Chen

Wound healing remains a significant global health challenge, affecting millions annually and imposing substantial economic burdens. Most commercially available biomaterials for wound management primarily address external symptoms, including hemostasis, exudation, scarring, and infection. Advanced biomaterials derived from endogenous molecules aim to better replicate the native wound microenvironment, promoting enhanced repair. Since wounds frequently occur on exposed skin, which is vulnerable to UVA radiation and requires protective yet invisible materials, traditional wound care products often lack these essential features. Inspired by natural UV protection mechanisms, a novel bioscaffold is developed using recombinant human collagen XVII (rhCOL17) crosslinked with porphyra-334 (P334) to improve wound healing under UVA exposure. The resulting rhCOL17-P334 conjugate integrates extracellular matrix (ECM)-like cues with UVA-shielding properties provided by P334. This conjugate is used to construct a transparent, injectable hydrogel combining gelatin methacryloyl (GelMA) and rhCOL17-P334 (GCP). GCP significantly inhibits UVA-induced fibroblast senescence and improves wound healing outcomes by targeting integrin α6β4 through rhCOL17. Its transparency facilitates convenient wound monitoring while also addressing the aesthetic requirement for invisibility. By combining UVA shielding with wound repair capabilities, GCP presents a promising platform for advanced wound management.

Fabrication of microspheres containing coagulation factors by reverse microemulsion method for rapid hemostasis and wound healing

Nan Wang,Shuying Zhao,Xiaoyong Tian,Shanyi Guang,Hongyao Xu

Traditional dressings, such as bandages, gauze, and cotton pads stick to new granulation tissue, thereby aggravating wound injury or causing secondary injury during replacement. Microspheres that are biodegradable and adaptable to various wound shapes are a good alternative to traditional dressings. In this work, a novel microsphere was prepared by reverse microemulsion method using sodium alginate and silk peptide (SP) as the aqueous phase. After cross-linking by Ca 2+ , calcium alginate (CA) and SP composite microspheres called CA/SP were prepared. By adjusting the SP content, the swelling rate of microspheres reached 1050 % and the pore diameter reached 19.59 nm. In addition, the introducing SP provided a stable loading site for thrombin (Th). This platform (called CA/SP@Th)—integrating rapid blood enrichment, calcium release , and Th catalysis—can ensure rapid hemostasis in a variety of bleeding models. Additionally, the SP present in this modality also promoted fibroblast proliferation, this increased the wound closure rate in a total cortex injury mouse model (more than 97 % within 15 days). Therefore, CA/SP@Th can be used as a multifunctional dressing for rapid hemostasis and wound healing.

Scarless wound healing programmed by core-shell microneedles

Zhang Ying,Wang Shenqiang,Yang Yinxian,Zhao Sheng,You Jiahuan,Wang Junxia,Cai Jingwei,Wang Hao,Wang Jie,Zhang Wei,Yu Jicheng,Han Chunmao,Zhang Yuqi,Gu Zhen

Effective reprogramming of chronic wound healing remains challenging due to the limited drug delivery efficacy hindered by physiological barriers, as well as the inappropriate dosing timing in distinct healing stages. Herein, a core-shell structured microneedle array patch with programmed functions (PF-MNs) is designed to dynamically modulate the wound immune microenvironment according to the varied healing phases. Specifically, PF-MNs combat multidrug-resistant bacterial biofilm at the early stage via generating reactive oxygen species (ROS) under laser irradiation. Subsequently, the ROS-sensitive MN shell gradually degrades to expose the MN core component, which neutralizes various inflammatory factors and promotes the phase transition from inflammation to proliferation. In addition, the released verteporfin inhibits scar formation by blocking Engrailed-1 ( En1 ) activation in fibroblasts. Our experiments demonstrate that PF-MNs promote scarless wound repair in mouse models of both acute and chronic wounds, and inhibit the formation of hypertrophic scar in rabbit ear models.

