Hydrogel-Transformable Antioxidant Poly-γ-Glutamic Acid/Polyethyleneimine Hemostatic Powder for Efficient Wound Hemostasis

Xiang Li,Wenli Han,Gao He,Jiahao Yang,Jing Li,Hongxia Ma,Shige Wang

Hemostatic powder, which can absorb large amounts of water and tends to produce repeated hydration with tissue, has been clinically proven as an ideal engineering material for treating wounds and tissues. We herein designed a polypeptide-based hemostatic powder. A water-soluble polypeptide, γ-polyglutamic acid (γ-PGA), was mixed with the polyethyleneimine (PEI), N-hydroxysuccinimide, and 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide. The solution of these polymers was lyophilized to harvest the γ-PGA/PEI powder (PP hemostatic powder). When deposited on a bleeding wound, the PP hemostatic powder can quickly absorb a large amount of blood and interstitial fluid, concentrate coagulation factors, coagulate blood cells, and eventually form a stable mechanical hydrogel. The wound bleeding time of the PP hemostatic powder group was 1.8 ± 0.4 min, significantly lower than that of the commercial chitosan hemostatic powder group (2.8 ± 0.4 min). The PP hemostatic powder was endowed with antioxidant capacity by introducing protocatechuic aldehyde, which can effectively inhibit inflammation and promote wound healing. Therefore, via preparation through a facile lyophilization method, the PP hemostatic powder is expected to find a wide application prospect as a qualified hemostatic powder. Keywords:hemostatic powder;hydrogel;γ-polyglutamic acid;polyethyleneimine;wound hemostasis

A Glycosylated and Catechol-crosslinked ε-Polylysine Hydrogel: Simple Preparation and Excellent Wound Hemostasis and Healing Properties

Teng Lin,Shao Zheng-Wei,He Yu-Shi,Lu Jia-Yu,Zou De-Rong,Feng Chuan-Liang,Dong Chang-Ming

Commercial tissue adhesives have been widely applied in wound hemostats and dressings while enhancing the hemostasis and healing capabilities is challenging to meet clinical needs. Herein, we designed the glucose- and catechol-functionalized derivatives from commercial ε -polylysine (EPL) and prepared the hydrogels by simple amidation and catechol-crosslinking reactions, which have larger swelling ratios of 220%–240%, suitable microporous size of about 6–8 µ m, and tissue adhesion strength of about 20–40 kPa. The hemolysis, cytotoxicity, and cellular double-staining assays indicate that those hydrogels had good biocompatibility and the H-3 hydrogel with higher glucose content gave a lower hemolysis ratio of 0.73%±0.14%. The blood-clotting index, blood cell attachment and adhesion studies showed those hydrogels had fast blood-coagulation, resulting in excellent hemostasis performance with a short hemostatic time of 38–46 s and less blood loss of 19%–34% in a liver hemorrhage model. A full-thickness rat-skin defect model further demonstrates that the H-3 hydrogel achieved fast wound healing with a wound closure of 70.0%±2.7% on postoperative day 7 and nearly full closure on day 14. Remarkably, the hydroproline level that denotes the collagen production reached a higher one of 7.24±0.55 µg/mg comparable to that in normal skins on day 14, evidencing the wound healing was close to completion in the H-3 treatment. Consequently, this work provides a simple method to construct a glycosylated and catechol-functionalized hydrogel platform from commercial EPL, holding translational potentials in wound hemostats and dressings.

Neutrophil extracellular traps-inspired DNA hydrogel for wound hemostatic adjuvant

Ye Rui,Zhu Ziyu,Gu Tianyi,Cao Dengjie,Jiang Kai,Dai Qiang,Xing Kuoran,Jiang Yifan,Zhou Siyi,Cai Ping,Leong David Tai,Yu Mengfei,Song Jie

Severe traumatic bleeding may lead to extremely high mortality rates, and early intervention to stop bleeding plays as a critical role in saving lives. However, rapid hemostasis in deep non-compressible trauma using a highly water-absorbent hydrogel, combined with strong tissue adhesion and bionic procoagulant mechanism, remains a challenge. In this study, a DNA hydrogel (DNAgel) network composed of natural nucleic acids with rapid water absorption, high swelling and instant tissue adhesion is reported, like a band-aid to physically stop bleeding. The excellent swelling behavior and robust mechanical performance, meanwhile, enable the DNAgel band-aid to fill the defect cavity and exert pressure on the bleeding vessels, thereby achieving compression hemostasis for deep tissue bleeding sites. The neutrophil extracellular traps (NETs)-inspired DNAgel network also acts as an artificial DNA scaffold for erythrocytes to adhere and aggregate, and activates platelets, promoting coagulation cascade in a bionic way. The DNAgel achieves lower blood loss than commercial gelatin sponge (GS) in male rat trauma models. In vivo evaluation in a full-thickness skin incision model also demonstrates the ability of DNAgel for promoting wound healing. Overall, the DNAgel band-aid with great hemostatic capacity is a promising candidate for rapid hemostasis and wound healing.