Biological Effects of EGCG@MOF Zn(BTC)4 System Improves Wound Healing in Diabetes

Song Li,Jing Yan,Qiangqiang Zhu,Xinxiang Liu,Senlin Li,Shenhou Wang,Xuanjun Wang,Jun Sheng

Tea contains high levels of the compound epigallocatechin gallate (EGCG). It is considered an important functional component in tea and has anti-cancer, antioxidant, and anti-inflammatory effects. The eight phenolic hydroxyl groups in EGCG’s chemical structure are the basis for EGCG’s multiple biological effects. At the same time, it also leads to poor chemical stability, rendering EGCG prone to oxidation and isomerization reactions that change its original structure and biological activity. Learning how to maintain the activity of EGCG has become an important goal in understanding the biological activity of EGCG and the research and development of tea-related products. Metal–organic frameworks (MOFs) are porous materials with a three-dimensional network structure that are composed of inorganic metals or metal clusters together with organic complexes. MOFs exploit the porous nature of the material itself. When a drug is an appropriate size, it can be wrapped into the pores by physical or chemical methods; this allows the drug to be released slowly, and MOFs can also reduce drug toxicity. In this study, we used MOF Zn(BTC)4materials to load EGCG and investigated the sustained release effect of EGCG@MOF Zn(BTC)4and the biological effects on wound healing in a diabetic mouse model. Keywords:EGCG;MOF;wound healing;diabetes

Bioinspired Design of Sericin/Chitosan/Ag@MOF/GO Hydrogels for Efficiently Combating Resistant Bacteria, Rapid Hemostasis, and Wound Healing

Meng Zhang,Dong Wang,Nana Ji,Shaoxiang Lee,Guohui Wang,Yuqi Zheng,Xin Zhang,Lin Yang,Zhiwei Qin,Yang Yang

Due to the spread of drug-resistant bacteria in hospitals, the development of antibacterial dressings has become a strategy to control wound infections caused by bacteria. Here, we reported a green strategy for in situ biomimetic syntheses of silver nanoparticles@organic frameworks/graphene oxide (Ag@MOF–GO) in sericin/chitosan/polyvinyl alcohol hydrogel. Ag@MOF–GO was synthesized in situ from the redox properties of tyrosine residues in silk sericin without additional chemicals, similar to a biomineralization process. The sericin/chitosan/Ag@MOF–GO dressing possessed a high porosity, good water retention, and a swelling ratio. The hemolysis rate of the composite was 3.9% and the cell viability rate was 131.2%, which indicated the hydrogel possessed good biocompatibility. The composite also showed excellent lasting antibacterial properties against drug-sensitive and drug-resistant pathogenic bacteria. The composite possessed excellent hemostatic activity. The coagulation effect of the composite may be related to its effect on the red blood cells and platelets, but it has nothing to do with the activation of coagulation factors. An in vitro cell migration assay confirmed and an in vivo evaluation of mice indicated that the composite could accelerate wound healing and re-epithelialization. In summary, the composite material is an ideal dressing for accelerating hemostasis, preventing bacterial infection, and promoting wound healing. Keywords:biomimetic synthesis;hemostasis;antibacterial activity;wound healing

Multifunctional electrospun asymmetric wettable membrane containing black phosphorus/Rg1 for enhancing infected wound healing

Liming Zhou,Nanbo Liu,Longbao Feng,Mingyi Zhao,Peng Wu,Yunfei Chai,Jian Liu,Ping Zhu,Rui Guo

Bacterial infection is one of the most frequent complications in the burn and chronic wounds. Inspired by natural existing superhydrophobic surface structures, a novel asymmetric wettable membrane was prepared using the electrospinning technique for facilitating the bacteria-infected wound healing. Herein, the prepared membrane consists of two layers: The hydrophobic outer layer was composed of poly (lactic-co-glycolic) acid (PLGA) and black phosphorus-grafted chitosan (HACC-BP), while the hydrophilic inner layer was composed by using a mixture of gelatin (Gel) with ginsenoside Rg1 (Rg1). Biological studies in vitro showed BP@PLGA/Gel (BP@BM) membrane with excellent antibacterial activity could significantly inhibit the adhesion of bacteria, and Rg1 could facilitate the migration and tube formation of human umbilical vein endothelial cells (HUVECs). Compared to Aquacel Ag dressing, the result in vivo revealed that the Rg1/BP@BM could facilitate better wound healing by triggering phosphoinositide 3-kinase (P-PI3K/PI3K) and phosphorylation of protein kinase B (P-AKT/AKT) signaling pathways, upregulating Ki67, CD31, α-SMA, and TGF-β1, and downregulating TNF-α, IL-1β, and IL-6, promoting M2 polarization (IL-10, CD206, and Arg-1) of macrophages, inhibiting M1 polarization (iNOS) of macrophages. These findings suggested that the asymmetric wettable membrane have the huge potential for wound healing.

Procedural Promotion of Wound Healing by Graphene-Barium Titanate Nanosystem with White Light Irradiation

Jianlin Wang,Zhaoyang Wen,Yumei Xu,Xin Ning,Deping Wang,Jimin Cao,Yanlin Feng

Background Wound healing is a continuous and complex process that comprises multiple phases including hemostasis, inflammation, multiplication (proliferation) and remodeling. Although a variety of nanomaterials have been developed to control infection and accelerate wound healing, most of them can only promote one phase but not multiple phases, resulting in lower efficient healing. Although various formulations such as nitric oxide releasing wound dressings were developed for dual action, the nanostructure synthesis and the encapsulation process were complex.Materials and Methods Here, we report on the design of graphene-barium titanate nanosystem to procedural promote the wound healing process. The antibacterial effect was assessed in Gram-negative Escherichia coli bacteria (E. coli) and Gram-positive Staphylococcus aureus bacteria (S. aureus), the cell proliferation and migration experiment was investigated in mouse embryonic fibroblast (NIH-3T3) cells, and the wound healing effect was analyzed in female BALB/c mice with infected skin wound on the back. Results Results showed that graphene-barium titanate nanosystem could generate abundant ROS to kill both E. coli and S. aureus. The growth curves, bacterial viability, colony number formation and scanning electron microscopy (SEM) images of E. coli and S. aureus all confirmed the antibacterial effect. Cell Counting Kit-8 (CCK-8) assay displayed that GBT possesses great biocompatibility. EdU assay showed that GBT plus white light irradiation significantly promoted the proliferation and migration of NIH-3T3 cells. Scratch assay found that GBT could achieve a fast scratch closure compared to the control. In vivo wound healing effect indicates that GBT can accelerate wound repair procedure. Conclusion GBT nanocomposite is capable of programmatically accelerating wound healing through multiple stages, including production of a large amount of ROS after white light exposure to effectively kill E. coli and S. aureus to prevent wound infection and as a scaffold to accelerate fibroblast proliferation and migration to the wound to accelerate wound healing.

Effects of a PDGF-Stem Cell-Hydrogel Compound on Skin Wound Healing in Mice

Jiaqi Lin,Ziwei Lin,Anqi Huang,Xinyi Wu,Wei Yan,Daojun Liu,Chiju Wei,Wencan Xu

Introduction The treatment of chronic refractory skin wounds still remains a serious clinical challenge. Stem cells and hydrogels are widely used in healing of skin wound of various types due to their superior bioactivities and biocompatibility. This study aimed to demonstrate the wound healing effect of a hydrogel compound loaded with enucleated stem cells expressing the platelet-derived growth factor (PDGF). Methods An injectable hydrogel was formulated using 22% poloxamer 407, 1% poloxamer 188 and 1% hyaluronic acid. A PDGF-B transgenic cell line of mouse bone marrow mesenchymal stem cells (BMSCs) was generated by lentiviral infection. Cells were enucleated and embedded in hydrogel. The healing effects of the compound was tested in a full-thickness skin wound model of Balb/c mice. The wound models were randomly divided into four groups: the control group applied with PBS buffer; the hydrogel group with hydrogel only; the BMSC group with hydrogel mixed with normal BMSCs; and the BMSC-PDGF group with hydrogel mixed with enucleated BMSCs expressing PDGF. Results Overexpression of PDGF-B in transgenic cell line of BMSCs was verified by RT-PCR, immunofluorescence staining and western blot. When enucleated, the viability measured by Calcein-AM staining reduced to 54.29% at 48 h. Conditioned medium was collected with or without hydrogel layered over cells. PDGF concentration measured by ELISA reached 14.66 ng/μL and 257.89 ng/μL respectively after 48-h cultivation, suggesting a possible slow releasing effect in the presence of hydrogel. When applied to the skin wound, the healing rates of the BMSC-PDGF group was significantly higher than that of the control group on day 3. BMSC-PDGF group had significantly more neovascularization and cutaneous appendages from day 7. The proliferation of collagen fibers in BMSC-PDGF group was significantly higher than the control group on day 3 and day 7. Finally, BMSC-PDGF group had significantly lower amount of the inflammatory factors TNF-α, IL-1β, IL-6, MMP-3 and MMP-9 than that of the control group on day 7. Conclusion PDGF-stem cell-hydrogel compound significantly improved wound healing and reduced wound inflammatory factor expression in Balb/c mice. This biomaterial-based approach provides a new powerful reference for the treatment of chronically wounded skin.

Borate and Disulfide Dual-Dynamic Hydrogel for ROS Dual-Scavenging

Xing Wang,Haohao Cui,Qingyun Ning,Huayang Feng,Xue Sun,Boyuan An,Zhanrong Li,Jianliang Shen,Jingguo Li

The excessive accumulation of reactive oxygen species (ROS) on wound surfaces poses considerable challenges to the wound healing process, as it exacerbates inflammation and heightens the risk of skin infections. Consequently, the development of multifunctional dressings that possess both ROS-scavenging and antimicrobial properties is of clinical significance for enhancing wound healing. To address these challenges, we synthesized a dual-scavenging hydrogel utilizing dual-dynamic crosslinking techniques. This hydrogel is composed of poly-lysine-grafted D-glucono-1,5-lactone, dithiodipropionic acid/lipoic acid, and dopamine (PLGDTD), in conjunction with borax. The multifunctional attributes of the hydrogel can be attributed to the incorporation of borate esters, dopamine, and disulfide bonds. Experimental results demonstrate that this hydrogel exhibits a robust antioxidant capacity and a pronounced inhibitory effect against Staphylococcus aureus and Escherichia coli . The results of cellular studies indicate that the hydrogel is capable of effectively scavenging ROS and mitigating oxidative stress, attributable to its dual-scavenging mechanism involving borate and disulfide bonds. This multifunctional hydrogel demonstrates significant potential as a candidate for enhancing wound healing associated with ROS. Graphical

Natural polyphenolic antibacterial bio-adhesives for infected wound healing

Hengjie Zhang,Yuqi Feng,Tianyou Wang,Jianhua Zhang,Yuxian Song,Jing Zhang,Yiwen Li,Dingzi Zhou,Zhipeng Gu

Bio-adhesives used clinically, commonly have the ability to fill surgical voids and support wound healing, but which are devoid of antibacterial activity, and thus, could not meet the particular needs of the infected wound site. Herein, a series of natural polyphenolic antibacterial bio-adhesives were prepared via simple mixing and heating of polyphenols and acid anhydrides without any solvent or catalyst. Upon the acid anhydride ring opening and acylation reactions, various natural polyphenolic bio-adhesives could adhere to various substrates (i.e., tissue, wood, glass, rubber, paper, plastic, and metal) based on multi-interactions. Moreover, these bio-adhesives showed excellent antibacterial and anti-infection activity, rapid hemostatic performance and appropriate biodegradability, which could be widely used in promoting bacterial infection wound healing and hot burn infection wound repair. This work could provide a new strategy for strong adhesives using naturally occurring molecules, and provide a method for the preparation of novel multifunctional wound dressings for infected wound healing.

Isolation and structural identification of the homogeneous polysaccharide CGPA1 from Calvatia gigantea with wound healing effects

Xiuhua Meng,Wenjian Zhao,Yanan Zhao,Juntong Guo,Xiaoqin Ding,Yanan Gai,Han Lv,Yiwei Wang,Jian Chen

Calvatia gigantea is a traditional Chinese medicinal fungus known for its diverse pharmacological effects, with polysaccharides being one of its main bioactive components. In this study, a novel homogeneous water-soluble polysaccharide (CGPA1) was successfully isolated and purified from the fruiting body of Calvatia gigantea . The molecular weight of CGPA1 was determined to be 11.69 kDa. Structural characterization was performed using a combination of high-performance gel permeation chromatography (HPGPC), Fourier-transform infrared spectroscopy (FTIR), methylation analysis, and nuclear magnetic resonance (NMR) spectroscopy. The analysis data revealed that CGPA1 is primarily composed of galactose, methyl-galactose, mannose, xylose, and glucose in a molar ratio of 60.01:27.16:10.34:1.83:0.67. The backbone structure predominantly consists of α-1,6-Gal-(1 → and α-1,6 (3-MeO)-Gal-(1→, with side chains of α-Gal and α-Man. Morphological analysis using SEM, XRD, and AFM, combined with Congo red experiments, suggesting that CGPA1 adopts an amorphous granular chain conformation. Both in vitro and in vivo tests confirmed that CGPA1 can promote the migration of MSF cells and accelerate the healing of acute injured wounds. It was found to facilitate skin re-epithelialization, and collagen deposition over the healing process.
